Description
但し、腫瘍細胞のアポトーシスを誘導する以外に、腫瘍壊死因子関連アポトーシス誘導リガンドはさらに自然免疫及び獲得免疫に係り、自身免疫性疾患に関与することが最近報告された。例えば、近年の報告は、腫瘍壊死因子関連アポトーシス誘導リガンドは胸腺の発育過程において負の選択を調節し、または胸腺細胞をアポトーシスし、且つ自己免疫性疾患、例えばI型糖尿病の誘発において重要な役割を果すことを述べている。また、腫瘍壊死因子関連アポトーシス誘導リガンドの受容体は全身において広範に発現し、さらに肝細胞死及び肝炎に関与するため、臨床において腫瘍壊死因子関連アポトーシス誘導リガンドを繰り返して、全身に大量に使用することは予想できない免疫機能への影響を招く恐れがある。
本発明の一態様では、腫瘍組織ターゲティングを有する腫瘍壊死因子関連アポトーシス誘導リガンド変異体であって、配列表における配列1のアミノ酸残基配列を有する蛋白質であり、遺伝子操作方法によって、CD13のリガンド、ジョイニングペプチド、腫瘍壊死因子関連アポトーシス誘導リガンドによって構成した腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体融合蛋白質を構築する。即ち、一般的な遺伝子操作方法を使用して、人工合成またはクローンによって、腫瘍壊死因子関連アポトーシス誘導リガンド変異体のコード遺伝子を構築し、可溶性型に組み換え・ 発現し、及び容易に分離・精製する。前記CD13リガンドはNGR配列を含有するポリペプチドであってもよく、好ましくは、環状構造を有し、NGR配列を含有するポリペプチド、例えばショートペプチドCNGRCである。
した腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体との間に、柔軟性構造を有し、側鎖アミノ酸を含まないショートペプチドを添加してもよい。なお、ショートペプチドは、1〜25個のアミノ酸残基からなり、主にグリシン、アラニン、セリンなどの、側鎖を含まないアミノ酸によって構成される。
前記CD13のリガンドを、腫瘍壊死因子関連アポトーシス誘導リガンドのN端に配置する場合、発現時にN端アミノ酸の分解が発生してCD13リガンドの機能に影響することを防止するために、CD13のリガンドの前に側鎖を含まないアミノ酸、主にアラニンまたはグリシンの配列を増加する。
前記CD13リガンドと腫瘍壊死因子関連アポトーシス誘導リガンド変異体とによって構成した腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体は、その腫瘍組織における分布を大幅に向上させ、その腫瘍組織における標的搬送を実現させ、その抗腫瘍効果を大幅に向上させるとともに、同時に、その使用量を顕著に低減することができる。
同時に腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体のcDNAのコードを公開したが、これによって腫瘍壊死因子関連アポトーシス誘導リガンドのdDNAを介してコードされるCD13リガンド及びジョイニングペプチドのDNA配列調製を促進することができる。この結果、前記構成の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体のcDNAを遺伝子治療に用いることができる。
本発明の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体は、ポリエチレングリコールと脂肪酸で酸性にして修飾する、または、抗体のFc断片またはアルブミンを加えた組み換えで修飾することができ、それによって、腫瘍壊死因子関連アポトーシス誘導リガンド変異体の半減期を延長し、薬物動態学的により良好な効果を達成する。
(1)より良好な腫瘍組織ターゲティング能:従来の腫瘍壊死因子関連アポトーシス誘導リガンドは、腫瘍組織に対する相対的な選択的作用を、主に腫瘍組織が発現する細胞死受容体4及び受容体5を使用して顕著に実現するが、一方、本発明の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体は、腫瘍組織が発現する細胞死受容体4及び受容体5を使用する以外に、さらに腫瘍組織がCD13を多量に発現する特徴を利用して、腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンドの、腫瘍組織に対する標的搬送をより有効に実現する。
(2)より高効率の抗腫瘍効果:腫瘍壊死因子関連アポトーシス誘導リガンドの、腫瘍組織における標的投与を実現するため、本発明の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体は、同様な用量の腫瘍壊死因子関連アポトーシス誘導リガンドと比べた場合、及び、腫瘍細胞表面のインテグリンαVβ3、αVβ5にターゲティングするTRAIL変異体RGD−L−TRAIL(特許文献1参照)と比べた場合、単独使用でも、あるいは、従来の化学治療、放射線治療、中医薬治療、生物治療などの方法との併用でも、ともにより強い抗腫瘍効果を示した。
(3)より低い使用量:腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガン
ド変異体が、同様な用量の腫瘍壊死因子関連アポトーシス誘導リガンドと比べてより強い抗腫瘍効果を有するため、使用時に、同程度な抗腫瘍治療効果を奏したい場合、腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体蛋白質の用量を大幅に低減することができる。腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体蛋白質の投与量の低減は、腫瘍壊死因子関連アポトーシス誘導リガンドの、腫瘍治療における潜在的な副作用を克服することができ、同時に、腫瘍患者の治療費用を削減でき、高い有効性、低毒性副作用、低コストの腫瘍治療効果を達成する。
(5)可溶性型の発現及び容易な分離精製:現在、腫瘍壊死因子関連アポトーシス誘導リガンドを大腸菌において発現する時に非生物活性の封入体生成物を形成しやすい。一方で、本発明の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の分子構造及び分子量は野生型の腫瘍壊死因子関連アポトーシス誘導リガンドと比べより複雑で大きい。従って、本発明は腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を低温下で培養及び誘導発現する発現方法を開発することにより、発現生成物に封入体が形成することを有効に防止可能とした。本発明は同時に、腫瘍壊死因子関連アポトーシス誘導リガンドが金属イオンをキレート化する生物機能を有することを利用し、金属アフィニティークロマトグラフィーとイオン交換クロマトグラフィーとを連結することによって、腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を有効に精製し、高純度の蛋白質生成物を得た。本発明は可溶性型の発現及び容易な分離精製方法を最適化することによって精製した生成物に、含有率の高い重合体型を最終的に得た。それによって、得られた生成物が非常に良好な生物活性を有することを証明した。含有率の高い重合体型の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を生産することは、本発明と同類の研究に対して顕著な相違点となる。本発明によって、高い重合体含有率の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を有効に発現し及び高効率に得る発現方法及び精製プロセスを提供できる。
(6)CD13リガンドのN端に側鎖を含まない1個のアミノ酸を結合することで、発現時にN端アミノ酸の分解を抑制し、CD13リガンドの機能に影響することを有効に防止する。
コンピュータ支援構造を使用してシミュレーション及び分子設計を行い、SGIコンピュータワークステーションにおいて、MSI社の分子設計ソフト(InsightII、Discoverなどモジュール)を使用して、腫瘍壊死因子関連アポトーシス誘導リガンドの結晶構造を元に、腫瘍壊死因子関連アポトーシス誘導リガンドのN端に対してCD13リガンドショートペプチドを付加して分子モデル構築及び分子設計を行い、ジョイニングペプチドのアミノ酸の長さを確定した。
前記分子設計に基づいて、まずジョイニングペプチドが5個のグリシンを選択して設計案とした。その理由は、コンピュータシミュレーションから、前記案のジョイニングペプチドが比較的短いので、腫瘍壊死因子関連アポトーシス誘導リガンドの蛋白質の構造、及びCD13との結合に対して共に大きい影響を与える可能性があったためである。その他の設計案のジョイニングペプチドの多くはCD13リガンドと腫瘍壊死因子関連アポトーシス誘導リガンドの分子表面とがある程度の距離を有するため、この相互作用及び影響が小さくなる。
また、ジョイニングペプチドがアラニンまたはグリシン、アラニン−グリシン−グリシン−セリン−セリン−グリシン−グリシン−グリシンであるショートペプチドの腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体、及び3回以上重複するグリシン−(アラニン−グリシン−グリシン−セリン−セリン−グリシン−グリシン−グリシン)3の25個のアミノ酸残基の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の発現を試してみたが、共に類似な結果を得た。以上の結果から、コンピュータ分子設計の正確性が証明された。 更に、上記結果を比較した場合、5個のシステインをもつ腫瘍壊死因子関連アポトーシス誘導リガンド変異体の発現レベルが最も高く(100 mg/L)、その他の変異体の発現レベルがやや低いが、共に約50−100 mg/Lのレベルであった。

coli BL21(DE3)において発現した。腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体NGR−L−TRAILの可溶性型発現を行うため、発現条件を以下の通りにした。一夜成長させた組み換え発現菌をLB培地に100倍希釈し、37℃で2.5時間培養し、その後さらに24℃で1〜2時間培養し、24℃下で0.5mMまでIPTGを加えて、その後細菌を24℃下で一夜誘導発現した。細菌を遠心した後、分裂分解液(50mMリン酸ナトリウム、0.5M塩化ナトリウム、1mMジチオトレイトール、pH7.6)に懸濁して超音波粉砕を行った。
従来の報告によれば、野生型腫瘍壊死因子関連アポトーシス誘導リガンド変異体が大腸菌において発現するときに、常に非生物活性の封入体生成物の形で存在する。本発明の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体は4個のシステイン(即ち2対のジスルフィド結合を形成した)が増加しており、該変異体が大腸菌において発現するときに野生型腫瘍壊死因子関連アポトーシス誘導リガンドと比べ非生物活性の封入体生成物をより形成しやすくなるので、精製もより難しくなる。しかし、本発明は発現条件及び分離精製過程を最適化することによって、腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を、可溶性型で発現させることに成功し、且つカチオン性樹脂層析及びニッケル金属アフィニティークロマトグラフィーによって、生物活性を有し、純度の高い腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体蛋白質を得ることができた。収量は100mg/Lであった。変性還元条件下の15%ポリアクリルアミゲル電気泳動銀染色法の分析(図1の1A)及び質量スペクトルで配列を測定した分析結果は共に発現生成物が間違いないことを証明した。
且つ本発明の発現条件及び精製プロセス下で、腫瘍壊死因子関連アポトーシス誘導リガンド及びその変異体は共に非常に高い含有率の重合体(図1の1B)を有したが、これはその他の腫瘍壊死因子関連アポトーシス誘導リガンドの発現及び精製における同種の操作ではほとんど見られなかった。
腫瘍壊死因子関連アポトーシス誘導リガンドとその腫瘍ターゲティング変異体NGRL−TRAIL、及びRGD−L−TRAIL変異体をそれぞれフルオレセイン(Sigma社)で標識を行い、標識した蛋白はSephadex−G25分子篩によって遊離フルオレセインを除去した。ヒト皮膚微小血管細胞はトリプシンによって消化された後、2%のウシ胎児血清を含有する氷冷リン酸緩衝液を用いて2回洗い、その後改めて懸濁し、1μgの標識蛋白を加え、4℃下で1時間育成した。実験はフルオレセインで標識したウシ血清アルブミンを対照とした。染色した細胞を3回洗浄した後フローサイトメトリー(Becton Dickinson社)を使用して結合能力を分析した。
さらに、フルオレセイン標識の腫瘍壊死因子関連アポトーシス誘導リガンドとその腫瘍ターゲティング変異体NGR−L−TRAIL、及びインテグリンαVβ3、αVβ5にターゲティングするTRAIL変異体RGD−L−TRAIL(特許文献1参照)の、ヒト皮膚微小血管内皮細胞と直接結合する能力を評価した。フローサイトメトリーの測定結果は、腫瘍壊死因子関連アポトーシス誘導リガンド変異体NGR−L−TRAILの、ヒト皮膚微小血管内皮細胞と結合する能力が、腫瘍壊死因子関連アポトーシス誘導リガンド及びインテグリンαVβ3、αVβ5受容体にターゲティングする変異体RGD−L−TRAILよりも遥かに高いことを示した。
細胞表面のCD13またはインテグリンの発現存在度は間接標識法でフローサイトメトリーを用いて分析測定した。消化後の細胞は2%ウシ胎児血清を含有する氷冷リン酸緩衝液で2回洗浄し、細胞を改めて懸濁した後氷上Anti−CD13モノクロナル抗体(eBioscience社)、Anti−αVβ3インテグリン抗体MAB 23C6(eBioscience社)、またはAnti−αVβ5インテグリン抗体MAB 1961(Chemicon International社)で1時間抗体コーティングし、陰性対照として、精製同型ラット免疫グロブリンG(eBioscience社)を採用した。1次抗体コーティング細胞を2回洗浄した後、緑色フルオレセイン結合のヒツジ抗ラット免疫グロブリンG1 (γ)(Caltag Laboratories社)を加えて、30分間2次抗体遮光標識した。その後、3回洗浄し、さらに4%ホルマリンを含有するリン酸緩衝液で固定し、最後にフローサイトメトリーにおいてCD13またはインテグリンαVβ3、αVβ5発現の存在比を測定分析した。なお、全ての染色実験は3回繰り返した。
ティングのよりよいターゲットであるかは、誰も詳細に研究及び対比してこなかった。本発明は、腫瘍新生血管及び腫瘍細胞表面CD13、及びインテグリンαVβ3、αVβ5の発現レベルを丁寧に比較し、且つ細胞及び動物モデルレベルにおいて、それぞれCD13にターゲティングするTRAIL変異体NGR−L−TRAIL及びインテグリンαVβ3、αVβ5にターゲティングするTRAIL変異体RGD−L−TRAILの抗腫瘍
活性を評価したところ、驚くべき、且つ予想と反する結果を得た。本発明はCOLO−205表面のCD13及びインテグリンαVβ3、αVβ5の発現状況を比較し、COLO−205表面ではほとんどCD13分子を発現せずに、但し低レベルのインテグリンαVβ3及び高レベルのインテグリンαVβ5を発現することを発見した(図3)。これを裏付けるデータとして、CD13にターゲティングするTRAIL変異体NGR−L−TRAILの、COLO205細胞のアポトーシス誘導活性がやや増加し(図4)、一方、インテグリンαVβ3、αVβ5にターゲティングするTRAIL変異体RGD−L−TRAILの、COLO−205細胞のアポトーシス誘導活性が10倍も向上した(COLO−205細胞に対して、TRAILの半数致死量が3.5ng/mLで、RGD−L−TRAILの半数致死量が0.37ng/mLである)。そのため、COLO−205細胞表面のターゲティング発現レベルからも、NGR−L−TRAIL及びRGD−L−TRAILの、COLO−205細胞半数致死量に対する増加レベルからも、RGD−L−TRAILは共にNGR−L−TRAILより優れていた。
腫瘍壊死因子関連アポトーシス誘導リガンドとその腫瘍ターゲティング変異体NGRTRAIL、及びRGD−L−TRAIL変異体に処理された細胞をトリプシンで消化した後、培養プレートから吸出し、リン酸緩衝液で2回洗浄し、300g遠心力下で5分間遠心し、上澄み液を除去し、その後100μL結合緩衝液を用いて改めて懸濁し、最終濃度が2μg/mLのAnnexin V−FITC(BD Pharmgen社)を加えて室温で育成し、10分間後400μLの結合緩衝液を補充し、細胞をフロー分析管内に転移し、管ごとに1μgのピリジニウムヨージド(Sigma社)を加えた。細胞を30分以内にフローサイトメトリーで分析した。各々の細胞系は少なくとも3回繰り返して実験した。
腫瘍細胞アポトーシス誘導活性を評価した(図4)。腫瘍細胞を一連の濃度分布のTRAIL変異体またはTRAILで誘導処理した後、Annexin V−FITCとPI双染色の方法を使用して、フローサイトメトリーで測定分析を行った。その結果、腫瘍細胞が異なると、TRAILに対して感受性が異なることが明示された。COLO−205細胞が最も感受性を有し、HCT−15細胞はその次であり、Hela細胞では、比較的感受性がなかった。但し、Hela腫瘍細胞において、腫瘍細胞アポトーシスを誘導するNGR−L−TRAILの活性は、顕著に向上し、NGR−L−TRAIL対Helaの細胞半数致死量(EC50)は約18.5ng/mLであり、TRAIL対Helaの細胞半数致死量(EC50)は、約145ng/mLであったことから、NGRショートペプチドの追加は、Hela細胞対TRAILの感受性を8倍も向上した。COLO−205及びHCT−15腫瘍細胞に対して、腫瘍細胞アポトーシスを誘導するNGR−L−TRAILの活性は、TRAILよりやや高かった。NGR−L−TRAIL及びTRAILが、これらの腫瘍細胞に対してアポトーシスを誘導する程度の差が、腫瘍細胞表面のCD13分子の発現及び存在比と、正に相関した。
カスパーゼ−8及びカスパーゼ−3の酵素活性は蛍光検出試薬キット(Oncogene社)を使用して測定した。実験はメーカ提供の方法を使用した。蛍光値は酵素標識固相免疫測定装置で測定し、蛍光パラメータは、励起波長400nm、発光波長505nmで行った。
5〜6週齢のメスヌードマウスは上海実験動物センターから購入した。ヌードマウス
に対してまず尾静脈へ100μgの、自然殺傷性細胞を阻害する特異抗体−精製Asialo GM−1抗体(和光純薬工業株式会社、日本)を静注した。24時間後、ヌードマウス背部の右上側に10万個のCOLO−205、HT−15またはHT−29結腸癌細胞を皮下接種した。腫瘍サイズが70mm3に達した時点で、ランダムに群を分け、治療を開始した。組み換え腫瘍壊死因子関連アポトーシス誘導リガンドとその腫瘍ターゲティング変異体NGR−TRAIL、及びRGD−L−TRAIL変異体蛋白を、腹腔を経由して投与し、1日1回投与で、連続で10〜14日間投与した。水溶性のカンプトテシンCP
T−11(11−ヒドロキシ−カンプトテシン、商品名:カンプト、Pharmacia/Upjohn社製品)は週ごとに1回、合計2回投与した。組み換え蛋白及びカンプトテシンは共にリン酸緩衝液によって希釈した。腫瘍サイズをノギスで計測し、次式で求めた。計算式:(長×幅)2/2、とした。
HCT−15モデルにおいて、同様な用量の処理下(400μg/日)で、NGR−L−TRAILは、野生型TRAILより腫瘍抑制効果が良好であり(p<0.05)、RGD−LTRAILよりも強く(p<0.05)、NGR−L−TRAIL(80μg/日)の投与量を、野生型TRAILの1/5までに下げた場合、その腫瘍抑制効果は野生型TRAIL(400μg/日)に相当した(両者の間に有意差はなかった)(図7)。80μg/日投与量のNGR−L−TRAILの腫瘍抑制効果は、すでに400μg/日のRGD−L−TRAILの腫瘍抑制効果と近い効果であった。治療過程において、NGR−LTRAILが、腫瘍の成長を抑制することは用量依存的であった。
以上の結果は、CNGRCペプチドをTRAILと融合した後TRAILのインビボ抗腫瘍活性を顕著に向上させ、さらに、NGR−L−TRAILの抗腫瘍効果がRGD−L−TRAILより顕著に強いことを明確に示した。
インビボ動物実験は、腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の治療効果が、腫瘍壊死因子関連アポトーシス誘導リガンド、ひいては、インテグリンαVβ3、αVβ5ターゲティングTRAIL変異体RGD−L−TRAILの治療効果よりも優れることを十分に証明した。
組み換え腫瘍壊死因子関連アポトーシス誘導リガンドと腫瘍ターゲティング変異体NGR−TRAIL、及びRGD−L−TRAIL変異体をそれぞれ放射性同位体125ヨウ素標識試薬キット(Pierce社)で標識した。標識結果:125I−腫瘍壊死因子関連アポトーシス誘導リガンドの比放射能が7.86μCi/μg蛋白、125I−腫瘍壊死因子関連アポトーシス誘導リガンド変異体の比放射能が7.49μCi/μg蛋白であった。ヌードマウスにCOLO−205結腸癌を接種した後、腫瘍サイズが400〜500mm3に到達した後ランダムに群を分け、野生型群と2種の変異体群とに分け、各群に5回の測定時間点を設け、それぞれ5、30、60、120、240分とし、各時間点で3匹の動物を用いた。各担腫瘍ヌードマウスに、100μLの、5μCi含有の標識蛋白質を注射した。各時間点で、マウスの腫瘍を剥離して且つ重量を測り、液体シンチレーション計数器において放射剤量を測定した。腫瘍組織における蛋白質の分布量の標識は、グラムごとの組織の測定放射量と注射放射量との百分率(%ID/g)で表示した。
腫瘍壊死因子関連アポトーシス誘導リガンド及びその腫瘍ターゲティング変異体NGR−L−TRAIL、RGD−L−TRAIL変異体、ウシ血清アルブミンを、緑色フルオレセインによって標識した。標識した蛋白を、ゲル分子篩Sephadex−G25を用い、遊離のフルオレセインを除去した。腫瘍が400〜500mm3の大きさに成長した担腫瘍マウスに対して500μgフルオレセインで標識した蛋白を尾静脈へ静注し、静注
後30分時点で、マウス腫瘍を手術して剥離し、腫瘍細胞単細胞懸濁液を調製し、生理食塩水で複数回洗浄し、癌細胞を6万個採取してフローサイトメトリー測定を行い、組み換え蛋白質の、腫瘍細胞表面における結合状況を分析した。
験は少なくとも3回繰り返し、細胞アポトーシス及び付着実験の結果は平均値及び標準偏差によって表示し、腫瘍サイズを平均値及び標準偏差で表示した。p<0.05は有意差があり、p<0.01は極めて有意差がある、それぞれ1つの及び2つの「*」でマークした。
なお、他器官におけるNGR−L−TRAILの分布がTRAILと類似し、その他の臓器において特異的に分布することはなかった(図10(a)、10(b))。体内におけるNGR−L−TRAILの代謝は、主に腎臓を経由して体外に排出された(図10(b))。そのため、動物体内腫瘍組織区域分布の増加及び動物モデルにおいて観察された抗腫瘍活性の増強は、設計した分子におけるNGR−L−TRAILがRGD−TRAILと比べより優れており、TRAIL蛋白により優れたターゲティング能を与え、最終的に抗腫瘍効果を向上できることを十分に証明した。
Claims (11)
- 腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体であって、CD13のリガンドペプチドと、ジョイニングペプチドと、腫瘍壊死因子関連アポトーシス誘導リガンドとによって構成された融合蛋白質であり、
CD13の前記リガンドペプチドは環状構造を有するCNGRCペプチドであり、前記ジョイニングペプチドは分岐側鎖を含まない1〜25個のアミノ酸残基から構成されることを特徴とする腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体。 - 前記腫瘍壊死因子関連アポトーシス誘導リガンド変異体を、ポリエチレングリコールと脂肪酸で酸性にして修飾した、または、抗体のFc断片またはアルブミンを加えた組み換えで修飾した腫瘍壊死因子関連アポトーシス誘導リガンド変異体である、ことを特徴とする請求項1に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体。
- 腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体のコード遺伝子を人工合成または遺伝子クローンによって構成し、一般的な遺伝子操作方法を使用して該コード遺伝子の遺伝子発現及び分離精製を実現する、ことを特徴とする請求項1または請求項2に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の作製方法。
- 請求項1または請求項2に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の大腸菌組み換え発現菌株を、35℃〜10℃の低温下で培養及び誘導発現を行う、ことを特徴とする請求項1または請求項2に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の大腸菌における可溶性型発現方法。
- 腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の分離精製方法において、請求項1または請求項2に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体をイオン交換クロマトグラフィー及び金属アフィニティークロマトグラフィーによって精製する、ことを特徴とする腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体の分離精製方法。
- CD13リガンド及びジョイニングペプチドの連続配列をコードするcDNAであって、前記コードが請求項1または請求項2に記載の腫瘍壊死因子関連アポトーシス誘導リガンド変異体から選ばれたcDNA。
- 請求項6のcDNAを含む遺伝子発現用担体。
- 請求項1または請求項2に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体を含有する腫瘍治療薬剤。
- 従来の腫瘍治療薬剤及び技術と併用される、請求項8に記載の腫瘍治療薬剤。
- 請求項6または請求項7に記載の腫瘍ターゲティング腫瘍壊死因子関連アポトーシス誘導リガンド変異体のcDNA及びその遺伝子発現用担体を適用して調製した腫瘍治療薬剤。
- 従来の腫瘍治療薬剤及び技術と併用される、請求項10に記載の腫瘍治療薬剤。
