Description
2段階からなる自動化方法の例は、単なる濾過、遠心分離法又は磁気分離単独ではなく、分離に続いて濾過を組合せるものであり、遠心分離機の試料ホルダの物理的寸法が固定されていることにより、遠心分離法に比べて、(1)総有核細胞数(TNC)回収率の増加、(2)RBC除去の増加及び(3)ある範囲の試料体積(例えば50〜300mLの血液)を扱う際の柔軟性の向上を実現する。サイズの固定された遠心分離機とは異なり、システム及び方法で使用されるフィルタは、試料体積に従って拡大縮小することができる。
材料:ヒトの末梢血を実験に使用した。この例で使用したデキストランT500は、Pharmacosmos社(デンマーク)から入手した。クエン酸ナトリウム二水和物はJ T Baker社から入手した。コハク酸ナトリウムはSigma社(米国ミズーリ州セントルイス)から入手した。
赤血球凝集の効率を、様々な濃度の無毒性促進剤の存在下で生体外にて測定した。1.5%デキストランT500と共に、促進剤を加えずにインキュベートした血液試料を対照とする。対照試料は、血液試料2.0mlと3%デキストランT500含有PBS2.0mlを混合することによって調製した。1.5%デキストランT500並びに促進剤として12.5mM、25mM、50mM、75mM及び100mMのクエン酸ナトリウムを含有する血液試料を試験試料とした。この試験試料は、血液試料2.0mlと、3.0%デキストランT500並びに25mM、50mM、100mM及び150mMのクエン酸ナトリウムをそれぞれ含有するPBS2.0mlを混合することによって調製し、各試験試料のデキストランの最終濃度は1.5%になり、それぞれの試験試料のクエン酸ナトリウムの最終濃度は12.5mM、25mM、50mM及び75mMとなった。対照及び試験セット用の試料を室温で20分インキュベートした。
Claims (10)
- 赤血球細胞及び有核細胞を含む生物材料を処理するためのシステムであって、
生物材料を収容して2種以上の異なる成分材料の層に分離させることのできる容器であって、成分材料の層の1つが赤血球細胞の層である、容器と、
前記容器から赤血球細胞の層を除去する抽出装置であって、前記容器内で操作中に赤血球細胞の層が沈降したレベルに位置する末端を有する収集ラインを備える抽出装置と、
残りの成分材料から有核細胞を分離するための濾過装置と、
前記容器と前記濾過装置の間で1種以上の成分材料を輸送する導管と、
前記容器と前記導管との間に位置する弁と、
前記抽出装置による前記容器から赤血球細胞の層の除去を自動的に開始するとともに、赤血球細胞の除去後に、前記弁を自動的に開いて前記容器内に残存する1種以上の成分材料を前記導管を介して前記濾過装置に輸送するための制御装置と
を備えていて、当該システムが前記成分材料を前記濾過装置に再循環するように構成されている、システム。 - 前記容器が、前記成分材料を沈降層に分離し、前記沈降層のうち1つ以上を吸引するのに適合している、請求項1記載のシステム。
- 所定の量の薬剤を前記薬剤受け器から選択的に除去し、前記所定の量の薬剤を前記容器に導入するための、薬剤受け器と流体連通する弁をさらに備える、請求項1又は請求項2記載のシステム。
- 前記抽出装置が、さらに、前記材料と前記薬剤の混合を容易にするために、所定の量の材料を、前記薬剤が前もって導入されている前記容器から吸引し、次いで前記吸引された材料を前記容器に戻すのに適合している、請求項1乃至請求項3のいずれか1項記載のシステム。
- 前記容器内における前記成分材料の1種以上の位置又はレベルを判断するための検知装置をさらに備える、請求項1乃至請求項4のいずれか1項記載のシステム。
- 請求項1乃至請求項5のいずれか1項記載のシステムを用いて、赤血球細胞及び有核細胞を含む生物材料を処理するための方法であって、
前記容器内に生物材料を用意し、
前記容器内の前記材料に凝集剤を加えて、前記材料を2種以上の異なる成分材料の層であって、成分材料の層の1つが赤血球細胞の層である層に分離せしめ、
前記抽出装置の収集ラインを通して前記容器から赤血球細胞の層を抽出し、次いで
前記弁を自動的に開いて前記容器に残った前記成分材料の1種以上を濾過装置に自動的に輸送し、
前記成分材料を前記濾過装置に再循環し、
得られた目標保持液を目標保持液受け器に回収する
ことを含む方法。 - 前記容器内の前記成分材料の1種以上の存在を検知することをさらに含む、請求項6記載の方法。
- 設定値に達するまで、前記成分材料の1種以上が抽出される、請求項6又は請求項7記載の方法。
- 前記生物材料が全血、臍帯血又は骨髄を含む、請求項6乃至請求項8のいずれか1項記載の方法。
- 前記容器内の前記材料に凝集促進剤を加えることをさらに含む、請求項6乃至請求項9のいずれか1項記載の方法。
