Description
本明細書および特許請求の範囲において使用する用語「含む」は、「〜を含むが、それに制限されない」という意味を持つ。また、この用語は、「実質的に〜で構成された(必須成分として〜を有する)」及び、「〜からなる」の多少限定的な表現をも含む。
本発明は、非神経(非ニューロン)SNAREタンパク質を切断(開裂)する(cleave)新規の遺伝子工学的ボツリヌス毒素(engineered botulinum neurotoxins(BoNTs))、及び、その使用方法を提供する。
他の実施形態において、本発明は、ボツリヌス毒素による治療を必要とする被験者を治療する新規の方法を提供する。一実施形態において、本発明は、前述のように、改変触媒ドメインの治療有効量を投与することによる、ヒト分泌疾患を患う被験者を治療する方法を提供する。
本発明の一実施形態では、本発明の改変触媒ドメインで被験者を治療するためのキットが提供される。一実施形態において、そのキットは、本発明の遺伝子工学的BoNTの形態、及び、その使用のための指示書を含む。一実施形態において、本発明の改変触媒ドメインは、生理学的な状態において使用するために、製造され、伝達され、保管される。好ましい実施形態において、そのキットは、標的化システムを含み得る。その改変触媒ドメインは、その標的化システムに既に結合しているか、又は、そのキットは結合のための指示書とともに標的化システムを含む。その代案として、キットは、融合たんぱく質を特定組織特異的な標的化分子に操作するために用いられるLC/E(K224D)をコードするDNAを含む。
以下の実施例は、本発明を説明するために設けたものであり、本発明の範囲を何らかの形で制限するものではない。ここに示したもの以外の本発明の様々な変更は、発明の詳細な説明及び実施例の記載から当業者にとって明らかであり、添付された特許請求の範囲に記載された技術的範囲に含まれる。
分子モデリング
分子モデリングは、スイス−モデル(SWISS-MODEL)を用いて行った。LC/E-SNAP25(146-202)複合体の構造は、参考文献(19)に記載されたように、LC/E (PDB:3d3x)の結晶構造を用いて得られた。LC/E(K224D)-SNAP23の構造葉、テンプレートとしてLC/E-SNAP25複合体構造を用いて、PyMolTMを用いてモデル化した。示したデータは、3回以上実施した実験の平均(値)である。
Clostridium botulinum serotype E Beluga (配列番号: 1)のLC/E(1~400)をコードするDNAを増幅して、pET-15bへサブクローンすることで、BoNT LC/E発現ベクターを作製した。トランスフェクション実験のために、LC/E(1-400)をpEGFPベクターへサブクローンして、CMVプロモーター下で発現されたEGFP-LC/E (1-400)融合たんぱく質を産生した。SNAP25(145-206)のたんぱく質同等物(相当物)、SNAP23(152-211), SNAP29(202-259)、及び、SNAP47(406-464)の発現ベクターは、cDNA鋳型:SNAP23 (ATCC 2900640, 配列番号:2), SNAP29(ATCC 10700609, 配列番号: 4)、及び、SNAP47(ATCC 10468826, 配列番号: 5)をPCR増幅し、pGEX-2Tへサブクローンすることによって、作製された。 部位特異的突然変異は、QuickChange (Stratagene)を用いて行った。たんぱく質の発現及び精製は、前述のとおり(32)、実施した。
SNAREたんぱく質の切断は前述のように行った。
含まれた反応物(10 μ1): 5 μMのヒトSNARE たんぱく質、10mMのTris-HC1 (ph 7.6、20 mM NaC1を有する)、並びに、指示量のLC/E及びLC/E(K224D)。反応物を37℃で10分間インキュベートし、SDS-PAGEにかけ、ゲルをクーマシィー(Coomassie)で染色した。SNAREたんぱく質切断量(amount of SNARE protein cleavage)を密度(濃度)測定(densitometry)によって決定した。
SNAP25イソフォームを用いて、Wt-LC/E and LC/E誘導体に対するKm及びkcatを決定した。LCの濃度を調整して、幾つかの基質濃度(1.5~18μMのSNAREたんぱく質)において<10%(の)基質を切断した。反応を37℃で10分間行い、SDS-PAGEにかけ、切断生成物の量(amount of cleaved product)を密度(濃度)測定によって計算した。Lineweaver-Burkプロット、SigmaPlot IX (Chicago, IL)を用いて、反応速度対基質濃度をミカエリス・メンテン式にあてはめた。
HeLa細胞を、10%子牛血清(newborn calf serum)を加えたMEM中6ウェルプレートにおいて培養した。準融合細胞(sub-confluent cells)を、Lipofectamine LTX (GIBCO/BRL)を用いて、0.5又は1.0 μgの指示プラスミドでトランスフェクトさせた。
たんぱく質伝達は、参考文献(33)の手法に変更を加えて行った。HeLa細胞を、30 μMのジギトニンを含む透過性バッファー1ml/ウェルで7分間透過処理し、指示LCを有する、及び、有しない透過性バッファーにおいてインキュベートした。
一晩インキュベートした後、トランスフェクトされ、たんぱく質伝達された細胞を、20ng/mlのTNF-αを含んだ2mlの無血清MEMにおいてインキュベートした。36時間後、1.5 mlの上清を収集し、13,000 g(1分)の遠心分離にかけ、そして、ELISAを用いて、分泌されたムチン及びIL−8について分析した。上清(150 μl)を50 μlの0.2 M Na2CO3 (pH 9.6)と混合し、96-ウェルプレートに加え、そして、4℃で一晩インキュベートした。プレートを洗浄して、1%(w/v)BSAを含んだ50 mM Na2CO3(pH 9.6)バッファーでロックした(固定した)。プレートを洗浄して、RTにおいて100μlのα-ムチンIgG(1/200希釈、 Abcam)又はα-IL-8 IgG (1/200希釈, Abcam)とともに1時間インキュベートした。プレートを洗浄して、RTにおいてα-mouse Horse Radish Peroxidase-接合抗体(conjugate antibody)(1:10,000希釈、Pierce) とともに1時間インキュベートした。プレートを洗浄して、RTにおいて100μlのUltra-TMB (Pierce)を用いて20分間現像し、100μlの1M H2SO4. で急冷させた。対照上清において分泌されたムチン及びIL−8に対する(相対的な)部分(画分)としてA450を表した。
TNF-αとともにインキュベートされた(36時間)細胞からの溶解物を、ウエスタンブロット法(19)に基づいて、α-SNAP23マウスIgG(Abcam, Cambridge, MA)を用いて、LC/E及びLC/E(K224D)仲介(介在)の内因性SNAP223の切断について分析した。
次の伝達方法に基づいて、変異BoNTを被験者に投与した。
分子生物学分野における通常の知識を有する者は、PCR等のプロトコールに基づいて、標的化分子に融合されたLC/E(K224D)のキメラを操作することができるであろう。
脂質系伝達システムの使用の分野における通常の知識を有する者は、脂質対たんぱく質マトリックス(lipid to protein matrix)を用いて、LCの有効な内在化のために脂質LC/E(K224D)比率を適合させることができるであろう。
本発明の遺伝子工学的BoNTは、一実施形態において、過剰分泌(hyper−secretory)及び非分泌性(non-secretory)疾病を治療するために、被験者に投与された。当業者は、有毛細胞白血病における抗毒素療法(immunotoxin therapy)の開発に用いられた戦略に従って、遺伝子工学的BoNTを用いた治療に対し被験者を識別(確認)し、その治療を行い(投与し)、その結果をモニターし、そして、その治療の有効性を決定することができるだろう。
以前の研究では、インビトロ切断(19)においてLC/Eに対する206アミノ酸たんぱく質、SNAP25の最小の最適なペプチドとして残基167〜186を同定した。SNAP25 (167-186)は、基質結合「B」領域、及び、活性部位「AS」領域を含む2つのサブサイト(sub−site)を含む(図1a)。LC/Eは、P3残基を認識して、SNAP25のP2及びP1’残基の配列(alignment)を促す。LC/EのS1’ポケットは、LC/EのF191とSNAP25(20)のP1’残基I181間の疎水性相互作用とともに、F191、T159、及び、T208 によって形成される。ベーシック(basic)S2ポケットは、K224を含むが、それ(K224)が、(予想)塩橋(salt bridge)を通じて、P2残基D179を認識する。LC/Eの活性部位ポケットへのSNAP25のP2及びP1’残基をドックさせる(dock)ことによって、切断のために切ることができる結合がそろえられる(19、20)。
ボツリヌス神経毒の基質特異性の理解によって、拡張された基質特異性を有するBONT/Eの新規の軽鎖誘導体の操作が可能となり、神経学的適用以外にBONTの臨床治療を広げられる証拠を提供する。気道粘液は、 繊毛運動によって気道からの外部の破片(即ち、異種の破片)、細菌、及び、ウイルスを捕まえて除去することで上皮層を保護するが、粘膜毛様体クリアランス(mucociliary clearance)(17、18)と呼ばれるプロセス、過剰な気道粘液の分泌、粘液の過剰分泌は、粘液の蓄積が呼吸器疾患の一因となる慢性閉塞性肺疾患及び喘息のようなヒトの臨床症状(病態)にかかわる粘液の蓄積を引き起こし得る。粘液の分泌は、エキソシトシス(17,18)のために分泌細胞原形質膜とムチン含有小胞のドッキング(docking)を調整する(統合する)Muncタンパク質、SNARタンパク質、及び、ミリストイル化アラニン豊かなCキナーゼ基質(myristoylated alanine-rich C kinase substrate:MARCKS)を含んだいくつかの分子が連係された、制御されたプロセスである。基質改変されたBoNTによるSNAP23の標的化は、過剰分泌症候群の分泌プロセスを減らすことができる。また、SNAP23特異的BoNTは、分泌過多の要素(成分)(28、29)を含む免疫疾患、炎症性疾患、及び、糖尿病を含むそれ以外の治療的適用の標的とされ得る。

Claims (9)

に対し少なくとも90%の配列同一性を有し、
(b)改変した前記人工ボツリヌス神経毒軽鎖が、BoNT/E軽鎖のリジンアミノ酸残基224の置換物、又は、リジン以外のアミノ酸残基によってBoNT/E軽鎖の位置224に対応するリジンアミノ酸残基の置換物からなり、
(c)前記改変した触媒ドメインがSNAP23を切断する
ことを特徴とする遺伝子工学的ボツリヌス神経毒軽鎖。 - 請求項1に記載の遺伝子工学的ボツリヌス神経毒軽鎖を含む遺伝子工学的ボツリヌス神経毒E。
- 前記触媒ドメインがSNAP23を切断する、請求項1に記載の遺伝子工学的ボツリヌス神経毒軽鎖。
- 前記触媒ドメインが、SNAP25を切断するが、SNAP29又はSNAP47を切断しない、請求項1に記載の遺伝子工学的ボツリヌス神経毒軽鎖。
- 残基224、又は、残基224に相当する残基が、アスパラギン酸又はグルタミン酸に改変された、請求項1に記載の遺伝子工学的ボツリヌス神経毒軽鎖。
- たんぱく質伝達システムに有用な標的分子を更に含む、請求項1に記載の遺伝子工学的ボツリヌス神経毒軽鎖。
- 請求項1に記載の改変触媒ドメインからなる医薬組成物。
- 請求項1に記載の改変触媒ドメインからなるSNAP23が介在するヒトの非神経性分泌疾患を治療するための医薬組成物。
- 前記ヒトの非神経性分泌疾患が、喘息、CF、慢性閉塞性肺疾患、胃酸流出及び炎症、サイトカイン成分にかかわる免疫疾患、又は、サイトカイン成分にかかわる癌からなる、請求項8に記載の医薬組成物。
