Description
本発明の主目的は、CD6のドメイン1(D1)と結合することができ、かつ、ALCAM結合を妨害することなくT細胞増殖を阻害するモノクローナル抗体を得ることである。
したがって、本発明は、CD6のドメイン1(D1)と結合することができ、ALCAM結合を妨害することなくT細胞増殖を阻害するモノクローナル抗体;モノクローナル抗体を使用して、炎症状態、例えば乾癬、関節リウマチ、又は患者における自己免疫応答、例えば多発性硬化症又は移植拒絶反応と関連する有害応答(adverse responses)、移植片対宿主病、1型糖尿病、乾癬、皮膚T細胞リンパ腫、甲状腺炎(thyroiditis)、及び他のT細胞媒介性自己免疫疾患を変調させる方法;免疫抑制剤と組み合わせてモノクローナル抗体を使用して、炎症状態、例えば乾癬、関節リウマチ、又は患者における自己免疫応答、例えば多発性硬化症又は移植拒絶反応と関連する有害応答、移植片対宿主病、1型糖尿病、乾癬、皮膚T細胞リンパ腫、甲状腺炎、及び他のT細胞媒介性自己免疫疾患を変調させる方法;並びに抗炎症性免疫応答を誘発することができる抗原、例えばインスリン、GAD、MOG、MBP及びHSP60と組み合わせてモノクローナル抗体を使用して、炎症状態、例えば、多発性硬化症又は移植拒絶反応、移植片対宿主病、1型糖尿病を変調させる方法に関する。

(2007) 4351-4361.)をT1hと競合させると、用量依存的な競合が観察され、両方が同じドメイン、すなわちドメイン1と結合することが示唆されている。
ゲノムDNA及びプラスミドDNAを鋳型として使用する多重PCR反応を使用して、T1hのヌクレオチド配列を決定した。ゲノムDNAを、T1hを発現するNSO細胞から調製した。アミノ酸配列を、ESI−TOF及びMALDI−TOFを使用して決定した。トリプシン、Lys−C及びGlu−Cを含む種々の酵素を使用したペプチドマッピングを使用して、最終的なアミノ酸配列を設定した(evolve)。図1に開示されている配列は、特許文献3及びそれに相当する特許文献4、並びに特許文献5及びそれに相当する特許文献6で既に公表されたT1hの配列とわずかに異なっている。下記の多数の実施例において述べるように、本特許で開示した配列がその標的CD6に対して完全な機能的活性を有することは、立証されている。
ELISA実験により、種々の濃度のALCAMの存在は、T1hがCD6−Fcと結合するのを妨げないことが明らかに示された。ALCAMとT1hとの間の競合が存在しないことは、これらの2つに関する結合ドメインが異なることを示唆している。
アポトーシスの特質の1つは、細胞膜の内部から外側へのホスファチジルセリン(phosphatidylserine)(PS)の転位である(J Biol Chem 1990. 265: 4923-4928)。アポトーシス細胞の膜の外葉上のホスファチジルセリンの分析を、アポトーシス細胞及びネクローシス細胞の識別のためにアネキシン−V−フルオレセイン及びヨウ化プロピジウム(PI)を使用することにより行った。アネキシンVは、ホスファチジルセリンに対して高い親和性を有するCa2+依存性リン脂質結合タンパク質である(J Immunol Methods 1995. 184: 39-51)。PIが明らかな(distinct)ネクローシス細胞と結合するのに対して、アネキシン−V−フルオレセインはアポトーシス細胞と結合する。この方法は、アポトーシス細胞及びネクローシス細胞の集団を区別する上で役立つ。初期アポトーシス集団はアネキシンV陽性であるのみであるが、後期アポトーシス集団はアネキシンV及びPIの両方について陽性である。
胸腺細胞、成熟T細胞、B−1サブタイプと呼ばれるB細胞のサブセット、及び幾つかの脳細胞は、CD6を発現する(J Exp Med 1991. 174: 949-952; J Immunol 1997. 158:
1149-1156)。実験のねらいは、T細胞及びB細胞のそれぞれにおいて受容体密度、及びCD6の発現の差異を定量化することである。
抗体が標的細胞又は感染細胞と結合する際には、抗体のFc部分が細胞(特にナチュラルキラー(NK)細胞)の表面上に存在するFc受容体と結合する。これは引き続きこれらの細胞の活性化を引き起こす。これらの活性化細胞は、サイトカイン及び細胞毒性顆粒を放出し、アポトーシスを誘発することにより細胞死を促進する。これは、抗体依存性細胞媒介性細胞毒性(ADCC)として知られている。
アラマーブルー(レサズリン)ベースのアッセイを使用して、細胞死滅を促進する抗体の能力を測定する。これは、抗体と細胞表面抗原との結合により、標的細胞溶解をもたらす補体を固定及び活性化することによって誘導される。レサズリンは、還元されると色が青色からピンク色へ変化する酸化還元活性色素である。最近のデータにより、レサズリンは細胞の細胞質に進入し、細胞質において蛍光生成物へと還元され、その後再び培地へ排出されることが示唆されている(Eur J Biochem 2000. 267: 5421-5426)。
リンパ球培養物における細胞分裂の刺激のためにフィトヘマグルチニン(PHA−M)を使用する。アカインゲンマメ(ファセオラス・ブルガリス(Phaseolus vulgaris))からのレクチン抽出物であるPHAは、強力な細胞凝集活性及び分裂促進活性を含有する(Proc Natl Acad Sci U S A 1982. 79: 1611-1615)。PHAは、各々が非共有的な力により共に保持される四量体である5種類のイソレクチン(L4E0、L3E1、L2E2、L1E3、L0E4)のファミリーを含有する。サブユニットは、白血球反応性(L)及び赤血球反応性(E)と称される、2つの異なる種類のものである。Lはリンパ球表面受容体に対して高い親和性を有するが赤血球表面受容体に対しては親和性をほとんど有さず、イソレクチンの分裂促進特性に関与する。Eは赤血球凝集特性に関与する。PHA−Pはタンパク質形態であり、PHA−Mはこれらのイソレクチンのムコタンパク質形態(form)である(J Biol Chem 1977. 252: 9018-9023)。
以前の実験をハイスループットプレートベースのアッセイに変更した。該アッセイではT1h又は非特異的IgG1アイソタイプ抗体hR3を含む又は含まない96ウェルディッシュ中で新たに回収したPBMCをインキュベートする。その後PHAを添加してリンパ球の増殖を3日間刺激する。アラマーブルーを使用することにより増殖を測定する。有意性の試験はT検定により行う。
T細胞受容体(TCR)及び同時刺激受容体による刺激が、急速な成長及び増殖の速度、並びにエフェクター機能の獲得を特徴とする静止(休止)状態から活性化状態へのT細胞分化のスイッチをトリガすることが知られている(Immunity 2007. 27: 173-178)。ヒトCD6受容体は、未熟胸腺細胞、成熟T細胞、及びB1aサブセットと呼ばれるBリンパ球のサブセット、慢性B細胞リンパ球性白血病、並びに脳の様々な領域で発現される105kDa〜130kDaの1型糖タンパク質である(J Immunol 2006. 177: 1152-1159)。CD6は、このファミリーに特徴的な3つの100個〜110個のアミノ酸長のシステインリッチドメインの細胞外領域の(of in)存在に基づくタンパク質受容体のスカベンジャー受容体システインリッチ(SRCR)スーパーファミリーのグループBのメンバーである(J Exp Med 1991. 174: 949-952)。入手可能な証拠により、CD6がリンパ球活性化及び分化プロセスの変調に関与するアクセサリー分子であることが示されている(J Immunol 2006. 177: 1152-1159)。胸腺細胞及び休止成熟T細胞上で、CD6は、TCR/CD3複合体(J Immunol 2004. 173: 2262-2270)、及びSRCRスーパーファミリーの近縁なメンバーであるCD5(J Biol Chem 2003. 278: 8564-8571)と、部分的に会合する。さらに、CD6は成熟免疫シナプス(IS)の中心部で蓄積し、そこでCD6はTCR/CD3及びCD5と共存する。CD6は、IS成熟に影響を与える初期T細胞−APC接触、またT細胞の増殖応答にも関与している(J Immunol 2006. 177: 1152-1159, Eur J Immunol 2004. 34: 930-940)。
IL2は、T細胞活性化に必要且つ十分なサイトカインである。IL2は、IL2により上方調節されるIL2受容体を介してシグナル伝達を行う。IL2は、T細胞発生、T細胞の免疫学的記憶、及び調節性T細胞の発生において重要な役割を有する(J Immunol 1995. 155: 1151-1164)。この実験から、IL2の最適濃度1.25ng/mlを、さらなる実験で使用した。
フローサイトメトリーは、大きさ及び色に基づき種々の粒子の識別を可能にする分析ツールである。BD(商標)CBAは、別個の蛍光強度を有する一連の粒子を利用して、複数の可溶性の分析対象を同時に検出する。BDA(商標)CBAは、フローサイトメトリーと組み合わせて、強力な多重アッセイを創出する。BDA CBAヒトTh1/Th2サイトカインIIを使用して、単一試料中のインターロイキン−2、インターロイキン−4、インターロイキン−6、インターロイキン−10、TNF及びインターフェロンγのレベルを定量的に測定する(J Immunol Methods 2001. 254: 109-118)。
抗原提示細胞上の外来性抗原を認識した後、T細胞は増殖し、免疫応答を開始する。これらのT細胞のうち幾つかは系で循環中の記憶T細胞に変化し、同じ抗原で再負荷されると、これらの細胞は増殖し免疫応答を開始する。たいていのヒトは破傷風トキソイドに対して免疫化されており、そのヒト由来のT細胞を同じ抗原で負荷すると、集団中の記憶T細胞が増殖する。実験結果により、破傷風トキソイドがT細胞の増殖を用量依存的に刺激するが、sT1hはこれらの細胞の増殖の阻害を示さないことが示されている。このことは、T1hが記憶T細胞増殖を阻害しないことを強力に示唆している。循環記憶T細胞増殖が影響されずT1h療法の患者が感染し易くならないため、このことはT1h療法にとって好ましい。
遺伝的に異なる個体由来のリンパ系細胞の混合物をインビトロで培養すると、該細胞は混合リンパ球反応(MLR)と称されている特徴的な応答を受ける(Proc Natl Acad Sci
U S A 1973.70: 2707-2710)。この反応は、初期増殖期、及び元の反応を刺激するのに使用した抗原を生じる標的細胞に対して特異的な細胞毒性を示すエフェクターリンパ球のその後の出現により典型的に表される。
成熟樹状細胞(抗原提示細胞)がナイーブPBMCの増殖を引き起こす同種(或る個体から採取された抗原提示細胞、及び別の個体から採取されたPBMCによる)混合リンパ球反応では、T1hは、この増殖を用量依存的に阻害する。データにより、作用様式は少なくとも2種類の主要な炎症促進性サイトカイン、すなわちIL6及びIFNγの下方調節を伴っていることが示された。
細胞株及び細胞培養:
HUT78(ATCCから入手したT細胞リンパ腫ヒト細胞株)を、標的細胞として使用する。細胞を補足イスコブDMEMで培養し、該培地は2mg/mlのNaHCO3、20mM Hepes、2mM L−グルタミン、及び20% FBS(Invitrogen)を含有する。

rhCD6FC/キメラ(R and D systems)(100μg/ml)をコーティング緩衝液で希釈し、100μlを96ウェルNunc−Maxisorpプレートの各ウェルに添加した。その後プレートを4℃で終夜インキュベートした。プレートをPBS/Tween20で3回洗浄した。その後200μlのブロッキング溶液(1×PBS中における2% BSA+0.1% Tween20)を添加し、37℃で1時間インキュベートした。インキュベーション後、プレートを再びPBS/Tweenで3回洗浄して、その後T1hモノクローナル抗体(0.2mg/ml)及びrhALCAMFc(R and D systems)を種々の濃度で添加した。その後これを37℃で1時間インキュベートした。その後プレートをPBS/Tweenで3回洗浄した。ウェルに、ブロッキング緩衝液で希釈した(1:20000)200μlの抗ヒトIgG(Fab)2ALPを添加し、37℃で1時間インキュベートした。プレートをPBS/tweenで3回洗浄した。200μlのp−ニトロフェニルホスフェート(PNPP)基質を各ウェルに添加し、発色するまで37℃でおよそ15分間インキュベートした。読取り値を、BIOTEK社のマイクロプレートリーダーを使用して405nmで取得した。実験により、種々の濃度のALCAMの存在は、T1hがCD6受容体と結合するのを妨げないことが示されている。ALCAMとT1hとの間の競合が存在しないことは、これらの2つに関する結合ドメインが異なることを示唆している(図2)。
T1hは、CD6のドメイン1(D1)と結合する。
ドメイン1と結合する抗体であるMEM98(Castro AA M et al, J of Immunol, 178
(2007) 4351-4361.)をT1hと競合させると、用量依存的な競合が観察され、両方が同じドメイン、すなわちドメイン1と結合することが示唆されている(図3)。
T1hは、HUT78細胞においてアポトーシスを誘導しない。
細胞を採取し、各35mmディッシュに1.5mlの3.3×105細胞/mlの菌液(最終細胞数:5×105個)を播種した。所要量の抗体をそれぞれのディッシュに添加して、最終濃度を5μg/mlとした。対照ディッシュには、抗体を添加しなかった。陽性対照として細胞を濃度1.2μg/mlのラパマイシンと共にインキュベートした。細胞を、5%CO2インキュベーター中、37℃で終夜インキュベートした。その後細胞をFACSチューブ(BD Falcon、カタログ番号:352054)に移し、1200RPMで5分間室温(RT)で遠心分離した。上清を廃棄し、2mlのPBS中に再懸濁し、1200RPM、RTで5分(minutes)間遠心分離した。上清を廃棄し、100μlのアネキシン−V−フルオレセイン標識溶液を添加し、RTで10分間〜15分間インキュベートした。細胞を2mlのPBSで洗浄し、1200RPMで5分間遠心分離した。その後上清を廃棄した。細胞を0.5mlのPBS中に再懸濁し、励起波長488nmでフローサイトメーター(3000個の細胞をゲーティングした)によりデータ取得した(acquired)。試料は、アネキシンVに関してFITCチャネルで、PIに関してPEテキサスレッドチャネルで、読み取った。ラパマイシン処理アームにおいてアネキシンV単独及びPI単独の試料を、補償が可能となるように分析した(run)。
正常個体由来のTリンパ球及びBリンパ球上におけるCD6受容体の発現の差異。
PBMCを、フィコールパック(Amersham、カタログ番号:17−14403−03)を用いた密度遠心分離により単離した。バフィーコートを健常ドナーから取得し、常に新たな状態で採取した。その後PBMCをPBSで洗浄した。その後PBMCを5mlの1×PBS中に1.5×106細胞/mlの細胞密度で再懸濁し、100ulを各falconチューブに添加した。10μLの結合抗体(1:10希釈)をそれぞれのチューブに添加し、ボルテックス処理し、その後4℃で30分間インキュベートした。インキュベーション後、2mlの1×PBSを各チューブに添加し、その後1200RPM、RTで5分間遠心分離した。上清を廃棄し、細胞を0.5mLの1×PBS中に再懸濁して、フローサイトメーターで細胞についてデータ取得した。FSC及びSSCで所要の集団(細胞数3000)に関して細胞(リンパ球)をゲーティングした。SPHERO(商標)(カタログ番号:RCP−30−5A)Rainbow calibration Particlesについても、同じ電位設定で上記チューブと共にデータ取得する。各ピークに関する蛍光平均チャネル数(すなわち相対輝度)を記録した。平均チャネル数を、以下の式を使用して相対チャネル数に変換した。
i. 相対チャネル数♯=(R/4)log(平均チャネル数♯×10)
ii. 式中、R=解像度(すなわち256)
a. FITCチャネル : y=134.95e0.0366x
b. PEチャネル : y=23.6e0.03736x
T1hは、穏やかなADCC(抗体依存性細胞媒介性細胞毒性)を引き起こす。
標的細胞株(HUT78/ダウジ)を採取し、PBSで2回洗浄した。細胞(2×105)を、1mlの濃度0.25μMのCFSE溶液(PE Cy5チャネル中に漏れないCFSEの濃度を選択するために、各細胞型に関してCFSEの用量設定が必要である)に再懸濁した。37℃で10分間正確に、細胞をインキュベートした。2mlのRPMI、5% FBSを添加して反応を停止した。2ml RPMI 5% FBS各々で細胞を2回洗浄して、CFSEを除去した。その後標的細胞調製物を1×105細胞/mlの細胞密度でRPMI培地中に再懸濁し、各falconチューブに100ul(細胞数104)を添加した。50ulの抗体(5ug/ml、10ug/ml)をそれぞれのチューブに添加し、RTで30分間インキュベートした。その後PBMC(エフェクター細胞)を、1:1、1:25、1:50及び1:100のT/E比でそれぞれのチューブに添加した。最終容積は、各チューブ中で培地により250ulとした。各チューブを1200rpmで2分間遠心分離して細胞間相互作用を促進し、ADCCが起こるように37℃でインキュベートした。ダウジ細胞をリツキサンと共に6時間インキュベートし、陽性対照として試料と共に試験した(run)。その後細胞を1×PBSで洗浄した。100ulの7AAD(6.25ug/ml)を添加し、4℃で15分間インキュベートした。細胞を1×PBSで洗浄し、1200RPM、4℃で5分間スピンダウンした。上清を廃棄し、細胞を500ulの1×PBS中で再懸濁した。およそ200事象/秒で細胞についてデータ取得し、フローサイトメーターのFITCチャネル及びPE Cy5チャネルで観察した。CFSE陽性細胞は、対数目盛の102及び103(the second and third decade)の間で蛍光を発し、PE Cy5チャネルへの漏出はなかった。計3000個のFITC陽性細胞をゲーティングし、この集団において、7AAD(PE Cy5陽性)を示す細胞の百分率を評価した(図7及び図8)。非特異的抗体対照の他に、1:1、1:25、1:50及び1:100の比のエフェクター/標的細胞(単独)もアッセイに含まれるものとした。アポトーシスの%は、FITC/PEcy5ドットプロット散布図(scatter)における右上の区分の細胞の%とした(図9)。
HUT78に対するT1hによるADCC。
実施例4に基づき、同じアッセイ手順でADCCアッセイを行って、HUT78に対するT1hの効果を検証した。細胞を4時間〜終夜インキュベートした。終夜のインキュベーションにより、最適且つ一貫性のある結果が得られた。
T1hは、補体依存性細胞毒性(CDC)を媒介しない。
プールした全血由来ヒト血清(最小3(minimum three))を無菌チューブに回収し、血液を室温で少なくとも4時間凝固させた。該血液を900gで20分間遠心分離する。血清を採取し、分注し、−80℃で保存した。標的細胞(Wil−2S/HUT−78)を希釈緩衝液で洗浄し、2×105細胞/mLに再懸濁した。抗体を希釈緩衝液で希釈し、所望の最終濃度の4×とした。補体を希釈し、所望の最終濃度の4×とした(すなわち、最終濃度が1:10である場合、1:2.5希釈)。各50μLの希釈した抗体、希釈した補体、及び50μLの細胞懸濁液(10000細胞/ウェル)を、96ウェル平底プレートの各ウェルに添加した。以下の対照ウェルを含むものとした:標的細胞+Ab単独(自発的細胞死)、標的細胞+血清のみ(バックグラウンドの溶解)、及び標的細胞+10% SDS(最大の細胞死に関する)。陽性対照は、種々の濃度のリツキサンで処理したWil−2S細胞であった。96ウェルプレートを37℃で2時間インキュベートした。50uL/ウェルのアラマーブルーを各ウェルに添加し、プレートを37℃で終夜インキュベートした。励起波長530nm、発光波長590nm及び感度=35で、Biotek社の分光光度計Synergy(商標)HTにより蛍光を測定する。結果は、リツキサンと比較してT1hがCDCを誘導しないことを示唆している(図13)。
T1hは、PHAに媒介されるリンパ球増殖を阻害する。
細胞株及び細胞培養:
同意を得た後で、標準的な静脈切開手順に従って全血を正常個体から抽出した。フィコールパック(Amersham、カタログ番号:17−14403−03)を用いた密度遠心分離により、PBMCを単離した。バフィーコートを健常ドナーから取得し、常に新たな状態で採取した。RPMI1640(Invitrogen)中で細胞を培養した。該培地は2mg/mlのNaHCO3、20mM Hepes、2mM L−グルタミン、及び10% FBS(Invitrogen)を含有する。
PBMCを採取し、PBSで洗浄した。細胞(7.5×106)を、1mlの、濃度2μMのCFSEを含むPBSに再懸濁した。37℃で10分間正確に細胞をインキュベートした。10mlのRPMI、10% FBSを添加して反応を停止した。10mlのPBSで細胞を2回洗浄した。その後細胞調製物を1.5×106細胞/mlの細胞密度で5mlのPBSに再懸濁し、各BD FACSチューブに200ulを添加した。200ulの所要の抗体及び非特異的抗体を様々な濃度(それぞれ、50ug/ml、25ug/ml、12.5ug/ml、及び6.25ug/ml)で添加し、37℃で30分間インキュベートした。2mlのPBSを各チューブに添加し、1200RPM、RTで5分間遠心分離して、未結合の抗体を洗い流した。1mlのRPMI、10% FBSを各チューブ中のペレットに添加した。1mlのPHA(20μg/ml)を含むRPMI 10% FBSをそれぞれのチューブに添加して、増殖を刺激した。チューブ中の総容積は2mlである。PHAの最終濃度は10μg/mlであった。チューブをボルテックス処理し、CO2インキュベーター中、37℃で3日間インキュベートした。細胞をPBSで洗浄し、1200RPM、4℃で5分間スピンダウンした。上清を破棄し、500ulの1×PBSに再懸濁した。計20000事象についておよそ200事象/秒でデータ取得し、FITCチャネルで観察した。

細胞単独とは、CFSE+細胞である。
T1hは、96ウェルプレートベースのアラマーブルーアッセイにおいても、PHAに媒介されるリンパ球増殖を阻害する。
PBMCを新たに回収し、4×105細胞/mlの濃度でRPMI 5% FBSに再懸濁した。50μlの細胞を各ウェルに添加した。100μlの2×濃度(20μg/ml)の抗体(T1h又はhR3)をそれぞれのウェルに添加した。100μlの培地を単独で非抗体ウェルに添加した。その後プレートを、37℃で30分間インキュベートした。50μlの4×濃度(20μg/ml)のPHAをそれぞれのウェルに添加して、リンパ球の増殖を刺激した。残りのウェルには、50μlの培地を添加して、全てのウェルの最終容積を200μl/ウェルとした。プレートを、CO2インキュベーター中、37℃で3日間インキュベートした。65μlのアラマーブルーを各ウェルに添加し、CO2インキュベーター中、37℃で終夜インキュベートした。励起波長530nm、発光波長590nm及び感度=35で、Biotek社の分光光度計Synergy(商標)HTにより蛍光を測定した。
T1hは、連結抗CD3、抗CD3+連結T1h、及び抗CD3+連結ALCAM−Fcにより誘導されるリンパ球の増殖を低減する。
50μlのコーティング緩衝液を96ウェルプレートに添加し、図18に示すように50ulの2×濃度(1ug/ml)の抗CD3抗体を行Aに添加した。第1の行Aから50μlの抗体希釈物を採取し、12チャネル電動ピペットを使用してプレートの下方へ行Eまで段階希釈した。50μlのコーティング緩衝液を、行A〜行Eの列1〜列4に添加し、50μlの2×(2μg/ml)濃度の抗体(T1h(列5〜列8)又はALCAM−Fc(列9〜列12))をそれぞれのウェルに添加して、ウェルの最終濃度を、抗体は一定濃度(1μg/ml)、及び抗CD3抗体は種々の濃度(行A〜行Eにおいて、1μg/ml、0.5μg/ml、0.25μg/ml、0.625μg/ml)とした(図18)。100μlのT1h又はhR3又はALCAM(1μg/ml)をそれぞれのウェルに対照として添加した(テンプレートに示したように)。培地対照、sT1h及びshR3に関して、3つのプレートで上記セットを行った。抗体がプレートと結合するように、プレートを4℃で終夜インキュベートした。
プレート温度を室温とし、コーティング緩衝液中の抗体をvaccusafe(IBS Integra BioSciences)を使用して吸引し、200μlのブロッキング溶液を各ウェルに添加することにより非特異的結合部位をブロッキングした。プレートを37℃で1時間インキュベートし、PBS/Tweenで2回洗浄した。第3の洗浄は、RPMI培地で行った。残留量の培地を慎重に除去した。
100μlのPBMC(細胞数30,000)を各ウェルに添加した。100μlの2×濃度の所要の抗体(T1h又はhR3)をそれぞれのプレートに添加した。100μlの培地を培地プレートに添加した。96ウェルプレートを、5%CO2インキュベーター中、37℃で72時間インキュベートした。65μlのアラマーブルー溶液を各ウェルに添加し、5%CO2インキュベーター中、37℃で終夜インキュベートした。96ウェル蛍光光度計(Gen5ソフトウェアを用いるSynergy HT)を使用して、励起波長530nm及び発光波長590nm(感度=35)で、蛍光を読み取った(図19)。
ナイーブPBMCにおけるIL2の用量設定。
100μlの培地を、全てのウェルに添加した。100μlの2×濃度のIL2(20ng/ml)を、第1の行のA2〜A10に添加した。第1の行から100μlを採取し、プレートの下方へ行Hまで段階希釈を行った。50μlのPBMC懸濁液(6×105細胞/ml)を全てのウェルに添加した。50ulの培地又は抗体(T1h又はhR3、10μg/ml)又はPHA(10μg/ml)をそれぞれのウェルに添加して、最終容積を200μlとした。プレートを、5%CO2インキュベーター中、37℃で5日間インキュベートした。65μlのアラマーブルー溶液を各ウェルに添加し、5%CO2インキュベーター中、37℃で終夜インキュベートした。96ウェル蛍光光度計(Gen5ソフトウェアを用いるSynergy HT)を使用して、励起波長530nm及び発光波長590nm(感度=35)で、蛍光を読み取った。この実験に基づき、その後の実験において1.25ng/mlのIL2濃度を選択することを決定した(図21)。
T1hは炎症促進性サイトカインの低減を引き起こし、この阻害は外来のIL2の存在下で部分的に除去される。
サイトカイン分析。
ヒトTh1/Th2サイトカイン標準の調製。
バイアル1本の凍結乾燥ヒトTh1/Th2サイトカイン標準を、0.2mlのアッセイ希釈液を用いて再構成して10×バルク標準を調製し、希釈する前に少なくとも15分間平衡化した。12mm×75mmのチューブ(BD Falcon、カタログ番号352008)にラベルを付し、以下の順序で配置した:1:2、1:4、1:8、1:16、1:32、1:64、1:128及び1:256。
捕捉用ビーズを個別にボトル充填した。PE検出試薬、標準及び試料と混合する直前にビーズ試薬(A1〜A6)をプールすることが必要である。実験に必要なアッセイチューブ(標準及び対照を含む)の数を決定した。各捕捉用ビーズ懸濁液を、混合前に数秒間ボルテックス処理した。分析対象の各アッセイチューブに関して、「混合捕捉用ビーズ」とラベルを付した単一チューブに各捕捉用ビーズの10μlの一定分量を添加した(例えば、10μlのIL−2捕捉用ビーズ×18本のアッセイチューブ=180μlのIL−2捕捉用ビーズが必要とされる)。
適当な容積のアッセイ希釈液を使用して、所望の希釈倍率(すなわち、1:2、1:10又は1:100)により試験試料を希釈した。混合捕捉用ビーズ及びPE検出試薬を含有する適当なアッセイチューブに試料を移す前に、試料希釈物を完全に混合した。
50μlの混合捕捉用ビーズ(混合ヒトTh1/Th2サイトカイン捕捉用ビーズの調製において記載された手順を使用して調製された)を、適当なアッセイチューブに添加した。アッセイチューブに添加する前に、混合捕捉用ビーズをボルテックス処理した。50μlのヒトTh1/Th2−II PE検出試薬をアッセイチューブに添加した。50μlのヒトTh1/Th2サイトカイン標準希釈物を対照アッセイチューブに添加した。50μlの各試験試料を、試験アッセイチューブに添加した。アッセイチューブを、RTで3時間インキュベートし、光への直接的曝露から保護した。1mlの洗浄緩衝液を各アッセイチューブに添加し、200×gで5分間遠心分離した。上清を慎重に吸引し、各アッセイチューブから廃棄した。300μlの洗浄緩衝液を各アッセイチューブに添加し、ビーズペレットを再懸濁した。試料をフローサイトメーターで解析した(図22及び図23)。
連結アッセイの手順、すなわち図18で言及されたプレートのコーティング及びPBMCの培養において記載されているようにアッセイを設定した。細胞を96ウェルプレートから採取しBD falconチューブに移した。2mlの1×PBSを各チューブに添加し、1200RPMで5分間遠心分離した。上清を廃棄し、100ulの1×PBSを細胞ペレットに添加した。100ulの細胞を各チューブ中で採取し、10μlの蛍光色素(Fluorochrome)、結合型抗CD4 Alexa又は抗CD25 PEを添加し、4℃で30分間インキュベートした。インキュベーション後、2mlの1×PBSを各チューブに添加し、その後1200RPM、RTで5分間遠心分離した。上清を廃棄した。細胞を0.5mLの1×PBSに再懸濁し、細胞についてフローサイトメーターでデータ取得する。FSC及びSSCで所要の集団(細胞数3000)に関して細胞(リンパ球)をゲーティングした。細胞の百分率と平均蛍光強度との両方を評価した。この実験は3回行った。1回の代表的な図を示す。結果は、各独立の実験で再現性を有していた(図24)。
実施例9の連結アッセイの手順において説明されたアッセイ設定では、96ウェルプレートからBD falconチューブに細胞を採取し、アポトーシスアッセイのために標識した。2mlの1×PBSを各チューブに添加し、1200RPMで5分間遠心分離した。上清を廃棄し、100ulの1×PBSを細胞ペレットに添加した。100ulのPI/アネキシンV標識溶液を添加し、4℃で30分間インキュベートした。インキュベーション後、2mlの1×PBSを各チューブに添加し、その後1200RPM、室温で5分間遠心分離した。上清を廃棄した。細胞を0.5mLの1×PBSに再懸濁した。細胞についてフローサイトメーターでデータ取得する。FSC及びSSCで所要の集団(細胞数3000)に関して細胞(リンパ球)をゲーティングする(図25)。
破傷風トキソイド媒介性T細胞増殖アッセイにおいては、T1hにより記憶T細胞は阻害されない。
PBMCを、フィコールパック(Amersham、カタログ番号:17−14403−03)、密度勾配遠心分離により単離した。バフィーコートを健常ドナーから取得し、常に新たな状態で採取した。その後PBMCをPBS(Invitrogen)で洗浄した。その後PBMCを、0.3×106細胞/mlの細胞密度で、5%FBSを添加した2mlのRPMI培地に再懸濁させた。その後細胞を、無菌BD FACS 5mlチューブ中で、T1h(10ug/ml)及び非特異的対照として使用するhR3の存在下又は非存在下で30分間インキュベートした。インキュベーション後、細胞をボルテックス処理し、100μlの細胞懸濁液をそれぞれのウェルに添加した。100ulの破傷風トキソイド(カタログ番号582231、CALBIOCHEM)(10ug/ml)希釈標準溶液(5% FBSを含むRPMI培地)をそれぞれのウェルに添加して、記憶T細胞増殖を刺激した。プレートを、CO2インキュベーター中、37℃で5日間インキュベートした。65μlのアラマーブルーを各ウェルに添加し、CO2インキュベーター中、37℃で終夜インキュベートした。励起波長530nm、発光波長590nm及び感度=35で、Biotek社の分光光度計Synergy(商標)HTにより蛍光を測定した(図26)。
T1hは、PBMC及びラージ細胞により媒介される混合リンパ球反応においてT細胞増殖を阻害する。
ラージ細胞/PBMC細胞を採取し、1×PBSに再懸濁した。8×105細胞/mlのラージ細胞/PBMCを、1mlのマイトマイシン(25ug/ml)に再懸濁した。細胞を、CO2インキュベーター中、37℃で30分間インキュベートした。インキュベーション後、5% FBSを含む2mlのRPMIを各チューブに添加し、1200RPM、RTで5分間遠心分離して、マイトマイシンを除去した。上清を廃棄し、再び5% FBSを含む2mlのRPMIを添加し、遠心分離した。上清を廃棄し、細胞をRPMI培地に再懸濁させた。
混合リンパ球反応(樹状細胞及び末梢血リンパ球(PBL):
MLRアッセイ:
MLRアッセイをDC:PBL=1:2比として行った。実験テンプレートを、陽性対照ピメクロリムス及び陰性対照hR3を含む3種類の濃度のT1hで設計した。テンプレートを示す(図29)。それに従い、プレート全体にわたり(across)段階希釈を行った。
インキュベーション後、プレートを、顕微鏡によりコンタミネーションに関して確認し、20ul(1ウェル当たりの総容積の10%)のアラマーブルーを各ウェルに添加した。読取り値を、4時間目に開始して終夜、種々の時間フレームで、感度35で取得した(図30)。
上清を各ウェルから回収し、各群について上清をプールした。サイトカインを、BD CBA Th1/Th2キットIIを使用してフローサイトメトリーで分析した(図31)。
1型糖尿病動物モデル。
NOD(非肥満糖尿病)雌マウスは、経時的なグルコースの増大により測定される、自発性自己免疫性T細胞媒介性糖尿病を発症することが知られている。このインスリン依存性真性糖尿病(IDDM)モデルは、薬剤の治療効果及び予防効果を研究するための優れたモデルである。このマウスを、CD6に対する代替抗体(T1hがヒトCD6と結合する場合と同様の様式でマウスCD6と結合するラットモノクローナル抗体)の治療上及び予防上の利益を研究するために使用した。
コラーゲン誘導性関節炎モデル。
コラーゲン誘導性関節炎(CIA)に関するマウスモデルは、動物モデルにおける関節リウマチを研究するのに特に有利である。モデルは、抗体を含む抗関節リウマチ分子の効果を研究するのにも歴史的に使用されている。
自己免疫性脳脊髄炎(EAE)
実験的自己免疫性脳脊髄炎(EAE)は、脳炎症の動物モデルである。EAEは中枢神経系(CNS)の炎症性脱髄性疾患であり、多発性硬化症の疾患を含むヒトCNS脱髄性疾患のモデルとして研究されている。
同種移植片。
皮膚移植片は、A系の近交系マウス由来の皮膚をマウスBに移植すると特異的に拒絶されることがあり、皮膚は3日目と7日目との間に最初に血管新生する。その後血管新生された皮膚はリンパ球、単球、好中球及び他の炎症性細胞で浸潤される。7日目〜10日目までに、移植された組織の血管新生の減少が生じ、10日後には目に見えるネクローシスが生じる。
Claims (6)
- 配列番号1に示される重鎖ポリペプチドと配列番号2に示される軽鎖ポリペプチドとを含む抗CD6モノクローナル抗体であって、
多発性硬化症、移植拒絶反応、1型糖尿病、関節リウマチ、乾癬、及び他のT細胞媒介性自己免疫障害を含む群から選択される自己免疫障害の治療に使用され、かつ、以下の特徴:
i)連結抗CD3により誘導されるナイーブPBMCの増殖を阻害すること;
ii)ラージ細胞が抗原提示細胞でありPBMCが増殖する一方向MLRを特異的に阻害すること;
iii)PBMCにより媒介される一方向自己MLRを特異的に阻害すること;
iv)IL2の抑制を媒介することによりT細胞増殖を阻害すること;及び
v)CD25カウント数及びCD4カウント数の低減によりナイーブT細胞増殖の阻害を引き起こすこと;のうち1つ又は複数を有する、抗CD6モノクローナル抗体。 - 免疫抑制剤と組み合わせて0.1mg/Kg/週〜25mg/Kg/週の薬学的有効用量でそれを必要とする被験体に前記抗体を投与することにより、多発性硬化症、移植拒絶反応、1型糖尿病、関節リウマチ、乾癬、及び他のT細胞媒介性自己免疫障害を含む群から選択される自己免疫障害の治療において使用される、請求項1に記載の抗CD6モノクローナル抗体。
- 連結抗CD3;連結抗CD3及び抗CD6の組合せ、又は連結抗CD3及びALCAMの組合せにより誘導されるナイーブPBMCの増殖を阻害する、請求項1または2に記載の抗CD6モノクローナル抗体。
- 炎症促進性サイトカインIL6及びINFγの下方調節に関与する、請求項1ないし3のいずれか1項に記載の抗CD6モノクローナル抗体。
- 配列番号1に示される重鎖ポリペプチドと配列番号2に示される軽鎖ポリペプチドとを含み、それを必要とする被験体に対して0.1mg/kg/週〜25mg/kg/週の薬学的有効用量で治療のために使用される、抗CD6モノクローナル抗体。
- インスリン、GAD、MOG、MBP及びHSP60を含む群から選択される抗炎症性免疫応答を誘発することができる抗原と組み合わせて投与される、請求項5に記載の抗CD6モノクローナル抗体。
