細胞株の起源および培養条件は、補足的方法において詳述される。すべての神経膠腫始原細胞(GIC)は、切除したばかりの腫瘍から確立され、最初の継代に使用された。すべての細胞は細菌汚染について常に検証された。Trpを含まないRPMI 1640 (Promocell) および透析されたFBS (Invitrogen)を用いて、Trpを含まない条件下で細胞を培養した。L-TrpおよびL-KynはSigma-Aldrich製とした。インターフェロンγ(IFNγ)は、Immunotools (Friesoythe, Germany)製とした。TCDDおよび3-メチルコラントレン(3-MC)はSigma-Aldrich製、3,4-ジメトキシフラボン(3,4-DMF)はAlfa Aesar(Karlsruhe, Germany)製とした。TDO阻害剤((E)-6-フルオロ-3-[2-(3-ピリジル)ビニル]-1H-インドール) 680C91は、ピペリジン存在下での6-フルオロインドール-3-カルボキサルデヒドとピリジン-3-酢酸との縮合によって合成した。
C57BL/6NおよびCD-1 nu/nuマウスは、Charles River (Sulzfeld, Germany)から購入した。Ahr欠損マウス(B6.129-AHRtm1Bra/J)は、Charlotte Esser (Duesseldorf, Germany)に提供していただいた。Ahr欠損マウスと齢の適合したC57BL/6Nは、Harlan Laboratories (Rossdorf, Germany)から得た。
TDOの発現は、ヒト腫瘍において免疫組織化学によって解析した。Trp分解に対するその関連性は、遺伝子のノックダウンによって、またはTDOの過剰発現によって判定した。TrpおよびKynは、細胞培養上清、ヒト血清、および異種移植組織においてHPLCで測定した。混合白血球反応、クロム放出、ELISpot、および腫瘍組織中の免疫細胞の染色を用いて、TDO活性の免疫効果を評価した。細胞周期の分析、マトリゲルおよびスフェロイド浸潤アッセイ、スクラッチアッセイ、スフィアフォーメーションアッセイ、およびコロニー形成能アッセイを用いて、TDO活性のオートクリン作用を分析した。すべての動物手順は、施設の実験動物研究指針に従い、政府機関により承認された。TDOのノックダウンあり、およびなしのヒト神経膠腫細胞のCD1nu/nuマウスへの同所移植、前記細胞のNK枯渇または野生型CD1nu/nuマウスへの皮下注入、ならびマウスのTdo機能を有するGL261およびTdo欠損GL261細胞の同系C57BL/6Nマウスへの皮下注入を行って、in vivoでのTDO活性のオートクリンおよびパラクリン作用を分析した。Kynで処理したヒト神経膠腫細胞のマイクロアレイ分析を行って、Kynにより活性化されるシグナル伝達経路を確認した。AHRトランスロケーションの分析、DREルシフェラーゼ分析および放射性リガンド結合アッセイによって、KynによるAHRの活性化が確認された。AHRの薬理学的阻害および安定したノックダウン(in vitroおよびin vivo)は、Kynの影響がAHR依存性であることを立証した。Tdo機能を有する腫瘍細胞およびTdo欠損腫瘍細胞のAhr+/+およびAhr-/-マウスへの注入を用いて、TDOを介した癌促進に対する宿主の影響の寄与を検討した。最後に、ヒト腫瘍組織の染色、マイクロアレイデータ、および臨床データを用いて、TDOがヒトの癌においてAHRを活性化するかどうか、ならびにこれが生存にどのような影響を及ぼすかを分析した。
HPLC分析は、フォトダイオードアレイ(PDA)検出およびLichrosorb RP-18カラム(250 mm x 4 mm ID、5μm、Merck、Darmstadt、Germany)を有するBeckman HPLCを用いて実施した。KynおよびTrp濃度は、3 x 105細胞の培地中で測定した。ヒト血清は、24人の膠芽細胞腫患者(女性10人、男性14人、年齢中央値53.5歳)ならびに24人の年齢および性別のマッチした健常対照(女性10人、男性14人、年齢中央値53.5歳)からインフォームドコンセント後に採取し、TrpおよびKyn濃度について分析した。U87異種移植片中のKyn濃度を測定するために、U87腫瘍を切除して秤量し、ただちに液体窒素中で凍結して加工処理した。
全RNAはQiagen RNeasyキットを用いて分離し、cDNAはApplied Biosystems逆転写キット(Foster City, CA, USA)を用いて合成した。QRT-PCRは、SYBR Green PCR Mastermixを用いてABI 7000サーマルサイクラーで行った(いずれもApplied Biosystems)。すべてのプライマーは、ゲノムDNAの増幅を除外するために、ゲノムDNA上の少なくとも1つのイントロンで隔てられた。PCR反応は、鋳型の脱落、ならびに融解曲線およびゲル分析によって、no-RTコントロールを含めることにより確認された。標準曲線は、それぞれの遺伝子について作製された。遺伝子発現の相対的定量は、閾値の比較によって決定した。すべての結果は、GAPDHに対して標準化された。

ON-TARGETplus siCONTROL Non-targeting Pool (D-001810-10- 05, Dharmacon) およびsiRNAなしのトランスフェクションを、ネガティブコントロールとして使用した。細胞は、Invitrogen製のリポフェクタミン(Lipofectamine)RNAiMAXを用いてトランスフェクトした。ノックダウン効率はqRT-PCRによって分析した。
U87ヒト神経膠腫細胞は、FUGENE HDトランスフェクション試薬(Roche, Mannheim, Germany)を用いて、sh-TDOまたはスクランブルコントロールを発現するpSUPER.puroプラスミド (OligoEngine, Seattle, WA, USA)でトランスフェクトした。トランスフェクションの72時間後に、培地を5μg/mlピューロマイシン(AppliChem GmbH)含有DMEMに交換した。別に指示のない限り、sh-TDOAを使用した。LN308神経膠腫細胞においてAHRをノックダウンするために、pSingle-tTS-shRNAベクターをClontech (CA, USA)より購入した。XhoI/HindIIIオーバーハングを有する、望ましいshRNA配列をコードするアニーリングされた二本鎖オリゴヌクレオチドを、XhoI/HindIIIクローニング部位を用いてベクター中にクローニングした。XhoI/HindIIIオーバーハングを含む、コントロール/AHRまたはTDO shRNAサイレンシングのための低分子ヘアピン配列は以下の通りとした:

組換えベクターをLN-308およびLN-18神経膠腫細胞にトランスフェクトし、クローン形質転換体を1 mg/mlネオマイシン(Sigma-Aldrich)によって選択した。2μg/mlドキシサイクリン(Sigma-Aldrich)を用いてノックダウンを誘導し、誘導の72時間後に細胞を分析した。他に特に指示しない限りsh-AHR1を使用した。
GL261細胞に、Tdo cDNA (NM_ 019911)を発現するpcDNA3.1 (-) (Invitrogen)、または空のベクターを、FUGENE HD試薬(Roche)を用いてトランスフェクトした。クローン形質転換体を1 mg/mlネオマイシン(Sigma-Aldrich)を用いて選択した。
3μmに切断した切片を、Ventana BenchMark XT(登録商標)免疫染色装置(Ventana)を用いてそれぞれの抗体とともにインキュベートして処理した。TDO染色の定量分析のために、染色された腫瘍細胞のパーセンテージ、および染色強度を、光学顕微鏡(Olympus BX51)を用いて、組織切片上の代表的な倍率視野(200x)において評価した。
免疫蛍光のために、TDO低発現の神経膠腫およびTDO高発現の神経膠腫の切片は、Ventana Cell Conditioner 1の中で30分間の熱による抗原回復の後、ウサギ抗TIPARP(1:50)およびマウス抗LCA(1:50)抗体とともに4℃にて一晩インキュベートした。その後、ロバ抗ウサギAlexaFluor 568 (1:500, Invitrogen)およびロバ抗マウスDyLight 488 (1:100, Jackson ImmunoResearch, West Grove, PA, USA)二次抗体を5時間作用させた。顕微鏡写真は、Olympus BX-50顕微鏡 (Olympus GmbH, Hamburg, Germany)で、Zeiss Axiocam MRm (Zeiss, Jena, Germany)を用いて撮影された。
96ウェルプレート内で、10% FBS、100 U/ml ペニシリン、および100μg/mlストレプトマイシンを含有するRPMI 1640中に神経膠腫細胞をシードした。シードの24時間後に、刺激細胞として2 x 105 放射線照射 (30 Gy) PBMC、ならびにレスポンダーとして非血縁ドナー由来の2 x 105 PBMCを添加した。6日MLRを行って、最後の18時間、培養物に[3H]-メチルチミジン(PerkinElmer, Waltham, MA, USA)を適用し、放射性核種の取り込みをシンチレーション計数により測定した。6人の非血縁PBMCドナーについて実験を繰り返した。
補足的方法に詳述されるように、デュアルルシフェラーゼ/ウミシイタケアッセイを行った。さまざまなTrp代謝産物に反応してDREによって駆動されるレポーター遺伝子活性化を分析するために、ウミシイタケルシフェラーゼを発現する、pGL3プロモーター発現プラスミドおよび対照プラスミドpRL-SV40を使用した(Promega, Heidelberg, Germany)。
マトリゲルアッセイにおいて、細胞の浸潤は、膜を超えて移動した細胞数を5つの独立した顕微鏡高倍率視野で計数することによって評価し、微小格子を用いて対照に対する侵襲のパーセンテージとして表した。スフェロイドアッセイでは、埋め込み後0、24、48、および72時間の時点で、各スフェロイドがカバーする領域の顕微鏡写真を撮った。定量のために、ImageJを用いて、指示された時点で浸潤神経膠腫細胞によってカバーされる平均面積を24時間間隔で測定し、0時間の面積と比較した。
免疫細胞の細胞傷害性の阻害は、標準化51クロム放出アッセイ(補足的方法)を用いて評価した。パーセント単位の特異的な溶解は次のように計算した:[51Cr放出実験値 - 最小放出] / [最大放出- 最小放出] x 100。この実験は、少なくとも4人の異なるPBMCドナーについて実施した。
樹状細胞(DC)を健常C57BL/6Nマウスの骨髄から分離し、20 ng/ml GM-CSF (Immunotools)を含有するRPMI 1640中で5日間培養した。担癌マウスから脾臓を摘出し、40μmセルストレイナーを通してすりつぶした。赤血球を溶解し、Pan T CellアイソレーションキットII(Miltenyi GmbH)を用いて、MACSによりT細胞を分離した。抗IFNγ抗体(Mabtech AB, Nacka Strand, Sweden)でコーティングしておいたELISpotプレート(Millipore)に2 x 105 DCをシードし、10μg GL261溶解物(PBS中で凍結融解サイクルを繰り返すことによって作製)を4時間適用した後、1 x 105 T細胞を添加した。36時間後、IFNγ産生T細胞を、ビオチン標識抗IFNγ抗体、ストレプトアビジン-ALPおよびBCIP/NBTPLUS (Mabtech)を用いて検出し、ImmunoSpot Analyzer (Cellular Technology Limited, Shaker Heights, OH, USA)を用いて定量した。
AHRトランスロケーションを検出するために、ウェル当たり7000個の、GFP標識AHRを有するTao BpRc1細胞を50μM Kynまたは50μM Trpに暴露し、PBS中3.7%ホルムアルデヒドの中で固定して、0.1%Triton X100中で透過性とし、1μg/ml Hoechst 33342 (Invitrogen)とともにインキュベートしてから、20x U-Apo 340対物レンズ(Olympus, NA 0.75)を用いてノンコンフォーカルモードで、BD Pathway(登録商標)Imager 855にてイメージングした。蛍光強度の分析はさらに、Attovisionソフトウェア(BD Biosciences)を用いて行った。それに加えて、LN-229神経膠腫細胞の核および細胞質画分中のAHRタンパク質含量をイムノブロッティングにより比較した。
11 Ci/mmolの比放射能を有するL-3H-Kynは、Quotient Bioresearch (Radiochemicals) Ltd. (Cardiff, UK)より入手した。Ahr高発現およびAhr欠損マウス由来のマウス肝サイトゾルを用いたL-3H-Kynによる結合アッセイを行った。特異的な結合は、Ahr高発現およびAhr欠損サイトゾル間の放射能の相違として定義された。
すべての動物手順は、施設の実験動物研究指針に従い、政府機関により承認された。ヒト神経膠腫細胞は、6匹の6-12週齢胸腺欠損マウス(CD1nu/nu)の右線条体に皮下注入されるか、または定位的に移植され、観察された。NK細胞の枯渇は、腫瘍細胞注入の2日前に開始して、ウサギ抗アシアロGM1抗体(Wako Chemicals, Duesseldorf, Germany)の隔週腹腔内注入によって実施した。対照は、ウサギIgG(Calbiochem, Darmstadt, Germany)を注入した。in vivoでAHRノックダウンを誘導するために、ドキシサイクリンを、スクロース含有飲用水中2 mg/mlの濃度でマウスに投与した。マウス神経膠腫細胞は、6-12週齢野生型C57BL6/NマウスまたはAHR-/- C57/Bl6マウスの右側腹部に皮下注入した。
図3のMRIスキャンは、通常の全身1.5 T MRIスキャナー(Symphony, Siemens, Erlangen, Germany)で特注の送信/受信小動物コイルを用いて行った。
U87神経膠腫細胞を100μM Kynで8時間または24時間処理した後、その細胞を集め、RNAeasy-Kit (Qiagen)を用いてRNAを分離した。RNAを補足的方法に詳述したようにマイクロアレイ分析に供した。4つの処理(8時間、24時間、Kyn処理、未処理)のそれぞれについて、2つのアレイをハイブリダイズして、Kyn処理サンプル対未処理サンプルの平均log2比を計算した。データのその他の解析は補足的方法に詳述している。臨床サンプルのために、マイクロアレイおよび臨床データは、Molecular BRAin Neoplasia DaTa (REMBRANDT)のリポジトリ(REpository)から入手した(補足的方法)。The Cancer Genome Atlas (TCGA)ネットワークの未処理の原発性膠芽細胞腫(n=362)のデータセットの中で生存分析を、R2マイクロアレイ分析のKaplan-Meier分析モジュールおよび可視化プラットフォーム (http://r2.amc.nl)を用いて行った。
データは、平均±平均誤差として表した。有意性の分析は、スチューデントのt検定で実施した。P値<0.05を有意と見なした。Ki67およびTDOの相関は、スピアマン(Spearman)の順位相関(SPSS, IBM, Somers, NY, USA)によって分析した。TDOとAHRとの相関 (図6c)、ならびにTDOとCYP1B1との相関 (図6d)は、スピアマンの順位相関(Sigmaplot)によって分析した。核の蛍光強度は、順位による一元配置ANOVA(p=<0.001)に続いてDunn法(p<0.05)によって分析した。
本発明の基礎となる研究によると、ヒト癌細胞株の選別は、脳腫瘍細胞、すなわち神経膠腫細胞株およびグリオーマ幹細胞(GIC)におけるTrpの構成的な分解および高マイクロモルレベルのKynの放出を明らかにしたが、ヒト星状膠細胞ではそうではなかった(図1a)。驚くべきことに、IDO1およびIDO2は、脳腫瘍において構成的Trp異化の原因とはならなかった。その代わりに、トリプトファン-2,3-ジオキシゲナーゼ(TDO)が、これは主に肝臓で発現され、全身のTrp濃度を制御すると考えられているが、ヒト神経膠腫細胞で強く発現され、Kyn放出と相関していた(図1b)。TDOの薬理学的阻害またはノックダウンは、神経膠腫細胞によるKyn放出をブロックしたが、IDO1およびIDO2のノックダウンは影響を及ぼさず(図1c、d)、したがって、TDOがヒト神経膠腫細胞において主要なTrp分解酵素であることが確認された。ヒト脳腫瘍標本において、TDOタンパク質レベルは悪性度とともに増加し、増殖指数と相関した(図1e-g)。前記のように((Miller 2004, Neurobiol Dis 15(3), 618)、健常なヒトの脳はニューロンに弱いTDO染色しか示さなかった(図1e)。TDO発現は神経膠腫に限定されず、本明細書の別項で言及される他のタイプの癌、肝細胞癌、結腸癌、乳癌、非小細胞肺癌(NSCLC)、卵巣癌、悪性黒色腫(脳転移)、および腎細胞癌などでも検出された。
抗腫瘍NK細胞応答の、TDOを介した阻害は、ヌードマウスでは機能的であるが、それがTDOノックダウン腫瘍の形成障害の原因となるかどうかを分析するために、皮下の腫瘍増殖をNK細胞の有無で比較した。NK細胞の枯渇は、対照およびTDOノックアウト腫瘍のいずれの増殖も強めたが、TDOノックアウトの増殖を対照の増殖にまでは回復させなかった(図2l)ので、機能性抗腫瘍T細胞およびNK細胞応答のない状態で、構成的TDO活性によって生成されるKynが、オートクリンの様式でヒト神経膠腫の悪性表現型を強めることが示唆される。
神経膠腫細胞に及ぼすKynのオートクリン作用の根底にある分子機序をよりよく理解するために、Kyn処理した神経膠腫細胞のマイクロアレイ分析を行って、KynによるAHR応答遺伝子の広範な誘導を明らかにした(図3a)。経路分析から、U87細胞のKyn処理によって8時間および24時間時点で最も強く誘導された、25個の遺伝子がすべて、直接または間接的にAHRによって制御されることが示された(図3a)。

- 前記フラボンまたはその誘導体が、3,4-ジメトキシフラボン、3’-メトキシ-4’-ニトロフラボン、4’,5,7-トリヒドロキシフラボン(アピゲニン)、または1-メチル-N-[2-メチル-4-[2-(2-メチルフェニル)ジアゼニル]フェニル-1H-ピラゾール-5-カルボキサミドである、請求項4に記載の医薬組成物。
- 前記植物性化合物またはその誘導体が、レスベラトロールもしくはその誘導体、エピガロカテキン、または没食子酸エピガロカテキンである、請求項4に記載の医薬組成物。
- 前記AHR阻害剤が、AHRタンパク質に特異的に結合して阻害する抗体である、請求項1に記載の医薬組成物。
- 前記AHR阻害剤が、AHRリプレッサータンパク質、または不活性なAHR核トランスロケーター(ARNT)である、請求項1に記載の医薬組成物。
- 前記AHR阻害剤が核酸阻害剤である、請求項1に記載の医薬組成物。
- 前記核酸阻害剤が、AHRをコードするポリヌクレオチドに特異的に結合し、リボザイム、アンチセンス分子、阻害剤オリゴヌクレオチド、アプタマー、マイクロRNA、および低分子干渉RNA(siRNA)からなる一群から選択される、請求項11に記載の医薬組成物。
- 天然AHRリガンド依存性癌の治療および/または予防のための医薬の製造におけるAHR阻害剤の使用であって、ここで前記天然AHRリガンドがキヌレニンである、前記使用。
- 前記AHR阻害剤が、請求項3〜12のいずれか1つに記載のAHR阻害剤である、請求項13に記載の使用。
- 前記癌が、脳腫瘍、好ましくは神経膠腫、黒色腫、結腸直腸腺癌、結腸癌、腎細胞癌、非小細胞肺癌(NSCLC)、乳癌、肝細胞癌、卵巣癌、頭頸部癌、膀胱癌、膵臓腺癌、中皮腫、および小細胞肺癌(SCLC)からなる一群から選択される癌である、請求項13に記載の使用。
