Description
・治療期間を通して、例えば、患者が睡眠中である夜間も含めて終日、治療効果を得るために望ましい血漿中濃度を保ち、患者に必要な一日の服用回数を減らすことで、患者の服用遵守を改良する。
・副作用を伴う場合にはピーク血漿中濃度を下げる。
・従来の急速放出システムによる連続投与の結果見られる血漿中濃度の変動を抑えることで、長期治療における副作用を抑える。
・薬剤が胃腸粘膜に対して局所的に作用する場合には、薬物送達システムが胃腸管運動により胃腸粘膜に送達されたとき、放出物を胃腸粘膜全体に広く空間的に分散させる。

を有する経口薬送達システムであって、
前記経口薬送達システムは、水性環境との接触により前記システムの予め選択された面の1つ以上から、完全または部分的に、前記剤皮が確実に除去され、さらに、前記面のうち少なくとも1つの面からは前記剤皮が除去されないよう設計される経口薬送達システム。
b.前記核を囲む剤皮と
を有する経口薬送達システムであって、
前記経口薬送達システムは被覆された錠剤形態であり、水性環境との接触により、前記錠剤の1つ以上の面から、完全または部分的に、前記剤皮が除去されるような特徴を有し、さらに、前記特徴は、剤皮が完全または部分的に除去されることが所望される前記錠剤の1つまたは複数の面のいずれでも選択することを可能とし、さらに、前記面のうち少なくとも1つの面からは前記剤皮が除去されないということである経口薬送達システム。
b.前記核を囲む剤皮と、
c.前記剤皮中の通路と
を含む経口薬送達システムであって、
前記経口薬送達システムは、前記システムが水性環境に接触した際に、部分的に、前記剤皮が確実に除去され、前記部分的に露出された面から有効成分が放出されるよう設計される経口薬送達システム。
b.前記核を囲む剤皮と
を有する経口薬送達システムであって、
前記経口薬送達システムは1つだけ面を有し、前記システムが水性環境に接触した際に、前記面から前記剤皮が部分的に除去されるよう設計される経口薬送達システム。
b.前記核を囲む剤皮と
を有する経口薬送達システムであって、
前記経口薬送達システムは少なくとも2つの面を有し、前記経口薬送達システムが水性環境に接触した際に、前記面のうちの1つの面から、前記剤皮が部分的に除去されるよう設計され、さらに、前記剤皮は最小面積を有する面とは異なる面から除去される経口薬送達システム。
b.前記核を囲む剤皮と
を有する経口薬送達システムを提供することにより、有効成分をその環境から単離する方法であって、
前記システムは、水性環境との接触により、前記剤皮が破裂し、前記有効成分が即座に、かつ急速に放出されるという特徴を有する方法。
(a)少なくとも1つの有効成分および製薬上許容可能な賦形剤を含む有効成分組成物を含む核と、
(b)前記核を囲む剤皮と
を有する経口薬送達システムであり、
前記経口薬送達システムは、水性環境との接触により、前記システムの予め選択された面の1つ以上から、完全または部分的に、前記剤皮が確実に除去され、さらに、前記面のうち少なくとも1つの面からは前記剤皮が除去されないよう設計される経口薬送達システムを提供する。
(a)少なくとも1つの有効成分および製薬上許容可能な賦形剤を含む有効成分組成物を含む核と、

を有する経口薬送達システムであって、
前記経口薬送達システムは被覆された錠剤形態であり、水性環境との接触により、前記錠剤の1つ以上の面から、完全または部分的に、前記剤皮が除去されるような特徴を有し、さらに、前記特徴は、剤皮が完全または部分的に除去されることが所望される、前記錠剤の1つまたは複数の面のいずれでも選択することを可能とし、さらに、前記面のうち少なくとも1つの面からは前記剤皮が除去されないというものである経口薬送達システムを提供する。
本発明の実施形態は、
(a)少なくとも1つの有効成分および製薬上許容可能な賦形剤を含む有効成分組成物を含む核と、
(b)前記核を囲む剤皮と
を有する経口薬送達システムであり、
前記経口薬送達システムは少なくとも2つの面を有し、前記経口薬送達システムが水性環境に接触した際に、前記面のうちの1つの面から剤皮が完全または部分的に除去されるよう設計され、さらに、前記剤皮は、最小面積を有する面とは異なる面から除去される経口薬送達システムを提供する。本発明の経口薬送達システムは、剤皮が除去される面について選択の幅がなく、一般的に面の下方領域を露出させる先行技術によるシステムより利点がある。
(a)有効成分および製薬上許容可能な賦形剤を含む核と、
(b)前記核を囲む剤皮と
を有する経口薬送達システムを提供することにより、有効成分はその環境から隔離され、このシステムは、水性環境との接触により、剤皮が破裂して即座に、かつ急速に有効成分を放出するための特徴を有する。このようなシステムは苦味を有する有効成分または粘膜表面を刺激する有効成分に有効である。またこのシステムは、周辺環境からの液体との接触による分解から保護する必要がある有効成分に有効である。
(a)有効成分組成物の第1の層、
(b)有効成分組成物の第2の層、および、
(c)膨潤性組成物の第3の層
を有し、第1の層および第2の層の有効成分組成物は、同一または異なる有効成分を含むことができる。膨潤性組成物は膨潤剤を含み、有効成分を含んでも、含まなくてもよい。
以下の表1に詳述されるとおり、本発明に従いコハク酸メトプロロールを含む経口薬送達システムを得た。
以下の表3に詳述されるとおり、本発明に従い塩酸ビュープロビオンの制御放出医薬組成物を得た。
Claims (10)
- 1つ以上の面を有する被覆された錠剤の形態である経口薬送達システムであって、
(a)有効成分組成物層と膨潤性組成物層とを含む核、及び
(b)その核を囲む剤皮を有しており、
前記有効成分組成物層は、少なくとも1つの有効成分および製薬上許容可能な放出速度制御賦形剤を含み、
前記膨潤性組成物層は、膨潤性賦形剤、ガス生成剤およびこれらの混合物からなる群から選択される膨潤剤を含み、該膨潤剤は、前記剤皮を破裂させるのに十分な量で存在し、その量は、前記膨潤性組成物の0.5〜95重量%の量であり、前記膨潤性組成物層は、前記剤皮の1つ以上の予め選択された面のごく近傍に位置し、前記有効成分組成物層は、前記剤皮の別の面の近傍に位置し、
前記剤皮は、前記剤皮の前記1つ以上の予め選択された面に、レーザーでの開孔による1つ以上の孔を含み、
前記剤皮は、水性環境との接触により、破裂し、前記錠剤の前記膨潤性組成物層のごく近傍にある前記1つ以上の予め選択された面から、完全に除去され、かつ、前記別の面からは前記剤皮が除去されない経口薬送達システム。 - 請求項1に記載の経口薬送達システムであって、前記剤皮が、前記有効成分組成物に対して不透過性を有する経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、前記有効成分組成物は2つ以上の層として存在し、かつ、前記膨潤性組成物は1つ以上の層として存在する経口薬送達システム。
- 請求項3に記載の経口薬送達システムであって、異なる層に存在する前記有効成分が同一であっても異なっていてもよい経口薬送達システム。
- 請求項3に記載の経口薬送達システムであって、前記有効成分組成物は第1の有効成分組成物および第2の有効成分組成物を含んでおり、上記第1の有効成分組成物は、急速放出組成物であり、第2の有効成分組成物は、第1の有効成分組成物と同一の有効成分を含む制御放出組成物である経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、前記膨潤性組成物は吸い上げ剤を含む経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、前記膨潤性組成物は浸透剤を含む経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、前記核が前記水性環境と接触した時点から60分以内に前記有効成分の放出が開始される経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、pH依存型ポリマーの外側剤皮をさらに有する経口薬送達システム。
- 請求項1に記載の経口薬送達システムであって、前記膨潤性組成物が、前記核の中の埋め込み型錠剤として存在する経口薬送達システム。
