Description
Mは水素又は1当量の陽イオンを表し、
R1は同一又は相異なるものであって、1以上のヒドロキシル基で任意に置換された直鎖又は枝分れC1〜C6−アルキル基或いは−(CH2)n−X−R2基(式中、nは1、2又は3であり、XはO又はSであり、R2は1以上のヒドロキシル基で任意に置換された直鎖又は枝分れC1〜C4−アルキル基である。)を表す。
Ardenkjaer−Larsen et al(2003)[Increase in signal−to−noise ratio of >10,000 times in liquid−state NMR.Proc.Natl.Acad.Sci.U.S.A.100,10158−10163]によって記載されたようにして、15mM OX063トリチルラジカル(Oxford Instruments社)を含む水に溶解した2−[13C]−フルクトース(Isotec社、マイアミズバーグ、米国オハイオ州)の4.0M溶液をHypersense装置(Oxford Instruments社)で過分極させた。凍結試料を1×リン酸塩緩衝食塩水(PBS)に溶解したところ、得られたpHは7.6であり、それを直ちに10mmのNMR管に移した。
フルクトースからフルクトース−6−リン酸への酵素転化のNMR試験のため、15mM ATP、50mM TRIS及び13mM MgCl2の存在下で過分極[2−13C]−フルクトースを400Uのヘキソキナーゼ(Sigma Aldrich社)と反応させた。輸送及び代謝に関する標識及び機構を下記に示す(図1)が、この酵素試験では、輸送要素を除去して酵素活性を独立に測定した。同様な1組の実験パラメーターを使用しながら、天然存在比炭素スペクトルを用いてフルクトース−6−リン酸に対応するピークを同定した。
C2フルクトース炭素に関して計算したT1を、11.7T及び3Tの両方について表1に示す。対象炭素のT1緩和は、磁気強度の減少に伴って僅かに減少している。フルクトースの異性体間におけるT1の有意差は存在しなかった。%分極はフルクトースの異性体に関して同様な値を示し、平均は12%程度であった。フルクトースに関して5.9〜7.8のpH範囲で観察したところ、T1のpH依存性は存在しなかった。
