Description
本発明は、HEV VLPに基づくペプチド/核酸(P/N)系に関する。HEV VLPに基づくP/N系を使用するという考えにより、MHCクラスIおよびII系を通した免疫応答の誘導に加えて、幾つかの独特な利点が提供される。1)P/N系によって経口ワクチン接種の可能性が提供される。実際、経口投与はストレスが少なく、かつ専門的な技術を必要としない。加えて、腸管を通してのワクチン送達は、全身性の注射よりも安全であるとみなされる。天然のHEV感染経路およびHEV VLPの構造は、胃内の低pH、タンパク質分解酵素の存在、ならびに粘膜表面に付随する物理的および化学的なバリアーの存在のような、消化管内における混乱した環境に抵抗するものである。2)HEV VLPは標準的な培養プロトコルにより産生することができ、精製したHEV−VLPの収率は50〜100μg/mlであり、これはその他のVLPに比較して約100倍大きい。3)HEV VLPは、アミノ酸免疫源を正二十面体粒子の形で送達する。すなわち侵入に成功した全ての粒子は、同じホスト細胞に60コピーの免疫源を送達する。4)HEVに対する免疫応答は、DNA投与およびペプチド免疫源ワクチン接種の両方に効果を示さないことが証明されている。5)HEVは室温で安定である。これら全ての特色を総合することにより、P/N系は、本発明者らの目的である有効かつ広い反応性を持つワクチンの開発を達成するであろうと予想される。
「E型肝炎ウイルス」または「HEV」は、i)水媒介性であり、感染性肝炎の原因となる;ii)血清学的な特性の点から、A型肝炎ウイルス(hepatitis A virus:HAV)、B型肝炎ウイルス(hepatitis B virus:HBV)、C型肝炎ウイルス(hepatitis C virus:HCV)、またはD型肝炎ウイルス(hepatitis D virus:HDV)とは区別される;iii)アメリカ合衆国培養細胞系統保存機関(American Type Culture Collection:ATCC)に受入番号67717により保管されているE.coli株に統合されるプラスミド、pTZKF1(ET1.1)に挿入されている、1.33kbのcDNAに相同なゲノム領域を含む、ウイルス、ウイルス型、またはウイルスのクラスを指す。
E型肝炎ウイルス(Hepatitis E virus:HEV)は、重篤な急性肝不全の原因となることが知られている。HEVは、ヘペウイルス科ファミリーのヘペウイルス属に属する。HEVは、約7.2−kbの一本鎖プラス鎖RNA分子を含む。RNAは3’ポリアデニル化され、三つの翻訳領域(open reading frames:ORF)を含む。ORF1は、ゲノムの5’側の半分に位置する、ウイルス性の非構造タンパク質をコードする。0RF2は、ゲノムの3’側の半分に位置する、タンパク質形成性のウイルスカプシドをコードする。ORF3は、ORF1のC末端およびORF2のN末端に重複する13.5−kDaのタンパク質をコードする。ORF3は膜および細胞骨格分画に関連する。

本発明はまた、本発明の改変HEVカプシドタンパク質により形成されたキメラVLP内に被包された異種性核酸を含んでなる、医薬組成物または生理的組成物も提供する。このような医薬または生理的組成物は、1以上の薬剤的もしくは生理的に許容される賦形剤または担体も含む。本発明の医薬組成物は、様々な薬物送達系における使用に適する。本発明における使用に適した処方は、Remington's Pharmaceutical Sciences, Mack Publishing Company, Philadelphia, PA, 17th ed. E型肝炎ウイルス(HEPATITIS E VIRUS:HEV)のカプシド構造
E型肝炎は、世界中で経腸感染非A型、非B型肝炎の大多数を占める。衛生状態が悪く、高い人口密度を有する国において、E型肝炎は、妊婦に実質的な死亡率(20%)を伴う水媒介性の流行病を引き起こす(Zhou et al., 2005. Vaccine 23:3157-65.)。原因媒介物は、ヘペウイルス科ファミリーのヘペウイルス属に属する、非エンベロープ型一本鎖プラス鎖RNAウイルスの、E型肝炎ウイルス(hepatitis E virus:HEV)である(Emerson et al., 2004. Hepevirus. Elsevier/ Academic Press, London.)。
IgGの産生および精製:8週齢の雌BALB/cマウスを、HEV VLP(100μg/ml)の腹腔内接種によって0および4週目に免疫した。4週間後に同量の抗原を投与して最終の追加免疫を行った。最終の追加免疫から3日後、基本的にはAdlerおよびFaineによる記載に従い(Adler et al., 1983. Pathology 15:247-50)、ポリエチレングリコール1500(50%[重量/容量])(Boehringer, Mannheim、独国)を用いて、マウスの脾臓細胞をP3U1マウスミエローマ細胞と融合させた。ハイブリドーマの増殖がみられたマイクロプレートウェルからの上清を、組み換えHEV VLPを抗原として用いた酵素結合免疫吸着測定法(ELISA)によりスクリーニングした。HEVに対する特異的な抗体を分泌するハイブリドーマを、それらがモノクローナルであるとみなされた後で、限界希釈法により3回サブクローン化した。上清中の抗体を、マウスモノクローナル抗体アイソタイピングキット(Amersham、リトル・チャルフォント、バッキンガムシャー州、英国)を製造者のプロトコルに従って用いて、アイソタイプ決定した。ハイブリドーマを、15%FCSを含んだPRMI−1640を用いて、静止したフラスコ(Nunc、ロスキレ、デンマーク)内で大量増殖させた。上清を回収し、抗体をHiTrapプロテインGアフィニティーカラム(Pharmacia Biotech AB、ウプサラ、スウェーデン)を用いて精製した後、−80℃に保管した。マウスモノクローナル抗体MAb224は免疫グロブリンG1(IgG1)アイソタイプを有した。
MAb224はHEV ORF2タンパク質の残基595〜601を認識する:抗体MAb224により認識されるエピトープを特徴付けるため、ウェスタンブロット法を行った。一連のC末端切断型ORF2タンパク質を10%SDS−PAGEを用いて分離し、MAb224によりブロットした(図1A)。残基112〜600、112〜596、および112〜589を含んでなる切断型ORF2タンパク質は、MAb224と相互作用できなかった。これらのデータにより、残基601がMAb224の結合に最も重要であることが示された。抗原性エピトープは通常3〜7aaの長さの短い領域であることを考えると、Mab224結合部位は、ORF2タンパク質のaa位置595〜601に位置するはずであり、かつSchofieldらにより報告された残基578〜607の中和エピトープ(Schofield et al, 2000. J Virol 74:5548-55.)内にある。MAb224により検出された最も低分子量のタンパク質は、ORF2タンパク質がN末端で分解した産物である。
Tn5またはSf9細胞のいずれかに切断型HEV ORF2タンパク質を発現させることにより、同様の構造を持つVLPの自己集合がもたらされる(Li et al, 2005. J Virol 79: 12999-3006.)。組み換えVLPおよび天然ウイルス粒子の両方は、実験動物において全身性および粘膜性の免疫応答(Li et al., 2001. Vaccine 19:3476-84.)、およびHEV特異的な中和免疫応答(Li et al., 2004. Vaccine 22:370-7.)を誘導できることから、抗原特性を共有する。以前に同定されたクライオEM三次元構造により、組み換えHEV VLPは、カプシド殻の上、〜43Åに放射状に突出した30の二量体の突起を持つ、T=1正二十面体粒子を形成することが示された(Li et al., 2005. J Virol 79: 12999-3006; Xing et al., 1999. Virology 265:35-45.)。突起は粒子の最も露出された部分であることから、これらは抗原性を確定するものであると思われている。本発明者らの結果は、HEV ORF2タンパク質配列の、その空間的な立体配置への関連についての最初の証拠を提供するものである。
結晶構造に基づく、E型肝炎ウイルス様粒子の生物学的および免疫学的特性
E型肝炎ウイルス(Hepatitis E virus:HEV)は、急性肝炎の原因媒介物である。カプシドタンパク質から成るHEV様粒子(HEV-like particles:HEV−LP)の結晶構造を、3.5−Åの分解能にて決定した。カプシドタンパク質は、突起および中央ドメインにおいて、カリシウイルス科およびトンブスウイルス科のファミリーメンバーとは非常に異なるフォールディングをを示すが、殻ドメインにおいては共通のフォールディングを共有する。5回軸におけるTyr−288は、HEV−LPの集合において鍵となる役割を果たし、芳香族アミノ酸残基は、構造的に関連するウイルスの間でよく保存されている。変異解析により、突起ドメインは、HEV感染に感受性である細胞への結合に関わり、幾つかの中和エピトープを有することが示された。これらの構造的および生物学的な知見は、HEVの集合および侵入の分子機構を理解するために重要であり、また、E型肝炎の防止的および予防的な方策の開発における手がかりも提供する。
遺伝子型3のHEV−LPの調製。遺伝子型1株のHEV−LPの場合について記載されたように(Li TC, et al, J Virol, 71 :7207-7213 (1997); Li TC, et al, J Virol, 79:12999-13006 (2005); Xing L, et al., Virology, 265:35-45 (1999))、遺伝子型3株由来の切断型カプシドタンパク質(アミノ酸112〜608)のゲノムを有する組み換えバキュロウイルスをポリヘドリンプロモーターの制御下で感染させることによって、大量のHEV−LPが培養上清中に分泌された。さらなる構造的、および生物学的解析のために、精製した遺伝子型3のHEV−LPを使用した。
切断型HEVカプシドタンパク質(アミノ酸112〜608)の昆虫細胞内での発現により、天然HEV粒子に同様の抗原性を保持したHEV−LPの集合がもたらされた(Li TC, et al., J Virol, 71 :7207-7213 (1997); Li TC, et al., Virology, 349:222-229 (2004))。T=1対称性のこの粒子の直径は270Åであり、患者の糞便検体に検出された天然ウイルス粒子の直径である320Åよりも小さい(Bradley D, et al., J Gen Virol, 69:731-738 (1988))。HEV−LPの内腔は、長さ7.8kbのウイルス性RNAを収容するには非常に小さく(Xing L, et al., Virology, 265:35-45 (1999))、粒子がヌクレオチドを含有しているという証拠はない(Li TC, et al., J Virol, 71 :7207-7213 (1997); Xing L, et al., Virology, 265:35-45 (1999))ことが報告されてきた。従って天然HEV粒子は、HEV−LPよりも多数の、および/または大きなサイズのカプシドタンパク質から構成されることが示唆される。T=3対称性の植物ウイルスの幾つかの場合には、カプシドタンパク質は、N末端塩基性領域の欠失(Hsu C, et al., Virology, 349:222-229 (2006); Kakani K et al., J Virol, 82:1547-1557 (2008))またはN末端領域もしくはN末端領域とSドメインの間のリンカードメインのいずれかにおけるアミノ酸置換(Kakani K et al., J Virol, 82: 1547-1557 (2008))によってT=1対称性の粒子へ集合することから、N末端塩基性領域はT=3からT=1対称性への移行の切り替えに重要な役割を果たしていることが示唆される。加えて、NVカプシドタンパク質の昆虫細胞内での発現により、T=3大粒子のみではなく、T=1対称性を持つと思われる小粒子の産生ももたらされた(White LJ, Hardy ME, Estes MK, J Virol, 71 :8066-8072 (1997))。構造的に関連したウイルスの、カプシド構造およびそれらのパッケージングについての多くの類似性に基づき、天然HEV粒子はT=3表面格子を有することが示唆される。MおよびPドメインに連結するプロリンリッチなヒンジの柔軟性により、カプシドタンパク質二量体の、T=3ウイルスに見出されるA/BおよびC/Cサブユニットの間での立体構造の切り替えが可能になる。天然HEVの構造は、HEV−LPのそれとはわずかに異なる可能性があるが、この研究でHEV−LPを用いて得られたデータは、HEVのウイルス粒子の構造、生活環、および病原性の理解に有用な情報を提供するはずである。Sドメインは、その他の幾つかの正二十面体ウイルスとゼリーロールフォールディングを共有する(Prasad BV, et al, Science, 286:287-290 (1999);Chen R et al, Proc Natl Acad Sci USA, 103:8048-8053 (2006); Morgunova E, et al., FEBS Lett, 338:267-271 (1994); Hogle JM, Chow M, Filman DJ, Science, 229:1358-1365 (1985); Tsao J, et al., Science, 251: 1456-1464 (1991))。Sドメイン間の相互作用において構築された五量体構造の中心に位置するTyr−288が置換されたカプシドタンパク質は、HEV−LPへ集合できないことが見出された。GeneBankから入手可能なデータを用いたアミノ酸配列のアラインメント解析により、Tyr−288はHEVの5遺伝子型の間で完全に保存されていることが示された。さらに、HEVカプシドタンパク質のTyr−288に対応する残基は、rNV(Phe−118)、SMSV(Tyr−330)、およびCARMV(Phe−145)の構造内に、これらの芳香族残基の位置は異なっていたものの、見出された。H−Iループに位置するHEVのTyr−288およびSMSVのTyr−330、ならびにD−Eループに位置するrNVのPhe−110は粒子表面の外側に露出し、一方でD−Eループに位置するCARMVのPhe−145は粒子内部に露出する。これらのデータにより、これらの残基の芳香族側鎖は、隣のサブユニットのそれとの疎水性相互作用に関与し、安定した粒子の集合を確実にすることが示唆される。細胞への侵入の間に、殻からのゲノム放出のため、ウイルス粒子構造の再構成が必要とされる。しかしながら、HEVの侵入および脱殻機構については不明なままである。五量体の中心は粒子の最も薄い領域であり、かつチャネル様の構造であるため(Xing L, et al., Virology, 265:35-45 (1999))、この領域もまた脱殻およびウイルス性RNAの放出に重要であり得る。ポリオウイルスの5回軸は、受容体分子への結合により開始されるウイルス粒子の立体構造変化を通した、ゲノムRNAの移行に関与することが提唱されてきた(Belnap DM, et al. J Virol, 74: 1342-1354 (2000); Bubeck D, Filman DJ, Hogle JM, Nat Struct Mol Biol, 12:615-618 (2005))。
HEV−LPの発現、精製、および結晶化。HEV遺伝子型3のORF2をコードする組み換えバキュロウイルスの2712株を昆虫細胞内に発現させた。HEV−LPを以前に記載されたように精製し(Xing L, et al., Virology, 265:35-45 (1999))、ハンギングドロップ蒸気拡散法により結晶化した。詳細をSI材料および方法に報告する。

カプシド突起ドメインにおける主な抗原性領域の構造的な性質決定
E型肝炎ウイルス(Hepatitis E virus:HEV)は、急性肝不全の原因となるヒトの病原体である。C末端から52アミノ酸、およびN末端から111アミノ酸を欠失させて昆虫細胞に発現させた場合、カプシドタンパク質は天然HEVウイルス粒子の抗原性を保持したT=1ウイルス様粒子(virus like particle:VLP)へ自己集合することができる。ここではクライオ電子顕微鏡およびイメージ再構成を用いて、抗HEVモノクローナル抗体が、カプシドタンパク質の突起ドメインに、VLPあたり60コピーのFab断片によって結合したことを特定した。HEV結晶構造の分子ドッキングにより、抗体Mab224の結合部位は、表面水平域の周縁における3個のPORF2表面ループを覆うことが明らかになった。この抗体結合部位は、キメラHEV VLPのクライオEM構造により決定されたように、挿入されたB細胞タグ、PORF2タンパク質のC末端に融合された11アミノ酸のエピトープの潜在的な位置から離れている。従って、T=1HEV VLPは、HEVおよび外来エピトープの両方に対する抗体を効果的に誘導する、強固な送達候補である。
抗HEVモノクローナル抗体の産生および精製

分離したFab断片を調製するため、パパイン切断を用いた。パパインの活性化のために還元L−システイン緩衝液を用い、MAb224とパパインを100:1のモル比で混合した。混合物を30℃にて一晩インキュベートした。ヨードアセトアミドを加えて反応を消失させ、産物をSDS−PAGEにより分析した。プロテインAカラムを製造者の指示に従って用い、Fab断片を精製した。Fc断片および未切断のMAb224をプロテインAに対する親和性によってカラム内に捕捉する一方、Fab断片をフロースルー画分中に回収した。
HEV VLPの産生および精製は、以前に記載されたプロトコルに従って行った(Li, T. C. et al., J Virol 71:7207-7213 (1997); Niikura, M. et al., Virology 293:273-280 (2002))。簡単に述べると、N−切断型ORF2タンパク質(野生型VLP用)、およびキメラORF2タンパク質(VLP/Cタグ用)を含むDNA断片を、バキュロウイルス転移ベクターpVL1393によりクローン化してpVLORF2を得た。組み換えバキュロウイルスをSf9昆虫細胞(理研細胞バンク、つくば市、日本)より産生し、次にM.O.I.>5にてTn5昆虫細胞に感染させた。感染した昆虫細胞を、EX-CELL(商標)405培地(JRH Biosciences、レネクサ、カンザス州)中で6日間、26.5℃にて培養した。10,000g、90分間の遠心分離によって細胞片を除去した後、培地を回収した。上清をBeckman SW32 Tiローターで、30,000rpmにて2時間遠心沈殿させ、ペレットを4.5mlのEX-CELL(商標)405に再懸濁した。ペレットはHEV VLPを含んでおり、これをCsCl密度勾配内でさらに精製した。白いウイルスバンドを回収し、遠心分離(Beckman TLA 55ローターで45,000rpm、2時間)によってCsClを除去するため、EX-CELL(商標)405で4倍に希釈した。VLPを100〜500μlの10mM カリウム−MES緩衝液に再懸濁し、4℃にて保管した。キメラVLP/Cタグを構築するため、単純ヘルペスウイルスの糖タンパク質D由来のB細胞タグエピトープ「QPELAPEDPED」を、アミノ酸608の後に挿入することにより、組み換えバキュロウイルスを調製した(Schofield, D. J. et al., Vaccine 22:257-267 (2003))。VLP/Cタグは、上記のプロトコルによって調製および精製した。
Fab断片とVLPとを、1:300を超えるモル比にて、4℃にて一晩インキュベートすることにより、VLP/Fab複合体を調製した。Sephacryl-300を含む短いゲル濾過カラムに試料を流すことによって、未結合のFab断片を除去した。OD280nmの読み取りに基づき、VLP/Fab複合体を含む画分を同定した。精製VLP/Fab複合体をアクリルアミドゲル(勾配8〜25%)に負荷し、Phast(商標)システム(Pharmacia)にて定電圧条件下で電気泳動を行うSDS−PAGEによって、Fab結合占有率を大まかに推定した。2%酢酸ウラニル(uranyl acetate:UA)による逆染色の後、電子顕微鏡(electron microscopy:EM)によって粒子の完全性を調べた。
試料調製およびクライオEMデータ収集は、以前に記載された、十分に確立されている手順に従った(Xing, L. et al., Virology 265:35-45 (1999))。簡単に述べると、3.5μlの試料を含んだ液滴を、グロー放電した自家製穴あきカーボングリッドに載せ、余分な液体を濾紙片で吸収して除去し、液体窒素で冷却した液体エタン内へ素早く入れた。次に凍結水和した検体グリッドを、続くデータ収集のために低温環境(−178℃)に検体を保持するGatan 626DHクライオホルダーと共にFEI CM-120電子顕微鏡へ移した。顕微鏡像を、デフォーカス範囲1000ないし3000nm、拡大率45,000xにて、低線量条件下(<10e−/Å2)でKodak SO 163フィルムに記録した。顕微鏡像を、適切な粒子濃度、至適な氷の厚さ、および最小の検体ドリフトという基準に基づいて視覚的に選択した。これらの基準を満たした顕微鏡像のみを解析した。
選択した顕微鏡像を、検体空間におけるピクセルあたり3.11Åに対応する14μlの走査ステップサイズで、Heidelberg Primescanner D8200(Heidelberg、独国)を用いてデジタル化した。粒子を、Robem(v2.14)を用いて用手で選別し、円形の平均イメージに対するそれら各々の相互関連によって集合させた。各顕微鏡像の非点収差およびデフォーカス値を、顕微鏡像内の全ての選択された粒子からのパワースペクトルを重ねることによって評価した。構造決定に用いたイメージの最初の零点は、約17〜20Å−1の範囲内に位置した。VLP/Cタグ、VLP/MAb224、およびVLP/MAb4それぞれの初期モデルを産出するため、セルフコモンラインアルゴリズムを用いた(Crowther, R. A. Philos Trans R Soc LondB Biol Sci 261:221-230 (1971))。粒子の起点および配向の精密化は、極フーリエ変換(polar Fourier transformation:PFT)アルゴリズムを繰り返し用いて実行した(Baker, T. et al, J. Struct. Biol. 116:120-130 (1996))。三次元再構築は、正二十面体非対称性単位内に均一に分布した配向を有する粒子のセットを、フーリエ−ベッセルアルゴリズムと組み合わせる一方で、5−3−2正二十面体対称に重ね合わせて算出した。三次元再構築の信頼性を試験するため、データセットを均等に二つの部分に分割し、二つの三次元再構築を算出した。フーリエシェル相関(Fourier shell correlation:FSC)を用い、フーリエ空間におけるこれら二つの再構築間の一致を評価することによって分解能を推定し;係数値0.5を基準として用いて、VLP/Cタグ、VLP/Mab224、およびVLP/Mab4の有効な分解能をそれぞれ17.5Å、18.5Å、および24Åと推定した。
プログラムO(Jones, T. A. et al., Acta crystallogr. Sect. A 47: 110-119 (1991))を用い、PORF2結晶構造の並進的および回転的な動きによって、クライオEM密度マップへの用手によるフィッティングを行った。最もよい適合を得るため、PORF2サブユニットの座標を剛体として扱った。フィッティングの結果を最適化するため、対称関連分子を生成し、サブユニット間の衝突により判断した。フィッティングを、クライオEM密度と、適合したPORF2の座標から算出された密度との間の相関係数(CC値)に基づいて評価した。cc値が80%に達したとき、フィッティングは安定であるとみなした。フィッティングにおける図はPyMOLプログラムを用いて作成し(DeLano, W. L. DeLano Scientific, Palo Alto, CA, USA (2002))、HEV VLPの表面ステレオ投影はRIVEMプログラムを用いて作成した(Xiao, C. et al., J Struct. Biol. 158:181-186 (2007))。
抗体Mab224の結合は、残基601〜608のタンパク質の存在に依存する
現在、ORF2エスケープ変異体を試験するための細胞培養系は利用できないため、モノクローナル抗体Mab224のPORF2への結合を、ウェスタンブロット法によって性質決定した。一連のC末端切断型PORF2タンパク質を、還元条件下で10%SDS−PAGEを用いて分離し、Mab224によってブロットした(図1A)。Mab224は、112〜660、112〜608、112〜602、および112〜601の領域を含むPORF2由来のペプチドを認識した。対照的に、残基112〜600、112〜596、および112〜589を含んでなるペプチドは、Mab224と反応しなかった。これらのデータにより、残基Leu601が、Mab224のPORF2タンパク質への結合に決定的であることが示唆される。Mab224により検出された、より小さな分子量のバンドは、Mab224の結合配列を含む分解したORF2タンパク質である。
クライオ電子顕微鏡像において、キメラHEV粒子およびFab結合VLP複合体の両方は、表面から伸びる尖った密度を持つ円形の輪郭を有していた(図1C)。これらは空の粒子に見え、本発明者らが以前にHEV天然VLPによって観察した形態に類似していることから(Li, T.-C. et al., J. Virol. 79:12999-13006 (2005); Xing, L. et al., Virology 265:35-45 (1999))、これらのVLPにRNA成分は欠如していることが示される。両VLPのサイズは、VLP/MAb224の表面からさらに伸びる余分な密度を考慮しないで、約27nmである(図1C、右)。
VLP/Mab224のクライオEM構造は615のイメージから再構成され、粒子あたり60サブユニットのT=1正二十面体対称を示した。各正二十面体2回軸に位置する、30の二量体突起スパイクが存在した(図14A)。各VLP粒子の周囲に60のFab断片が観察され、Pドメインの側面から突き出ていた。Fab密度は、スパイク表面から〜57Åの高さとして測定された。Fab断片に対応する密度は、HEV VLPのそれにほぼ等しい規模であったことから、60の結合部位の大部分または全てはFab分子によって占有されていることが示される。HEV VLPの密度マップ(Xing, L. et al, Virology 265:35-45 (1999))を用いて、VLPカプシドに対応する密度をクライオEMマップから除くことにより、Mab224抗体の結合部位を特定するために用いるFab相違密度マップを作成した(フットプリント)。
11アミノ酸のB細胞タグをPORF2のC末端に組み入れてPORF2融合タンパク質を構築し、そこからキメラHEV VLP(VLP/Cタグ)が集合した(図1c)。個々の粒子の全782イメージを、VLP/Cタグの最終的な三次元構造モデルを再構成するために用いた。以前に発表されたVLPのクライオEM構造に一致して、VLP/Cタグの表面もまた、二つの異なる層としての正二十面体殻および突起スパイクに分けることができた(図16A)。スパイク突起は正二十面体殻から外部へ向かい、PORF2二量体から構成される。二つの隣接するPドメインの上端表面間で測定した各2回軸間の距離は〜76Åであり、Tn5昆虫細胞から得られたVLPおよびSf9昆虫細胞から得られたVLPにおいて測定された結果に一致する。キメラVLP/Cタグ内に、有意なRNA密度は見出されなかった。
HEV T=1 VLPは、非ヒト霊長類に抗HEV抗体を誘導する組み換えウイルス様粒子である(Li, T. -C. et al., Vaccine 22:370-377 (2004))。これは、経口投与を通した外来エピトープ(Niikura, M. et al., Virology 293:273-280 (2002))、およびDNAワクチン(Takamura, S. et al., Gene Ther 11:628-635 (2004))を送達するための抗原担体としても使用できる。従って、抗体認識部位における構造解析が、VLPに基づく抗ウイルス戦略の開発には必須である。この目的のため、本発明者らは、抗体Mab224(VLP/Mab224)および抗体Mab4(VLP/Mab4)と複合したHEV VLPの構造、ならびにPORF2 C末端にB細胞タグを有するキメラHEV VLP(VLP/Cタグ)の構造を決定した。PORF2結晶構造のドッキングにより、HEV抗原性ドメインについての空間的な情報、および外来エピトープ挿入をより良く設計するための構造ガイダンスが提供される。
HEVにおける抗原性の性質および中和機構を特徴付けることは、細胞培養における適切な複製系が不足しているという事実のために困難である。従って、HEV免疫学に対する我々の理解は、主に大腸菌(Escherichia coli:E. coli)内で発現された組み換えタンパク質、および昆虫細胞内で発現された組み換えタンパク質またはHEV−VLPを用いた研究に基づいている。全ての証拠により、PORF2のC末端領域がHEVの免疫応答に関与していることが示され、HEVの中和エピトープは立体構造的であることが示唆された。後に、HEV中和抗体の誘導に必要とされる最小ペプチドはPORF2の148残基、アミノ酸459〜607と同定された(Zhou, Y. H. et al., Vaccine 23:3157-3165 (2005))。このペプチド領域はPドメインに一致することが、PORF2の結晶構造で明らかにされた。本発明者らのクライオEM構造によって、Fab断片の全体がスパイクに付着することが明らかになり、Pドメインが主にHEVの抗原性を有することが実験的に示された。Mab4は、チンパンジーのcodaライブラリーからファージディスプレイにより単離されたORF2タンパク質に対するチンパンジー抗体である(Schofield, D. J. et al., J Virol 74:5548-5555 (2000))。これは天然HEVウイルス粒子およびアミノ酸597〜607を含む組み換えPORF2ペプチドに結合する(Schofield, D. J. et al., Vaccine 22:257-267 (2003))。本発明者らはHEV/Mab4の構造のフィッティングを行ったが、Fab断片に対応する密度はアミノ酸606の位置を網羅したものの、決定的なMab4の結合配向を提供するためにはMab4の密度は弱すぎた(データ未提示)。ウェスタンブロットにおいて、Mab224がアミノ酸601〜608を欠如したペプチドと反応しないように見える理由は不明であるが、Mab224の結合部位は、変異誘発によって以前に判定された決定的な抗原性残基に一致する。荷電した残基であるE479、K534のアラニンへの、同様に疎水性アミノ酸Y485およびI529のアラニンへの点突然変異により、中和抗体による反応性が選択的に抑止され得ることが見出された(Li, S. et al, PLos Pathog. 5:el000537 (2009))。変異体の別のセットにより、抗体が認識する残基と同じ領域が、ループAB、CD、およびEFにわたって広がることが示唆された(Yamashita, T. et al, Proc Natl Acad Sci USA 106:12986-12991 (2009))。この中和部位は受容体結合部位と部分的に重複し、ここで結合した抗体がHEV VLPの細胞表面への付着を阻止する空間的な障害を作り出す(Yamashita, T. et al., Proc Natl Acad Sci USA 106: 12986-12991 (2009))。両実験で使用した抗体は中和活性を有し;それ故に、モノクローナル抗体Mab224は、その結合部位がHEVを優位に中和する表面の一部であることから、おそらく中和抗体である。
ウイルス様粒子(Virus-like particles:VLPs)は、ウイルス粒子の全体構造を模倣した高度に組織化されたカプセルであることから、小分子、ペプチド抗原性エピトープ、およびその他の疾病を標的とする異種起源のDNAワクチンを同時に輸送するための強固な積荷の一つである。このアプローチは、外来エピトープの結合における合理的な設計を可能にする、VLPについての優れた構造情報に依存する。以前に結晶構造が未知の状態で、PORF2において6の挿入部位が選択された。それらはNおよびC末端、ならびに4の内部部位である。内部部位は、残基A179、R366、A507、およびR542の後に位置する。A179およびR336の部位に挿入を有する融合タンパク質は、VLP産生に完全に失敗し、A507およびR542に挿入を有するものは、ペプチド発現には影響することなく、VLP産生を大きく減少させた(Niikura, M. et al., Virology 293:273-280 (2002))。結晶構造により、これらの内部挿入は、不適切な空間的位置にあることが明らかになった。残基A179はα−へリックスの中央のS1ドメインに位置し、一方でα1へリックスの完成度がS1ドメインの完全性に必要であることから、その2回軸に関連する隣接するサブユニットと相互作用する(図16C)。R366はS2ドメイン内に位置し、その3回軸に関連する隣接するサブユニット由来の残基E386との静電気的な相互作用を促進する(図16D)。Pドメインに位置するものの、R542の側鎖は二量体の接触面内にあり、二つの単量体の疎水性相互作用を導く(図16D)。R542の後への挿入により、二つのPドメインの配向が誤配列され、PORF2タンパク質の二量体相互作用が弱くなる可能性がある。Pドメインにおける残基A507の役割は、長いプロリンリッチなヒンジに対する角度を固定することにより、Pドメインの配向を維持することである(図16C)。4残基のうちのいずれもVLPの表面に露出していないが、それらの幾つかは個別のPORF2サブユニットの表面にある(図16)。従って、これらの部位への外来配列の挿入により、個々のタンパク質の発現への干渉は誘導されないが、HEV VLPの集合への障害は起こる。結晶構造により、C末端はVLPの表面に露出し、N末端はVLPの中心へ向かうことが明らかになった。従って、これらの二つの部位における挿入によりVLPの集合は阻害されないが、C末端がVLPの外表面に露出していることから、粗大な外来抗原配列を係留するためにはより適している(図17)。
E型肝炎ウイルス粒子サイズ粒子の、RNA依存性集合経路に対する構造的基礎
E型肝炎ウイルス(Hepatitis E virus:HEV)はヒトに急性肝不全を誘導し、妊婦における致死率は高い。HEV天然カプシドの集合過程を理解するための、抗HEVについての研究が必要とされている。ここで本発明者らは、比較構造解析のために、N末端の111残基を有するかまたは有さないHEVカプシドタンパク質それぞれを昆虫細胞内に発現させることによって、大ウイルス粒子サイズ、および小T=1のカプシドを産生した。ウイルス粒子サイズのカプシドはT=3正二十面体格子を示し、RNAを含まないT=1カプシドとは対照的にRNA断片を含む。しかしながら、両方のカプシドは共通の十量体構成を共有していた。in vitro集合により、HEVカプシドタンパク質は十量体の中間体を形成する内在的な能力を持つことがさらに示された。本発明者らのデータにより、RNA結合は、HEV天然カプシドの集合に必須の外的因子であることが示唆される。
HEV−VLPの産生ならびにin vitroでの解体および再集合
以前に記載されたプロトコルに従い、HEV−VLPを産生および精製した(Li, T. C. et al., J Virol 71, 7207-7213 (1997))。簡単に述べると、遺伝子型3 HEV−ORF2タンパク質の残基14ないし660をコードする組み換えバキュロウイルスを構築した(Liらにより投稿準備中)。Tn5細胞に組み換えバキュロウイルスを5のM.O.Iにて感染させ、6日間培養した。上清を回収し、複数回の超遠心分離法、およびそれに続くCsCl密度勾配における分離によってVLPを精製した。最終ペレットを、10mM カリウム−[2−(N−モルホリノ)エタンスルホン酸](MES)緩衝液、pH6.2、に再懸濁した。解体および再集合実験には、少量のサンプル(20〜40μl)の透析が可能であることから自家製の透析装置を用いた。精製したVLPを、EDTA(10mM)およびDTT(20mM)を含む緩衝液に対する、様々なpHでの透析によって破壊した。VLPの解離後、Tris−HCl緩衝液(pH7.5)中の150mM NaClを加え、試料を二価イオンのCa2+(20mM)の存在下で1時間インキュベートした後、電子顕微鏡で観察した。
ブルックヘブン国立研究所STEM施設(Brookhaven National Laboratory STEM facility)において、TMVを内部コントロールとして走査型TEMを行った。VLPとTMVの混合物を液体窒素中で素早く凍結し、データ収集の間、汚染を排除し、かつ質量の減少を少なくするために、−150℃に維持した。検体を0.25nmのサイズで40kegの電子ビームにより走査し、大角および小角検出器の両方に対し、10のプリアンプ増幅率によってイメージを収集した(Wall, J. S. et al., Methods Cell Biol. 53, 139-164 (1998))。イメージを10Åのピクセルサイズで記録し、PCMass29プログラム(Brookhaven National Lab http://www.biology.bnl.gov/stem)によって解析した。バックグラウンドを標準化した後、プログラムにより提供されるMSV殻モデルによってVLPの質量を選択した。TMVおよびHEV−VLPの質量測定は、常に同じイメージから行った。HEV−VLPの質量はMDa(粒子あたりの質量)において測定し、TMVの質量はKDa/Å(単位長さ当たりの質量)において測定した(Wall, J. S. and Simon, M. N., Methods MoI Biol. 148, 589-601 (2001))。
イメージ再構成のためのクライオEMデータの収集は、200kVにおいて、以前に詳述された手順に従って操作されるJEOL JEM-2100F TEMによって行った(Xing, L. et al., Virology 265, 35-45 (1999))。簡単に述べると、E330Kカプシドまたは再集合したORF2複合体を含む3μlの溶液を、穴あきのカーボンフィルムコートされた銅グリッド上に置き、余分な液体を除去した後、液体エタン内へ素早く入れた。VLPをガラス状の氷の薄層内に包埋し、Gatan 636クライオトランスファーシステムを用いてEM内に移した。検体を50,000xの倍率で観察し、関心のある範囲をTVIPS CCDカメラ(TemCam-F415)で記録した。顕微鏡像を、検体空間における2.0Åのピクセルサイズ、および0.7〜3.5Åのデフォーカスレベルによって記録した(図26a)。無非点収差またはドリフトの無いデジタルイメージを、後のイメージプロセシングのために選択した。次に個々のHEV T=3 VLPイメージを四角で囲み、正二十面体粒子のために確立されたソフトウェアパッケージを用いて処理した(Baker, T. S., and Cheng, R. H. (1996) J Struct Biol 116, 120- 130; Ji, Y. et al., J Struct Biol 154, 1-19 (2006))。総数7720の個々のイメージが処理され、それらのデフォーカスレベルは主に1.0〜2.5Åに分布していた。
以前に記載された方法に従い、VLPの結晶化を行った(Wang, C. Y. et ah, Acta Crystallogr. Sect. F Struct. Biol. Cryst. Commun. 64, 318-322 (2008))。結晶を液体窒素中で直接急速冷凍して、X線回折実験を行った。HEV−VLPの結晶についての全てのx線実験は、兵庫県、日本のSPrin-8で行った。単位細胞内の粒子配向を、自己回転機能(Blow, D. M. et al., J MoI Biol 8, 65-78 (1964))により決定し、粒子位置は、クライオEM構造をモデルとする並進探索により決定した。非対称の結晶単位は一個の粒子を含むため、結果的に60回非結晶学的対称(non-crystallographic symmetry:NCS)の平均化を実行した。データIの初期位相を得るため、クライオEM構造(Xing, L. et al., Virology 265, 35-45 (1999))を用い、NCS平均化のために用いるエンベロープ(マスク)を作成した。位相は、正二十面体対称エレメントにより平均化した実空間電子密度および溶媒平滑化により改良した。分解能は次第に8.3Åに拡大した(R因子=0.21、相関係数[correlation coefficient:CC]=0.92)。この構造をデータIIの位相決定に用い、位相は改良されて3.8Åの分解能へ拡大した(全体的なR因子およびCCはそれぞれ0.18と0.97であった)。
走査透過型電子顕微鏡(Scanning transmission electron microscopy:STEM)
遺伝子型3のORF配列を昆虫細胞に発現させ、ウイルス粒子サイズのHEV−VLPを回収した。このVLPは、直径〜40nmの球状のイメージとして投影され、T=1 VLP(直径27nm)よりも大きかった(図18a)。クライオ電子顕微鏡像では、HEV−VLPのイメージはスパイク様の特色を施されており、対照的に均一であった(図18b)。
大HEV−VLPのクライオEM構造により、完全な正二十面体殻上に90の突起スパイクが明らかにされ(図19a)、これはT=3 正二十面体対称およびSTEM質量測定の結果に一致する。三次元密度マップから測定されたように、VLPの全体的な直径は410Åであり、中心腔の半径は170Åであった(図19c)。単層カプシドは180コピーのORF2タンパク質を含み、これは幾何学的環境によって三つの独特な単量体にグループ化された。単量体AおよびBは、各5回軸の周辺で二量体スパイクを形成したが(A−B二量体)、2回軸関連の二つのC単量体は、各正二十面体2回軸においてスパイクを形成した(C−C二量体)(図19b)。大HEV T=3 VLPにおけるORF2タンパク質の表面格子は、カリシウイルスのカプシド配置と同様であった。A−B二量体に比較して、HEVC−C二量体の形態は、おそらく正二十面体殻に向かって突出するドメインの角度の柔軟性のために、よく定義されていない。
切断型の遺伝子型1カプシドタンパク質(残基112〜608を含むPORF2)の結晶構造は、三つのドメインS1、S2、およびPに分けることができ、残基555〜560を網羅する領域についてはよく解明されていない。残基118〜317により形成されるS1ドメインは、多くのT=3ウイルス性カプシドタンパク質に観察される、ゼリーロールモチーフを有する古典的な8本鎖βバレルへ折り畳まれる(図20a)(Rossmann, M. G. and Johnson, J. E., Ann Rev Biochem 58, 533-573 (1989); Harrison, S. C, Curr Opin Struct Biol 11, 195-199 (2001))。ユニークなことには、三つのさらなる短いαへリックスが、S1ドメインにおけるE鎖とF鎖の間およびG鎖とH鎖の間に観察された。カプシド殻は主に、S1ドメイン間でのサブユニット間相互作用によって安定化される。残基318〜451から成る折り畳まれたS2ドメインは、B’鎖とC’鎖の間のαへリックスを有するねじれた逆平行βシートであった(図20a)。残基452〜606から構成されるPドメインは、逆平行βシートのF”A”Bb”およびBa”E”D”C”から構成されるβバレルへ折り畳まれ(図20a)、長いプロリンリッチなヒンジ(残基452〜461のPTPSPAPSRP)を通じてS2ドメインに結合した(図20a)。S2およびPドメインの両方はS1ドメインの上に存在するが、HEVクライオEMマップの突起スパイクはPドメイン密度のみを含むことが、カリシウイルスとの明確な違いである(図25)。PORF2二量体は、主にPドメイン間であるという理由から、単量体間に最も大きな埋没表面領域(buried surface area:BSA)(5,900Å2)を有する(図20b)。BSAは3回軸および5回軸周辺それぞれで、二つの隣接するPORF2サブユニットについて3,400Å2および1600Å2である(図20b)。さらに、3回軸周辺の第3の分子のBSA(9,500Å2)は、5回軸周辺のもの(4,700Å2)よりもかなり広い。
T=1とT=3集合の間でのORF2タンパク質移行の機構を理解するため、T=1の十量体と六量体とをT=3のクライオEM密度マップへドッキングした。T=1 VLPの十量体は、5回軸周辺の5の二量体に対応する、10の隣接するPORF2単量体から成ったが、一方でT=1六量体は、3回軸周辺の3の隣接する二量体に対応した。六量体とは異なり、PORF2十量体の座標は、5回軸頂点におけるT=3密度マップの屈曲(図21a)およびドメイン分離(図21b)に非常によく適合した。3回軸におけるT=3カプシドの屈曲は、二量体の一つがクライオEM密度マップの外部に張り付いているように見えることから、PORF2六量体の座標とは一致しなかった(データ未提示)。それに加えて、S2/S1ドメインに対する、C−C二量体のPドメインの配向は、A−B二量体のそれとは90゜異なっていた(図22)。このことにより、T=3正二十面体におけるA−B二量体の間の分子相互作用は、T=1正二十面体の集合における二量体−二量体相互作用に一致するが、A−B二量体とC−C二量体の間の相互作用は、T=3集合に対して独特であることが示唆される。
T=3 VLP集合におけるORF2十量体の役割を理解するため、HEV−VLPの自己集合過程をin vitroで解析した。キレート剤(EDTA)および還元剤(DTT)の組み合わせが、高アルカリ環境(pH10)において、ORF2タンパク質を変性させることなくT=3 VLPを解体することが見出された(データ未提示)。解体溶液への20mM CaCl2の添加によってORF2二量体の星型の複合体への会合が誘導され、リフォールディングされたVLPは見出されなかった(図21c)。星型の複合体について調べたところ、二つの対立する頂点間の距離が、TMVの直径に近い〜18nmであることが見出された(図21c)。このサイズは、PORF2十量体について測定したものと一致した。従って、星型の複合体は、外見のみではなくサイズもORF2十量体に似ていた(二量体の五量体)。3回軸周辺の全体的なBSAは5回軸周辺のものよりも大きかったが、PORF2六量体に適合し得るいずれの複合体も見出せなかった。
E型肝炎ウイルスは、急性肝不全の原因となるヒトの病原体である。その他の肝炎ウイルスと同じように、HEVは現在利用できる細胞技術によって増殖させることはできない。遺伝子型3 HEVのカプシドタンパク質は、T=1 VLPへ自己集合するアミノ酸112〜608を含むPORF2タンパク質として、およびT=3 VLPを形成するアミノ酸1〜608を含むORF2タンパク質として、昆虫細胞内に発現させることができる。
急性E型肝炎ウイルスの改変されたヌクレオペプチドカプシドは、経口ワクチンとしての機能的なモジュール性を有する
E型肝炎ウイルス(Hepatitis E virus:HEV)は水媒介性のウイルス性媒介物であり、主に糞口経路を通して伝染することから、低pHならびに胃および消化管に付随する消化酵素に抵抗性である。HEVの感染はヒトに急性肝炎を引き起こし、妊婦における死亡率は30%に達する(Naik, S.R. et al, Bull World Health Organ, 70 (5), 597-604 (1992))。現在、国際的なヌクレオチド配列データベースの協働(Khuroo, M.S. & Khuroo, M.S., Curr Opin Infect Dis, 21, 539-543 (2008))において入手可能なHEVのゲノム配列は1,600であり、これらは4の遺伝子型に分類される(Jameel, S., Expert Rev Mol Med, 1999, 1-16 (1999))。特に、知られた単一の血清型のみが認識されていることから、HEVの免疫優性ドメインは高度に保存されていることが示唆される。第2の翻訳領域においてコードされる主要なカプシドタンパク質のORF2は、最も免疫原性であり、かつ防御的な液性免疫応答の誘導に関与することが報告された(Li, T. et al., Vaccine, 22, 370-377 (2004); Purdy, M.A. et al., J Med Virol, 41 (1), 90-94 (1993); Riddell, M.A., Li, F., & Anderson, D.A., J Virol, 74 (17), 8011-8017 (2000))。アミノ酸112〜608を網羅する組み換えHEVカプシドタンパク質(Recombinant HEV capsid protein:PORF2)は、昆虫細胞に発現させるとウイルス様粒子へ自己集合することができる(Li, T. C. et al., J Virol, 71 (10), 7207-7213 (1997))。免疫電子顕微鏡によって示されるように、HEVウイルス粒子はエンベロープを持たない直径320〜340Åの正二十面体粒子であるが(Balayan, M., Andiaparidze, A., Savinskaya, S., & et. al., Intervirology, 20, 23-31 (1983))、自己集合したウイルス様粒子の直径は270Åであることが、その三次元再構成から決定された。HEV−VLPはHEVの天然の免疫原性を有し、非ヒト霊長類への経口投与によって全身性および粘膜性の抗HEV抗体を誘導することができる(Li, T. et al., Vaccine, 22, 370-377 (2004))。HEV−VLPは、60コピーのPORF2タンパク質から構成されるT=1正二十面体粒子であると判定されたが、HEV VLPを、E型肝炎に対する直接の粘膜ワクチンまたは粘膜表面において免疫応答を活性化するための送達担体のいずれかとしてより良く利用するために、アミノ酸配列についての詳細な構造情報がなお必要とされる。
DNAワクチンを被包するHEV−VLP
ワクチンプラットフォームに関する本発明は、単にカプシド内に含まれたプラスミドDNAを送達するのみではなく、有効性を最大限にするためのアジュバントおよびブースターの両方としてのペプチドエピトープを、ウイルス様粒子(virus-like particles:VLP)の表面に付着させ得る。このことにより、ヘルパーT細胞、および重要なことにはウイルス感染のためのMHCクラスI限定の細胞傷害性Tリンパ球の両方による、細胞性免疫の誘導が実行可能となる。この試みは、VLPがペプチドまたはDNAいずれかの一つの型の抗原を輸送するために用いられる際、付着された抗原が、DNAワクチンの有効性を増強するためのアジュバントおよびブースターの両方として機能するため、一般的なVLP送達系よりも強力であるとみなされる。全体的な産生には、潜在的に致命的なインフルエンザウイルスの扱いが必要とされず、産生時間が著しく短縮されることにより、進行中の大流行では最高に注目される。DNAワクチンのカプシド内への包含は、全てin vitroで行われることから、M2eを結合したHEV VLPは、逆遺伝学により産生された潜在的な大流行ウイルスのRNA部分を含むDNAプラスミドを封入するためのエンベロープとして、前もって産生することができる。
様々なキメラHEV−VLPコンストラクト
様々な異種性ペプチドを、HEV ORF2タンパク質の残基483〜490、残基530〜535、残基554〜561、残基573〜577、残基582〜593、または残基601〜613の予め選択された領域内のHEV ORF2の一部分に挿入した。これらのコンストラクトに使用できる異種性エピトープには、HIV−V3、flu−M2、HSVおよびレオ(下痢ウイルス)が含まれるが、これらに限定されない。(1)残基483〜490の領域内に挿入されたHIV V3ペプチドによるHEV−VLP;(2)残基530〜535の領域内に挿入されたHIV V3によるHEV−VLP;(3)残基554〜561の領域内に挿入されたHIV V3ペプチドによるHEV−VLP;(4)残基582〜593の領域内に挿入されたHIV V3ペプチドによるHEV−VLP;および(5)残基601〜613の領域内に挿入されたHIV V3ペプチド、またはflu−M2ペプチド、または単純ヘルペスウイルスペプチドによるHEV−VLPを含む、様々なコンストラクトが作製された。
