Description
本発明は、抗原特異的T細胞応答を刺激するための方法に関する。
抗原(Ag)特異的T細胞応答の研究は、厄介な技術的課題をもたらす[Kern, Trends Immunol. 26:477, 2005]。これはAg特異的画分が一般的に、末梢血中に非常に低い頻度で示されるという事実に主に起因し、この特徴がその検出を困難なものとしている[Mallone, Clin.Immunol. 110:232, 2004]。この検出はCD4+T細胞を考える場合にはさらにより問題のあるものとなる。なぜなら、これらの画分は、しばしば、そのCD8+の同等物よりもさらにより低い頻度でしか存在しないからである[Homann, Nat.Med. 7:913, 2001; Seder, Nat.Immunol. 4:835, 2003]。
本発明者らは、適切なサイトカイン混液および培養条件を使用して、未分画全血または末梢血単核球(PBMC)試料からの直接的な成熟樹状細胞と共にAg特異的T細胞を共培養することによって、Ag特異的T細胞応答を刺激することが可能であることを発見した。
a)樹状細胞(DC)の分化を誘導する培地中において血液試料またはPBMC試料を培養する工程;
b)場合により、前記DCを成熟させる工程;
(Agを、工程a)および/またはb)の最中に加える)
を含む、被験体から単離した前記血液試料またはPBMC試料中において抗原(Ag)特異的T細胞応答を刺激するための方法を提供する。
本発明は、以下の工程:
a)DCの分化を誘導する培地中において血液試料またはPBMC試料を培養する工程;
b)場合により、前記DCを成熟させる工程;
(Agを、工程a)および/またはb)の最中に加える)
を含む、被験体から単離した前記の血液試料またはPBMC試料中においてAg特異的T細胞応答を刺激するための方法に関する。
本発明の方法は、DCの分化を誘導する培地中における血液試料またはPBMC試料の培養工程を含む。
本発明の方法によると、工程a)の最中に血液試料またはPBMC試料のDCを濃縮した後、前記DCを工程b)の最中に成熟させることができる。
理論によって練ることなく、本発明の方法にかけられた血液試料またはPBMC試料は、種々の成熟段階のDC(単球、未成熟DC、成熟DC)と、他の細胞の中でもとりわけT細胞との共培養液を含むと考えられる。
刺激されたT細胞の検出のための方法は、当業者には公知である。以下に記載した手順は、適切な方法のいくつかの例を提供する。しかしながら、当業者はAgに応答するT細胞の刺激を評価するのに適した任意の方法を使用することができることを容易に解釈することができる。
この手順を、以下の実施例1に詳述する。
培養上清中に放出されたサイトカインを、種々の技術、例えば酵素結合イムノソルベントアッセイ(ELISA)、BDサイトメトリックビーズアッセイ、Biorad Bio-Plexアッセイおよびその他の技術によって測定する。
この手順を用いて、特異的なペプチドエピトープを認識するAg反応性T細胞を、市販されている試薬(例えばProImmune MHCクラスII Ultimers)または社内で作成した試薬(例えば、Dr. G.T. Nepom, Benaroya Research Institute, Seattle, USAからの) [Novak et al., J.Clin.Invest. 104:R63, 1999]のいずれかを使用して検出する。
この手順を用いて、Ag特異的T細胞応答を、Ag認識後に膜上に露出する活性化マーカーのその異なる発現によって検出する。
Miltenyi Biotechによって開発されたこのシステムは、そのサイトカイン応答に従ってAg特異的T細胞を可視化するためのELISpotに代わる有効なものである。さらに、それは、対象のT細胞の直接的な選別およびクローニングを可能とする(以下参照)。
この手順は、近年、詳細に記載されている[Chattopadhyay et al., Nat.Med. 11:1113, 2005; Frentsch et al., Nat.Med. 11: 1118, 2005]。それはAg特異的CD4+T細胞の検出に限定される。
この手順[Betts et al., J.Immunol.Methods 281:65, 2003]は、細胞障害能を有するAg特異的CD8+T細胞の可視化を可能とする。
この手順は、Ag認識後のその増殖によりAg特異的T細胞(CD4+およびCD8+)を検出する[Mannering et al., J.Immunol.Methods 283:173, 2003]。
本出願において記載したAg特異的T細胞応答を刺激するための方法は、迅速で、効率的で、特異的で、多用途な手順である。要約すると、伝統的な方法と比較しての利点は以下の通りである:
1.より高い感度;
2.未分画PBMCまたはさらには未分画血液(新鮮または凍結のいずれか)を使用することができる。事前の精製工程の必要がなく、このことがこの技術をより簡単にし、そして血液容量の点における負担をより少なくさせている;
3.事前の長期の増殖が必要でない;
4.タンパク質Agまたは組織もしくは細胞の調製物のAgを使用する場合には、特異的なエピトープの限定されたセットに対するレパートリーよりもむしろそうしたAgに対する全T細胞レパートリーを検出することができる;
5.HLA拘束性の制限がない;
6.ペプチドAgとも適合性;
7.T細胞活性化の種々の解読値と適合性
8.同じ刺激技術を使用して、Ag特異的CD4+T細胞を増殖させ、続いて選別し、そしてさらなる特徴付けのためにCD4+T細胞株およびクローンを生成することができる。

実施例1:PBMC由来の促進された共培養DC(acDC)は、共培養T細胞のAg特異的応答(タンパク質Agによる刺激)を増幅する
材料および方法:
0日後に、全PBMC(2.5×106個の細胞/ウェル)を、48ウェルプレートの、1,000U/mlのGM−CSF、500U/mlのIL−4(両方共にR&D Systemsから)、および関連タンパク質Ag(10μg/ml)の補充されたAIM−V培地(Invitrogen)に播いた。試験したタンパク質抗原は破傷風トキソイド(TTX)、M. tubercolosis精製タンパク質誘導体(PPD)、6価ワクチンのインファンリックスヘキサ(GlaxoSmithKline)、プロインスリン(PI)、グルタミン酸デカルボキシラーゼ(GAD)、インスリンC−ペプチド、プレPIリーダー配列、ミエロペルオキシダーゼ、プロテイナーゼ3であった。陰性対照として使用したタンパク質の一例はウシ血清アルブミン(BSA)である。

慣用的な7日後のDCのための類似の成熟混液が記載されている[Luft et al., Int.Immunol. 14:367, 2002]。さらに、本発明者らは、低用量のIL−7(0.5ng/ml)を使用して応答をさらに増幅させた。
このプロトコールは、IL−10基礎(非刺激)分泌の大きな増加に因り、IL−10ではなく例えばIFN−γを産生するAg特異的CD4+T細胞応答を検出するのに適していた。

acDCおよび慣用的な(7日後の、単球由来の)DC(未成熟または成熟のいずれか)の表現型を、HLA−DR、CD14、CD80、CD86、CD11cに特異的なmAbで染色することによって決定した。エンドサイトーシス活性をデキストラン−FITCと共にインキュベーションし、続いて取り込まれた蛍光を評価することによって評価した。全ての細胞を、FACSAriaフローサイトメーター(BD)で分析した。
acDCの特徴付けにより、慣用的な7日後のDCのそれと同一な表現型が判明した。CD14のダウンレギュレーションは、HLA-DRおよび共刺激分子の発現増加と平行して起こったが、デキストラン取り込みは成熟時に減少した。
材料および方法:
実施例1に記載したのと同じ実験を、タンパク質Agによる刺激の代わりに、ペプチド抗原刺激を使用して行なった。試験したペプチドAgの例は、インフルエンザマトリックス(MP)58〜66、インフルエンザヘマグルニチン(HA)306〜318、GAD555〜567、GAD114〜123、PIB10〜18であった。陰性対照として使用したペプチドの例はピルビン酸デヒドロゲナーゼ(PD)5〜13およびコラーゲンII(CII)261〜273であった。
タンパク質AgでパルスされたacDC培養液中に誘起されたシグナルは専ら、CD4+T細胞を起源としていた。なぜなら、これらの細胞を除去した時に前記シグナルが完全に消失したからであった。Ag特異的CD8+T細胞応答は、より長い培養期間(7日間)かけてしか誘起することができなかった。7日後のDCを使用して全PBMCまたはCD4枯渇PBMCを刺激した場合に同じことが当てはまり、従って、acDCに特異的な欠陥特徴は除外される。このCD4特異的刺激は、CD8+T細胞の非効率的な活性化に起因せず、しかしむしろ、内部移行Agの交差提示を誘起するのに最適ではない培養条件に起因していた。実際に、タンパク質Agではなくペプチドエピトープを使用した場合、CD4+およびCD8+T細胞応答の両方がトリガーされ、そして両方が、単球と比較してacDCによって有意に増幅された。従って、本発明者らは、特異的なペプチドエピトープを認識するAg特異的T細胞を増殖および検出するための、acDC培養技術の改変形を綿密に作った。この場合、対象のペプチドを1日後に炎症誘発刺激と共に加える。この改変形は、所与のエピトープに対して特異的なCD4+およびCD8+T細胞の両方の検出を可能とする。
サイトカイン混液およびタンパク質/ペプチドAgを、実施例1(タンパク質Agについて)または実施例2(ペプチドAgについて)のように、予めPBMC精製または血液希釈を全く行なっていない新しく採血したヘパリン処理した血液試料中に直接的に加えた。48時間の培養終了時に、血漿および/またはPBMCを回収し、そしてELISA(R&D)、サイトメトリックビーズアレイ(BD)、もしくはBio−Plex(Biorad)アッセイを使用した血漿中のサイトカイン測定によって、または赤血球溶解後の細胞画分に対するMiltenyiサイトカイン捕捉アッセイによって、Ag特異的T細胞応答について分析した。またこの場合、acDCにより引き起こされる培養液中において誘起されたAg特異的応答は、単球により引き起こされる培養液中において誘起されるものよりも高かった。使用する成熟プロトコールに依存して、これは、IFN−γ、IL−10、IL−2、IL−6、IL−13、TNF−α、G−CSF、IL−1βを含む試験した多くのサイトカインについて該当した。
acDC培養系はまた:1)さらなる機能的な特徴付け(例えばRT−PCR技術による)のためのAg特異的T細胞の選別;並びに2)さらなる分析のためのT細胞株およびクローンを生成するために適している。
1型糖尿病(T1D)は、インスリン産生β細胞をターゲティングするT細胞により媒介される自己免疫疾病である。その発症率は着実に増加している(フランスにおいては1年当たり100,000中15人までが新たに診断される;1年あたり3〜4%の発症率の増加)。その独特な疫学から(それは主に小児および若年成人に生涯にわたり罹患する)、それは慢性的で費用がかかり衰弱させる疾病であり、重度の合併症(心臓血管疾病、腎症および末期腎疾病、網膜症および盲目)に至る。
アブストラクト
抗原(Ag)特異的T細胞の検出はしばしばアッセイ感度によって制限される。それ故、本発明者らは、in situにおいて樹状細胞(DC)の誘導および成熟を促進することによって(促進された共培養DC、acDCと呼ぶ)、ヒトおよびマウスの末梢血単核球(PBMC)中におけるAg処理およびT細胞への提示を増強するためのアプローチを考案した。未分画PBMCまたは全血を、タンパク質またはペプチドAgおよびサイトカイン混液と共に48時間かけてインキュベーションし、acDCを迅速かつ連続的に誘導、パルスおよび成熟させた。同様に、Agを処理および/または隣接するT細胞に提示し、これによりT細胞活性化に至る複数の工程を圧縮し、そして時間、操作および血液必要量を最小限とした。誘起されたT細胞応答は、種々の解読値(サイトカイン分泌、増殖、CD137アップレギュレーション、ヒト白血球Agマルチマーの結合)によって検出されたようにAg特異的であった。acDCに基づいたアッセイは、ウイルス、腫瘍および自己免疫疾病などの種々の設定においてT細胞応答をモニタリングするための価値ある適用を見出し得る。
種々の外来抗原および自己抗原(Ag)に対する応答を開始する上でのT細胞の中心的な役割にも関らず、例えば感染症または自己免疫疾病における免疫により媒介される過程の慣用的な診断検出は、主に、専らではなくても、抗体(Ab)応答の測定に依拠する。しかしながら、Absは基礎にある病態を常に媒介または反映しているわけではなく、そして免疫過程が主にT細胞により媒介されている場合にはより情報は少なくなるであろう(1)。現在までのAg特異的T細胞アッセイの唯一の信頼できる臨床的適用は、M. tuberculosis感染の診断においてであった(2)。さらに、T細胞免疫を効果的に測定する重要性は診断適用を超える(3)。T細胞のモニタリングは、ウイルスもしくは腫瘍特異的な免疫をブーストすることを、または自己組織(4)もしくは移植組織(5)に対する免疫を消失させることを目的とした、免疫調節療法を評価するためにも必要とされる。補充タンパク質(例えば凝固因子)(6)またはワクチン(7)の免疫原性能を評価するためのT細胞スクリーニングツールも等しく要求される。
acDCはAg特異的T細胞応答を増幅する
acDCが共培養T細胞のAg特異的応答を増幅し得るかどうかを調べるために、acDCを、GM−CSFおよびIL−4によって24時間かけてPBMC混合物内において誘導した。同時に、異なるタンパク質Ag[例えば、破傷風トキソイド(TTX)、M. tuberculosis精製タンパク質誘導体(PPD)、6価ワクチンまたは無Ag]を培養開始時から加えた。さらに24時間の成熟の後、非接着細胞を、抗インターフェロン(IFN)−γ捕捉Abでコーティングされた酵素結合イムノスポット(ELISpot)プレートに移し、そして追加のAgまたはサイトカインの補充を行なうことなくさらに6時間かけて培養した(図解については、図7を参照されたい)。
再現性を、同じPBMC調製物(すなわち、分析レベルにおけるアッセイ間変動性、これは採血および処理によって導入される差異を除外する;図2c)および同じ個体からの異なる時機に採取した血液に由来するPBMC調製物(すなわち、分析前レベルおよび分析レベルにおけるアッセイ間変動性、これは採血および処理に起因する差異を含む;図2d)からの異なる凍結細胞アリコートを試験することによって、再現性を評価した。両方の場合において、アッセイ変動度は10%未満であった。顕著には、新鮮な試料と凍結した試料との間の変動(6.9%)も小さかった(図2e)。
Ag特異的T細胞を選択および増殖するために、細胞に結合するダイであるカルボキシフルオレセインジアセテートスクシンイミジルエステル(CFSE)の希釈液を、増殖の解読値として使用し(14)、そしてacDC手順と併用した(図3a)。PBMCを最初にCFSEで標識し、その後、acDCにより増幅されたELISpot手順において使用した。それらをELISpotプレートから回収し、そしてさらに操作を行なうことなくさらに6〜8時間かけて培養した。その後、増殖している(CFSE低度)細胞をフローサイトメトリーによって同定し、単一細胞へと選別し、そして抗CD3Ab、IL−2およびIL−4を用いての3回の刺激を通してさらに増殖させた(15)。代表例を図3b〜dに示す。TTX特異的IFN−γELISpot応答が検出され(441個のIFN−γスポット形成細胞(SFC)/106個のPBMC;0.044%)(図3b)、これにより3.0%のTTX特異的CFSE(低度)画分が得られ、これは約68倍の増殖に対応する。acDC条件は、サイトカインの非存在下における慣用的な増殖よりも優れ、これは10倍少ないTTX特異的細胞を生じた(0.29%、p<0.001)。バックグラウンド増殖の有意な増加は全く観察されなかった(図3c)。TTX特異的CFSE(低度)画分(分裂細胞)を選別およびクローニングし、TTXvs対照でパルスしたDCの想起アッセイによって評価したところ、TTX特異的T細胞クローンが生成された(図3d)。
次に、本発明者らは、タンパク質およびペプチドAgでパルスされたacDCによるT細胞刺激を比較した。タンパク質Agを使用した場合、IFN−γELISpotによって追跡される48時間のacDC培養液中に誘起される応答は、専らCD4+T細胞を起源とした。なぜなら、刺激開始時または終了時のいずれかにおけるその枯渇は完全に応答を消失させたからである(図4a)。同じことがmoDCにも該当し、このことは、この特徴が、acDCに特有ではなかったことを実証する。弱いCD8+T細胞活性化は、48時間の培養中に取り込まれたAgの非効率的な交差提示に起因する可能性が高かった。従って、タンパク質Agを、HLAクラスIIまたはクラスI拘束性ペプチドエピトープによって置換した場合、CD4+およびCD8+T細胞応答がそれぞれトリガーされ(図4b)、ここでもCD4+およびCD8+T細胞の枯渇によって確認された(示さず)。さらに、両方のタイプの応答が、慣用的なPBMCと比較してacDCによって有意に増幅され(それぞれCD4+およびCD8+T細胞について3.2倍および6.3倍;p<0.05)、このことは、タンパク質およびペプチドAgの両方におけるacDC技術の有用性を実証する。
本発明者らは、acDC増幅が、精製PBMCを使用した他の機能的T細胞解読値にも適用されるかどうかをさらに調べた。IFN−γ分泌を、CD45(免疫細胞の表面に結合する)およびIFN−γに対する二重特異的mAbを用いての捕捉アッセイ(Miltenyi)によって検出した(図5a)。サイトカインの非存在下において有意なTTX特異的IFN−γ分泌は検出されなかった。しかしながら、IFN−γは、acDC増幅後のCD4+およびCD8+T細胞上の両方に検出され、バックグラウンドの増加は検出されなかった。IFN−γ+T細胞の数は、ELISpotによって検出されたものよりも高く、これは何故CD8+T細胞応答も可視化されたのかを説明し得る。T細胞活性化マーカーであるCD137の表面発現を用いて類似の結果が得られた(16、17)(図5b)。両方の解読値はまた、Ag特異的T細胞の下流の選別およびクローニングに適合性であった(データは示さず)。
最後に、本発明者らは、acDC増幅が、全血刺激後に回収した血漿でのAg特異的な大容量のサイトカイン分泌を検出できるかどうかを探索した。この目的を達成するために、ヘパリン処理した全血を、acDC混液(GM−CSF/IL−4、次に抗CD40/IFN−α)およびAgと共に上記のようにインキュベーションした。48時間後、血漿上清をサイトカイン測定のために回収した。いくつかのサイトカインが、Ag刺激時に有意な増加を示した(図6a)。マクロファージ炎症タンパク質(MIP)−1αを除いて、正味の(すなわちバックグラウンドを差し引いた)Ag特異的シグナルは、全てのマーカーについて「サイトカインなし」の条件よりもacDCにおいての方がより高かった(シグナル増幅の中央値、6.1倍;3.6〜41.9の範囲;p<0.001)。重要なことには、基礎分泌はacDC暴露時に増加せず、そしていくつかの場合には減少さえし、Ag特異的増幅効果を示す。CD137のアップレギュレーションについて本発明者らは、全血およびPBMC中におけるサイトカインの検出の感度を比較した(図6b)。サイトカインは、全血よりもPBMCを用いての方がより高い感度で検出され、濃度の中央値は、PBMCよりも約4倍高かった(1.0〜34.0の範囲;p<0.001)。
DCの治療能は、疾病に関連したAgに対する免疫原性または免疫寛容誘発T細胞応答を誘導するために活発に探索されている(18、19)。しかしながら、その強力なAg処理および提示特性にも関わらず、DCは、T細胞診断のためには活用されていない。これは、近づける循環DCの頻度が低いことによって、並びに単球および他の前駆体から開始してDC型APCを生成するのに必要とされる多くの血液容量によって示される拘束におそらく起因する。acDC技術は、慣用的な実験適用に受け入れられる短くて簡単な方法でAg特異的T細胞応答を増幅する手段を提供することによってこの欠陥を充足する。acDCとmoDCとを並べて比較することにより、表現型および刺激能の点の両方において顕著な類似性が判明した。acDCの顕著な利点は、未分画PBMCまたは全血のより生理学的設定において48時間以内にin situで生成されることである。さらに、試料必要量は最小限であり、僅かに106個のPBMC(約1mlの血液)または250μlの全血が必要とされるだけである。これは、特に小児におけるT細胞応答の長期モニタリングにおいて重要な考慮すべき事柄であり、そしてT細胞エピトープのためのペプチドライブラリーをスクリーニングする際の明確な利点である。
抗原。以下のAgを使用した:ウシ血清アルブミン(BSA; Sigma, Lyon, France)、TTX(Dr. Rino Rappuoliの親切なる贈り物、Novartis, Siena, Italy)、M. tubercolosis PPD(Tubertest, Sanofi Pasteur, Lyon, France)、6価ワクチン(インファンリックスヘキサ、GlaxoSmithKline, Rixensart, Belgium)およびKLH(Sigma)。Ag純度はSDS−PAGEによって確認され、そしてエンドトキシン濃度はLumulus溶解液アッセイ(Lonza, Saint Beauzire, France)によって0.035EU/μg未満であった。ペプチドFlu MP58〜66およびFlu HA306〜318は、95%超で純粋であった(GL Biochem, Shanghai, China)。

Claims (19)
- 以下の工程:
a)樹状細胞(DC)の分化を誘導する培地中において血液試料または未分画末梢血単核球(PBMC)試料を培養すること;
b)前記DCを成熟させること
ここで、抗原(Ag)を、工程a)および/またはb)の間に加える
を含む、被験体から単離した血液試料または未分画PBMC試料中においてAg特異的T細胞応答を刺激するための方法。 - 以下の工程:
a)DCの分化を誘導する培地中において被験体から得た血液試料または未分画PBMC試料を培養すること;
b)前記DCを成熟させること;
c)T細胞応答を検出すること
ここで、1つ以上の疾病に関連したAgを、工程a)および/またはb)の間に加える
を含む、前記被験体において疾病を診断するためにT細胞応答を検出する方法。 - 以下の工程:

- 以下の工程:
a)DCの分化を誘導する培地中において血液試料または未分画PBMC試料を培養すること;
b)前記DCを成熟させること;
