(ii) J6CF株由来の5’非翻訳領域、coreタンパク質コード配列、E1タンパク質コード配列、E2タンパク質コード配列、p7タンパク質コード配列及びNS2タンパク質コード領域のN末端から16位のアミノ酸残基までのアミノ酸配列をコードする配列と、JFH-1株由来のNS2タンパク質コード領域のN末端から17位のアミノ酸残基以降のアミノ酸配列をコードする配列、NS3タンパク質コード配列、NS4Aタンパク質コード配列、NS4Bタンパク質コード配列、NS5Aタンパク質コード配列、NS5Bタンパク質コード配列及び3’非翻訳領域を5’側から順番に連結したHCVキメラゲノム
(4)上記HCVキメラゲノムは、配列表の配列番号2に示される塩基配列からなる核酸(ただし、該核酸がRNAの場合は、塩基配列中のチミン(T)をウラシル(U)に読み替える。)である、(3)記載の抗HCV抗体。
本発明の一の態様は、HCV粒子を抗原とし、かつHCV感染の阻害活性を有する抗HCV抗体又はその断片である。
本発明の「抗HCV抗体」は、後述するHCVキメラゲノムから産生されるHCV粒子を抗原として誘導される抗体であって、HCV粒子における立体構造をエピトープとして認識して結合し、かつそのHCVの宿主細胞への感染を阻害する活性を有する、いわゆる中和抗体である。本発明の抗HCV抗体は、ポリクローナル抗体又はモノクローナル抗体を含む。好ましくはモノクローナル抗体である。本明細書において「モノクローナル抗体」とは、単一の免疫グロブリン、又はそのフレームワーク領域(Frame work region;以下、「FR」とする)及び相補鎖決定領域(Complementarity determining region;以下、「CDR」とする)を含み、抗原であるHCV粒子に特異的に結合し、かつそれを認識することのできるポリペプチドをいう。上記免疫グロブリンには、IgG、IgM、IgA、IgE、及びIgDの各抗体クラスが知られているが、本発明の抗体は、いずれのクラスであってもよい。好ましくはIgGである。

Sel. 17:21-27)。このようにダイアボディは二価の抗体断片であるが、それぞれの抗原結合部位は、同一エピトープと結合する必要はなく、それぞれが異なるエピトープを認識し、特異的に結合する二重特異性を有していてもよい。例えば、一方の抗原結合部位が、配列番号18、20及び22で示されるアミノ酸配列を含むCDR(それぞれP18-9EのVHにおけるCDR1、CDR2及びCDR3に相当)を包含するVHと、配列番号25、27及び29で示されるアミノ酸配列を含むCDR(それぞれP18-9EのVLにおけるCDR1、CDR2及びCDR3に相当)を包含するVLからなり、他方の抗原結合部位が、配列番号32、34及び36で示されるアミノ酸配列を含むCDR(それぞれP19-7DのVHにおけるCDR1、CDR2及びCDR3に相当)を包含するVHと、配列番号39、41及び43で示されるアミノ酸配列を含むCDR(それぞれP19-7DのVHにおけるCDR1、CDR2及びCDR3に相当)を包含するVLから構成されていてもよい。トリアボディ、及びテトラボディは、ダイアボディと同様に一本鎖Fv構造を基本としたその三量体、及び四量体構造を有する。それぞれ、三価、及び四価の抗体断片であり、多重特異性抗体であってもよい。さらに、上記「その断片」は、ファージディスプレイライブラリーを用いて同定された抗体断片(例えば、McCafferty et al., 1990, Nature Vol.348:552-554参照)であって、かつ抗原結合能力を有しているものが含まれる。この他、例えば、Kuby, J., Immunology, 3rd Ed., W.H. Freeman & Co., New York (1998)も参照されたい。
本発明における抗HCV抗体は、HCVキメラゲノムから産生されるHCV粒子を抗原とすることを特徴とする。
本発明において抗原として使用する感染性HCV粒子は、細胞培養系で調製することができる。この基本技術は、WO04104198A1、WO06022422A1、WO06096459A2、Wakita, T. et al., Nat. Med. 11:791-796,2005、Lindenbach, BD. et al., Science 309:623-626,2005及びPietschmann, T. et al., Proc. Natl. Acad. Sci. USA. 103:7408-7413, 2006に記載されている。以下、感染性HCV粒子の調製について具体的に説明をする。
HCV粒子の調製には、上記「1−2.抗原としてのHCV粒子」の項で記載したHCVキメラゲノムを使用することができる。例えば、配列番号2で示される塩基配列を有する核酸を使用してもよい。また、使用するHCVキメラゲノムは、RNA又はDNAのいずれであってもよい。ただし、核酸がRNAの場合には、上記塩基配列中の「チミン(T)」を「ウラシル(U)」に読み替えるものとする。本明細書中、他の塩基配列についても、以下同様とする。
HCV粒子は、完全長のHCVキメラゲノムRNAのcDNAを転写プロモーターの下流に発現可能なように連結した発現ベクター(例えば、T7プロモーターの制御下にHCVキメラゲノムを挿入したベクター)からHCVキメラゲノムRNAを合成し、このゲノムRNAを宿主細胞に導入することで調製することができる。ゲノムRNAの合成は、当該分野で公知の技術を使用すればよい。例えば、前述の発現ベクターを用いる場合であれば、in vitro RNA合成法によりゲノムRNAを合成することができる。in vitro RNA合成法を利用する各種キットは、ライフサイエンス関連の各メーカーから市販されているので(例えば、Ambion社のMEGAscript T7 kit)、それらを利用して合成してもよい。
合成したHCVキメラゲノムRNAを導入する宿主細胞は、HCV粒子形成を許容する細胞であれば特に限定はしない。例えば、Huh7、HepG2、IMY-N9、HeLa、HEK293等の培養細胞、より好ましくはHuh7等の肝由来培養細胞、さらに好ましくはHuh7の派生株であるHuh7.5、Huh7.5.1が挙げられる。本発明において「派生株」とは、ある培養細胞から誘導された異なる細胞株をいう。また、Huh7、HepG2、IMY-N9、HeLa又はHEK293においてCD81遺伝子及び/又はClaudin1遺伝子を発現させた細胞も使用することができる。
HCVキメラゲノムRNAを上記宿主細胞に導入する方法は、公知の任意の方法を用いることができる。例えば、リン酸カルシウム共沈殿法、DEAEデキストラン法、リポフェクション法、マイクロインジェクション法、エレクトロポレーション法が挙げられる。好適にはリポフェクション法及びエレクトロポレーション法が、さらに好適にはエレクトロポレーション法である。
調製されたHCV粒子が感染能を有しているか否かは、当該分野で公知のウイルス感染の評価方法によって確認することができる。例えば、HCVキメラゲノムRNAを導入した細胞の培養によって得られる上清を、さらに新たなHCV許容性細胞(例えば、Huh7)に添加した後、適当な時間(例えば、48時間)で培養する。その後、その細胞を十分に洗浄し、抗core抗体等のHCV特異的タンパク質を認識する抗体で免疫染色して感染細胞数をカウントするか、又は上記細胞からタンパク質を抽出し、抽出物をSDS-ポリアクリルアミドゲルにて電気泳動した後、ウェスタン・ブロッティング法にてHCV特異的タンパク質を検出することで判断できる。
得られたHCV粒子は、抗原として使用する前に、精製することが好ましい。HCV粒子の精製は、上記HCVキメラゲノムRNAを導入した細胞を培養して得られる感染性HCV粒子を含む溶液(ウイルス液)から細胞及び/又は細胞残渣を除去し、その後、カラムクロマトグラフィ及び密度勾配遠心をそれぞれ単独で、又は任意の順番で組み合わせて行うことによって達成できる。細胞及びその残渣の除去は、遠心及び/又はフィルター等を用いて行えばよい。必要に応じて、上記残渣を除去したウイルス液を、さらに分画分子量100,000〜500,000の限外濾過膜を用いて10〜100倍程度に濃縮することもできる。
本発明の抗HCV抗体の作製において、抗原として使用するHCV粒子の不活化処理は、ヒト以外の動物に投与する場合は必須ではない。しかし、操作する術者への感染防止の点から不活化することが好ましい。また、上記HCVキメラゲノムから産生されるHCV粒子をワクチンとしてヒトに投与する場合には、この不活化処理は必須となる。HCV粒子を不活化する方法としては、ホルマリン、β−プロピオラクトン、グルタルジアルデヒド等の不活化剤を、例えば、感染性HCV粒子を含むウイルス液に添加して十分に混合することにより達成できる(Appaiahgari et al., Vaccine, 22:3669-3675, 2004)。また、感染性HCV粒子を含むウイルス液に紫外線を照射することによりHCVの感染性を迅速に失わせることもできる。紫外線照射による不活性化方法は、HCVを構成するタンパク質等への影響が少ない点で好ましい。紫外線の線源には、一般に市販されている殺菌灯、特に15W殺菌灯を用いることができるが、それに限るものではない。好ましくは感染性HCV粒子を含む溶液を20mW/cm2の紫外線で室温にて5分以上照射することにより、感染性HCV粒子を不活化することができる。また、本不活化方法は精製・未精製等の状態により限られるものではない。
1−4−1.動物への免疫
本発明の抗体は、HCV粒子を動物に投与することで免疫反応により誘導させて得ることができる。免疫に用いる動物としては、ハイブリドーマを作製することが可能な抗体産生細胞を産生できる非ヒト動物であれば、特に限定しない。例えば、非ヒト哺乳動物、より具体的には、マウス、ラット、ハムスター、モルモット、ウサギ、ヤギ、ロバ、ヒツジ、ラクダ及びウマ等を用いることができる。本発明においてはマウスを用いる方法を例に挙げ、以下に示す。
抗HCVモノクローナル抗体を作製する場合、その抗体を産生するハイブリドーマを、例えば、以下に記載する方法によって作製することができる。
上記ハイブリドーマが生産する抗体のHCV感染阻害活性の有無については、以下で例示する方法、及び後述の実施例に述べる感染性HCV粒子を用いた方法により評価することができる。例えば、まず、上記抗HCVモノクローナル抗体産生ハイブリドーマを培養後、培養上清の一部を抗体サンプルとして採取する。この抗体サンプルと前述の感染性HCV粒子とを混合し、37℃にて1時間反応させる(混合サンプル)。次に、前日に96ウェルプレートに5×103個/ウェルで培養したHuh7細胞に上記混合サンプル50μLを加え、37℃にて2.5時間培養する。培養後、培養液及び混合サンプルを除去し、PBSにて細胞を洗浄後、再び新鮮培地を加えて培養を続ける。48時間後、培養液を除去し、PBSで1回洗浄後、ISOGEN(ニッポンジーン社)を100μL加えて、細胞からRNAを調製する。RNAを定量後、HCVゲノムRNA量を測定する。HCV RNAの定量的RT-PCRによる検出は、Takeuchiらの方法(Gastroenterology, 116:636-642,1999)に従いHCV RNAの5'非翻訳領域のRNAを検出すればよい。
上記で選択されたハイブリドーマを無血清培地、例えば、Hybridoma-SFM(インビトロジェン社)に馴化させ、培養した上清を抗HCV抗体サンプル(抗HCVモノクローナル抗体サンプル)とすることができる。培養には、フラスコ、シャーレ、スピナーカルチャーボトル、ローラーボトルあるいは高密度培養フラスコCELLine(ベクトンデッキンソン社)を使用することができる。
抗HCVヒト化抗体では、CDRが良好な抗原結合部位を形成することができるヒト抗体のFRが選択される。必要に応じて再構成ヒト抗体の相補性決定領域が適切な抗原結合部位を形成するように、抗体のV領域におけるFRのアミノ酸を置換してもよい(Sato, K., et al., Cancer Res. 53:851-856, 1993)。
本発明の抗HCV抗体を選択するためには、HCVのタンパク質を担体に固定(固相化)し、次にサンプル抗体を加え、抗体/抗原複合体を形成するのに十分な時間及び条件下で反応させる。次に、形成した複合体を検出するため、二次抗体、すなわちシグナルとなる酵素、色素若しくはラジオアイソトープを結合させたサンプル抗体を認識する抗体と接触させ、第2の混合物を形成させる。第2の混合物を抗体/抗原複合体を形成するのに十分な時間及び条件下で反応させる。これによって、HCVタンパク質を認識する抗体の存在を酵素、色素若しくはラジオアイソトープのシグナルを介して検出する。
本発明に係るより好適なHCV感染阻害活性を有する抗体は、HCVの、好ましくはJ6CF株の、E1タンパク質及びE2タンパク質の複合体の立体構造、すなわち、本発明に係る好適な抗体は、E1タンパク質及びE2タンパク質の複合体の三次構造の全部又は一部をエピトープとして認識することができる。E1タンパク質及びE2タンパク質は、宿主細胞表面上の受容体(HCV受容体)との結合に関与するエンベロープタンパク質であり、HCVは、このHCV受容体を介して宿主細胞に感染する。CD81は、このHCV受容体の一つとして同定されており、HCVの感染に必須の因子の一つであることが報告されている(Akazawaら、J. Virol. 81: 5036-5045、2007)。したがって、細胞への感染能を維持しているHCVは、CD81と結合できるE1タンパク質及びE2タンパク質の複合体の立体構造を保持していると考えられる。
上記方法により選択されたハイブリドーマ細胞株が産生する本発明の抗体のエピトープを解析するためには、HCVのタンパク質を用いたエンザイムイムノアッセイ(EIA)、ウェスタン・ブロッティング又はドットブロッティング等を使用することができる。抗HCVモノクローナル抗体の一次スクリーニングとして本エピトープ解析をすることで、特定のHCVタンパク質を標的とした抗HCVモノクローナル抗体を効率よくスクリーニングすることができる。
本発明の他の態様は、HCV感染阻害剤である。
本発明のHCV感染阻害剤は、医薬組成物に使用することができる。本発明の医薬組成物は、医薬としての本発明の抗HCV抗体の他に医薬的に許容可能な担体を含有することができる。
本発明のHCV感染阻害剤を含む医薬組成物は、投与単位形態で投与することが好ましく、経口投与、組織内投与(例えば、皮下投与、筋肉内投与及び静脈内投与)、局所投与(例えば、経皮投与)又は経直腸的投与で投与することができる。それ故、HCV感染阻害剤の剤形は、投与方法に適する形態であることが好ましい。例えば、組織内投与の場合、血流を介した注射が好ましく、それ故、剤形は液体である。
本発明のさらなる態様は、HCVの検出方法及びそれに使用するHCV検出試薬である。本発明は、本発明の抗HCV抗体を用いて試料中のHCV粒子を免疫学的検出方法により検出することができる。
本発明の他の態様は、本発明の抗HCV抗体を含み、上記「3.HCV検出方法」によってHCVを検出するためのHCV検出用キットである。本発明の検出用キットは、標識二次抗体、標識の検出に必要な基質、陽性対照や陰性対照、試料の希釈や洗浄に用いる緩衝液、及び/又は使用説明書を含むことができる。
劇症肝炎の患者から分離したHCV JFH-1株(遺伝子型2a)のゲノムRNA全領域を逆転写して得たcDNA(ゲノムcDNA)をpUC19プラスミドのT7 RNAプロモーター配列の下流にクローニングして得られたプラスミドDNA(pJFH-1)を、Wakita, T. et al., Nat. Med., 11 (2005) p.791-796及び国際公開WO 2004/104198に記載の方法に準じて作製した。pJFH-1中に挿入されているJFH-1株由来ゲノムcDNAの塩基配列は、配列番号1に示す通りである。このpJFH-1を、EcoRIで消化後、さらにBclIで部分消化し、EcoRI部位から最初のBclI部位までの断片(約2840bp)を切除したプラスミドDNA断片を調製し、それを精製した。
実施例1で産生させたウイルス粒子は、以下3段階の工程を用いて精製した。
中空糸カートリッジHollow Fiber Cartridge(GE Healthcare社:500kDaカットオフ、モデル番号UFP-500-C-8A、以下「Hollow Fiber」とする)を利用して、上記実施例1で得たHCV粒子を含む培養上清を60倍に濃縮した。
Ultra-clear 25×89mm遠心管(ベックマンコールター社:カタログ番号344058)に、冷却した60%ショ糖を含むTNE緩衝液(10mM Tris-HCl(pH 7.4)、150mM NaCl、1mM EDTA)を3mL加え、20%ショ糖を含むTNE緩衝液を7mL重層した。さらに、20%ショ糖を含むTNE緩衝液の上に、サンプルを25mL重層した。SW-28(ベックマンコールター社)を用いて、28,000回転で4時間、4℃で超遠心を行った。
溶出画分をAmicon Ultra-15 Centrifugal Filter Units(排除分子量:100kDa、ミリポア社)及びTNE緩衝液を用いてバッファ置換を行うとともに濃縮を行った。こうして得た濃縮液を、後述の免疫工程において感染性HCV粒子を含むウイルス液として使用した。
上記実施例2で得たウイルス液中のHCVを、紫外線照射にて不活化した。紫外線の線源としては東芝社製GL-15を使用した。精製した感染力価1×106ffu/mLのHCV粒子含有溶液をシリコンコーティングされたポリエチレン製エッペンチューブ(アシスト社製)に入れ、紫外線の照射強度が20mW/cm2となるような距離に置き、5分間UV-Cを照射した。
アジュバントとして動物用アジュバントであるMPL+TDM(Sigma社:Sigma Adjuvant System、カタログ番号S6322)を用いた。実施例3に示した不活化J6/JFH-1-HCV粒子(HCV coreタンパク質 2pmol相当量)を含んだ溶液100μLに対して、等量のMPL+TDMを加え、エマルジョンを形成させた。エマルジョンの形成はビーカーに適量の水を準備しておき、混合液を水面に一滴落として液が分散しないことにより確認した。Balb/cマウス(7週齢、雌)をエーテル麻酔し、上記のように調製した不活化J6/JFH-1-HCV粒子を含むエマルジョンを腹腔内投与することにより免疫した。
(1)感染性HCV様粒子(HCVpp)の作製
HCVppの作製は、Bartoschらの方法に準じて行った(文献:Bartosch, B. et al.(2003) J. Exp. Med., 197, 633-642)。これは、レトロウイルスのGag-polを発現するベクター、HCVのエンベロープタンパク質を発現するベクター及びレポーター遺伝子を発現するレトロウイルスパッケージングベクターの3種類の発現ベクターを動物細胞に導入し、発現させることにより、レポーター遺伝子がパッケージングされ、HCVエンベロープタンパク質をそのウイルスの表面に発現するシュードウイルスを作製する方法である。
実施例4におけるHCV粒子投与前の正常マウス血清及び不活化HCV粒子(J6/JFH-1-HCV粒子)で免疫したマウスから初回免疫49日後に採血した200μLの血清をそれぞれ1mLのプロテインGセファロースカラム(GEヘルスケア社)にアプライし、PBSでカラムを洗浄後、0.1Mグリシン緩衝液(pH3.0)にて1mL/画分で溶出した。溶出後、直ちに1M Tris-HCl(pH9.5)を添加し、中性に戻した。溶出されたタンパク質のピーク画分をIgG画分とし、この画分のHCV感染阻害活性を測定した。具体的には、本実施例「(1)感染性HCV様粒子(HCVpp)の作製」の工程で得たJ6CF HCVpp溶液に、IgGの最終濃度が10、30、100、200μg/mLとなるようにDMEM培地(10% FCS(invitrogen社)、1% MEM 非必須アミノ酸溶液(invitrogen社)、10mM HEPES-Tris(pH7.3)、1mM ピルビン酸ナトリウムを含むDMEM)にてIgG画分サンプルを希釈して加え、37℃で30分インキュベーションした。前日に96ウェルプレートに1x104細胞/ウェルで培養したHuh7.5.1細胞の培地を捨てた後、IgG画分サンプルを加えてインキュベーションしたウイルス液を50μL/ウェルで加えて、37℃で3時間インキュベーションした。ウイルス液を捨てた後、PBS 100μL/ウェルで1回洗浄し、200μL/ウェルの培地を加えて37℃で72時間インキュベーションした。培地を捨てた後、25μL/ウェルのDMEM培地(血清無添加)と25μL/ウェルのSteady-Glo(Promega社:カタログ番号E2520)を加えて添付の説明書に従い細胞を溶解した。細胞の溶解液40μL/ウェルを白色の96ウェルプレート(住友ベークライト社:カタログ番号MS-8496W)へ移し、ARVO X4(PerkinElmer社)を用いて発光強度を測定した。
マウス骨髄腫細胞株SP2/0(ECACCより入手)を5×10-5Mの2-メルカプトエタノール、100単位/mLのペニシリン、100μg/mLのストレプトマイシン、及び10%FCS(インビトロジェン社)を含有するDMEM培地(インビトロジェン社)にて培養し、対数増殖期のSP2/0細胞を得た。その細胞を、血清を含まないDMEM培地にて3回洗浄した。
実施例6で作製したハイブリドーマが十分に増殖した時点で、培養上清を回収し、以下の通りスクリーニングを行った。
J6CF株のE1タンパク質及びE2タンパク質は、以下に示すようにして調製した。遺伝子型2aのJ6CF株由来ゲノムcDNA(GenBankアクセッション番号AF177036)を鋳型として、J6CF全長タンパク質配列(J6CF株ゲノム配列にコードされる連続したタンパク質配列;GenBankアクセッション番号AF177036にも記載のアミノ酸配列)のN末端の開始メチオニンを1位とした場合に192位から352位までに相当する、膜貫通領域を含まないE1タンパク質をコードする遺伝子を、Advantage GC2 PCRキット(タカラバイオ社)、並びにJ6E1dTM-s(配列番号3:CACAAGCTTGCCGAAGTGAAGAACATCAGT)及びJ6E1dTM-as(配列番号4:GCTCTAGATTAATGAGCCCCGCTAATGATGTC)を用いてPCR法で増幅した。増幅したDNA断片をpCR-TOPO(インビトロジェン社)中にクローニングし、3つのクローンについて塩基配列の解析を行った。正しい塩基配列のインサートを有するクローンを「pTOPO-J6E1dTM」と名付けた。
J6CF株由来のE1タンパク質及びE2タンパク質がそれぞれ1μg/mLとなるようにPBSで希釈した。このE1タンパク質及びE2タンパク質の混合タンパク質溶液をイムノプレート(NUNC社)の各ウェルに50μL加え、4℃にて一晩静置し、タンパク質をプレートに固定した。タンパク質溶液を除き、各ウェルに150μLのブロックエース(大日本製薬)を加え室温で4時間ブロッキングした。
上記(2)で作製したJ6CF株由来のE1タンパク質及びE2タンパク質が固定されたプレートを0.1%Tween20(SIGMA社)を含むPBSで4回洗浄し、実施例6で得られた各ハイブリドーマ上清サンプルを各ウェルに50μL加え、室温で1時間、プレートミキサーにて振とうさせながら反応させた。反応後、0.1%Tween20(SIGMA社)を含むPBSで4回洗浄した後、各ウェルに、0.1%Tween20を含むPBSで10,000倍に希釈したHRP標識抗マウスIgG抗体(SIGMA社)を50μL加え、室温で1時間、振とうさせながら反応させた。反応後、0.1%Tween20(SIGMA社)を含むPBSで4回洗浄し、ペルオキシダーゼ発色キット(住友ベークライト社)を用いて発色させ、マルチスキャン(タイターテック社)にて450nmでの吸光度を測定し、陽性クローンを選抜した。
実施例5の「(1)感染性HCV様粒子(HCVpp)の作製」の工程で得たJ6CF HCVpp溶液にハイブリドーマ上清を加え、37℃で30分インキュベーションした。前日に96ウェルプレートにて1×104cells/ウェルで培養したHuh7.5.1細胞から培地を除去した後、ハイブリドーマ上清を加えてインキュベーションしたウイルス液を50μL/ウェルで加えて、37℃で3時間インキュベーションした。ウイルス液を捨てた後、PBS 100μL/ウェルで1回洗浄し、200μL/ウェルの培地を加えて37℃で72時間、再びインキュベーションした。培地を捨てた後、25μL/ウェルのDMEM培地(血清無添加)と25μL/ウェルのSteady-Glo(Promega社:カタログ番号E2520)を加えて添付の説明書に従い細胞を溶解した。細胞の溶解液40μL/ウェルを白色の96ウェルプレート(住友ベークライト社:カタログ番号MS-8496W)へ移し、ARVO X4(PerkinElmer社)を用いて発光強度を測定した。J6CF HCVpp溶液にDMEM培地(ハイブリドーマ上清と等量)を混合したウイルス液で上記の試験を同時に行ったときの発光強度を100%感染として、ハイブリドーマ上清と混合した時の発光強度を%で表し、感染率(%)を求めた。
実施例8で得られたハイブリドーマ細胞株から産生されるHCV感染阻害モノクローナル抗体の特性解析は、以下の通り行った。なお、ハイブリドーマ細胞株P18-9E(寄託番号:FERM BP-11263)から産生されるモノクローナル抗体を「モノクローナル抗体P18-9E」とし、ハイブリドーマ細胞株P19-7D(寄託番号:FERM BP-11264)から産生されるモノクローナル抗体を「モノクローナル抗体P19-7D」とした。
マウス抗体のサブクラスの解析については、ハイブリドーマの培養上清を用いてMouse MonoAB ID KIT(インビトロジェン社)を用いて行った。モノクローナル抗体P18-9E のアイソタイプの解析の結果、H鎖がγ2b、L鎖がκであることが判明したことからIgG2bであることが示された。モノクローナル抗体P19-7Dのアイソタイプの解析の結果、H鎖がγ1、L鎖がκであることが判明したことからIgG1であることが示された。
無血清培地Hybridoma SFM(インビトロジェン社)にて、ハイブリドーマをコンフルエントになるまで培養した後、培養液を遠沈管に回収し、1500rpmで5分間遠心した。培養上清をProsep-G(ミリポア社)に添加し、20ベッドボリュームのPBSで洗浄し、続いて1ベッドボリュームの0.1Mグリシン-HCl(pH3.0)にて6画分溶出した。溶出後、直ちに1M Tris-HCl(pH9.5)を添加し、中性に戻した。NanoDrop(NanoDrop Technologies社, ND-1000)を用いて各フラクションの280nmの吸光度を測定し、タンパク質が含まれるフラクション(OD280nm>0.1)を回収し、アミコンウルトラ50K(ミリポア社)を用いて濃縮しながらPBSにbuffer置換し、0.22μmフィルター濾過した。濃縮サンプルの20μLを還元下及び非還元下にてSDS-ポリアクリルアミドゲル電気泳動を行い、クマシーブリリアントブルーにて染色してIgGであることを確認した。サンプルの280 nmの吸光度を測定し、溶液に含まれる抗体量(10mg/mL IgG=13.7 OD換算)を算出した。
A. 感染性HCV様粒子(J6CF HCVpp)に対するHCV感染阻害活性
実施例5の「(1)感染性HCV様粒子(HCVpp)の作製」の工程で得たJ6CF HCVpp溶液に、精製モノクローナル抗体P18-9E又は精製モノクローナル抗体P19-7Dを加え、37℃で30分インキュベーションした。精製モノクローナル抗体P18-9E又は精製モノクローナル抗体P19-7Dは、それぞれ、最終濃度が100μg/mL又は300μg/mLとなるように、PBSにて希釈して用いた。
HCVccウイルス液として、上記実施例2の(1)の工程で得たJ6/JFH1-HCV粒子液(HCV粒子を含む培養上清の濃縮液)(以下、「J6/JFH1 HCVcc」とする)を用いた。
JFH1 HCVcc溶液にPBS(希釈した精製モノクローナル抗体と等量)を混合したウイルス液で上記の試験を同時に行ったときの蛍光強度を100%感染とし、精製モノクローナル抗体と混合した時の蛍光強度を%で表し、感染率(%)を求めた。
J6CF株のE1タンパク質の1〜162位のアミノ酸(配列番号7)、J6CF株のE2タンパク質の1〜337位のアミノ酸(配列番号8)について、10個の連続するアミノ酸をN末端から3アミノ酸ずつずらして設計したアミノ酸配列からなるペプチド群を合成した。各ペプチドのN末端はビオチン化し、C末端はグリシンアミドとした(JPT社に合成委託)。
HCV粒子に対するマウスモノクローナル抗体のV領域をコードするDNAを次の様にしてクローン化した。
上記(5)で塩基配列を決定し、プラスミド中にクローン化したDNA(マウスモノクローナル抗体のVH及びVLをそれぞれコードするcDNA)について、以下の通り解析を行った。
モノクローナル抗体P19-7Dの L鎖ではMuIgκVL5'-Gの5’プライマーで増幅されたPCR産物をクローン化した4クローンについて解析した結果、V領域の塩基配列2箇所で、4クローン中1クローンで置換がみられたが、3クローンの塩基配列は同一であった。各箇所で3クローンが示すほうの塩基を選択し、モノクローナル抗体P19-7Dの L鎖cDNAの塩基配列を決定した(配列番号12)。
A.エンザイムイムノアッセイ(EIA)を用いたHCV 様粒子(HCV-VLP)に対する(HCV-VLP)モノクローナル抗体の反応性の検討
(a)HCV様粒子(HCV-VLP)の作製
HCV様粒子(HCV-VLP)とは、ウイルスゲノムを含まない中空粒子である。HCVのE1タンパク質及びE2タンパク質を発現させた293T細胞から、MembranePro(登録商標) Reagent(Invitrogen社)を用いて、HCVのE1タンパク質及びE2タンパク質をその表面に発現した中空粒子(HCV-VLP)を作製することができる。
実施例7の(1)に記載の方法で、J6CF株由来のE1タンパク質及びE2タンパク質(組換えタンパク質)をそれぞれ作製した。
96ウェルプレート(Nunc社)に、組換えタンパク質であるE1タンパク質及びE2タンパク質の各50 ngを混合したものを50μL/ウェルとなるようにPBS(-)で希釈して添加し、固相化した。同様に、「a.HCV様粒子(HCV-VLP)の作製」において作製したHCV-VLPを500ng/ウェルとなるように添加し、4℃で一昼夜インキュベーションして固相化した。抗原溶液をデカントで除去し、milliQ水で5倍希釈したブロッキングワン(ナカライテスク社)を200μL/ウェルとなるように添加して、室温で1時間インキュベーションすることでブロッキングを行った。ブロッキング溶液をデカントで除去し、0.05%(v/v)Tween 20(Sigma社)を含むPBS(-)(以下、「洗浄液」とする)で2回洗浄した。洗浄液で希釈したモノクローナル抗体溶液(P18-9E又はP19-7D)を50μL/ウェルとなるように添加した。陰性対照のウェルには洗浄液のみを50μL/ウェルとなるように添加した。室温で約1.5時間インキュベーションした後、洗浄液で3回洗浄した。次に、洗浄液で3,000倍に希釈したHRP標識抗マウスIgG抗体(アマシャム社)を50μL/ウェルとなるように添加し、室温で1時間インキュベーションした後、洗浄液で4回洗浄した。ペルオキシダーゼ発色キット(スミロン社)の製品の添付文書に従って発色液を調製し、50μL/ウェルとなるように添加した。室温で15分間インキュベーションした後、キットに同梱の反応停止液を50μL/ウェルとなるように添加した。その後、マイクロプレートリーダー(Model 680, Bio-Rad社)で450 nmの吸光度を測定した。各ウェルの吸光度より陰性対照のウェルの吸光度を減算し、得られた値を各溶液の反応値とした。
(a)protein A/Gの固定化
表面プラズモン共鳴測定装置Biacore S51(GE社)用センサーチップのうち、Series S sensor chip CM-5(GE社)をBiacore S51に装着し、チップ上に超純水にて1×に希釈したアッセイバッファーであるHBS-EP(GE社)を30μL/minの流速で流した状態にした。内部温度を25℃に設定した。アミンカップリングキット(GE社)中のEDC(N-ethyl-N'-(3-dimethylaminopropyl)carbodiimide hydorochloride)及びNHS(N-hydroxysuccinimide)を、それぞれ100mM及び400mMとなるよう調製した。Acetate 4.0(GE社)にて50μg/mLのprotein A/G(Pierce社)を調製した。調製したEDC、NHS及び同キット中のEthanolamine溶液を用いてprotein A/Gをアミンカップリング法にて固定化した。一連の固定化操作は10μL/minの流速で行い、protein A/Gの添加時間は10分間に設定した。これにより同一フローセルのスポット1、2双方にprotein A/Gをほぼ等量固定化した。
HBS-EPにて20μg/mLに調製した、精製モノクローナル抗体P18-9E及びmouse IgG(Sigma社)を10μL/min の流速で8分間上記のチップ上に流した。Protein A/Gとそれぞれの抗体のFc部位の親和性を利用して、チップ上のスポット1にモノクローナル抗体P18-9E、スポット2にmouse IgGをキャプチャーした。
HBS-EPにて30、100、300μg/mLに調製したHCV-VLPを10μL/minの流速で上記のチップ上にそれぞれ3分間流し、表面プラズモン共鳴リアルタイム計測により、VLPの結合反応をモニターした後、10分間HBS-EPを流し解離反応を測定した。
解析にはS51 Evaluationを用いた。測定値は共鳴レスポンスの単位であるRU(Resonance Unit)で表される。Mouse IgGに対するHCV-VLPの反応を非特異吸着として、モノクローナル抗体P18-9Eに対する反応から差し引くことで補正を行った。
その結果、モノクローナル抗体P18-9Eに対して、VLP濃度依存的かつ抗原抗体反応様の緩やかな結合及び解離が観察された。この結果からも、モノクローナル抗体P18-9Eは、HCV-VLPに結合し、HCV粒子表面のE1タンパク質及びE2タンパク質からなる複合体であるエンベロープ立体構造を認識することが示唆された。

- C型肝炎ウイルス粒子のE1タンパク質及びE2タンパク質からなる複合体の立体構造をエピトープとして認識し、C型肝炎ウイルスに対して感染阻害活性を有する、抗C型肝炎ウイルス抗体であって、
以下の(a)又は(b)である前記抗体。
(a)重鎖可変領域が配列表の配列番号18、20及び22に示されるアミノ酸配列を含む相補性決定領域を含み、かつ、軽鎖可変領域が配列番号25、27及び29に示されるアミノ酸配列を含む相補性決定領域を含む抗体
(b)重鎖可変領域が配列表の配列番号32、34及び36に示されるアミノ酸配列を含む相補性決定領域を含み、かつ、軽鎖可変領域が配列番号39、41及び43に示されるアミノ酸配列を含む相補性決定領域を含む抗体 - 前記E1タンパク質及びE2タンパク質のアミノ酸配列が、それぞれ配列表の配列番号7及び配列番号8で示されるアミノ酸配列を含む、請求項1に記載の抗C型肝炎ウイルス抗体。
- 前記C型肝炎ウイルス粒子は、C型肝炎ウイルスのJ6CF株及びJFH-1株のゲノムの一部を連結して構成されるC型肝炎ウイルスキメラゲノムから産生され、前記C型肝炎ウイルスキメラゲノムは、以下の(i)又は(ii)である、請求項1又は2記載の抗C型肝炎ウイルス抗体。
(i)J6CF株由来の5’非翻訳領域、coreタンパク質コード配列、E1タンパク質コード配列、E2タンパク質コード配列及びp7タンパク質コード配列、並びにJFH-1株由来のNS2タンパク質コード配列、NS3タンパク質コード配列、NS4Aタンパク質コード配列、NS4Bタンパク質コード配列、NS5Aタンパク質コード配列、NS5Bタンパク質コード配列及び3’非翻訳領域を5’側から順番に連結したC型肝炎ウイルスキメラゲノム
(ii)J6CF株由来の5’非翻訳領域、coreタンパク質コード配列、E1タンパク質コード配列、E2タンパク質コード配列、p7タンパク質コード配列及びNS2タンパク質コード領域のN末端から16位のアミノ酸残基までのアミノ酸配列をコードする配列、並びにJFH-1株由来のNS2タンパク質コード領域のN末端から17位のアミノ酸残基からC末端のアミノ酸残基までをコードする配列、NS3タンパク質コード配列、NS4Aタンパク質コード配列、NS4Bタンパク質コード配列、NS5Aタンパク質コード配列、NS5Bタンパク質コード配列及び3’非翻訳領域を5’側から順番に連結したC型肝炎ウイルスキメラゲノム - 前記C型肝炎ウイルスキメラゲノムは、配列表の配列番号2に示される塩基配列中のチミン(T)をウラシル(U)に読み替えた塩基配列からなる、請求項3記載の抗C型肝炎ウイルス抗体。
- 前記感染阻害活性は、C型肝炎ウイルス粒子の宿主細胞表面への結合を阻害する活性である、請求項1〜4のいずれか一項記載の抗C型肝炎ウイルス抗体。
- 寄託番号がFERM BP-11263であるハイブリドーマ細胞株により産生される、請求項1〜5のいずれか一項記載の抗C型肝炎ウイルス抗体。
- 寄託番号がFERM BP-11264であるハイブリドーマ細胞株により産生される、請求項1〜5のいずれか一項記載の抗C型肝炎ウイルス抗体。
- ヒト化抗体である、請求項1〜5のいずれか一項記載の抗C型肝炎ウイルス抗体。
- 寄託番号がFERM BP-11263である、ハイブリドーマ細胞株。
- 寄託番号がFERM BP-11264である、ハイブリドーマ細胞株。
- 請求項1〜8のいずれか一項記載の抗C型肝炎ウイルス抗体を有効成分とするC型肝炎ウイルスの感染阻害剤。
