Description
本発明の赤レタスの系図
以下の系図に従って、本発明のレタスを獲得した。すなわち、1986年に、品種ピッポ(Pippo)(Rijk Zwaan社;赤色)の植物体と、品種ブロンドマレシェール(Blonde Maraichere)(Caillard社;緑色)の植物体との間で交雑株を作製した。
本発明の赤レタスの遺伝解析
本明細書で使用される場合、遺伝子座(locus)(複数形:loci)は、ある遺伝子が位置する染色体上の特定位置と定義される(Griffiths AJF、Miller JH、Suzuki、DT、Lewontin RC、Gelbart,WM、「An introduction to genetic analysis.」、第6版、1996年、WH Freeman and Company社、米国ニューヨーク所在)。所与の植物体の赤色化の原因となっている遺伝子座の数は、この植物体と、緑色品種であるシャープシューター(Sharp Shooter)との交雑株の子孫を遺伝解析することによって確定できる。
レタスの色、アントシアニン、およびクロロフィルの測定
1.試料の調製および分析
以下の仕様を有する分光光度計UltrospecIII(Pharmacia社)を用いた。
−モノクロメーター:ホログラフィック回折格子(1200列/mm)を有するツェルニーターナー分光計
−波長精度:±1nm
−波長再現精度:±0.5nm
−検出器型:単一固体シリコンフォトダイオード
−バンド幅:5nm
赤色(アントシアニン)および緑色(クロロフィル)は生化学的方法で測定する。1つは総アントシアニンの尺度である523nmの吸光度を測定するためであり、1つは総クロロフィル(クロロフィルaおよびb)の尺度である665nmの吸光度を測定するためである2つの抽出物を調製する。
結球した成熟レタス植物を採取し、芯が残されるまで、外葉を除去する。レタスの芯は分析に用いる。植物体の頭頂における葉の小さな先端(最大10%)を除いて、収穫前に、芯葉が日光に直接さらされることがないようにするべきである。芯は、長さ1cm以上の葉を少なくとも10枚含有する。芯の最も古い葉は凹面であるべきである。
3グラムの粉末を、50ml管4本に量り入れる。2本の管をアントシアニンの分析に用い、2本の管をクロロフィルの分析に用いる。
