Description

合成後、ナノ結晶は一般に、それらの成長の制御を可能にする有機配位子で安定化された形態で得られる。当該ナノ結晶は、配位子を、一端がナノ結晶の表面との親和性を有し、他端が水性媒体との親和性を有する二官能性配位子で置き換えることによって、生体適合性水性媒体に可溶化することができる。
従って、生物学および医学分野、特に医用画像処理の分野における特定の必要性に適したナノ結晶製剤を提供する必要がある。
最後に、生体内分布を撮像または処置される領域に向けることができる製剤を提供することが有益である。
本発明によれば、特定の安定な配合を有し、かつヒトへの注射用に認可されている成分だけを用いて調製されているナノエマルションの油性分散相にナノ結晶を封入することが提案されている。
両親媒性脂質は好ましくはリン脂質である。可溶化脂質は、有利には、少なくとも1種の脂肪酸グリセリド、好ましくは12〜18個の炭素原子を含む少なくとも1種の飽和脂肪酸グリセリドを含む。さらに、ナノ結晶製剤の油相は、少なくとも1種の油を含んでいてもよい。
第2の側面によれば、本発明は、少なくとも1つの連続水相および少なくとも1つの分散油相を含むナノ結晶製剤を調製する方法であって、
(i)少なくとも1種の可溶化脂質、両親媒性脂質および好適な量のナノ結晶を含む油相を調製する工程と、
(ii)共界面活性剤を含有する水相を調製する工程と、
(iii)ナノエマルションを形成するのに十分な剪断力の影響下で油相を水相に分散させる工程と、
(iv)そのように形成されたナノエマルションを回収する工程と、
を含む方法に関する。
特に、ナノエマルションは、有利には、貯蔵中の優れたコロイド安定性(3ヶ月超)、ナノ結晶を封入する良好な能力ならびに分散相における濃度の上昇を示す。投与中、生体への該ナノ粒子の静脈内注射後に、長い血漿内寿命も観察された(ステルス性(stealthy character))。
連続水相中の共界面活性剤濃度が上昇するため、ナノエマルションは非常に小さな直径を有する分散相も示す。従って、3〜12nmの平均径を有するナノ結晶を封入しているナノエマルションは、典型的に50nmより小さく、好ましくは30〜40nmの平均径を有する。
本明細書における意味範囲内で、「ナノエマルション」という用語は、少なくとも2つの相、一般に油相および水相を有する組成物であって、分散相の平均的大きさが、1ミクロン未満、好ましくは10〜500nm、特に20〜100nm、最も好ましくは20〜70nmである組成物を意味する(論文C. Solans, P. Izquierdo, J. Nolla, N. Azemar and M. J. Garcia-Celma, Curr Opin Colloid In, 2005, 10, 102-110を参照)。
「生物学的配位子」という用語は、特定の方法で一般に細胞の表面に配置された受容体を認識する任意の分子を意味する。
第1の側面によれば、本発明は、ナノエマルション形態のナノ結晶からなる特定の製剤に関する。
当該ナノ結晶は、それらが一般に親油性の親和性である場合、ナノエマルションの油性分散相に封入されている。
特に、無機材料は、金および銀などの金属、酸化鉄などの酸化物(特に、磁鉄鉱、磁赤鉄鉱、酸化ガドリニウムおよび酸化ハフニウム)であってもよい。
発光半導体ナノ結晶によって吸収および/または放射される光の波長は、ナノ結晶の大きさだけでなく、半導体ナノ結晶のエネルギーギャップにも依存する。従って、エネルギーギャップが目的の用途において有益な波長(例えば、可視波長または近赤外波長)に対応する化合物が選択される。
従って、同じナノ液滴中に複数の同一または異なるナノ結晶を封入するナノエマルションを得ることができる。異なるナノ結晶の封入によって、多機能性ナノエマルションを得ることができる。従って、様々な量および異なる波長で光を放射する発光半導体ナノ結晶の封入によって、光学「バーコード」を有するナノ粒子を得ることができる。
安定なナノエマルションを形成するために、一般に、組成物中に少なくとも1種の両親媒性脂質を界面活性剤として含める必要がある。界面活性剤の両親媒性により、油滴を水性連続相中で安定化させることができる。
特定の一態様では、両親媒性脂質の全部または一部は、反応性官能基(例えば、マレイミド、チオール、アミン、エステル、オキシアミンまたはアルデヒド基)を有してもよい。反応性官能基が存在することにより、官能性化合物を界面でグラフト結合させることができる。反応性の両親媒性脂質は、分散相を安定化させている界面に形成された層に組み込まれており、そこでは、例えば、水相に存在する反応性化合物に結合しやすい。
上記量の両親媒性脂質は、有利には、得られたナノエマルションの分散相の大きさを制御するのに有用である。
可溶化脂質は、両親媒性脂質の可溶化を可能にする両親媒性脂質との十分な親和性を有する脂質である。可溶化脂質は、好ましくは室温で固体である。
使用される油は、好ましくは8未満、さらにより好ましくは3〜6の親水−親油性バランス(HLB)を有する。有利には、油は、エマルションの形成前に、化学的または物理的な修正もせずに使用される。
一般に、上記油は、存在する場合、油相中に1〜80重量%、好ましくは5〜50重量%、特に10〜30重量%の量で含有されている。
エマルションの分散相のための油相は、単に成分を混合し、かつすべての成分が溶解するまで、必要に応じてそれらを加熱することによって調製してもよい。
また、共界面活性剤は、当該ナノエマルションの目的の用途に加えて他の効果を有してもよい。特に、共界面活性剤は、対象とする配位子を運ぶようにグラフト結合されていてもよい。
好ましい態様では、連続相は、グリセリン、糖、オリゴ糖もしくは多糖、ゴム、さらにはタンパク質、好ましくはグリセリン、のような増粘剤も含む。実際、より粘度の高い連続相の使用によって乳化を容易にし、よって、超音波処理時間を短縮させることができる。
当然のことながら、水相は、適量の着色剤、安定剤および防腐剤などの他の添加剤をさらに含んでいてもよい。
分散相のゼータ電位の絶対値は、好ましくは20mV未満(すなわち、−20〜20mV)である。実際、ナノ粒子のゼータ電位の絶対値が増加すると、体内、特に、肝臓、脾臓、肺、さらに腎臓にまでナノ粒子が蓄積してしまう。しかし、画像処理の分野での用途においては、逆のもの、すなわち静脈内注射後に長い血漿内寿命を有するステルス製剤(stealthy formulation)が求められる。
上記ナノエマルションは、剪断力の影響下で好適な量の油相および水相を分散させることによって容易に調製することができる
本発明に係る方法では、最初に異なる油性成分および無機ナノ結晶を混合してエマルションの分散相のための油性プレミックスを調製する。混合は、場合により、適当な有機溶剤溶液にその成分または完全な混合物のうちの一方を入れることによって容易にすることができる。次いで、有機溶剤を蒸発させて分散相のための均質な油性プレミックスを得る。
好適な態様によれば、ナノエマルションの分散相は、有益な分子(例えば、生物学的配位子)によって界面でグラフト結合されている。この種のグラフト結合プロセスによって、特定の細胞(例えば、論文S. Achilefu, Technology in Cancer Research & Treatment(癌の研究および治療における技術), 2004, 3, 393-408に記載されているような腫瘍細胞)または特定の人体器官を認識することができる。
コンディショニング前に、例えば、濾過または透析によって、エマルションを希釈および/または殺菌してもよい。この工程によって、エマルションの調製中に形成され得る凝集体も除去することができる。
[使用方法]
本発明に係る製剤は、ヒトまたは動物へのナノ結晶の投与のために、そのままで、あるいは、例えば希釈により目的の用途に合わせて使用してもよい。
また、本発明は、光線療法または熱療法の治療方法であって、それを必要としている哺乳動物(好ましくはヒト)に上に定義したとおりの治療的有効量の製剤を投与することを含む治療方法、および哺乳動物(好ましくはヒト)に上述した製剤を投与することを含む診断法にも関する。
図1は、PBSで10回希釈されたナノエマルションP1、P2およびP3の蛍光スペクトルを示す。
図3は、実施例2に従って処置を受け、かつ24時間後に屠殺されたマウスの器官の蛍光反射画像(心臓(A)、肺(B)、脳(C)、皮膚(D)、筋肉(E)、腎臓(F)、副腎(G)、膀胱(H)、腸(I)、脾臓(J)、膵臓(K)、脂肪(L)、胃(M)および子宮−卵巣(N))を示す。
図5は、温度T=10℃およびT=60℃についての、製造後のナノエマルションの2種類の1H−NMRスペクトルを示す(実施例3)。
蛍光半導体ナノ結晶を封入するナノエマルションの調製
1)分散相のプレミックスの調製
1回分のプレミックス1gを以下の方法で調製した。大豆油(Sigma−Aldrich社)50mg、半合成グリセリド(Gattefosse社からSuppocire(登録商標)NCという商品名で販売されている)450mgおよび大豆レシチン(75%ホスファチジルコリン含有、Lipoid社からLipoid(登録商標)S75という商品名で販売されている)400mgを適当な容器に入れた。これらの化合物をクロロホルムに溶解し、次いで、低圧で蒸発させた。残留した混合物を50℃で乾燥して粘性油の形態のプレミックスを得、これを冷却するとすぐに凝固した。
1μMのデカン溶液で販売されている、近赤外線範囲(705nm)内の光を放射し、かつ10±2nmの平均径を有するCdTe−ZnS系コア/シェル型ナノ結晶(整理番号Q21761MPでInvitrogen社から販売されているQdots 705 ITK Organic)を以下のようにヘキサン溶液に移した。体積で4倍量(4×)のメタノール/イソプロパノール溶液(体積比75:25)を、1倍量(1×)の半導体ナノ結晶に添加した。得られた混合物をボルテックスで混合した後、3000rpmで5分間遠心分離した。次いで、上澄みを除去し、半導体ナノ結晶を1倍量(1×)のヘキサンに再分散させた。
約70℃の温度で、先の工程で調製した上記プレミックス0.334gを回収し、そこに、上記のとおりに得られた半導体ナノ結晶の分散系0μL(P0)、125μL(0.125nmol)(P1)、250μL(0.250nmol)(P2)および500μL(0.500nmol)(P3)を添加した。次いで、溶媒を低圧で蒸発させ、得られた混合物を50℃で乾燥して粘性油を得、これを冷却するとすぐに凝固した。この溶液をボルテックスによって均質化し、乳化段階の前に70℃に維持した。
1回分の連続相1.666gを以下のように調製した。グリセリン0.050g、50モルのエチレンオキシドを有するステアリン酸ポリオキシエチレン(ICI Americas社からMyrj(登録商標)53という商品名で販売されている)0.228gおよびリン酸緩衝液(PBS)から連続相を形成して、最高1.666gの混合物を生成した。この溶液を、乳化前に50℃まで加熱した。
上で得られた1回分の連続相を、3)で得られた1回分の分散相0.334gに添加し、上の表2に示す組成の混合物を得た。混合物を、全エネルギー6000ジュールを使用して5分間、直径が3mmの円錐形プローブを備えたAV505(登録商標)超音波処理器(Sonics Newtown)を用いて乳化した。非常に安定であり、かつ透明なナノエマルション(P0)、(P1)、(P2)および(P3)が得られ、この色の濃さは、ナノ結晶の濃度によって増加した。
得られたナノエマルションの、0.1MのPBS中に1:1000の比率で希釈された試料からZetasizer(Malvern Instruments社)を用いて測定された分散相の平均径は、30±10nmであった。
実施例2
蛍光画像処理のための蛍光半導体ナノ結晶を封入するナノエマルションの使用
実施例1に従って調製した1回分のナノエマルション(P3)100μLを、リン酸緩衝液100μLを添加して二回希釈した。次いで、得られた1回分200μL(0.025nMのナノ結晶)を、6〜8週齢の雌のヌードマウス(フランスのマルシーレトワール(Marcy l'Etoile)所在のIFFA−Credo社)の尾(尾静脈)に静脈内注射し、無菌条件下に置いた。
ナノエマルションの安定性の確認
可溶化脂質によってナノエマルションに与えられる安定性を実証するために以下の実験を行なった。
Suppocire(登録商標)NC(Gattefosse社)(可溶化脂質)255mg、大豆油(Sigma Aldrich社)(油)85mg、Myrj52(登録商標)(ICI Americas社)(共界面活性剤)345mg、Lipoid(登録商標)s75(レシチン、両親媒性脂質)65mgおよびリン酸緩衝液(PBS)を含むナノエマルションを、実施例1の手順に従って調製した。
Suppocire(登録商標)NC(Gattefosse社)(可溶化脂質)150mg、大豆油(Sigma Aldrich社)(油)50mg、Myrj53(登録商標)(ICI Americas社)(共界面活性剤)228mg、Lipoid(登録商標)s75(レシチン、両親媒性脂質)100mgのおよびリン酸緩衝液(PBS)を含むナノエマルションを、実施例1の手順に従って調製した。
Myrj53(登録商標)(ICI Americas社)(共界面活性剤)228mg、Lipoid(登録商標)s75(レシチン、両親媒性脂質)100mg、リン酸緩衝液(PBS)1600μL、表3に示す量のSuppocire(登録商標)NC(Gattefosse社)(可溶化脂質)および大豆油(Sigma Aldrich社)(油)を含む3種類のナノエマルションを、実施例1の手順に従って調製した。
Claims (12)
- ナノエマルション形態の金属、金属酸化物または半導体のナノ結晶製剤であって、
該製剤は、連続水相、及び、該ナノ結晶を封入する少なくとも1つの分散油相、を含んでなり、
該油相は、
少なくとも1種のリン脂質と、
少なくとも10重量%のC12脂肪酸、少なくとも5重量%のC14脂肪酸、少なくとも5重量%のC16脂肪酸、及び少なくとも5重量%のC18脂肪酸を含む飽和脂肪酸グリセリド類の混合物からなる、少なくとも1種の可溶化脂質と、を含み、
該水相は共界面活性剤を含む、ナノ結晶製剤。 - 少なくとも1種の可溶化脂質が、0%〜20重量%のC8脂肪酸、0%〜20重量%のC10脂肪酸、10%〜70重量%のC12脂肪酸、5%〜30重量%のC14脂肪酸、5%〜30重量%のC16脂肪酸、及び5%〜30重量%のC18脂肪酸を含む飽和脂肪酸グリセリド類の混合物からなる、請求項1に記載のナノ結晶製剤。
- ナノ結晶が発光性である、請求項1または請求項2に記載のナノ結晶製剤。
- 油相が少なくとも1種の油をさらに含む、請求項1〜3のいずれか1項に記載のナノ結晶製剤。
- 共界面活性剤が、エチレンオキシド単位またはエチレンオキシドおよびプロピレンオキシドの両方の単位から形成された少なくとも1つの鎖を含む、請求項1〜4のいずれか1項に記載のナノ結晶製剤。
- 共界面活性剤が、複合化合物である、ポリエチレングリコール/ホスファチジルエタノールアミン(PEG/PE)、脂肪酸とポリエチレングリコールのエーテル、脂肪酸とポリエチレングリコールのエステル、およびエチレンオキシドとプロピレンオキシドのブロック共重合体から選択される、請求項5に記載のナノ結晶製剤。
- 分散相が官能化されていることを特徴とする、請求項1〜6のいずれか1項に記載のナノ結晶製剤。
- 請求項1〜7のいずれか1項に記載の金属、金属酸化物または半導体のナノ結晶製剤を調製する方法であって、
(i)好適な量のナノ結晶と、リン脂質と、少なくとも1種の可溶化脂質とを含む油相を調製する工程と、
(ii)共界面活性剤を含有する水相を調製する工程と、
(iii)ナノエマルションを形成するのに十分な剪断力の影響下で水相に油相を分散させる工程と、
(iv)そのように形成されたナノエマルションを回収する工程と、
を含む方法。 - 剪断力の影響が超音波処理によって生成される、請求項8に記載の方法。
- 油相を、その成分の全てまたは一部を適当な溶媒の溶液に入れ、その後に該溶媒を蒸発させることによって調製する、請求項8または請求項9に記載の方法。
- 医用画像処理、温熱療法または光線療法に使用される、請求項1〜7のいずれか1項に記載のナノ結晶製剤。
- 請求項1〜7のいずれか1項に記載のナノ結晶製剤を含有する、哺乳動物を診断するための製剤。
