Description
核酸増幅反応は、反応媒体中に最初に存在する核酸配列からの新たな核酸分子の合成を可能にするポリメラーゼの作用を含む一連の酵素反応からなる。
i)消耗品の表面を気相のエチレンオキシドで処理し、ついで
ii)液相または気相の過酸化水素で表面を処理することを含む。
1.5ml ポリプロピレンチューブからなる幾つかの消耗品を既知量のDNA、RNAまたは微生物で並行して汚染した。ついで、これらの消耗品をガンマ線で滅菌し(0〜60kGyの線量)、または気相のエチレンオキシドで処理した。滅菌後、残留核酸の存在をPCR(DNAの場合)およびTMA(RNAの場合)により判定し、滅菌されていない対照消耗品と比較する。
プロトコール1で使用したのと同じタイプの幾つかの消耗品を同じ既知量のDNA、RNAまたは微生物で汚染した。ついで、これらの消耗品を、液体溶液中の過酸化水素(それぞれ、3%および30% H2O2)での処理により処理した。該方法の終了時に、60℃の温度での真空下の乾燥の工程により、該過酸化水素を蒸発させる。処理後、残留核酸の存在をPCR(DNAの場合)およびTMA(RNAの場合)により判定し、未処理サンプルと比較した。
プロトコール1で使用したのと同じタイプの幾つかの消耗品を同じ既知量のDNA、RNAまたは微生物で汚染した。ついで、それらの消耗品の一部を、本発明に従い、気体エチレンオキシドで、ついで3%または30% 過酸化水素で処理した。他の一部を、気体エチレンオキシドの代わりにガンマ線で処理し、ついで過酸化水素で処理した。どちらの場合も、60℃の温度での真空乾燥工程により過酸化水素を蒸発させた。処理後、残留核酸の存在をPCR(DNAの場合)およびTMA(RNAの場合)により判定し、未処理サンプルと比較した。
幾つかのフィルターを含む幾つかの複雑な消耗品を同じ既知量の微生物(ジー・ステアロテルモフィルス(G.stearothermophilus))で汚染した。ついで、それらの消耗品の一部を、本発明の方法に従い、気体エチレンオキシド、ついで約90%の気体過酸化水素で処理した。対照として用いた該消耗品の他の一部は、処理しなかった。残留核酸(DNA)の存在をPCRにより判定した。図4は、種々のPCRサイクル中に増幅されたDNAの量を示す。該処理サンプルにおける増幅の非存在が認められうる。
Claims (14)
- 消耗品の表面上に存在する増幅可能な残留核酸の汚染除去方法であって、以下の工程:
i)消耗品の表面を気相のエチレンオキシドで処理し、ついで
ii)25%〜60%(v/v)の濃度で50℃〜70℃の温度まで加熱した液相の過酸化水素で、または気化させ30%を超える濃度にした気相の過酸化水素で該表面を処理することを特徴とする方法。 - 工程i)におけるエチレンオキシドを70%〜90%(v/v)の濃度で使用する、請求項1記載の方法。
- 工程ii)における過酸化水素が、30%(v/v)を超える濃度の液相である、請求項1または2記載の方法。
- 液相の過酸化水素を60℃を超える温度に加熱する、請求項3記載の方法。
- 液相の過酸化水素が、工程ii)において、汚染除去すべき表面と少なくとも40分間にわたって接触している、請求項2または3記載の方法。
- 過酸化水素での処理を気相で行う、請求項1記載の方法。
- 過酸化水素を気化させ、50%を超える濃度にする、請求項6記載の方法。
- 過酸化水素を気化させ、80%を超える濃度にする、請求項7記載の方法。
- 過酸化水素を60℃未満の温度で電場を適用することによりプラズマ状態にする、請求項7及び8に記載の方法。
- 工程i)と工程ii)との間に蒸発工程を更に含む、請求項1〜9のいずれか1項に記載の方法。
- 分子生物学における消耗品が、部分的に不織性の包装内に予め包装されている、請求項1〜10のいずれか1項に記載の方法。
- 工程i)およびii)のどちらかの前に真空を適用する、請求項1〜11のいずれか1項に記載の方法。
- 工程i)と工程ii)との間に残留エチレンオキシドを除去する、請求項1〜12のいずれか1項に記載の方法。
- 工程ii)の後で消耗品の表面から残留過酸化水素を除去する、請求項1〜13のいずれか1項に記載の方法。
