Description
本発明は、個体のHIV−1ウイルス感染の予防及び治療に関するものである。
ヒトHIV感染の約90%は、HIV−1ウイルスによって起こる。ヒト免疫不全ウイルス1型(HIV−1)は、ウイルス複製時に生じる突然変異の蓄積によって起こる、及び組換え事象によっても起こる、顕著な遺伝的変異性によって特徴づけられる。HIV処置の化学療法の長期にわたる失敗は、特に、HIV−1ウイルス株の高い変異原活性によって説明される。異なる経過の抗レトロウイルス療法後に、及び多剤療法(HAART)後であっても、患者から耐性ウイルス変異体が速やかに生じることがこれまでに示された。これらの耐性ウイルスは、それらのタンパク質コンフォメーション及び構造に特異的変化を有する。通常、HIV−1が現行処置を回避する原因となるそのような突然変異は、代々のウイルス世代にわたり維持され、処置条件下で選択を受ける結果として蓄積する。

本発明は、主として、抗HIV−1免疫原性組成物又はワクチン組成物であって、特に広域抗HIV−1中和抗体の誘導を可能にする特異的抗原性ペプチドを含む抗HIV−1免疫原性組成物又はワクチン組成物を提供する。

a)複製コンピテントHIV−1ウイルスを既知量の被験抗体と接触させることによって被験混合物を調製する工程、
b)工程a)の終わりに得られた被験混合物と共にPHA活性化CD4+T細胞をIL−2の存在下でインキュベーションする工程、及び
c)工程b)の終わりに得られたCD4+T細胞培養物中のp24活性を決定する工程、及び
d)工程a)で添加された既知量の抗体に関して工程c)で決定されたp24活性を、工程a)が抗HIV抗体の不在下で行われた同じ試験で見出されたp24活性と比較することによって、被験抗体のIC50値を決定する工程

本明細書において広く論じられ、実験的に例示されたように、式(I)で示されるペプチドを含む免疫原性組成物を個体に免疫処置することで、抗HIV−1広域中和抗体の生成が誘導される。これらの抗HIV−1広域中和抗体自体は、活性抗HIV−1剤として使用することができる。
本明細書の実施例に開示されるように、式(I)で示されるペプチドに対する抗体は、HIV−1感染患者から見出すことができる。

実施例の材料及び方法
1. ウイルスの調製
突然変異体を、QuickChange II XL部位特異的突然変異誘発キット(Stratagene)を用いてプラスミドpNL4.3から調製し、その後にDNA配列決定により検証した。野生型及びアラニン突然変異型3S/gp41 pNL4.3プラスミドをリポフェクタミン(Invitrogen)によってOptima培地中で293T細胞にトランスフェクションした。トランスフェクションの48時間後に、無細胞上清を回収し、HIV主要コアタンパク質p24の濃度はVidas Ag p24 IIキット(Biomerieux)を用いた。ウイルス上清を−80℃で保存する。
コンジュゲーションされていない、及びKLH−コンジュゲーションされた、野生型及びアラニン突然変異型の精製3S/gp41合成ペプチド(図1)をCovalabs(Villeurbanne, France)から購入した。フロイント不完全アジュバントの存在下でKLH連結ペプチド20mgをマウスに2回IV注射し、各注射の間隔は約2週間であった。記載のように(Vieillard et al AIDS 2006)、野生型又はアラニン突然変異型3S/gp41ペプチド100ngを4℃で一晩コーティングしたMaxisorpプレート(Nunc)を用いることによって、酵素結合免疫吸着アッセイ(ELISA)によって血清を力価測定した。
健康ドナーからの全血試料由来の白血球は、「Etablissement Francais du sang」(Hopital Pitie-Salpetriere, Paris, France)からのバフィーコート遠心分離によって得た。CD4+T細胞をCD4磁気マイクロビーズ(Miltenyi)で精製した。フローサイトメトリー分析から、純度95%を超えるCD4細胞であることが実証された。10%ウシ胎仔血清(FCS)を補充したRPMI-1640 Glutamax培地(Invitrogen)中で細胞を1μg/ml PHA−L(Murex)で3日間活性化し、次に3日毎に添加した100IU/mlプロロイキン(Proleukin)−2(Chiron)と共に培養した。
精製された活性化CD4+T細胞、MT2細胞及びJurkat細胞(107個)をp24抗原等価物100ngと共に37℃でそれぞれ17時間及び2時間インキュベーションした。次に、細胞をPBS/EDTA中で2回洗浄し、106個/mLに再懸濁した。ウイルスの生成を感染後2日毎にELISAによってモニタリングし、Vidas Ag p24 IIキット(Biomerieux)でp24濃度を測定した。感染MT2細胞上のシンシチウム形成を96時間後に標準位相差顕微鏡法によって判断した。
A. Morris及びO. Schwartz(Pasteur Institute Paris, France)から親切に贈与してもらったHela P4C5にCD4+及びCCR5+を共トランスフェクションしたが、その細胞は、ルシフェラーゼ及びβ−ガラクトシダーゼ遺伝子に連結したHIV−1末端反復配列(LTR)の組み込み型コピーを含有した(Amara et al J. Exp. Med. 1997)。細胞を1日前に播種し、p24が4ng/mlの野生型又はアラニン突然変異型3S/gp41 NL4.3ウイルスを感染させた。感染48時間後に、Hela−P4C5細胞溶解液中のβ−ガラクトシダーゼ活性を、CPRG β−ガラクトシダーゼキット(OZ biosciences)を用いて測定した。
製造業者の説明書にしたがって、マウス抗血清由来のポリクローナルIgG画分をNabプロテインA/G Spinキットを用いて単離し、次にZebaスピン脱塩カラムで脱塩した(どちらもThermo Scientific製)。免疫グロブリン画分をCentricon遠心フィルターユニット(Millipore)で濃縮し、次にBCAタンパク質アッセイキットを用いて定量した(Thermo Scientific)。精製Abを初濃度20μg/mlに続く5回の2倍希釈で試験した。
(1)Abの効力(50%[IC50]及び90%[IC90]の阻害を達成するために必要な濃度)を比較するために、本発明者らは、最初にCD4+T細胞−p24アッセイを用いた。感染性複製コンピテントウイルス(200TCID50)を様々な濃度のIgと共に30分間インキュベーションし、続いてIL2の存在下でPHA活性化精製CD4+T細胞を添加した。p24測定(Vidas Ag p24 II kit, Biomerieux)のために、感染開始から7日後に二つ組の試料を回収した。
(2)様々な精製Abのディファレンシャルな中和活性の可能性を評価するために、本発明者らは、それらが異なるX4株(NL4.3、BRU、NDK)及びR5株(JR−CSF、YU−2)からのHIV−1侵入、又はROD HIV−2を阻害する能力を、Clarisse Berlioz-Torrent(Institut Cochin, Paris, France)から親切に提供されたHela−P4C5細胞及び/又はTZM−bl細胞を用いて測定した(NIH AIDS Research and Reference Reagent Program)。感染性複製コンピテントウイルス(200TCID50)を様々な濃度のIgと共に30分間インキュベーションし、続いてHela P4C5細胞を添加した。感染48時間後に、Hela−P4C5溶解液中のβ−ガラクトシダーゼ活性を、CPRG β−ガラクトシダーゼキット(OZ biosciences)を用いて測定し、TZM−bl溶解液中のルシフェラーゼ活性を、Britelite Plus試薬(Perkin Elmer)を用いて測定する。TCIDアッセイにおけるプラスの値を決定するために、バックグラウンドの2.5倍のカットオフ値を適用した。データをTCID50及びTCID95で表現した。
生存ウイルス又は熱不活性化ウイルスが17時間存在した、又は37℃で4時間3S/gp41ペプチドで処置された、精製CD4+T細胞にFACS分析を行った。試料を各抗NKp44L抗体1μgと共に、又は融合タンパク質(NKp30−Ig、NKp44−Ig、又はNKp46−Ig)と共に、PBS/BSA1%の存在下で4℃で2時間インキュベーションした。次に、細胞をPBS/BSA1%中で洗浄し、特異的二次抗体(PBS/BSA1%で1:100希釈)と共に4℃で30分間インキュベーションし、最終的に抗CD4 mAbで直接染色し、既述(Vieillard et al PNAS 2005)のようにBD CellFix溶液(BD Bioscience)を用いて固定した。CD4+細胞の少なくとも10,000回の事象をFACSCanto(BD Biosciences)又はNavios(Coulter)で検出した。
野生型又はアラニン突然変異型3S/gp41 NL4.3ウイルスに感染していないか、又は感染したCD4+T細胞を、自己IL2活性化自己PBMC及び抗CD107a mAb(H4A3; Becton Dickinson)の存在下で1:1の比で再懸濁した。1時間キュベーション後に、既述(Beziat et al PLoS One 2010)のように、3mMモネンシンを添加して追加的な3時間のインキュベーションに供した。NK細胞を抗CD3、抗CD56、及び抗NKp44 mAbで染色し、FACSCanto(BD Biosciences)又はNavios(Coulter)で分析した。少なくとも5,000回のCD3−CD56+事象を分析した。
ウイルス生活環に果たす3S/gp41モチーフの役割を描写するために、NL4.3 HIV−1株由来のgp41タンパク質の3Sモチーフ内の位置S613からS618の間の残基に、単一の点置換を個別に導入した。これらの単一置換がウイルスの感染性に効果を発揮するかどうかを検討するために、各ウイルス調製物からp24当量100ngの抗原となるようにMT−2細胞を誘発した。図2Aに、3S/gp41モチーフの置換効果は、ウイルス生成の完全に近い消失から、野生型モチーフを発現しているウイルスについての無効までの範囲であることを示す。実際に、S613A突然変異型ウイルスは、MT−2に感染するその完全な能力を保ったが、K617Aは、野生型に観察されたレベルのまだ66±10%であり、残りの突然変異体は22%未満であった。W614A及びS618A突然変異体は、MT−2細胞において感染を樹立する能力がないと思われる(図2A)。これらのデータは、これらの突然変異体がシンシチウム形成する能力で確認され、W614A及びS618A突然変異型ウイルスのいずれかに感染したMT−2細胞では、野生型に比べてこれらの巨細胞構造が完全に不在であった(図2B)。これらのデータから、3S/gp41モチーフのW614及びS618の両方の位置がウイルス感染時に重大な役割を果たしうることが示唆される。
本発明者他によって以前に示されたように、HIV−1感染は、NKp44Lを含めたある種のNKRリガンドの発現を誘導する(Vieillard et al 2005; Ward et al 2007)。本発明者らは、3S/gp41置換を含有するウイルスも、精製CD4+T細胞上のNKp44L発現を誘導する能力があるかどうか判定したかった。野生型ウイルスに感染後、高い頻度の細胞がNKp44Lを発現した(56.2%)。S613A又はK617A突然変異体に感染したCD4+T細胞でも類似の結果が得られ、それぞれ61.0及び67.6%の細胞がNKp44Lを発現していた。より興味深いことに、他の突然変異体;W614A、S615A、N616A、及びS618Aで発現は検出されなかった。以前のデータに一致して(Vieillard et al 2005; Ward et al 2007)、NKp30リガンド及びNKp46リガンドの発現は、置換の位置がどこであれ、野生型及びアラニン突然変異型3S/gp41ウイルスのどちらに感染した細胞でも誘導されなかった(図3A)。
本発明者らは、ある3S/gp41突然変異を含有するNL4.3ウイルスに感染したCD4+T細胞上にNKp44Lを見出したので(図2A)、本発明者らは、このリガンドを発現しているターゲット細胞の方が、NK細胞傷害性に対して感受性が高い可能性を検討した。野生型又は様々な突然変異型ウイルスのいずれかに感染した精製CD4+T細胞を自己IL2活性化NK細胞と共にインキュベーションして、それらの脱顆粒能を決定した。図3Cに、未感染細胞(4.2%)に比べて、野生型ウイルス(28.8%)、又はS613A(29.5%)及びK617A(29.9%)突然変異体の存在下でNKp44+NK細胞上でのCD107aの高発現が検出されたことを示すが、これは、CD4+ターゲットT細胞上でのNKp44Lの高発現を示しているデータと整合が取れている(図3A)。対照的に、CD4+T細胞上でのNKp44Lの誘導が観察されない残りの突然変異体では、脱顆粒のレベルは、未感染細胞で得られたこのレベルにほぼ近いままである(図3C)。熱不活性化ウイルス粒子(図6B)及び合成ペプチド(図4D)の存在下で類似の結果が得られた。
したがって、本発明者らは、野生型及び各アラニン突然変異型3S/gp41ペプチドをマウスに免疫処置して、特異的抗体を発生させた。それらの全てについて、遊離ペプチド捕捉後のELISAでそれらの特異的3S/gp41配列に対して強い免疫応答が見られた(データは示さず)。興味深いことに、血清交差反応性の大きさはペプチド成分に依存し;抗血清は、それらがNKp44Lの発現を誘導する能力に直接比例して、各合成ペプチドと相互作用する。したがって、野生型と、S613A及びK617A突然変異体との間に強い血清交差反応性が観察されたが、他方でこの効果は、他の突然変異体では、独特な置換によって誘導される推定上のコンフォメーション改変に一致して顕著に減少する。
近年、HIV感染患者において広域反応性Nab活性が経時的に生じると思われることが実証された。置換された3S/gp41モチーフに対するbNAbがHIV感染で誘発される頻度に関する情報を得るために、本発明者らは、多様な臨床特徴を有するHIV−1感染患者から得られた血漿106試料の中和活性を分析した。評価した106試料のうち、55/106(52%)は、検出可能なレベルの抗3S−WT Abを有し、その値は15〜325AU/mlの間の範囲であった。既述のように(Vieillard et al AIDS 2006)、Spearman検定から、抗3S Abの量とCD4カウントとの間に高度に有意な関係が明らかである(p<0.0001)(データは示さず)。予想外なことには、特異的にW614A突然変異型3S/gp41モチーフに対する抗体がELISAにより血漿106試料中5試料で検出された(4.7%)。それらのそれぞれについて、高レベルのAbが検出可能であった(120〜250AU/mlの範囲)。これらの特異的抗体は、IgGアイソタイプであり(データは示さず)、高CD4カウント(822±89CD4/mm3)及び検出不能のウイルス量(20コピー/ml未満)を有する患者だけから検出された(表3)。注目すべきは、特異的にW614A突然変異型3S/gp41モチーフに対するこれらのAbは、野生型モチーフに対する抗体と同様に、健康なドナーの血漿からは検出不能である(データは示さず)、(Vieillard et al AIDS 2006)。
