Description
(1) セルロース系物質とポリ酸と水性媒体とを含む混合物にマイクロ波を照射することを特徴とする、セルロース系物質の分解方法。
(2) セルロース系物質とポリ酸と水性媒体とを含む混合物にマイクロ波を照射することを特徴とする、セルロース誘導体の製造方法。
(3) セルロース系物質を(1)の方法により分解して分解物を得る前処理工程と、
前処理工程により得られた分解物を原料として用いて酵素糖化を行う酵素糖化工程と
を含む、セルロース系物質の酵素糖化方法。
水10mLに10mgのケイ酸タングステン(ポリ酸)(Alfa Aesar Tungstosilicicacid hydrate (H4SiO4・12WO3・xH2O), Reagent Grade)を溶解し(ケイ酸タングステン濃度:2.99×10-4 mol/l)、10mgのセルロース(Nacalai Tesque (code 07748-75))を分散させた。この分散溶液を家庭用の電子レンジで3〜20分間マイクロ波照射(2450 MHz, 500W)した。
(a)セルロース、(b)ポリ酸不存在条件で20分間マイクロ波照射したセルロース、(c)ポリ酸共存下で20分間マイクロ波照射したセルロース、(d)ポリ酸共存下で20分間マイクロ波照射したセルロースを水で洗浄したもの、それぞれについてX線回折図を得た。結果を図2に示す。
更に、(a)セルロース、(b)ポリ酸不存在条件で20分間マイクロ波照射したセルロース、(c)ポリ酸共存下で20分間マイクロ波照射したセルロース、(d)ポリ酸、それぞれについて赤外吸収(IR)スペクトルを得た。結果を図3に示す。
ポリ酸共存下でマイクロ波照射したセルロース分散溶液中の糖濃度を経時的に分析した。糖濃度(水溶性糖と還元糖)の測定は、一般的な測定法であるフェノール硫酸法とソモギネルソン法によって行った。
水90mLに200mgのケイ酸タングステン(ポリ酸)(Alfa Aesar Tungstosilicicacid hydrate (H4SiO4・12WO3・xH2O), Reagent Grade)を溶解し、100mgのセルロース(Nacalai Tesque (code 07748-75))を分散させた。この分散溶液を家庭用の電子レンジで10分間マイクロ波照射(2450 MHz, 500 W)して黒色粉体を得た。
ビーカーに10-2 mol/l、10-3 mol/lおよび2.99×10-4 mol/l のケイ酸タングステン水溶液10 mlをそれぞれ調製し、それぞれに10mgのセルロースを分散させた。得られた混合物を市販の電子レンジに設置し、マイクロ波を照射した。ケイ酸タングステン濃度以外の条件、使用装置等は実施例1と同じである。2.99×10-4mol/l のケイ酸タングステン水溶液を用いた処理は、実施例1での処理に相当する。
10-2 mol/lのケイ酸タングステン(Alfa Aesar Tungstosilicicacid hydrate (H4SiO4・12WO3・xH2O), Reagent Grade)水溶液100 mlに100 mgのセルロース(Nacalai Tesque (code 07748-75))を分散させ、市販の電子レンジに設置し、11.5分間マイクロ波(2450 MHz, 500W)を照射した。得られた生成物(残渣)に100 mLになるように精製水を加えて残渣を溶解させた。得られた試料を、高速液体クロマトグラフ(HPLC)分析に供した。
結果を図7に示す。上記条件によるセルロースのマイクロ波照射処理により、グルコースが分解物として選択的に生じることが確認された。
Claims (3)
- セルロース系物質とポリ酸と水性媒体とを含む混合物にマイクロ波を照射することを特徴とする、セルロース系物質の分解方法(ただし、前記混合物が触媒としてモリブデン酸イオン及び過酸化物を含有する場合と、前記混合物中で前記ポリ酸が固体である場合とを除く)。
- セルロース系物質とポリ酸と水性媒体とを含む混合物にマイクロ波を照射することを特徴とする、セルロース誘導体の製造方法(ただし、前記混合物が触媒としてモリブデン酸イオン及び過酸化物を含有する場合と、前記混合物中で前記ポリ酸が固体である場合とを除く)。
- セルロース系物質を請求項1の方法により分解して分解物を得る前処理工程と、
前処理工程により得られた分解物を原料として用いて酵素糖化を行う酵素糖化工程と
を含む、セルロース系物質の酵素糖化方法。
