Description
(A)細胞培養
NB324K、及びHEK293T細胞を、それぞれ最小必須培地(MEM)及びダルベッコ改変イーグル培地中で培養した。なお、培地は、5%(MEM)、又は10%(DMEM)ウシ胎仔血清と、100ユニット/mlペニシリン、100μg/mlストレプトマイシン、2mM L-グルタミン(すべてGibco, Invitrogen, Karlsruhe, Germany製)とを添加したものである。すべての細胞は、5%CO2雰囲気、95%湿度で、37℃に維持された。
Pol III H1 shRNA発現カセットをpSR19(19)、及びΔpSR19(20)プラスミドにクローニングした。前者は全H-1PV wtゲノムを含み、後者はヌクレオチド2022〜2135(パルボウイルスNS2タンパク質のC末端領域をコードする)を欠いた欠失バージョンである。前記HPAI制限酵素部位(NCBI参照配列NC_001358.1によるヌクレオチド4687〜4693)を前記クローニングに使用した。前記カセットを鋳型DNAとしてpSilencer 3.1ベクター(Ambion, Life Technologies, Grand Island, NY, USA)と下記のプライマとを使用するPCRによって増幅した。

すべてのクローンをEscherichia coli 株SURE(Invitrogen, Darmstadt, Germany)中で増やし、DNAをシークエンシングによって確認した。
すべてのウイルスをHEK293T細胞中で産生した。前記細胞は、T75培養フラスコ内で培養し、12.5%の培養密度のウイルスプラスミドの4〜10μg/フラスコで一過的に形質転換された。4日後、細胞をそれらの培地とともに収集し、3回の凍結、及び解凍サイクルで溶解させ、細胞残滓を遠心分離によって除去した。産生されたウイルスを、感染性NB324K細胞によって更に増幅し、イオジキサノール勾配遠心分離によって更に精製した(21)。ウイルス力価測定を、(22)に従って、qPCRとプラークアッセイによって行い、ウイルスゲノム(Vg)、又はプラーク形成単位(PFU)/mlで表した。
図2に記載の実験において、HEK293T細胞を、10 cmディシュで増殖させ、その製造業者の指示に従ってFuGENE HD (Roche, Mannheim, Germany)を使用して約40%の培養密度でトランスフェクションした。2μgのDNAを無血清培地中で、20ng/μlの最終濃度にまで希釈し、その後、7.5μlのFugeneをその混合物に添加した。室温で30分間のインキュベーション後、前記混合物を細胞に液滴によって添加した。
図4に記載の実験において、1.5×105のNB324K細胞を6-ウェルプレートに播種し、特に銘記されない限り、MOI 5 (PFU/細胞)で、H-1PV wt、H-1PV-silencer-GFP、又はH-1PV-silencer-controlで感染させる前に一晩インキュベーションした。感染の12時間後、細胞をMOI 80緑色蛍光トランスダクションユニット(GFU)細胞で使用されるEGFPタンパク質を発現するアデノウイルスで重複感染させ、プロセシング前に更に72時間増殖させた。
ウイルス細胞溶解活性を、その製造業者の指示に従ってLDHアッセイ(CytoTox 96: Promega, Mannheim, Germany)によって測定した。NB324K細胞を、96ウェルプレート(2,500細胞/ウェル)内で50μlの培地中に播種した。24時間後、細胞を、MOI 5(pfu/細胞)で前記ウイルスを含む追加の50μlの培地を添加することによって感染させた。感染の72時間後、細胞をLDH放出測定のために処理した。比色変化を、492nmでマイクロタイターリーダーを使用して測定した。
パルボウイルス感染NB324K細胞からのshRNA抽出を、その製造業者プロトコルに従ってmirVana miRNA単離キット(Ambion, Life technologies, Darmstadt, Germany)を使用して行った。shRNAの検出を、前記mirVana miRNA単離キット(Ambion)をそのインストラクションマニュアルに記載されているように使用して行った。
全RNAをRNeasy Mini RNA精製キット(Qiagen, Hilden, Germany)を使用して細胞から単離した。cDNA合成を、HotStartTaq DNA ポリメラーゼを添加して(+RT)又はその添加無しで(-RT)、ランダムヘキサマープライマ(Promega)を使用して、その製造業者のプロトコルに従ってQuantiTect Probe RT-PCR Kit (Qiagen)を使用して行った。
Power SYBR Green PCR Master Mix(Applied Biosystems, Germany)を使用したTaqMan ABI Prism 7600 Sequence 検出システム(Applied Biosystems, Germany)を使用してQRT-PCRを行った。RNAコントロールの各サンプルを正規化するために、ハウスキーピング遺伝子GAPDHをコントロール遺伝子として使用した。PCRを下記のプライマを使用して行った。

H-1PV-sil-GFP, H-1PV-sil-コントロール、及びH-1PV野生型で感染された細胞を、トリプシン処理しPBSで二回洗浄した。前記細胞ペレットを、50mM Tris-HCl pH 8, 200 mM NaCl, 0.5% NP-40, 1mM DTT, 10%グリセロール、及びプロテアーゼインヒビターの混合物(Roche, Diagnostics, Mannheim Germany)から成る500μlの細胞溶解緩衝液中で細胞溶解し、氷上で20分間維持した。遠心分離(10,000 rpm×10分)後、上清を収集し、タンパク質量をBCAアッセイ(Perkin Elmer)により、その製造業者のマニュアルに従って使用して測定した。全細胞抽出物(30μg)を12% SDSゲル上でロード、及び分離し、ウエットブロッティング(Invitrogen, X-Cell Sure Lock)によってHypbond-Pメンブラン(GE Healthcare)上にトランスファーした。それらのメンブランを、PBS、0.05% Tween 20、5%ノンファットドライミルク中で室温にて1時間ブロッティングした。そのブロットを下記の一次抗体と一晩4℃でインキュベーションした。 GFPウサギ(Santa Cruz Biotechnologies, Heidelberg, Germany)、β-Tubulinマウス(Sigma Life Science, Hamburg, Germany)。メンブランの洗浄後、ペルキシダーゼ共役ヤギ抗ウサギ、又はヤギ抗マウス抗体(Santa Cruz Biotechnology, Heidelberg, Germany)を室温で1時間添加した。その後、メンブランを洗浄し、Western Lightning Plus-ECL検出キット(Perkin Elmer, Rodgan, Germany)で可視化した。
NB324K細胞を、6ウェルプレート中で成長させ、その後、上述したように感染させた。72時間後、細胞を1×PBSで二回洗浄し、4℃で4%のパラホルムアルデヒド(PFA)で固定し、PBSで再び洗浄した。Hoechst 33342色素を使用して核染色を行った。固定された細胞を、Leica DMIL蛍光顕微鏡で調べた。GFP強度を、ImageJソフトウエア(23)を使用して定量化した。
shRNA発現カセットを持つ(harbouring)複製可能H-1PVウイルスを作り出すための戦略が考えられた。shRNA発現のために、他のカセットの挿入に耐える可能性が最も低いと考えられるPV(最大で300bp)の制限されているDNAパッケージング能力により、前記RNAポリメラーゼIII H1プロモーター(総サイズ170〜180塩基)が使用された。挿入によってなんらかのウイルスORFが破壊されることを回避するために、前記カセットをVP遺伝子(カプシドタンパク質をコードする)の下流側のH-1PV未翻訳領域、即ち、ヌクレオチド4683(前記パルボウイルスゲノム内のHpaI制限酵素部位)に組み込むことを決定した。適当な突出を有するアニーリングされたオリゴヌクレオチド使用することによる前記カセットへのshRNAのクローニングを容易にするためにユニークな制限部位を導入した(図1)。前記カセットの挿入のために、二つの異なるパルボウイルスバックボーン、即ち、全長H-1PVゲノムを含むpSR19 (19) と、ヌクレオチド2022〜2135を包含する(encompassing)欠失を含むH-1 dr(20)を使用した。この欠失は、パルボウイルス複製とヒト細胞の感染に干渉せず、従って、外来導入遺伝子の挿入のためのわずかに大きな遺伝子空間を提供する。前記新規プラスミドを、pH-1PV Silencer 1とpH-1PV Silencer 2と命名した。ここで提示される結果は、pH-1PV Silencer 2(以下、pH-1silと略称する)に関するものであるが、他方のプラスミドも使用した場合も類似の結果が得られた。前記新規プラスミドの遺伝子サイレンシングを誘導する有効性をテストするために、高感度緑色蛍光タンパク質(Enhanced Green Florescent Protein (EGFP))(pH-1sil-GFP)をコードする遺伝子に対するshRNAとコントロールshRNA(既知の何れの遺伝子配列も認識しないスクランブルされたshRNA配列)(pH-1sil-control)を導入し、二つのプラスミドをHEK293T細胞に一過的にトランスフェクションした。ポジティブ、及びネガティブコントロールとして、市販のプラスミド、即ち、同じEGFP、又はスクランブルされたshRNAを有するpSilencer 3.1(Ambion, Austin, Texas, USA)を使用した。トランスフェクションの16時間後、細胞を、前記EGFP遺伝子を有する組み換えAd 5ウイルスで感染させた。その後、細胞を、蛍光顕微鏡下で分析した。前記pSilencer 3.1プラスミドと同様、pH-1sil-GFPはEGFPのサイレンシングにおいて非常に効率的であり、その発現を60%以上も低減した(図2)。
次に、H-1PV-silウイルスがshRNAを発現する能力を実証した。NB324K細胞を、H-1PV-sil-GFPとH-1PV wt(ネガティブコントロールとして使用)で感染させた。shRNA発現用のポジティブコントロールとして、前記細胞をpSilencer 3.1 shRNA-EGFPベクターでトランスフェクションした。72時間後、前記細胞のshRNA-GFP含有量を分析した。H-1PV-sil-GFP感染細胞において高レベルのshRNAが検出された(図5A)。
次に、H-1PV-silencerのEGFP発現をノックダウンする能力を調べた。NB324K細胞をH-1PV-sil-GFP、又はH-1PV-sil-controlウイルスで感染させ、12時間後、EGFPタンパク質を発現するAd5で重複感染させた。RNA抽出のために処理される前に、更に72時間細胞を増殖させた。定量的リアルタイムPCRは、H-1-sil-GFPで感染された細胞におけるEGFPの発現が、コントロールウイルスに見られる発現と比較して80%以上劇的に低減したことを示した(図5B)。

Claims (18)
- 細胞中の標的遺伝子の発現をダウンレギュレートするためのげっ歯類パルボウイルスであって、前記細胞中のRNAポリメラーゼによって認識可能なプロモーター又はプロモーター領域の制御下で前記パルボウイルスゲノムの未翻訳領域内に発現可能な標的特異性核酸を含み、ここで、前記標的特異性核酸の転写物がRNAiであり、そして、前記細胞中で複製及び増殖可能であることを特徴とするパルボウイルス。
- 前記標的遺伝子は、病原遺伝子である請求項1に記載のパルボウイルス。
- 前記病原遺伝子は、(a)動物に対する病原性を有するウイルス遺伝子、(b)ヒトのガンの原因となる、または発ガンに関与している遺伝子、(c)ガン遺伝子、(d)抗アポトーシス遺伝子、(e)腫瘍細胞成長、転移、血管新生、若しくは化学療法耐性に関係する遺伝子、(f)免疫調節遺伝子、又は(g)サイトカイン、成長因子、酵素、若しくは転写因子をコードする遺伝子である請求項2に記載のパルボウイルス。
- 宿主細胞中で複製、及び増殖する能力を有する請求項1〜3の何れか一項に記載のパルボウイルス。
- LuIII、マウス微小ウイルス(MMV)、マウスパルボウイルス(MPV)、ラット微小ウイルス(RMV)、ラットパルボウイルス(RPV)、ラットウイルス(RV)、又はH1(H-1PV)である請求項1〜4の何れか一項に記載のパルボウイルス。
- 前記標的特異性核酸は、パルボウイルスVP遺伝子の下流側の未翻訳領域に挿入される請求項1〜5の何れか一項に記載のパルボウイルス。
- 前記標的特異性核酸は、野生型H-1PVゲノムのヌクレオチド4683に挿入される請求項6に記載のパルボウイルス。
- 前記細胞中のRNAポリメラーゼによって認識可能なプロモーター又はプロモーター領域は、RNAポリメラーゼII(Pol II)、又はIII(Pol III)プロモーターである請求項1〜7の何れか一項に記載のパルボウイルス。
- 前記RNAポリメラーゼ III(Pol III)プロモーターは、RNAポリメラーゼ III H1プロモーターである請求項8に記載のパルボウイルス。
- 前記標的特異性核酸は、shRNAである請求項1〜9の何れか一項に記載のパルボウイルス。
- 前記標的特異性核酸は、少なくとも15ヌクレオチドの長さを有する請求項1〜10の何れか一項に記載のパルボウイルス。
- (a)動物に対する病原性を有するウイルス遺伝子、(b)ヒトのガンの原因となる、または発ガンに関与している遺伝子、(c)ガン遺伝子、(d)抗アポトーシス遺伝子、(e)腫瘍細胞成長、転移、血管新生、若しくは化学療法耐性に関係する遺伝子によって引き起こされる疾患、又は(f)免疫調節遺伝子、若しくは(g)サイトカイン、成長因子、酵素、若しくは転写因子をコードする遺伝子の異常発現に関連する疾患の治療における使用のための請求項1〜11の何れか一項に記載のパルボウイルス。
- 前記使用は、腫瘍の治療のためである請求項12に記載の使用のための請求項1〜11の何れか一項に記載のパルボウイルス。
- 前記使用は、脳腫瘍の治療のためである請求項13に記載の使用のための請求項1〜11の何れか一項に記載のパルボウイルス。
- 前記腫瘍の細胞は、化学療法、及び/又は放射線療法耐性である請求項13又は14に記載の使用のための請求項1〜11の何れか一項に記載のパルボウイルス。
- 静脈内(i.v.)、腫瘍内、又は気管支内投与される請求項12〜15の何れか一項に記載の使用のための請求項1〜11の何れか一項に記載のパルボウイルス。
- 請求項1〜11の何れか一項に記載のパルボウイルスを含む細胞。
- 請求項17に記載の細胞を含むトランスジェニック非ヒト動物。
