Description
(a)ミルク試料に存在するアンギオゲニンが捕捉剤と相互作用してアンギオゲニン−捕捉剤複合体を生成するように、ミルク試料をアンギオゲニンと相互作用する捕捉剤に接触させる工程、
(b)前記複合体をミルク試料から分離する工程、
(c)前記複合体中の捕捉剤からアンギオゲニンを遊離させる工程、及び
(d)工程(c)からアンギオゲニンを回収し、アンギオゲニン濃縮生成物を得る工程を含む方法を提供する。
第一の態様の一実施形態においては、前記捕捉剤は支持体に固定化されている。
第一の態様の更なる実施形態においては、前記捕捉剤は抗体である。
(a)アンギオゲニンと相互作用する抗体を固定化した支持体にミルク試料を添加し、ここでミルク試料に存在するアンギオゲニンが支持体上の抗体と相互作用してアンギオゲニン−抗体複合体を生成する工程、
(b)前記抗体と相互作用しないミルク試料に存在する成分を前記支持体から洗い落すことにより、ミルク試料から前記複合体を分離する工程、
(c)前記複合体中の抗体からアンギオゲニンを遊離させる工程、及び
(d)工程(c)からアンギオゲニンを回収し、アンギオゲニン濃縮生成物を得る工程を含む方法を提供する。
第一又は第二の態様の一実施形態においては、前記方法はイムノアフィニティークロマトグラフィーを含む。
(a)液相のミルク試料を、前記ミルク試料の成分をそのサイズに基づいて分離可能な第二の相に添加する工程、及び
(b)前記ミルク試料の他の成分から分離されたアンギオゲニンを回収することによりアンギオゲニン濃縮生成物を得る工程を含み、
工程(a)に先立ち、前記ミルク試料をレンネット処理又は酸沈澱に付さないか、或いは前記ミルク試料がホエイ又はホエイ分画でない方法を提供する。
(a)液相のミルク試料を、前記ミルク試料の成分をそのサイズに基づいて分画に分離可能な第二の相に添加する工程、及び
(b)これらのアンギオゲニンを含む分画を同定し、該分画を回収することによってアンギオゲニン濃縮生成物を得る工程を含む方法を提供する。
(a)流動する水性ミルク試料にミルク流を横断する方向に電場を印加する工程、
(b)電場が印加されたミルク流の分画を回収する工程、
(c)これらのアンギオゲニンに関して濃縮された分画を同定し、該分画を回収することによってアンギオゲニン濃縮生成物を得る工程を含む方法を提供する。
試料可溶化溶液(SSS):7Mウレア、2Mチオウレア及び1%3−(4−ヘプチル)フェニル3−ヒドロキシプロピルジメチルアンモニオプロパンスルホネ−ト(C7BzO)。
Claims (10)
- ミルク試料からアンギオゲニン濃縮生成物を得る方法であって、
(III)
(a)液相のミルク試料を、前記ミルク試料の成分をそのサイズに基づいて分離可能な第二の相に添加する工程、及び
(b)前記ミルク試料の他の成分から分離されたアンギオゲニンを回収することにより、アンギオゲニン濃縮生成物を得る工程を含む方法;
(IV)
(a)液相のミルク試料を、前記ミルク試料の成分をそのサイズに基づいて分画に分離可能な第二の相に添加する工程、及び
(b)これらアンギオゲニンを含む分画を同定し、該分画を回収することによってアンギオゲニン富化産物を得る工程を含む方法;又は、
(V)
(a)流れている水性ミルク試料にミルク流を横断する方向に電場を印加する工程、
(b)電場が印加されたミルク流の分画を回収する工程、及び
(c)これらアンギオゲニンに関して富化された分画を同定し、該分画を回収することによってアンギオゲニン富化産物を得る工程を含む方法であり、
(III)、(IV)及び(V)の方法中の工程(a)に先立ち、前記ミルク試料を少なくとも1分間70℃超〜75℃に加熱して、試料中でアンギオゲニンよりも低い温度で変性する他のタンパク質とラクトペルオキシダーゼの量を低減させる方法。 - 方法(III)において、前記第二の相は半透過性相である、請求項1に記載の方法。
- 方法(III)において、前記ミルク試料はシリンジ、圧縮ガス、ポンプ、遠心力又はそれらの組み合わせによって加えられる力によって前記半透過性相を強制透過させられる、請求項1に記載の方法。
- 方法(III)において、前記第二の相は半透過性膜である、請求項1に記載の方法。
- 方法(IV)において、前記第二の相はサイズ排除樹脂である、請求項1に記載の方法。
- 方法(V)において、前記方法は変性条件下で行い、及び又は前記水性ミルク流はpH勾配をもたらす緩衝液媒体中で行う、請求項1に記載の方法。
- 前記勾配はpH8〜11の範囲である、請求項6に記載の方法。
- さらに前記アンギオゲニン濃縮生成物を一以上のアンギオゲニン濃縮工程に付す工程を含む、請求項1〜7のいずれかに記載の方法。
- 前記一以上のアンギオゲニン濃縮工程は、カチオン交換クロマトグラフィー、フリーフロー電気泳動を含む電気泳動、サイズ排除クロマトグラフィー及び限外濾過からなる群から選択される、請求項8に記載の方法。
- 前記ミルク試料は全乳、スキムミルク、バターミルク、ホエイ、ホエイ分画及び初乳から選択される、請求項1〜9のいずれかに記載の方法。
