Description
短鎖脂肪酸(SCFA)は、この発酵による主要な最終生成物であり、大腸機能の重要な特徴、すなわち液体および電解質吸収の刺激、筋肉収縮および内臓灌流の調節を促進する(Topping&Clifton、2001年)。主要なSCFAの1種である酪酸は、結腸細胞において正常な表現型を促進し、正常に制御された結腸細胞の増殖を強化し、結腸直腸癌の危険性を低下させる役割を演じ得る。後者の悪性度が、豊かな工業国における早期罹患の実質的な原因である。小腸での澱粉消化性の低下によるさらなる結果は、循環血中へのグルコースの取込速度を低下させる可能性と、それによるインスリン要求性の低下である。これは、罹患危険性の実質的要因として明らかになりつつある血糖指数(GI)として測定される。食物繊維に起因する作用の多くは、実際にはRSによるらしいということも明らかになりつつある(Topping&Clifton、2001年)。RSの摂取は富裕病の危険性が高い集団では低く、RSの含量および作用を高めるためのコンビニエンスフードの改良は、その集団レベルでの保健のために栄養を改善する有効な手段であるとみなされている。この改良は、元来RSが豊富な豆や全粒米(玄米)などの特定食品の消費を促すことにより、実行し得る。別の手法は、コンビニエンスフードを添加成分としてのRSで強化することである。
当業者であれば、本明細書に記載の発明が、明記された以外の変更や改変を受けることは認識されよう。本明細書に記載の発明には、このような変更/改変がすべて含まれるものと理解されたい。本発明には、本明細書に個別または包括的に言及または指示した、すべてのこのようなステップ、特徴、組成物および化合物、ならびに前記ステップまたは特徴のいずれか2つ以上のありとあらゆる組合せも包含される。
本発明は、改変した小麦植物から得られる穀粒から作製した製品が、十分な難消化性澱粉および/または低下した血糖指数を有することにより、食品または飲料品中に取り入れるか、あるいは哺乳動物の消化管へ送達することのできる量を摂取した場合、健康上の利益を提供するという知見から得られている。健康上の利益は、腸の健康もしくは代謝健康またはその双方に関係し得る。
i)腸内容物のpH低下、
ii)腸内容物中の全SCFA濃度または全SCFA量の増加、
iii)腸内容物中の1種または複数のSCFAの濃度または量の増加、
iv)糞便体積の増加、
v)下痢を伴わない、腸または糞便の全水分体積の増加、
vi)便通の改善、
vii)プロバイオティック細菌の1種または複数種の個数または活性の増加、
viii)糞便胆汁酸排泄の増加、
ix)腐敗性産物の尿濃度の減少、
x)腐敗性産物の糞便含量の減少、
xi)正常な結腸細胞の増殖量増加
xii)腸炎個体の腸における炎症の低下、
xiii)尿毒症患者における糞便または大腸での、尿素、クレアチニンおよびリン酸のいずれか1種の量の低下、あるいは
xiv)前記項目の任意の組合せ
を含んでもよい。
i)食後のグルコース変動の安定化、
ii)改善された(低下した)血糖反応、
iii)食後の血漿インスリン濃度の低下、
iv)改善された血液脂質プロファイル
v)血漿LDLコレステロールの低下、
vi)尿毒症患者における尿素、クレアチニンおよびリン酸の1種または複数の血漿濃度の低下、
vii)異常血糖反応の改善、または
viii)前記項目の任意の組合せ
を含んでもよい。
本発明は、改変小麦の産生および動物、特に哺乳類の食事中へのその導入により、いくつかの指標で測定した場合、腸が改善されるという知見に基づいている。さらに、その改変小麦で作製した食品は、難消化性澱粉(RS)量の増加、血糖指数(GI)の低下などの特質を示したので、食品の消費により代謝健康も改善される。その小麦植物は、澱粉生合成が、特に穀粒澱粉中のアミロース比率を高めるように改変されている。
本発明は、本明細書に記載の小麦植物から得られる、改変小麦粒またはそれ由来の改変澱粉で製造した食物、飲料または医薬製剤も提供する。本明細書では、穀粒は、本質的に成熟した穀粒と定義する。これには、商業的施設で収穫した穀粒も含まれる。一実施形態では、改変澱粉は、少なくとも部分的には、小麦粒の内胚乳の発生中にSBEIIa活性が低下した所産である。穀粒は、総澱粉のパーセントとして増加した比率のアミロースを含み得る。これは、野生型植物の穀粒と比較した際、澱粉中でのアミロペクチン比率の減少として決定してもよい。野生型小麦澱粉は、約18〜30%のアミロースおよび70〜80%のアミロペクチンを有する。本発明に使用するための穀粒は、少なくとも30%のアミロースを含み、好ましくは少なくとも50%(w/w)のアミロースを含んだ澱粉を含む。さらなる実施形態では、SBEIIa、SBEIIb双方の活性が、内胚乳の発生中に減少する。アミロース含量の増加は、光学顕微鏡下で観察した際の異常な澱粉顆粒形態もしくは顆粒の複屈折の減少、または当分野で既知の他の方法により証明し得る。特定の実施形態では、穀粒の澱粉中アミロース比率は、例えばMorrisonおよびLaignelet(1983年)の方法などの分光法とし得るヨウ素滴定法により、または高速液体クロマトグラフィー(HPLC、例えばBateyおよびCurtin、1996年)により測定される。
別の態様では、本発明は、本明細書に記載の小麦植物の穀粒から得られる改変小麦澱粉であって、アミロース比率が増加し、アミロペクチン比率が減少した澱粉を用いて製造した食物、飲料または医薬製剤を提供する。本明細書で使用する場合、用語「澱粉」とは、穀粒の総澱粉、またはアミロース対アミロペクチン比を変える何らかの分別をする前の澱粉を指す。澱粉は、少なくとも部分的に精製されていてもよく、すなわち、穀粒の少なくとも1つの他成分から分離されたものである。本明細書で使用する場合、「実質的に精製された澱粉」とは、その組成物の乾重量の少なくとも95%(w/w)が澱粉であることを意味する。精製澱粉は、製粉工程、例えば、澱粉をタンパク質、油および繊維から分離することを含む湿式製粉工程により、穀粒から得てもよい。製粉工程の初期生成物は、澱粉顆粒の混合物または組成物であり、したがって本発明は、本明細書に記載の改変澱粉を含むこのような顆粒を包含する。
本発明は、好ましくはSBEIIa活性の低下した小麦植物から得られる、本明細書に記載の改変小麦または改変澱粉で製造される食物、飲料および医薬製剤も包含する。一実施形態では、小麦植物では、SBEIIa以外の少なくとも1種の澱粉生合成酵素が変化、好ましくは減少している。SBEIIaおよびSBEIIb活性の低下した植物は、SBEIIa活性の低下した植物とSBEIIb活性の低下した植物との交配、またはSBEIIa、SBEIIb両遺伝子の発現を阻害する分子をコードする移入遺伝子の導入によって作製し得る。本明細書で明らかなように、小麦中でのSBEIIa遺伝子とSBEIIb遺伝子との密接な連鎖のために、両活性の低下した植物は、小麦ゲノムの1つがコードするSBEIIaおよびSBEIIbアイソフォームを欠いた変種を特定し、このような変種の交配により、少なくとも2つのゲノムがコードするアイソフォームの減少した植物を作製することによっても、作製し得る。このような食物製造には、粉、練り粉、または商業的食品製造の成分となりそうな他の製品の製造が含まれよう。
本明細書では、食物繊維とは、健常者の小腸で吸収されずに大腸に移動する炭水化物および炭水化物消化物である。これには、難消化性澱粉、β−グルカン、ならびに他の可溶性および不溶性炭水化物ポリマーが含まれる。それは、大腸中で常在ミクロフローラにより、少なくとも部分的に発酵できるような部分の炭水化物を含むと考えられている。
難消化性澱粉とは、健常者の小腸で吸収されずに大腸中に移動する、澱粉および澱粉消化物の総称と定義される。したがって、難消化性澱粉は、小腸中で消化、吸収される生成物を除外する。難消化性澱粉には、物理的接近不能澱粉(RS1形)、難消化性顆粒(RS2形)、老化澱粉(RS3形)および化学修飾澱粉(RS4形)が含まれる。本発明の澱粉の改変澱粉構造および特に高アミロース含量のため、食物中で消費する際、難消化性澱粉が増加する。この澱粉は、消化するにはやや接近し難いRS1形であってもよい。V複合体結晶化度で測定した場合の澱粉−脂質会合も、難消化性澱粉の含量に寄与するようである。食物または他の製品中に存在する難消化性澱粉の含量は、実施例13に記載のようにin vitroで、または実施例16に記載のようにin vivoで測定するのが好ましい。
血糖指数(GI)は、澱粉を含む食物の消化速度に関係しており、血糖濃度の変動に対して、試験食物の効果と白パンまたはグルコースの効果とを比較したものである。血糖指数は、食後の血清糖濃度および血糖恒常性を得るためのインスリン要求に対する、対象食物の効果見込みの尺度である。本発明の食物が提供する重要な一特性は、血糖指数の減少である。さらに、食物は、低い最終消化率を有し、その結果相対的に低キロジュールとなり得るものであり、または低エネルギー密度食物と表現してもよい。低カロリー製品は、製粉した小麦粒から製造される粉を含有することに基づくであろう。このような食物は、満腹、腸健康の促進、グルコースおよび脂質の食後血清濃度の減少、ならびに低代謝エネルギー食品の提供という作用を有し得る。
パンにおいては、粉または全粒形態の改変小麦澱粉は、非改変の粉または全粒を10%(w/w)以上、好ましくは少なくとも30%、一層より好ましくは非改変の粉または全粒を少なくとも50%代替し得る。したがって、その配合は、例えば、粉90部、改変小麦澱粉10部、脂肪2部、塩2部、改良剤1部、酵母2.5部となろう。パンの製造は、迅速パン生地法、または当業者に公知の他の技法によってでもよい。
改変小麦澱粉は、澱粉質系パスタ製品中に取り入れてもよい。パスタ組成物に用いる改変小麦澱粉の量は、澱粉質材料の全重量に対して10〜40%(w/w)、またはそれより高い範囲、より特別には15〜35%の範囲とし得る。適当な他の澱粉質材料は、当業者によって容易に選択されよう。
経口送達用組成物は、例えば錠剤、トローチ剤、水性または油性懸濁液、顆粒、散剤、乳濁液、カプセル、カプレット、ジェルキャップ、シロップ、またはエリキシル剤の形態であってもよい。経口投与組成物は、1種または複数の任意選択の作用物質、例えば、フルクトース、アスパルテーム、サッカリンなどの甘味剤、はっか油、ウィンターグリーン油、チェリーなどの香味剤、着色剤および保存剤を含むことにより、医薬として口当たりのよい製剤を得てもよい。その上、錠剤または丸剤形態の場合、組成物をコーティングすることにより、消化管中での崩壊および吸収を遅らせ、その結果長期間にわたり持続作用を示してもよい。浸透活性な推進化合物を囲む選択透過膜も、本発明の経口投与化合物にとって適切である。このような後半の基台(platform)においては、カプセルを囲む環境からの液体が、推進化合物により吸収され、その化合物は膨潤して、作用物質または作用物質組成物を開口部を介して押しのける。このような送達基台(platform)は、即時放出製剤のスパイク状プロファイルとは反対に、本質的にゼロ次の送達プロファイルを示すことができる。グリセロールモノステアレートやグリセロールステアレートなどの時間遅延物質も使用してもよい。経口組成物は、マンニトール、ラクトース、澱粉、ステアリン酸マグネシウム、サッカリンナトリウム、セルロース、炭酸マグネシウムなどの標準的な媒体を含むことができる。このような媒体は、好ましくは医薬等級である。
SBEIIa、SBEIIbまたは他の澱粉生合成もしくは修飾遺伝子の発現および/または活性は、小麦植物中に1つまたは複数の遺伝的変異を導入することによって、変化し得る。本明細書で使用する場合、「遺伝的変異」とは、小麦植物のゲノムにおける任意の遺伝的変化を意味しており、その変化が、この文脈では対象遺伝子の発現または活性に影響する。遺伝的変異には、点変異、挿入、置換、反転、複製、転位、好ましくは欠失などの変異、および1種または複数の移入遺伝子のゲノム中への導入が含まれる。

SBEIIa、SBEIIbまたは他の澱粉生合成もしくは修飾遺伝子の発現および/または活性は、1種または複数の移入遺伝子を小麦植物に導入することによって変更し得る。本明細書で言及する場合の「移入遺伝子」は、生物工学分野での通常の意味を有しており、組換えDNAまたはRNA技術により産生または変更され、しかも対象とする生物または細胞、好ましくは小麦細胞中に導入された遺伝子配列を包含する。移入遺伝子は、生物または細胞由来の遺伝子配列、例えばアンチセンス配列を包含し得る。移入遺伝子は、通常、前記生物または細胞に由来しない外因性核酸を含む。「トランスジェニック」とは、移入遺伝子、あるいはその生物もしくは細胞またはその前駆体に導入された遺伝子配列を含んだ、生物または細胞を指す。「非トランスジェニック」とは、ゲノム中に移入遺伝子がまったく存在しないことを指す。移入遺伝子は、好ましくは、安定な遺伝のために生物または細胞のゲノム中に組み込まれる。
小麦などの植物中の遺伝子活性を変更する、特に特異的に減少させるための遺伝子操作手法は、当分野で周知である。こうした方法には、標的遺伝子のRNAの少なくとも一部と配列が相補的であり、それとハイブリッドを形成できる、ヌクレオチドを含んだ適切なアンチセンス分子を発現させるための遺伝子構築体の導入が含まれる。アンチセンス分子は、標的遺伝子のmRNAの翻訳もしくはプロセッシングまたは安定性を妨害し、それによって遺伝子の発現を不活性化すると考えられている。アンチセンス配列を考案する方法は、当分野で周知であり、こうした方法の例は、米国特許第5190131号、欧州特許明細書第0467349−A1号、欧州特許明細書第0223399−A1号および欧州特許明細書第0240208号に見出すことができるが、これらのものは参照により本明細書に組み込まれている。植物におけるアンチセンス法の使用は、Bourque(1995年)およびSenior(1998年)により総説された。Bourqueは、植物系においてアンチセンス配列を用いた遺伝子不活性化の多数の例を列挙している。彼女は、酵素活性の部分的阻害でその系の測定可能な変化がおそらく起こると見込まれるので、100%の阻害達成は必要とはいえないとも明言している。Senior(1998年)は、アンチセンス法は今や植物中での遺伝子発現を操作する非常によく確立された技法であると明言している。
使用し得る別の分子生物学的手法は、共抑制である。共抑制の機構は十分に理解されていないが、転写後遺伝子発現抑制(PTGS)を伴うと考えられており、その点でアンチセンス抑制の多数の例と酷似し得る。それには、遺伝子またはその断片の余分なコピーを、その発現用プロモーターに関してセンス方向に植物中へ導入することが含まれる。そのセンス断片の大きさ、標的遺伝子領域との対応度、および標的遺伝子との配列同一度は、アンチセンス配列に対して前記した通りである。いくつかの例では、遺伝子配列の余分なコピーは、標的植物遺伝子の発現を妨害する。共抑制手法を実施する方法に関しては、WO 97/20936の特許明細書および欧州特許明細書第0465572号を参照されたい。
小麦植物中に遺伝子変異を導入するために用いられそうなさらなる方法は、二重鎖または二本鎖RNA媒介遺伝子発現抑制である。この方法にもPTGSが伴われる。この方法では、不活性化しようとする標的遺伝子と相同な少なくとも一部が二本鎖のRNA産物(複数も)の合成を誘導する、DNAが導入される。したがって、DNAはセンス、アンチセンスの両配列を含んでおり、両配列は、RNAに転写されると、ハイブリッド形成により二本鎖RNA領域を形成できる。好ましい実施形態では、センスおよびアンチセンス配列は、RNAに転写されるとスプライシングで除去されるイントロンを含む、スペーサー領域によって隔てられている。この配置は、遺伝子発現抑制の効率を高めることが示された。前記二本鎖領域は、DNA領域1つまたは2つから転写された1個または2個のRNA分子を含み得る。この二本鎖分子が存在するために、その二本鎖RNAだけでなく、標的植物遺伝子由来の相同RNA転写物も破壊する、植物内因系からの反応が誘発され、標的遺伝子の活性が効果的に低減または消去される。この技法を実施する方法に関しては、オーストラリア特許明細書第99/29514−A号およびWO 99/53050の特許明細書を参照されたい。特定の実施形態では、ハイブリッド形成するセンスおよびアンチセンス配列の長さは、少なくとも19個の連続ヌクレオチド、少なくとも30個、少なくとも50個、少なくとも100個、少なくとも200個、少なくとも500個、または少なくとも1000個のヌクレオチドである。全遺伝子転写物に対応する全長配列を使用してもよい。特定の一実施形態では、その長さはヌクレオチド100〜2000個の範囲である。さらなる実施形態では、標的転写物に対するセンスおよびアンチセンス配列の配列同一度は、少なくとも85%、少なくとも90%または95〜100%である。RNA分子が、その分子を安定化させるように機能し得る無関係な配列を含み得ることは当然である。RNA分子は、RNAポリメラーゼIIまたはRNAポリメラーゼIIIのプロモーターの制御下で発現してもよい。後者の例には、tRNAまたはsnRNAのプロモーターが含まれる。二本鎖RNA分子は、連結した複数の遺伝子からの配列を含み、それにより多重遺伝子を標的としてもよい。
小麦中での遺伝子発現の所望する不活性化を担う遺伝子変異は、1個または複数のリボザイムをコードする核酸分子を含んでもよい。リボザイムは、1個またはしばしば2個のハイブリッド形成配列により規定される特定部位で、他のRNA分子を切断できる酵素機能または触媒機能を有するRNA分子である。そのRNA分子の切断で、標的遺伝子の発現が不活性化される。リボザイムは、遺伝子不活性化に寄与し得るアンチセンス分子としても作用し得る。リボザイムは、ハイブリッド形成配列間に1個または複数の触媒ドメイン、好ましくはハンマーヘッド型またはヘアピン型のドメインを含有する。RNAseP、I群またはII群イントロン、および肝炎δウィルスの各型を含めた、他のリボザイムモチーフを使用してもよい。欧州特許明細書第0321201号および米国特許第6221661号を参照されたい。トランスジェニック植物において遺伝子を不活性化するためのリボザイムの使用は、例えばWegenerほか(1994年)によって示された。
本発明は、RNAまたはDNA、好ましくはDNAを含み、遺伝子阻害分子をコードする分離核酸分子も提供する。ある種の実施形態では、核酸分子は、アンチセンス、センス(共抑制)、二本鎖RNAまたはリボザイム分子をコードし、分子は、小麦SBEIIa遺伝子配列を標的とし、小麦粒の内胚乳中でその発現を不活性化する。本発明は、プロモーター、エンハンサー、ターミネーターまたはポリアデニル化配列などの1種または複数の調節要素を含んだ、単離核酸分子を含む、またはコードする遺伝子構築体も提供する。このような要素は当分野で周知である。遺伝子構築体は、植物、特に小麦などの単子葉植物において、移入遺伝子の発現を補助するイントロン配列も含んでもよい。用語「イントロン」は、転写されるが、タンパク質をコードせず、翻訳前にスプライシングによりRNAから除去される遺伝子断片を意味するものとして、通常の意味で使用される。イントロンは、移入遺伝子が翻訳産物をコードする場合は、5'−UTRまたはコード領域中に組み入れてもよく、そうでない場合は、転写領域中のいずれにあってもよい。
別の実施形態では、本発明の移入遺伝子または他の遺伝子構築体は、小麦の内胚乳で調節的または構成的発現を起こし得る転写開始領域(プロモーター)を含んでいる。プロモーターは組織特異的であり、内胚乳中で選択的または排他的に発現を起こし得る。プロモーターは、内胚乳特異的(高分子量グルテニンプロモーター、小麦SSIプロモーター、小麦SBEIIプロモーター、小麦GBSSプロモーターなど)、または内胚乳に非特異的なプロモーター(ユビキチンプロモーター、またはCaMV35Sもしくは強化35Sプロモーター)のいずれから選択してもよい。プロモーターは、温度、光、ストレスなどの因子によって調節し得る。通常であれば、プロモーターには、発現しようとする遺伝子配列の5'が提供されよう。構築体は、nos3'やocs3'ポリアデニル化領域または転写ターミネーターなどの、転写を高める他の要素も含んでもよい。例示したこのようなDNA領域は、適切な選択マーカー遺伝子配列および他の要素を含むベクター、またはこうした配列を含むベクターと同時形質転換を受けるベクター中に組み込まれることになろう。
小麦などの単子葉植物の形質転換法、すなわち外因性核酸の導入によりその植物に遺伝子変異を導入し、プロトプラストまたは未熟植物胚から植物を再生する方法は、当分野で周知であり、例えば、Beckerほか、1994年、Chengほか、1997年、Heほか、1994年、Hessほか、1990年、Nehraほか、1994年、Vasilほか、1992年、Vasilほか、1993年、Weeksほか、1993年、Weirほか、2001年、オーストラリア特許出願第75460/94号、欧州特許出願第709462号、国際特許公報 WO93/04178、WO89/12012、WO94/13822およびWO99/14314を参照されたい。所望のヌクレオチド配列または遺伝子構築体と選択マーカーを担持するベクターは、組織培養した植物もしくは外植片の再生可能小麦細胞、またはプロトプラストなどの適切な植物系の中へ導入してもよい。選択マーカー遺伝子は、小麦細胞に抗生物質または除草剤耐性を賦与し、あるいはマンノースなどの基質の利用を可能とする場合がある。選択マーカーは、小麦細胞にアスラム、ジェネティシンまたはハイグロマイシン耐性を賦与するのが好ましい。再生可能小麦細胞は、好ましくは、未熟胚の胚盤、成熟胚、これら由来のカルス、または分裂組織から得られる。
小麦内胚乳においてSBEIIa酵素または他の澱粉生合成酵素の活性低下を起こす遺伝子変異の導入は、個々の遺伝子内またはその遺伝子の調節配列内での適当な変異によっても実現し得る。本願に関しては、「誘発変異」とは、化学的、放射線的または生物学的な変異誘発、例えばトランスポゾンもしくはT−DNAの挿入の結果ともいえる、人工的に誘発した遺伝子変異である。遺伝子の阻害度は、産生する澱粉の特性をある程度決定するであろう。変異は、切断またはヌル変異の場合もあり、このような変異は、澱粉の性質に相当程度の影響を及ぼすことが知られているが、澱粉構造の変化は、アミロペクチンシンターゼ活性を十分に低下させることにより、小麦の澱粉または穀粒において注目すべき特性をもたらす、漏出変異体からも生じるであろう。他の染色体再配置も有効となり得るものであり、こうしたものには、挿入、欠失、反転、複製または点変異を含めてよかろう。本明細書で使用する場合の「ヌル変異」とは、例えば、遺伝子活性がもはや検出できない場合のような、対象遺伝子の完全な、またはほぼ完全な活性喪失を生じる変異を指す。
実施例1.材料と方法
炭水化物の定量および分析
澱粉は、Shulmanほか(1991年)の方法を用いて小麦穀粒より単離した。澱粉含有量は、Megazyme(Bray、Co Wicklow、アイルランド共和国)から得た総澱粉分析キットを用いて定量した。
内胚乳中の総澱粉シンターゼ活性は、タンパク質の抽出およびLibessartほか(19995)またはCaoほか(1999年)が記載した方法による分析により測定できる。分析には14CラベルADPG基質を使用し、単量体の澱粉ポリマー中への取り込みを測定する。抽出液中の澱粉シンターゼの個々のアイソフォームは、以下の通りゲル電気泳動で分離し、ゲル中でアッセイ(ザイモグラム)し得る。発生中の種子からの抽出液は、50mMリン酸カリウム緩衝液、pH7.5、5mM EDTA、20%グリセロール、10μMペファブロック(Pefabloc)および0.05mMジチオスレイトール(DTT)を使用して調製し得る。緩衝液中で種子をパルプ状になるまで粉砕した後、混合物を14,000gで4℃、15分間遠心分離し、上清を抜き出す。上清中のタンパク質濃度はCoomassie蛋白試薬または他の標準的な方法を使用して測定することもできる。タンパク質抽出物を未変性ゲルにかける場合は抽出液の保存は−80℃で行う。変性ゲル電気泳動のためには、100μlの抽出液をSDSおよびβ−メルカプトエタノールと混合し、混合液を沸騰水で4分間インキュベートしタンパク質を変性させる。電気泳動は、4.5%ポリアクリルアミド濃縮ゲルを上部に重ねた8%ポリアクリルアミド分離ゲルを使用する標準的な変性ポリアクリルアミドゲルで行われる。電気泳動後、タンパク質は、ゲルを40mMトリス−塩酸緩衝液に、緩衝液交換の度に少なくとも100mlの緩衝液を使用して30分ごとに緩衝液を交換しながら最低2時間浸すことにより再生できる。非変性ゲルの場合は、SDSおよびβ−メルカプトエタノールを使用する変性過程は省略し、ゲル中のSDSも省略する。トリス−グリシン(25mMトリス、0.19Mグリシン)、0.133M硫酸アンモニウム、10mM MgCl2 670μg/ml BSAおよび1mM ADPG基質を含む澱粉シンターゼアッセイ緩衝液を使用して澱粉シンターゼバンドを検出した後、澱粉産物を検出するために2%KI、0.2%I2ヨウ素溶液で染色することもできる。
小麦のSBEIIaまたはSBEIIb遺伝子のどちらかの発現を低減すべく二重鎖RNA(dsRNA)構築体を作成した。そのような構築体において、SBEIIaまたはSBEIIb遺伝子の一部に相当する好ましい核酸配列は、発現したRNAが塩基対合可能で二重または二本鎖RNAを形成し得る相補領域を含むように、プロモーターに対しセンスおよびアンチセンス方向の両方に見出された。センスおよびアンチセンス配列間のスペーサー領域は、形質転換植物中のRNAの一部として転写されると、スプライシングにより除かれて締まった「ヘアピン」二重構造を形成すると思われるイントロン配列を含有していた。イントロンを含有することは、二重鎖RNA構築体によって与えられる遺伝子発現抑制の効率を増すことが分かってきた(Smithほか、2000年)。好ましい核酸を、高分子量グルテニン(HMWG)プロモーター配列(Dx5サブユニット遺伝子のプロモーター、受入番号X12928、Andersonほか、1989年)およびアグロバクテリウム(Agrobacterium)由来ノパリンシンターゼ遺伝子(nos3')のターミネーター配列に連結した。これにより、dsRNA配列の内胚乳特異的発現が提供された。
virプラスミドpAL4404およびpSB1を有する無害化したアグロバクテリウムトゥメファシエンス(Agrobacterium tumefaciens)LBA4404株に小麦の形質転換のための遺伝子構築体をエレクトロポーレーションによって導入し、続いてスペクチノマイシンを含む培地上で選択した。形質転換されたアグロバクテリウム(Agrobacterium)株を、27℃、2日間YEP固化培地上でインキュベートした。次いで細菌を回収し、小麦接種のため、650nmにおける光学密度2.4に400mMアセトシリンゴンを含むTSIM1(100mg/lミオイノシトール、10g/lグルコース、50mg/l MES緩衝液pH5.5であるMS培地)に再懸濁した。
形質転換は、以下の1種または複数の方法で測定した:1個または複数の移入遺伝子についてのPCR分析。PCR分析は、StaceyおよびIssac(1994年)によって記載されたミニプレップ法を使用し、新鮮葉試料1〜2cm2から抽出されたゲノムDNAについて行った。PCR反応は、例えば、SBEIIa遺伝子からの断片(462塩基対)を増幅すべく設計されたプライマーSBEIIa−For:5'−CCCGCTGCTTTCGCTCATTTTG−3'[配列番号4]およびSBEIIa−Rev:5'−GACTACCGGAGCTCCCACCTTC−3'[配列番号5]を用いて、またはSBEIIb(505塩基対)についてのSBEIIb−DupFor5'−AGATGTGAATGGCTGCTTGCTG−3'[配列番号6]およびSBEIIb−DupRev5'−CAGGTCGACCATATGGGAGAGC−3'[配列番号7]を用いて行った。反応条件は以下の通りであった。すなわち、「ホットスタート」(94℃、3分間)の後、変性(95℃、30秒間)、アニーリング(55℃、30秒間)、伸長(73℃、2分間)を30サイクル後、73℃(5分間)を1サイクル行った。
澱粉顆粒形態
T0形質転換小麦植物から得た成熟T1種子由来澱粉顆粒の形態を、光学顕微鏡法によって観察した。ds−SBEIIaで独立に形質転換した25本のT0植物およびds−SBEIIbで独立に形質転換した12本の植物の各々からのそれぞれ10個の穀粒を分析した。各内胚乳を澱粉顆粒を放出させるために軽く押しつぶし、それを水中に分散させて、光学顕微鏡の下で視覚化した。分析したds−SBEIIaの25系統のうち、12個は変形顆粒を含む穀粒を有していたが、視覚的観察では種子ごとに変形レベルは様々であった。これに対し、ds−SBEIIbの12系統はいずれも、光学顕微鏡法下で観察した場合、内胚乳中の澱粉顆粒の顕著な変形は観察されなかった。
温室内で等しい条件下で生育させたds−SBEIIa形質転換植物からの個々の穀粒を秤量した(表3)。植物50.1b、58.2a、61.2aおよび109からのひどく変形した顆粒を有する穀粒は、同じ条件下で生育した野生株植物の穀粒と比較して平均重量で顕著な低下はなかった。したがって、澱粉生産は高度に変形した澱粉顆粒を有する種子においてでも実質的に低減する様子は見られなかった。このデータは、内胚乳中のSBEIIa活性が低下した野外生育小麦の収穫量がほぼ正常であることも示唆する。
独立に形質転換した5系統からなるds−SBEIIa形質転換T1植物13体、および独立に形質転換した3系統からなるds−SBEIIa形質転換植物9体からの種子(T2)に対して、内胚乳中のSBEIIaおよびSBEIIbタンパク質発現を非変性PAGEおよびウェスタンブロットによって分析した。先に記載の通り、ds−SBEIIa植物はすべて異常な澱粉顆粒形態を有する系統からのものであり、一方ds−SBEIIb系統はすべて正常の顆粒形態を有するものであった。SBEIIa検出のために用いた抗体は、ウサギ由来の3KLHであり、これは、成熟SBEIIaのN末端配列に相当するアミノ酸配列AASPGKVLVPDESDDLGC[配列番号8]を有する合成ペプチドに対して誘導されたものであり、使用の際に1:5000に希釈した。SBEIIbの検出のために用いた抗体はR6であり、これは、成熟SBEIIbのN末端推定配列に相当するアミノ酸配列AGGPSGEVMIGC[配列番号9]を有する合成ペプチドに対して誘導されたものであり、使用の前に1:6000に希釈した。使用した二次抗体は、GAR−ホースラディッシュペルオキシダーゼ複合体であった(1:3000希釈)。免疫反応バンドは、Amersham ECL検出システムを用いて検出した。
トランスジェニック小麦穀粒中のアミロースおよびアミロペクチンレベル
6個のプールしたT1種子試料由来の澱粉中アミロース含有量を実施例1に記載した通り定量した。プールした種子試料は、以下の通りトランスジェニック小麦系統から得た:

植物においてアミロース含有量を増加させる既知の機構が3種ある。i)GBSS活性の増大、例えば、GBSSの過剰発現がアミロース含有率の増加した米澱粉をもたらすことが近年報告された(Itohほか、2003年)。ii)澱粉シンターゼおよびイソアミラーゼの活性を抑制することによりアミロペクチンの合成を減少させ、アミロースの正味含有率を増加させる方法、例えばトウモロコシsugary−2、su1およびdu−1(Gaoほか、1998年)についておよび小麦Sgp−1(Yamamoriほか、2000年)変異体について35〜45%のアミロース含有量が、またはSSIIa活性を欠失した大麦品種(Morellほか、2003年)において70%より多いアミロースが報告された。本明細書に示す通り、アミロース含有量を増加させる第3の機構は、澱粉分枝酵素の活性を抑制するものであり、小麦中のSBEIIaおよびSBEIIbの減少によりアミロース含有率が>70%である澱粉となり、澱粉顆粒形態、澱粉組成、および澱粉の微細構造が同時に変化した。この結果は、SBEIIbの減少が高アミロース澱粉に必要とされたトウモロコシ(Garwoodほか、1976年)および米(Mizunoほか、1993年)における以前の知見と対照的であった。前記のhp−SBEIIa構築体の結果は、穀粒中の少なくともSBEIIaを含む澱粉分枝酵素の活性を抑制することで、高アミロース表現型が得られることを実証した。
SBEIIaの活性低下を引き起こす小麦SBEIIa遺伝子の変異は、変異誘発、例えばガンマ線照射またはエチルメタンスルホン酸(EMS)などの薬剤を使用する化学的変異誘発のいずれかによって達成することができる。ガンマ線変異誘発のために、種子に60Co源から20〜50kRの線量の照射をしてもよい(ZikiryaevaおよびKasimov、1972年)。EMS変異誘発は、Mullinほか(1999年)によれば種子をEMS(0.03%、v/v)で処理することにより行ってもよい。B+D二重ヌル背景においては、変異体穀粒はアミロース含有量の増加または澱粉顆粒形態の変化に基づいて同定され、前記の方法によって確認することもできる。SBEIIb活性を保持するSBEIIaの変異体は再び変異誘発することができ、子孫はSBEIIaに加えてSBEIIb活性の欠失についてスクリーニングできる、またはSBEIIa変異体はSBEIIb変異体と交配して変異を組み合わせることにより内胚乳において実質的にSBEII活性が欠失した非トランスジェニック小麦品種を生産することができる。
SBEIIa/SBEIIb阻害遺伝子構築体を含有するトランスジェニック小麦植物の穀粒における澱粉中アミロース含有量をセファロースカラム分離法により定量した。方法において、澱粉分子はその分子量に基づいてカラム上で分離された。次いで、分離した画分をStarch Assay Kit(Sigma)を用いて製造者の説明書に従ってアッセイした。
澱粉試料の鎖長分布は澱粉をイソアミラーゼで脱分枝後、蛍光ラベル糖鎖電気泳動(FACE)により測定した。トランスジェニック種子由来の澱粉におけるDP6−11、DP12−30およびDP31−60の鎖長の百分率を、非トランスジェニックの対照と比較して表6に示す。
ds−SBEIIaおよびds−SBEIIbトランスジェニック系統由来の澱粉のゲル化温度を含む物性を、Perkin Elmer Diamond示差走査熱量計を用いて分析した。各澱粉約20mgを、1:2の比率で、すなわち含水率66.7%で、水に混合し、DSC panに封入した。1分間当たり10℃の加熱速度で、0から150℃まで試験および対照試料を加熱した。データは装置で利用できるソフトウェアを使用して分析した。
動物の飼育
若年成体の雄Sprague Dawleyラットを使用した。ラットは、University of Adelaide Animal Resource Facilityから購入し、Animal Services Unit of CSIRO Health Sciences and Nutritionにおいて、気温(22±1℃)および照明(0800〜2000時に点灯)を制御した室内で底部が金網製の標準的なケージ内で群別に飼育した。
到着後ラットは7日間非精製の市販の食飼に慣れさせた。その後体重を量り、無作為に6匹ずつ、平均体重が同じになるように、食飼処置用の2群に分け、精製食飼に移行させた。AIN93G(American Institute of Nutrition、1993年)仕様書に基づいて標準的な材料から調製した基礎食飼の成分を表8に示す。食飼は、主要栄養素についてバランスをとり、タンパク質200g/kg、炭水化物550g/kg(450gの澱粉および100gのショ糖として)、脂質70g/kgおよび非澱粉性多糖(NSP)90g/kgを含んでいた。加工した小麦のふすま、サフラワーオイルおよびカゼインを好ましい主要栄養素プロフィールを得るために使用した。低アミローストウモロコシ澱粉を2種の食飼の澱粉含有量を等量(45g/100g)にするために使用した。高アミロース小麦食飼は576g/kgの新規(高アミロース)小麦を全粒として含有していた。他の処置食飼は32%〜48%の間の全粒(低アミロース小麦)または澱粉(低アミロースまたは高アミローストウモロコシ)をそれぞれ含有していた。食飼は様々な成分を少量の水と共に遊星型ミキサーを使用して混合することにより調製することもできる。次いで混合物は、押出成形によりペレット(例えば直径8mmで長さ1〜2cm)にし、40℃、16時間乾燥し、密封した容器に入れて4℃で保存することもできる。他方、本実験で実施したように食飼は粉状に調製し、飲料水と同様に自由に摂食できるようにした。
初期体重は群相互の違いはなかった(総平均193g;n=12、プール後のSE=3)。食飼は良好に摂食され、この時期のラットに適正な食物消費および体重増加(平均6.5g/日)の速度を維持した。食飼処置は、低および高アミロース小麦においてそれぞれ平均278g(SE=7、n=6)および282g(SE=6、n=6)であった最終体重に影響を及ぼさなかった。代謝ケージ試験期中の1日当たり食物摂取量は、2群のそれぞれにおいて平均20g/日(P>0.05)だった。
ラットをハロタンで麻酔し、腹腔を開き、盲腸および結腸の内容物を回収、秤量し、分析まで−20℃で保存した。盲腸内消化物の湿潤物はその一部の一定量を凍結乾燥し、乾燥の前後で秤量することにより定量した。実験から得られた以下のデータを1群当たり6匹の観察で、平均±標準誤差(SE)で示した。それらはt−検定により分析され、P<0.05の値を有意性の基準値として採用した。
消化物および排泄物試料をSCFA分析用に特定体積の内部標準(ヘプタン酸)と共に希釈し、標準的ガラス電極を使用するpH測定のために完全に混合した。次いでスラリーはさらなる分析を待つため凍結保存した。合計および主要な個々のSCFAを分析するために、スラリーを融解し、遠心分離し、気液クロマトグラフィー(GLC)による定量のため低温、真空での微量蒸留により濃縮した。
高アミロース小麦のような穀粒を食事の中に最も効果的に普及させる方法の1つはパンを通じてである。高アミロース小麦が容易にパンに混合できることを示すため、およびパン製造品質の保持を可能とする要因を検討するために、小麦粉試料を製造し、分析し、パンを焼くことに使用した。最初は少量だけの高アミロース穀粒が入手可能であったため、パン生地の混合および焼き上げは小規模(10〜15g)に実行した。このような方法は十分量の穀粒が入手可能の際は容易に商業レベルの規模に拡大することができる。
小麦穀粒は一晩、水分量16.5%に調整し、BRI Ltd、オーストラリアの実験用Buhler製粉機またはQuadromat Junior製粉機を使用して製粉し、続いて篩に掛けて最終粒径150μmを得た。試料のタンパク質および水分含有量は、赤外線反射率(NIR)によりAACC Method 39−11(1999年)に従って、またはDumas法およびエアオーブンによりAACC Method 44−15A(AACC、1999年)に従って測定された。
小麦粉の最適水分吸収値は、ミクロZアームミキサーで混合当たり4gの小麦粉試料を用いて定量された(Grasほか、2001年;Bekesほか、2002年)。一定の角速度(高速および低速ブレードに対する軸速度はそれぞれ96および64rpm)がすべての混合において使用された。混合は3回ずつ、1回当たり20分間実施された。小麦粉に水を加える前に固体内容物のみで攪拌しベースラインを自動的に記録(30秒間)した。水の添加は自動水ポンプを使用して一段階で実行された。以下のパラメーターを個々の混合実験から平均をとることにより定量した。WA%−吸水量は500Brabender Unit(BU)において定量した;パン生地稠度;パン生地形成時間(DDT):ピーク抵抗時間(秒)。
改変小麦粉を用いて最適パン生地混合パラメーターを測定するために、ミクロZアームミキサーで決定した吸水量に対応する様々な吸水量の試料をCSIRO 10gミキソグラフ試作機中でパン生地の全質量を一定に保って混合した。各小麦粉試料のそれぞれに対し、以下のパラメーターを記録した:MT−混合時間(秒);PR−ミキソグラフピーク抵抗(任意単位、AU);BWPR−ピーク抵抗でのバンド幅(任意単位、AU);RBD−抵抗破壊率(%);BWBD−バンド幅破壊率(%);TMBW−最高バンド幅時間(s);およびMBW−最高バンド幅(任意単位、A.U.)
パン生地伸長性パラメーターを以下の通り測定することもできる:パン生地を10gミキソグラフ試作機でパン生地形成ピークまで混合することができる。1cm/sでの伸長検査は、形状改良Kieffer式パン生地/グルテン伸長性装置を備えたTA.XT2iテクスチャ分析機で行ってもよい(Mannほか、2003年)。伸長検査用パン生地試料(〜1.0g/検査)は、Kieffer成形機で成形し、30℃、90%RHで45分間放置した後、伸長検査をし得る。R_MaxおよびExt_Rmaxは、Exceed Expertソフトウェア(Smewing、1995年;Mann、2002年)の補助を得てデータから決定し得る。
改変小麦から得た小麦粉の吸水特性を測定した(図4)。この高アミロース粉を割合の様々な対照粉と混合することにより、高アミロース小麦粉は対照粉よりも高い吸水性を有し、このことは、50.3x/6/(60.1%アミロース)と85.2c(81.0%アミロース)との比較で認識されるように、澱粉中のアミロース含量と正に相関していた。この結果は、澱粉顆粒径および形状の変化が吸水特性に影響を与えたことを反映し得るが、しかし、非澱粉性多糖含有量の変化などの高アミロース穀粒における他の変化もまた吸水量に影響を与えることもできると考えられる。
実施例12に記載の通りに製造されたパンを含む食物試料の血糖指数(GI)を以下の通りin vitroで測定した。
高アミロース小麦粉(>30%アミロース(w/w))で製造されたパンのGIおよびRS含有量を定量した。対照として、Sunco系統とSGP−1三重ヌル変異体との交配から得られた小麦系統(Yamamoriほか)からもまたパンを製造した。倍加半数体植物を、交配の子孫から得て、3個のSGP−1変異対立遺伝子について分離したホモ接合系統個体群を供給するために栽培した。各系統における変異対立遺伝子の存在は、各系統から単離されたDNAについての遺伝子特異的(SSIIa遺伝子)PCR反応により決定された。したがって、ゲノムA、BおよびD上のSSIIa遺伝子に対するそれぞれのヌル対立遺伝子の寄与が、個別におよび可能な組合せそれぞれについてアッセイ可能であった。各系統由来の穀粒を前記の小規模法によるパン製造に使用した。in vitro GIおよびRS測定の結果を図7および図8に示す。
小腸において消化され、または消化されないと予想される食品中の澱粉量を推定する際に、in vitroアッセイは有用であった。それにより、食物のin vivoでのGIおよびRS含有量を正確で確実に予測するとみなされる値が得られた。重要なことには、食物は「消費時点」として分析されたものであるが、この消費時点は、食物加工法がRS含量に有害な低下効果を及ぼし得ることを考慮すると重要である。改変小麦のパンの場合は、生理的な機能性(特に高RSおよび低GI)が調理および保存中に破壊されなかったこと、および消費時点でも恐らくその機能性が存在していると思われることが、その結果より示された。
血糖代謝異常
腸の健康改善以外に、本発明は、正常血糖の促進、および血糖調節障害の治療、予防またはリスク低下に適すると考えられる方法および組成物を提供する。これは改変小麦澱粉を含む食物の潜在的血糖指数の低下が観察されたことに基づく。これは、例えば競技者のような健康な対象においておよび外科手術または化学療法を受けている患者など易感染性の個人の両方において有用であることができる。特に、日中または夜間に最適な血糖レベルを維持する方法を求めている糖尿病患者において非常に有用となり得る。個人の血糖レベルは、運動、薬物または外科的治療によって、疾患または多数の疾患を含む症候群もしくは代謝障害によって乱れるまたは変化し得る。実施例は、競技者、化学療法で衰えた患者、絶食患者およびグルコース代謝の乱れまたは変化による疾患または障害を受けている患者、もしくはこれらのおよび他の疾患または障害を治療中の患者を含む。さらに実施例はヒトの他に例えばペット、家畜または競走馬などのような動物を含む。
腎不全では、糸球体ろ過速度の低下がおこり、腎臓が血液のホメオスタシスを維持することができない。水分の保持は浮腫を引き起こし、水素イオン濃度が増大することもあり、アシドーシスが発症することもあり、窒素性老廃物が蓄積することもあり、血液および組織において尿毒症と呼ばれる状態が発生することもある。尿毒症の例には、アンモニア、尿素、クレアチニン、フェノール、インドール、ならびにより大きな分子が挙げられる。血清クレアチニン、血中尿素窒素(BUN)、尿酸、およびN−メチルグアニジン(NMG)、グアニジノこはく酸(GSA)などのグアニジン化合物の濃度は有意に変化する。
in vitro消化試験および動物食飼供給試験は、改変小麦澱粉から製造された食物は消化時に比較的遅くグルコースを放出すること示した。人間の場合もこれが当てはまるかを確証するためには、食物のGIを測定しその値を野生株澱粉から製造した対応する食物の値と比較する食事供給試験を志願者について実行することになろう。GIは、炭水化物を含む人間の食物を食後の血糖反応に応じて重量比較に基づいてランク付けする。これは、世界的に十分認知されている相当な臨床的および実用的有用性を有する。本研究の特定の目的は、改変全粒を使用して製造した2種の一般的なパン食品、パンおよびマフィンの血糖指数(GI)を決定することである。本研究では、対照炭水化物と比較して改変食物が志願者の血糖値を上昇させる程度を評価し、これを標準小麦粉を使用して製造された同種のパン食品のGI値と比較することになろう。
血糖反応曲線(濃度対時間)を描き、曲線下の面積を台形規則を適応して幾何学的に算出する。空腹時濃度より下方の面積はGI算出においては無視する。グルコースを参照食物として用い、定義によりそのGIは100である。
個々の被験者における検査食物のGI=
(検査食物に対する血糖反応曲線の積分面積)/(対照に対する血糖反応曲線の積分面積)×100
改変小麦から製造した食物が有意に低いGIを有するかどうかを決定するために分散分析を使用する。標準小麦から製造した全粒粉食物は非常に高いGI(>70)を有すると期待されることから対応する改変小麦を含有する食物は、69以下、最も可能性が高くは60より低い低GIを記録すると期待される。
難消化性澱粉(RS)は、上部腸での消化に抵抗性で大腸に進行する澱粉である。量的に、それは大腸細菌叢の主な発酵基質源であり、そこで澱粉を大腸壁の健康および代謝保護に決定的だと考えられている代謝産物に転換する。特に、特定型の難消化性澱粉の細菌発酵は、機能不良となった細胞を排除することにより大腸ガンを予防することもできるプログラムされた細胞死および関連する過程を促進するその能力によって考慮すべき注目を引いている主要な有機酸である酪酸塩のようなSCFAの生成に助力する。
回腸人工肛門袋の内容物を午前7時に回収しドライアイス上で凍結する。基礎食の食物を朝食(午前7時30分)、モーニングティー(午前10時30分)、昼食(午後12時30分)、アフタヌーンティー(午後3時30分)および夕食(午後6時30分)として摂る。回腸人工肛門の内容物を午前7時から午後11時まで2時間ごとに回収し、凍結する。

摂食された種々の割合の澱粉は上部消化器官において分解されない。非消化(レジスタント)澱粉は、それにより多数の恩恵があるとされる結腸(大腸)に進行し、腸の領域に存在する細菌複合集団の作用を通じて大きな影響をもたらす。利用される澱粉については、結腸の細菌は、種々の決定的な健康関連機能を提供する有機酸を作り上げる。機能には、腸に並ぶ細胞にさらに必要なエネルギー源を提供し、腸粘膜の成長を促進および制御し、および腫瘍性形質転換を受けた細胞の増殖を停止することが含まれる。この食事を摂る個人は、結腸直腸ガンおよび大腸の特定の他の重篤な変性疾患について高リスクであると考えられ、恩恵的細菌性代謝産物はたびたび供給不足である。個体群研究は澱粉消費の増大に伴い(および難消化性澱粉の含有により)大腸ガン退縮の証拠を示す。見地における難消化性澱粉の予防効果は食物繊維のそれより大きいようである。全身の健康にも難消化性澱粉による恩恵が見られるが、この場合の恩恵は小腸における生理学的な作用を通じてもたらされる。難消化性澱粉に富む食物を摂取することにより、エネルギー摂取量および血糖指数が減少する。体重減少もまた難消化性澱粉が誘導した基礎代謝率の増加を通じて促進されることができる。

Claims (29)
- 哺乳動物の腸の健康または代謝健康の1種または複数の指標を改善するために前記動物の消化管に送達される、食品または医薬品の調製のための小麦植物由来の穀粒の使用であって、
前記小麦植物が、(i)内胚乳中の、SBEIIaタンパク質およびSBEIIbタンパク質、SBEIIa酵素活性およびSBEIIb酵素活性、またはその双方のレベル減少と、(ii)野生型小麦胚乳に比して、内胚乳中のSBEIIa遺伝子発現、SBEIIa酵素活性、またはその双方のレベル減少を起こす遺伝子変異とを含み、
前記遺伝子変異が、(iii)SBEIIa遺伝子の変異、または(iv)SBEIIa遺伝子発現を阻害するアンチセンス分子または二重鎖RNA分子である導入核酸、を含み、
前記穀粒の澱粉中のアミロース比率が少なくとも50%であり、
内胚乳中の前記SBEIIaとSBEIIbタンパク質、SBEIIaとSBEIIb酵素活性、またはその双方のレベルの低下が、野生型の小麦植物の胚乳中のタンパク質または活性のレベルと比較して少なくとも40%である使用。 - 前記穀粒の澱粉中のアミロース比率が少なくとも60%である、請求項1に記載の使用。
- 前記穀粒のアミロペクチンが、アミロペクチンのイソアミラーゼ脱分枝後に測定した場合に、野生型穀粒のアミロペクチンに比して、減少比率の4〜12dp鎖長分画を有する、請求項1または請求項2に記載の使用。
- 前記穀粒の澱粉が、示差走査熱量計で測定した場合、野生型穀粒の澱粉に比してゲル化温度の増加を示す、請求項1から3のいずれか一項に記載の使用。
- 前記穀粒の澱粉が、示差走査熱量計で測定した場合、野生型穀粒の澱粉に比してゲル化温度の減少を示す、請求項1から3のいずれか一項に記載の使用。
- 前記穀粒の澱粉が、少なくとも2%の難消化性澱粉を含む、請求項1から5のいずれか一項に記載の使用。
- 前記小麦植物が、Triticum aestivum ssp.aestivumまたはTriticum turgidum L.ssp.durumと交配できる小麦属の一員である、請求項1から6のいずれか一項に記載の使用。
- 前記食品または医薬品のうち少なくとも一部は、全粒、または製粉、挽き割り、精白、ロール、粗挽き、パーボイルもしくは荒挽きをした粒の形態で前記動物に供給される請求項1から7のいずれか一項に記載の使用。
- 前記食品または医薬品は、前記穀粒に由来する、粉、全粒小麦、セモリナ、または実質的に精製されたデンプンである成分を含む食品又は飲料の形で動物に供給される請求項1から7のいずれか一項に記載の使用。
- 前記食品または医薬品は、別の供給源からの、粉、全粒小麦、セモリナ、または澱粉を含む食品または飲料の形態で前記動物に提供される請求項1ないし9に記載の使用。
- 前記食品または医薬品は、食物または飲料の形態で前記動物に供給され、かつ、前記デンプンを前記食物または飲料の調製前もしくは調製中に、少なくとも60℃の温度に10分間加熱する、請求項1から10のいずれか一項に記載の使用。
- 前記食品または医薬品は、経口的に送達される請求項1から11のいずれか一項に記載の使用。
- 前記動物が人間である、請求項1から12のいずれか一項に記載の使用。
- 消化管に送達される穀粒由来の澱粉の量が1日当たり少なくとも10gである、請求項13に記載の使用。
- 腸の健康の1つまたは複数の改善指標が、
i)腸内容物のpH低下、
ii)腸内容物中の全SCFA濃度または全SCFA量の増加、
iii)腸内容物中の1種または複数のSCFAの濃度または量の増加、
iv)糞便体積の増加、
v)腸または糞便の全水分体積の増加、
vi)便通の改善、
vii)プロバイオティック細菌の1種または複数種の活性の個数の増加、
viii)糞便中への胆汁酸排泄の増加、
ix)腐敗性産物の尿濃度の減少、
x)腐敗性産物の糞便含量の減少、
xi)正常な結腸細胞の増殖量増加
xii)腸炎のある個体の腸における炎症の低下、
xiii)尿毒症患者における、糞便または大腸での、尿素、クレアチニンおよびリン酸のいずれか1種の量の低下、あるいは
xiv)前記項目の任意の組合せ
を含む、請求項1から14のいずれか一項に記載の使用。 - 1つまたは複数の前記改善指標が、
i)腸内容物のpH低下、
ii)腸内容物中の全SCFA濃度または全SCFA量の増加、
iii)腸内容物中の1種または複数のSCFAの濃度または量の増加、
iv)糞便体積の増加、
v)腸または糞便の全水分体積の増加、
からなる群から選択される、請求項15に記載の使用。 - 代謝健康の1つまたは複数の改善指標が、
i)食後のグルコース変動の安定化、
ii)改善された血糖反応、
iii)食前の血漿インスリン濃度の低下、
iv)改善された血液脂質プロファイル
v)血漿LDLコレステロールの低下、
vi)尿毒症患者における尿素、クレアチニンおよびリン酸の1種または複数の血漿濃度の低下、
vii)異常血糖反応の改善、あるいは
viii)前記項目の任意の組合せ
を含む、請求項1から16のいずれか一項に記載の使用。 - 治療または予防するために前記食品または医薬品を使用する際の1つ又は複数の状態が、便秘、下痢、過敏性腸症候群、クローン病、結腸直腸癌、憩室疾患、潰瘍性大腸炎、高血液LDLコレステロール、腎疾患もしくは他の疾患に起因する尿毒症、または糖尿病である、請求項1から17のいずれか一項に記載の使用。
- 前記動物が人間であり、前記食品または医薬品中の澱粉の送達量が1日当たり5〜30グラムである、請求項13から18のいずれか一項に記載の使用。
- 小麦植物の穀粒に由来する、少なくとも1%の難消化性小麦澱粉を含む食物、飲料または医薬製剤であって、
前記小麦植物が、(i)内胚乳中の、SBEIIaタンパク質およびSBEIIbタンパク質、SBEIIa酵素活性およびSBEIIb酵素活性、またはその双方のレベル減少と、(ii)野生型小麦の胚乳に比して、内胚乳中のSBEIIa遺伝子発現、SBEIIa酵素活性、またはその双方のレベル減少を起こす遺伝子変異と、を含み、
前記遺伝子変異が、(iii)SBEIIa遺伝子の変異、または(iv)SBEIIa遺伝子発現を阻害するアンチセンス分子または二重鎖RNA分子である導入核酸、を含み、
前記穀粒の澱粉中のアミロース比率が少なくとも50%であり、
内胚乳中の前記SBEIIaとSBEIIbタンパク質、SBEIIaとSBEIIb酵素活性、またはその双方のレベルの低下が、野生型の小麦植物の胚乳中のタンパク質または活性のレベルと比較して少なくとも40%である食物、飲料または医薬製剤。 - 腸の健康または代謝健康の1つまたは複数の指標を改善するために、哺乳動物の消化管へ送達するために使用する際の、請求項20に記載の食物、飲料または医薬製剤。
- 前記穀粒に由来する少なくとも2%の難消化性澱粉を含む、請求項20または21に記載の食物、飲料または医薬製剤。
- 前記難消化性小麦澱粉を少なくとも3%含む、請求項20から22のいずれか一項に記載の食物、飲料または医薬製剤。
- 前記澱粉を少なくとも60℃で少なくとも10分、1回または複数回加熱し、その加熱を前記食物、飲料または医薬製剤の調製前または調製中に行う、請求項20から23のいずれか一項に記載の食物、飲料または医薬製剤。
- パン、ケーキ、ビスケット、朝食用シリアル、麺類、ソース、スープ、パスタ、即席麺、およびパンケーキやケーキミックスなどのプレパックミックスからなる群から選択される食物である、請求項20から24のいずれか一項に記載の食物、飲料または医薬製剤。
- 医薬として許容可能な賦形剤をさらに含む医薬製剤である、請求項20から25のいずれか一項に記載の食物、飲料または医薬製剤。
- 錠剤、カプセル、顆粒剤、散剤、シロップおよび懸濁剤からなる群から選択される形態の経口投与に適した、請求項26に記載の医薬製剤。
- 前記澱粉を粉、全粒小麦または穀粒の形態でパンの調製中に添加し、粉、全粒小麦または穀粒の形態の非改変小麦澱粉と混合したパンであり、
食物、飲料または医薬製剤中の全澱粉の少なくとも5%が、内胚乳中の、SBEIIaおよびSBEIIbタンパク質、SBEIIaおよびSBEIIb酵素活性、または双方のレベル低下と、遺伝子変異とを含む穀粒由来である請求項25に記載の食物、飲料または医薬製剤。 - 直ちに販売できるように、またはバルク形態で包装されている、請求項20から28のいずれか一項に記載の食物、飲料または医薬製剤。
