アルギナーゼは金属活性化型の水酸化物イオンを含有するマンガン金属酵素であり、この水酸化物イオンは加水分解又は水和反応を触媒する金属酵素での重要な求核体である。アルギナーゼは自然に存在するアルギニンをオルニチン及び尿素へと変換する。この酵素は細菌及びヒトを包含する多くの生きている生物中に存在する(Jenkinson他、1996年、Comp Biochem Physiol B Biochem Mol Biol、114:107〜32頁)。
アルギナーゼは治療薬として用いられ、様々な適応症のために非経口的に投与され得る。しかしながら非経口的に投与されたアルギナーゼはタンパク質であり、免疫原性があって薬理学的半減期は短いものであり得る。その結果として、患者の中でタンパク質を治療的に有用な血中レベルに達させるのは難しいことがある。これらの問題はタンパク質をポリエチレングリコール(PEG)などのポリマーにコンジュゲートすることにより克服し得る。
アミノ酸欠如治療はいくらかのがんの治療に有効な手段である。正常細胞はアルギニンを要求しないが、多くのがん細胞系はこのアミノ酸に対して栄養要求性がある。多種の証拠は、アルギニン分解酵素による又はアルギニン欠乏培地を用いる試験管内でのアルギニン枯渇が広い範囲のがん細胞の迅速な破壊を導くことを示している(Scott他、2000年、Br J Cancer、83:800〜10頁)。しかしタンパク質である酵素の直接的な使用は免疫原性、抗原性及び短い循環半減期の問題がある。
ウイルス感染症は毎年数百万人にも上る死亡の主たる原因の1つで、肝炎ウイルス及びヒト免疫不全ウイルス(HIV)を包含するいくつかのウイルスに直接的に起因し得るものである。しかしながら、現在の抗ウイルス治療にはいくつかの問題がある。第一に、有効な抗ウイルス薬は比較的少ない。現存する抗ウイルス薬の多くは有害な又は望ましくない副作用を引き起こす。最も有効な治療(ワクチン接種など)は単一のウイルス株のみに対して高度に特異的である。しばしばウイルスは薬剤又はワクチンのいずれかに対して耐性となるように変異を起こす。従来技術に関連した問題を持たないウイルス複製を阻害する方法への需要がある。
後天性免疫不全症候群(AIDS)は致命的な疾病であり、その報告症例は過去数年の間で劇的に増加した。AIDSウイルスは1983年に初めて同定された。AIDSウイルスはいくつかの名称及び頭字語により知られている。AIDSウイルスは三番目に公知になったT−リンパ指向性のウイルス(HTLV−III)で、免疫系の細胞内で複製する能力を有し、重度の細胞破壊を引き起こす。AIDSウイルスはレトロウイルスで、複製の際に逆転写酵素を用いるウイルスである。2つの別個のHIVファミリー、即ちHIV−1及びHIV−2が今までに記述されている。本明細書の中で頭字語“HIV”はヒト免疫不全ウイルスを一般的に呼ぶのに用いられる。HIV複製はアルギニン依存的と思われており、したがってアルギニンの枯渇はHIV複製を阻害する。
ヒトアルギナーゼIの遺伝子配列は図1a(配列番号1)に示される。6×Hisタグ付加ヒトアルギナーゼI(HAI)の遺伝子はポリメラーゼ連鎖反応(PCR)によりpAED4/HAIプラスミドから生成され、以下のオリゴヌクレオチドを用いることで5’末端においてNdeI認識部位、3’末端においてBamHI認識部位が生成された。
PCR産物はNdeI及びBamHIで切断され、pET3a発現プラスミドベクター(Strategene)中にサブクローニングされた。
バチルス・カルドベロックスアルギナーゼの遺伝子配列は図1c(配列番号3)に示される。6×Hisタグ付加バチルス・カルドベロックスアルギナーゼ(BCA)の遺伝子はpUC57/BCAプラスミドからNdeI及びBamHI制限酵素を用いて切り出された。挿入断片はpET3a発現プラスミドベクター(Strategene)中にサブクローニングされた。
プラスミドpET3a/HAIはQuikChange(登録商標) site−directed mutagenesisキット(Strategene)に従って部位指定的な変異誘発のための鋳型として用いられた。Cys168及びCys303残基に対するコドンは、以下の変異誘発プライマー(それぞれ配列番号19、20、21及び22)を用いることでそれぞれSer168及びSer303に対するコドンに変異した。

6×Hisタグ付加ヒトアルギナーゼIをコードする変異アルギナーゼ遺伝子を含有するプラスミドを内部に有する大腸菌BL21−DE3は37℃で一晩、80μg/mLのアンピシリンを含有するLB培地中で培養された。接種菌液は1:25に希釈されてOD600が約0.8になるまで振とうフラスコ中で培養されるか、又は接種菌液は1:10に希釈されてOD600が約15になるまで発酵槽中で培養された。細胞は次いで0.4mMのIPTGで4時間誘導された。細菌細胞は遠心分離により回収され、50mM Tris、0.1M NaCl、10mM MnCl2、pH7.4中に再懸濁され、高圧ホモゲナイゼーションにより破砕された。
図5aは6×Hisタグ付加ヒトアルギナーゼI変異体の単一鎖mPEG−マレイミド(20kDa)でのCys45特異的なモノペグ化についてのコンジュゲーション手順を示し、これは“HAI−PEG20”と呼ばれる。マレイミドの二重結合はスルフヒドリル基とアルキル化反応を起こして、安定なチオエーテル結合を形成する。図5bは6×Hisタグ付加バチルス・カルドベロックスアルギナーゼ変異体の単一鎖mPEG−マレイミド(20kDa)でのCys161特異的なモノペグ化についてのコンジュゲーション手順を示し、これは“BCA−PEG20”と呼ばれる。6×Hisタグ付加アルギナーゼ1gは0.02Mリン酸ナトリウム、0.5M NaCl、pH7.4中で、Millipore Tangential Flow Filtrationシステム(500mL)を用いて10K(カットオフ)膜(Millipore)により透析濾過された。アルギナーゼ濃度は最終的に約2mg/mLに希釈された。還元剤のトリス(2−カルボキシエチル)ホスフィン、TCEPが1モルのアルギナーゼに対してモル量で10モル超、還元のために添加され、溶液は室温で4時間、穏やかに撹拌された。1モルのアルギナーゼに対してモル量で20モル超のmPEG−マレイミド又はmPEG−MAL(20kDa)(Sunbright)が還元されたアルギナーゼに対して添加され、4℃で一晩撹拌された。
アルギナーゼ遺伝子を含有する大腸菌BL21−DE3株は−80℃で保存された。回分発酵及び流加回分発酵のための種菌液を調製するため、前述の株の凍結ストック100μLが80mLの発酵培地を含有する250mLフラスコ中に移された。細菌培養液は37℃、pH7.0でオービタルシェーカーの中で回転数250rpmで培養された。培養はOD600nmが5.5〜6.0に達したおよそ8〜10時間時で停止された。種菌液12mL(1%)は1200mLのオートクレーブされた強化発酵培地を含有する2L発酵槽中に導入された。回分発酵は温度37℃で行われた。pHは水酸化ナトリウム及び塩酸を添加することにより7.0に維持された。溶存酸素レベルは空気を1〜4L/分で導入すること及び発酵槽の撹拌速度を300〜1200rpmに調整することにより、30%より高い空気飽和度に調節された。イソプロピル−β−D−チオガラクト−P(IPTG)100mMは、バチルス・カルドベロックスアルギナーゼ(BCA)タンパク質発現の誘導物質であるが、OD600nmがおよそ11.0であった5時間時に発酵ブロス中に終濃度0.5mMで導入された。IPTG誘導の後、発酵はOD600nmがおよそ16.4であった9時間時まで続けられた。発酵細胞はBCAの分離及び精製のため、IPTG誘導後4時間時に収集された。前述の株はおよそ105mg/L発酵培地の量の活性型BCAを生産した。発酵の時間経過は図6a1にプロットされる。温度、撹拌速度、pH及び溶存酸素値などのパラメーターの変化を示す、この回分発酵の履歴プロットは図6b1に表される。
高細胞密度培養での流加回分発酵は37℃、pH7.0で行われ、溶存酸素は全発酵プロセスの間、30%より高い空気飽和度に維持された。種菌液を調製するための手順は上記の回分発酵のそれと同様であった。発酵は初期には回分培養戦略で、5mL(1%)の種菌液を500mLのオートクレーブされた強化発酵培地を含有する2L発酵槽中に導入することにより開始された。溶存酸素は回分培養期間中の増殖相の間、30%前後の空気飽和度に徐々に減少した。いったん溶存酸素レベルが80%より高く上昇したら、これは炭素源の枯渇を表しており、PO2 stat流加回分戦略が強化培地供給を追加して開始された。この戦略の中で、供給速度は60%を下回る溶存酸素レベルを維持するように調製され、これは最小限ではあるが適切な量の炭素源を発酵プロセスの間、提供した。イソプロピル−β−D−チオガラクト−P(IPTG)100mMはOD600nmがおよそ100であった18時間時に発酵ブロス中に終濃度0.5mMで導入された。IPTG誘導の後、発酵はOD600nmがおよそ186.8であった28時間時まで続けられた。発酵細胞はBCAの分離及び精製のため、IPTG誘導後10時間時に収集された。前述の株は発酵培地1リットルあたりおよそ1489.6mg量の活性型BCAを生産し、これは異なる型のアルギナーゼについての他の全ての報告された収率より高い。発酵の時間経過は図6a2にプロットされる。温度、撹拌速度、pH及び溶存酸素値などのパラメーターの変化を示す、この回分発酵の履歴プロットは図6b2に表される。
下の表1は回分発酵と流加回分発酵との結果を比較している。比較は、培養液のOD600、細胞乾燥重量及び培養液1リットルあたりのBCA収率の点で流加回分発酵が回分操作より大きく優れていることを実証している。
アフィニティーニッケルイオンカラムクロマトグラフィーは6×Hisタグ付加部位指定的ペグ化アルギナーゼをmPEG−MAL(20kDa)から分離するのに以下に述べられるように用いられた。コンジュゲーションの最終産物はバッファーA(0.02Mリン酸ナトリウム、0.5M NaCl、pH7.4)で平衡化されたキレート化FFセファロース(GE Healthcare)カラム(5.0cm×9cm、総体積176mL)上にロードされた。カラムはカラム体積の5倍のバッファーAで洗浄され、遊離mPEG−MAL(20kDa)が除去された。ペグ化アルギナーゼは30%から100%までの塩勾配の、カラム体積の5倍のバッファーB(0.02Mリン酸ナトリウム、0.5M NaCl、0.5Mイミダゾール、pH7.4)を用いて溶出された。溶出液のタンパク質含量は280nmの波長でモニターされた。カラムは流速20mL/分で溶出され、ペグ化アルギナーゼ画分が回収された。プールされた画分はPBSバッファー(Gibco)中で透析濾過され、4〜6mg/mLに濃縮された。動物試験の前に、タンパク質薬剤の中のエンドトキシンはQ−filter(Sartoris)を用いて除去された。
Cys45ペグ化ヒトアルギナーゼI及びCys161ペグ化バチルス・カルドベロックスアルギナーゼの試験管内での細胞毒性は、異なるヒトがん細胞(黒色腫、肝細胞癌、胃腺癌、結腸直腸腺癌、膵臓癌、膵臓腺癌及びT細胞白血病)中での標準MTTアッセイにより試験された。
Cys45ペグ化ヒトアルギナーゼI及びCys161ペグ化バチルス・カルドベロックスアルギナーゼの薬力はBALB/c正常マウスを用いて試験された。試験は薬物動態試験(後述)と一緒に行われた。それ故、プロトコールは同じままである。再度、回収された血液サンプルは速やかに13,200rpmで5分間遠心分離され、血漿層が回収されてAmino Acid Analyzer(Biochrom30、Biochrom Ltd.、England)を用いたさらなる解析が行われた。
ペグ化されていないヒトアルギナーゼI(HAI)及びCys45ペグ化ヒトアルギナーゼI(HAI−PEG20)の肝臓がんに対する生体内での抗腫瘍有効性が次いで試験された。
乳がんに対するCys161ペグ化バチルスアルギナーゼ(BCA−PEG20)の生体内での抗腫瘍有効性が次に決定された。
胸腺欠損のヌードBALB/cマウス(6〜8週齢)は滅菌条件下、12時間の明暗周期で、オートクレーブされた飼料を自由摂取で供給されて飼育された。マウスは実験開始前に少なくとも1週間馴化された。各ヌードマウスはA549ヒト肺がん細胞5×106個をマトリゲル増殖補助剤と共に右の腋窩に皮下注入された。触知可能な直径約5mmの腫瘍が増殖したら、マウスは3つの異なる群にランダムに分けられた(表5)。薬剤又は対照溶媒(PBS)は0日目から始まって週1回、腹腔内に注入された。腫瘍寸法(L:長径、W:それに直交する直径)及び体重は毎月曜日、水曜日及び金曜日にノギスで測定された。腫瘍体積は式(1/2×L×W2)で計算され、腫瘍体積の増加倍率(相対的腫瘍体積)が0日目を参照して計算された。
結腸直腸がんに対するペグ化されていない(BCA)及びCys161ペグ化バチルス・カルドベロックスアルギナーゼ(BCA−PEG20)の生体内での抗腫瘍有効性は次のように決定された。
マウス転移乳がん細胞系(4T1)の細胞1×105個は6〜8週齢の野生型BALB/cマウスの4番鼠径部乳房脂肪パッド内に同所性に注入された。腫瘍が平均5mmに達した時、マウスは2つの異なる処理群に分割された(表7)。BCA−PEG20(250U/マウス)又は対照溶媒(PBS)は0日目から始まって週に2回、腹腔内に注入された。体重は毎週測定された。3週間後、マウスは屠殺されて肺転移について分析された。肺転移の数はPBSで洗浄した後に解剖顕微鏡下で数えられた。
Cys45ペグ化ヒトアルギナーゼI(HAI−PEG20)のヒト免疫不全ウイルス(HIV)に対する50%阻害濃度(IC50)は、HIVに対するその効果の尺度として決定された。
異なるアルギニン枯渇酵素の抗がん効力に対する試験管内でのがん細胞培養試験は様々ながんのタイプについて行われた。
野生型アルギナーゼの金属補因子はマンガン(Mn2+)である。驚くべきことに、マンガンをコバルトで置き換えることで酵素の活性は劇的に亢進されることが本発明により発見された。バチルス・カルドベロックスアルギナーゼ(BCA)又はヒトアルギナーゼI(HAI)のいずれかが先に述べられたように発現された。精製方法は、10mMの金属イオン(CoSO4又はMnSO4)がニッケルアフィニティークロマトグラフィー前の添加の代わりにニッケルアフィニティークロマトグラフィーからの精製タンパク質溶出液に添加されたこと以外は前述と同じであった。アルギナーゼ酵素を含有する溶出された画分は10mMの金属と50〜55℃で15分間インキュベートされた後、0.45μmのシリンジフィルターを通して濾過が行われた。次いで溶液は限外濾過により保存バッファーと交換された。
驚くべきことに、BCAの20位は酵素活性を改善するために他のアミノ酸で置き換えられ得ることが本発明により発見された。野生型配列中の20番目のアミノ酸残基であるバリンはプロリン(又は例えばセリン若しくはグリシンといった他の任意のアミノ酸で、BCAの特異的活性を改善するもの)で部位指定的な変異誘発(例えばコドンGTT[バリン]をCCG[プロリン]に)により置き換えられた。変異体遺伝子は詳細な試験のためにクローニングされ、発現されて精製された。代表的なかかる変異体酵素はバリンをプロリンで置換することで作られたが、これは“BCA変異体V20P”又は“bcArg V20P変異体”と呼ばれる。BCA変異体V20P及びMn2+又はCo2+を伴うBCAの定常状態反応速度はリン酸ナトリウムバッファー中でpH7.4、25℃で測定され、図17に示された。Mn2+を伴うBCA変異体V20P(BCA変異体V20P Mn2+)及びCo2+を伴うBCA変異体V20P(BCA変異体V20P Co2+)のKm値はそれぞれおよそ1.29mM及び0.18mMである。Mn2+を伴うBCA(BCAWTMn2+)のKmはおよそ3.2mMである。それ故、補因子としてCo2+を伴うBCA変異体V20P[Km=0.18mM]は、アルギニンを枯渇させるのにBCA(BCAWTMn2+)[Km=3.2mM]よりも一層効率的な薬剤である。

Claims (6)
- 遺伝的に改変したヒトアルギナーゼに共有結合的に結合したPEG部分を有するPEG−アルギナーゼコンジュゲートを含む、アルギニン依存性疾患を治療するための医薬組成物であって、
該ヒトアルギナーゼの遺伝的改変は該PEG部分を共有結合的に結合させるための単一のアミノ酸位置を有するアミノ酸配列を形成するものであり、
該遺伝的に改変したヒトアルギナーゼのアミノ酸位置は、該PEG−アルギナーゼコンジュゲートの血清循環半減期を純粋の非改変ヒトアルギナーゼの血清循環半減期を越えて増加させ、該PEG−アルギナーゼコンジュゲートの免疫原性を純粋の非改変ヒトアルギナーゼの免疫原性を下回るように減少させるように配置させるものであり、
該PEG部分の結合位置は、結果として生じた折りたたまった遺伝的に改変したPEG−コンジュゲート化したヒトアルギナーゼタンパク質内の活性部位から該PEG結合が活性部位に干渉しない程度に十分に遠いものであり、
該遺伝的に改変したヒトアルギナーゼが配列番号6で示されるアミノ酸配列からなり、該PEG部分の結合位置がCys 45 である、上記医薬組成物。 - 遺伝的に改変したバチルス・カルドベロックス(Bacillus caldovelox)アルギナーゼに共有結合的に結合したPEG部分を有するPEG−アルギナーゼコンジュゲートを含む、アルギニン依存性疾患を治療するための医薬組成物であって、
該バチルス・カルドベロックスアルギナーゼの遺伝的改変は該PEG部分を共有結合的に結合させるための単一のアミノ酸位置を有するアミノ酸配列を形成するものであり、
該遺伝的に改変したバチルス・カルドベロックスアルギナーゼのアミノ酸位置は、該PEG−アルギナーゼコンジュゲートの血清循環半減期を純粋の非改変バチルス・カルドベロックスアルギナーゼの血清循環半減期を越えて増加させ、該PEG−アルギナーゼコンジュゲートの免疫原性を純粋の非改変バチルス・カルドベロックスアルギナーゼの免疫原性を下回るように減少させるように配置させるものであり、
該PEG部分の結合位置は、結果として生じた折りたたまった遺伝的に改変したPEG−コンジュゲート化したバチルス・カルドベロックスアルギナーゼタンパク質内の活性部位から該PEG結合が活性部位に干渉しない程度に十分に遠いものであり、
該遺伝的に改変したバチルス・カルドベロックスアルギナーゼが配列番号8で示されるアミノ酸配列からなり、該PEG部分の結合位置がCys 161 である、上記医薬組成物。 - 前記PEG部分と前記遺伝的に改変したヒトアルギナーゼ又は遺伝的に改変したバチルス・カルドベロックスアルギナーゼの比が1である、請求項1又は2に記載の医薬組成物。
- 前記PEG部分が一本鎖又は分岐鎖ポリエチレングリコールである、請求項1又は2に記載の医薬組成物。
- 薬学的に許容される担体、賦形剤又は補助剤をさらに含む、請求項1又は2に記載の医薬組成物。
- 前記アルギニン依存性疾患が、アルギニン依存性の癌又はHIV、C型肝炎ウイルス及びB型肝炎ウイルスからなる群より選択されるウイルスによる感染症である、請求項1又は2に記載の医薬組成物。
