適切なチアゾールアミン(1.0当量)及びピリジン(1.2当量)のジクロロメタン(5体積)撹拌及び冷却(0℃)溶液に、5分間に亘って臭化ブロモアセチル(1.2当量)を添加し、得られた混合物を室温に昇温し、次いで、室温で更に2時間撹拌した。反応混合物をジクロロメタン(50mL)及び水(50mL)で希釈した。層を分離した。水層をジクロロメタン(2×50mL)で抽出し、合わせた有機層を水(2×50mL)で洗浄し、次いで、食塩水(50mL)で洗浄し、乾燥させ(Na2SO4)、濾過した。濾液を減圧下で濃縮した。溶媒留去後に得られた残渣を、石油エーテル中5%〜10%酢酸エチルを用いてシリカゲルカラムクロマトグラフィーによって精製して、オフホワイトの固体として所望の生成物を得た。
方法1
アセトフェノン誘導体(1.0当量)の氷酢酸(5体積)溶液に0℃の液体臭素(1.0当量)を添加し、反応混合物を室温で2時間撹拌した。前記反応混合物を水で希釈し、酢酸エチルで抽出し、食塩水で洗浄し、Na2SO4で乾燥させた。濃縮して得られた粗生成物を無水THF(10体積)に溶解させ、チオ尿素(2.0当量)を添加し、一晩還流させた。前記反応混合物を酢酸エチルで希釈し、チオ硫酸ナトリウム溶液で洗浄し、有機層を1NのHClで処理して、アミンの塩を形成させた。沈殿した塩を濾取した。次いで、前記塩をNaHCO3飽和溶液で処理して、アミンを再生成させた。混合物をジクロロメタン(2×50mL)で抽出し、合わせた有機抽出物を水及び食塩水で洗浄した。減圧下で溶媒留去して、2−アミノ−4−アリール−チアゾール誘導体を得た。
アセトフェノン誘導体(1.0当量)、チオ尿素(2.0当量)、及びヨウ素(1.0当量)の無水エタノール(5体積)溶液を24時間還流させた。反応混合物を酢酸エチルで希釈し、層を分離した。有機層をチオ硫酸ナトリウム溶液で洗浄して、ヨウ素を除去した。酢酸エチル溶液を1NのHClで処理し、沈殿した塩を濾取した。上記方法1に記載の通り遊離アミンを再生成させた。
方法A:
無水DMF(10mL/g)中ピロロ[3,2−d]ピリミジンジオン(中間体1〜6、1.0当量)又はピラゾロ[4,3−d]ピリミジンジオン(中間体9〜10、1.0当量)とNaH(1.5当量)との撹拌混合物に、0℃の2−ブロモ−N−フェニル−1,3−チアゾール−2−イルアセトアミド(1.1当量)を添加し、反応混合物を室温に昇温し、30分間撹拌した。反応混合物を一晩80℃に加熱した。この時間後、前記反応混合物を減圧下で濃縮し、クロロホルム中2%メタノールを用いることによりシリカゲルカラムクロマトグラフィーによって残渣を精製して生成物を得た。
(1,3,7−トリメチル−2,4−ジオキソ−2,3,4,7−テトラヒドロ−1H−ピロロ[2,3−d]ピリミジン−5−イル)酢酸(中間体7、1.0当量)又は2,5,7−トリメチル−4,6−ジオキソ−4,5,6,7−テトラヒドロ−2H−ピラゾロ[3,4−d]ピリミジン−3−イル)酢酸(中間体8、1.0当量)の1,2−ジクロロエタン撹拌溶液に、EDCI(1.2当量)、HOBt(0.3当量)、及び4−ジメチルアミノピリジン(0.1当量)を添加し、混合物を10分間〜15分間室温で撹拌した。次いで、適切なアミン(1.0当量)を添加し、混合物を同温度で48時間撹拌した。減圧下で溶媒留去し、得られた残渣をメタノールで希釈し、30分間室温で撹拌した。分離した固体を濾取した。イソプロパノール又はメタノールから再結晶化させることにより固体生成物を更に精製して所望の生成物を得た。

TRPA1受容体の活性化阻害を、アリルイソチオシアネート(AITC)によって誘導される放射性カルシウムの取り込み阻害として測定した。試験化合物をDMSOに溶解させて、10mMの原液を調製し、次いで、0.1%のBSA及び1.8mMのCaCl2を含む普通培地を用いて所望の濃度に希釈した。反応におけるDMSOの最終濃度は、0.5%(v/v)であった。10%のFBS、1%のペニシリン−ストレプトマイシン溶液、400μg/mLのG−418を含むF−12 DMEM培地中で、ヒトTRPA1を発現しているCHO細胞を増殖させた。実験日に1ウェル当たり約50,000細胞となるように、アッセイの24時間前に前記細胞を96ウェルプレートに播種した。10分間試験化合物で前記細胞を処理し、次いで、3分間最終濃度30μMのAITC及び5μCi/mLの45Ca2+を添加した。前記細胞を洗浄し、1%のTriton X−100、0.1%のデオキシコレート、及び0.1%のSDSを含有するバッファを用いて溶解させた。液体シンチレータを添加した後、Packard Top countで溶解物の放射活性を測定した。試験アンタゴニストの不在下で得られた最大反応の百分率として、濃度反応曲線をプロットした。GraphPad PRISMソフトウェアを用いて非線形回帰分析によって濃度反応曲線からIC50値を計算した。

で表される化合物、又はその薬学的に許容しうる塩。 - 請求項1〜3のいずれかに記載の化合物から選択される1以上の化合物と、1以上の薬学的に許容しうる賦形剤、担体、希釈剤、又はこれらの混合物とを含む医薬組成物。
- 対象におけるTRPA1機能に関連する疾患又は症状の予防又は治療に使用するための組成物であって、請求項1〜3のいずれかに記載の化合物を含む組成物。
- 疼痛、慢性疼痛、複合性局所疼痛症候群、神経因性疼痛、術後痛、関節リウマチ痛、変形性関節症痛、背痛、内臓痛、癌痛、痛覚過敏、神経痛、偏頭痛、ニューロパシー、糖尿病性ニューロパシー、坐骨神経痛、HIV関連ニューロパシー、ヘルペス後神経痛、線維筋痛症、神経損傷、虚血、神経変性、脳卒中、脳卒中後痛、多発性硬化症、呼吸器疾患、喘息、咳、COPD、炎症性障害、食道炎、胃食道逆流性疾患(GERD)、過敏性腸症候群、炎症性腸疾患、骨盤過敏症、尿失禁、膀胱炎、火傷、乾癬、湿疹、嘔吐、胃十二指腸潰瘍、及び掻痒から選択されるTRPA1機能に関連する疾患又は症状の症候の予防又は治療に使用するための請求項5に記載の組成物。
- 慢性疼痛、神経因性疼痛、関節リウマチ痛、変形性関節症痛、COPD、咳、又は喘息に関連する疾患又は症状の症候の予防又は治療に使用するための請求項5に記載の組成物。
