Description
本願は、2007年8月2日に出願された米国仮特許出願第60/963,271号の利益を主張する。米国仮特許出願第60/963,271号の内容は、その全てが参考として本明細書に援用される。
(発明の分野)
本発明は、一般的には、RANTES(正常なT細胞が発現した活性化に応じて調整され、分泌される(Regulated upon Activation,Normal T−cell Expressed,and Secreted))に結合する完全なヒトモノクローナル抗体、ならびにそれを使用する方法に関する。
いくつかの実施形態では、抗体は、モノクローナル抗体またはその抗原結合断片である。いくつかの実施形態では、抗体は完全なヒトモノクローナル抗体またはその抗原結合断片である。いくつかの実施形態では、抗体は、例えば、IgG1アイソタイプのようなIgGアイソタイプである。
本発明は、例えば以下の項目を提供する。

本明細書中で使用される場合は、用語「抗体」は、免疫グロブリン分子、および免疫グロブリン(Ig)分子の免疫活性部分、すなわち、抗原に特異的に結合する(抗原と免疫反応する)抗原結合部位を含む分子をいう。
用語「抗原結合部位」または「結合部分」は、抗原結合に関与している免疫グロブリン分子の一部をいう。抗原結合部位は、重(「H」)鎖と軽(「L」)鎖のN末端可変(「V」)領域のアミノ酸残基によって形成される。重鎖と軽鎖のV領域の中の3つの大幅に異なるストレッチは「超可変領域」と呼ばれ、「フレームワーク領域」または「FR」として知られているより保存されている隣接しているストレッチの間に差し込まれている。したがって、用語「FR」は、免疫グロブリンの超可変領域の間に、そしてそれに隣接して本来見られるアミノ酸配列をいう。抗体分子の中では、軽鎖の3つの超可変領域と重鎖の3つの超可変領域は、抗原結合表面を形成するように三次元空間の中で互い相対的に配置される。抗原結合表面は、結合した抗原の三次元表面に対して相補的であり、重鎖と軽鎖のそれぞれの3つの超可変領域は「相補性決定領域」または「CDR」と呼ばれる。個々のドメインへのアミノ酸の配置は、Kabat Sequences of Proteins of Immunological Interest(National Institutes of Health,Bethesda,Md.(1987および1991))またはChothia & Lesk J.Mol.Biol.196:901−917(1987),Chothiaら、Nature 342:878−883(1989)の定義にしたがう。
huRANTES抗体は、例えば、以下に提供される実施例に記載される手順を使用して作製される。
本発明に従い、そしてRANTESに関して本明細書中で生産され、特性決定される抗体の活性に基づいて、抗体部分を超えた他の治療手段(modality)の設計が、促進される。そのような手段としては、進歩した抗体治療薬(例えば、二重特異性抗体)、免疫毒素、および放射標識された治療薬、ペプチド治療薬の作製、遺伝子治療(特に、イントラボディ)、アンチセンス治療薬、ならびに低分子が挙げられるが、これらに限定されない。

本発明の完全なヒト抗RANTES MAbは、診断処方物および予防処方物において使用される。1つの実施形態では、RANTESアンタゴニスト(例えば、本発明のhuRANTES MAb)は、虚血、虚血に伴う臨床的適応症、再潅流損傷、および/または上記自己免疫疾患の1つを発症するリスクがある患者に投与される。虚血、虚血に伴う臨床的適応症、再潅流損傷、および/または上記自己免疫疾患の1つに対する患者または臓器の素因は、遺伝子型マーカー、血清学的マーカー、または生化学的マーカーを使用して決定され得る。
ヒトRANTESのクローニング、発現、および精製
クローニング
ヒトRANTES(GenBankアクセス番号M21121)の成熟タンパク質または他のケモカインをコードする遺伝子を、PCR増幅によって発現プラスミドpET43(Novagen Madison,WI)にクローニングした。X因子プロテアーゼ切断部位の配列を、NusAのC末端に導入した。AviTag(Avidity,Denver CO)ビオチニル化部位の配列を、ケモカインコード配列のC末端に導入した。pET由来プラスミドを、ビオチンリガーゼ遺伝子をコードするpACYC184−BirAプラスミドでの細菌株Origami Bの同時形質転換に使用した。哺乳動物細胞中での発現のために、関連するケモカインをコードする遺伝子を、pEAK8発現ベクター(Edge Biosystems,Gaithersburg,MD)中にcDNAからクローニングした。AviTagビオチニル化部位をタンパク質のC末端に導入し、その後ろには、ビオチンリガーゼをコードするBirA遺伝子の発現を可能にする内部リボソーム侵入部位(IRES)が続いた。この構築物は、1つの部位でインビボでビオチニル化されたケモカインの分泌型を発現することができる。
発現構築物を有している細菌の一晩の培養を、50μg/mLのアンピシリン、10μg/mLのカナマイシン、5μg/mLのテトラサイクリン、20μg/mLのクロラムフェニコール、および50μMのビオチンを含むTerrific培養液(InvitroGen)の中に、1:30に稀釈した。培養物を、OD600=0.7に到達するまで震盪させながら37℃でインキュベートした。その後、IPTGを、1mMの最終濃度になるように添加し、37℃で15分間、そして25℃で一晩インキュベートした。
PEAK細胞を、10%のFCS、2mMのL−グルタミン(Sigma)、25μg/mLのゲンタマイシン(Sigma)を補充したDMEM(Sigma)中で培養し、37℃、5%のCO2でインキュベートした。PEAK細胞を、Mirusトランスフェクション試薬を使用して、改変型pEAK8ベクターでトランスフェクトした。ピューロマイシン(Sigma)を、トランスフェクトされた細胞集団を選択し、維持するために、細胞接着後に1μg/mLで添加した。ビオチン(Sigma)を生産用バッチ(production batches)に対して50μMで添加した。トランスフェクトされたPEAK細胞の上清に由来するビオチニル化ケモカインは、化学走査アッセイにおいて活性があることが示された。
細菌ペレットを、ベンゾナーゼヌクレアーゼ(Benzonase Nuclease)とプロテアーゼ阻害剤Complete EDTA−free(Roche)を含むBugbuster(Novagen)の中に再懸濁し、4℃で1時間インキュベートした。可溶性画分と不溶性画分を、4℃で15分間、10,000gでの遠心分離によって分離させた。可溶性タンパク質画分と不溶性タンパク質画分を、SDS−PAGE(Novex gels,InvitroGen)によって分析した。可溶性画分を、緩衝液A(Tris−HCl 100mM(pH8.0)、NaCl 600mM、CaCl2 10mM、イミダゾール40mM)で1/2に稀釈し、緩衝液B(Tris−HCl 50mM(pH8.0)、NaCl 300mM、CaCl2 5mM、イミダゾール20mM)中に予め平衡化させた50%(v/v)Ni−NTAアガロース(Qiagen)と混合した。上記混合物を、穏やかに震盪させながら室温で30分間インキュベートした。遠心分離後に得られたビーズを、Poly−Prepクロマトグラフィーカラム(Biorad)上に充填し、5倍容量の緩衝液Bで3回洗浄し、そして緩衝液C(Tris−HCl 50mM(pH8.0)、NaCl 200mM、CaCl2 5mM、イミダゾール400mM)で溶出させた。タンパク質を含む溶出画分をプールし、PD−10カラム(Amersham)を使用して脱塩させた.NusA−ケモカイン融合タンパク質をX因子(Novagen,Madison,WI)によって、1mgのタンパク質を25UのX因子とともに30℃で24時間まで、切断緩衝液(Tris−HCl 50mM(pH8.0)、NaCl 200mM、CaCl2 5mM)中でインキュベートすることによって切断した。いくつかの融合タンパク質については、最適な切断のためのパラメーターはわずかに異なったが、インキュベーション時間(4時間〜24時間)および/または温度(25℃〜37℃)を変化させることによって容易に決定することができた。切断されたタンパク質をSDS−PAGEによって分析し、活性を化学走査によって試験した。
ヒトscFvライブラリーのスクリーニング
ヒトscFvライブラリーの構築と処理のための一般的な手順は、その全体が引用により本明細書中に組み入れられるVaughanら著(Nat.Biotech.,1996,14:309−314)に記載されている。ヒトscFvのライブラリーを、以下の手順に従ってhuRANTESに対してスクリーニングした。
Cambridge Antibody Technology(Cambridge,UK)から得たscFvファージライブラリー(1012Pfu)のアリコートを、3%(w/v)のスキムミルクを含むPBSで、室温で1時間、回転式混合機上でブロックした。次いで、ブロックしたファージを、ストレプトアビジン磁気ビーズ(Dynal M−280)上で室温で1時間、回転式混合機上で脱選択化(deselect)した。次いで、脱選択化したファージを、インビボでビオチニル化されたhuRANTES(100nM)と共に室温で2時間、回転式混合機上でインキュベートした。この選択工程を、NusA−huRANTESビオチニル化融合タンパク質、またはタンパク質分解的切断によって融合体から切り離されたビオチニル化huRATESのいずれかに対して行った。ビーズを磁気スタンドを使用して捕捉し、続いて、PBS/0.1%のTween 20で4回洗浄し、そしてPBSで3回洗浄した。その後、10mlの指数関数的に増殖しているTG1細胞に、ビーズを直接的に添加し、37℃で1時間、ゆっくりと震盪(100rpm)させながらインキュベートした。感染したTG1のアリコートを、選択の出力(selection output)を既定するために段階的に希釈した。残りの感染したTG1を3000rpmで15分間回転させ、0.5mlの2×TY−AG(100μg/mlのアンピシリンと2%のグルコースを含む2×TY培地)中に再懸濁させ、2×TYAG寒天培地バイオアッセイプレート上に拡げた。30℃で一晩のインキュベーションの後、10mlの2×TYAGを上記プレートに添加し、上記細胞を表面から剥がし取り、そして、50mlのポリプロピレンチューブに移した。17%のグリセロールの最終濃度が得られるように、50%のグリセロールを含む2×TYAGを上記細胞懸濁液に対して添加した。上記選択ラウンドのアリコートを−80℃で維持した。
先の選択ラウンドから得られた細胞懸濁液のうちの100μlを、20mlの2×TYAGに添加し、0.3〜0.5のOD600に達するまで37℃で攪拌(240rpm)しながら増殖させた。その後、上記培養物を、3.3×1010MK13K07ヘルパーファージで過剰感染(supre−infect)させ、37℃で1時間インキュベートした(150rpm)。次いで、上記細胞を2000rpmで10分間遠心分離し、培地を除去し、ペレットを20mlの2×TY−AK(100μg/mlのアンピシリン;50μg/mlのカナマイシン)中に再懸濁させることによって、培地を交換した。その後、上記培養物を一晩30℃で増殖させた(240rpm)。
単一クローンを、1ウェルあたり150μlの2×TYAG培地(2%のグルコース)を含むマイクロタイタープレートに採取し、37℃で(100〜200rpm)5〜6時間増殖させた。10の感染多重度(MOI)(すなわち、上記培養物中で個々の細胞について10個のファージ)が得られるように、M13KO7ヘルパーファージをそれぞれのウェルに添加し、37℃で(100rpm)1時間インキュベートした。増殖後、プレートを3,200rpmで10分間遠心分離した。上清を注意深く除去し、細胞を150μlの2×TYAK培地中に再懸濁し、30℃で一晩増殖させた(120rpm)。ELISAのために、上記ファージを、5%スキムミルク粉末を含む150μlの2×濃度のPBSを添加し、続いて、室温で1時間インキュベートすることによってブロックした。次いで、上記プレートを3000rpmで10分間遠心分離し、ファージを含む上清をELISAに使用した。
ELISAプレート(Maxisorb,NUNC)を、PBS中の2μg/mlのNusA−Rantesとともに一晩コーティングした。対照プレートを、2μg/mlのNusAでコーティングした。その後、プレートを3%スキムミルク/PBSで、室温で1時間ブロックした。上記予めブロックしたファージの上清を移す前に、プレートをPBS 0.05% Tween 20で3回洗浄し、室温で1時間インキュベーションした。次いで、プレートをPBS 0.05% Tween 20で3回洗浄した。それぞれのウェルに対して、(HRP)結合型抗M13抗体(Amersham,1:10,000に希釈した)を含む、50μlの3%スキムミルク/PBS。室温で1時間のインキュベーションの後、上記プレートを、PBS 0.05% Tween 20で5回洗浄した。次いで、反応を停止させるために50μlのTMB(Sigma)と50μlの2NのH2SO4を添加することによって、ELISAを明らかにさせた。吸収強度を450nmで読み取った。
単一クローンを、1ウェルあたり150μlの2×TYAG培地(2%のグルコース)を含むマイクロタイタープレートの中に置き、30℃(120rpm)で一晩増殖させた。翌日、5μlの培養物を、45μlのdH2Oを含む別のプレートに移し、混合した。次いで、上記プレートを−20℃で凍結させた。解凍後、この懸濁液のうちの1μlを、標準的なPCRプロトコールを使用し、以下のpCANTAB6に特異的なプライマーを用いるPCR増幅に使用した。:mycseq、5’−CTCTTCTGAGATGAGTTTTTG−3’(配列番号197)およびgene3leader、5’−TTATTATTCGCAATTCCTTTAGTTGTTCCT−3’(配列番号198)。
個々のクローンを、1ウェルあたり0.9mlの2×TYAG培地(0.1%のグルコース)を含む深型ウェルマイクロタイタープレート(deep well microtiter plate)に接種し、37℃で5〜6時間(250rpm)増殖させた。次いで、0.02mMのIPTGの最終濃度となるように、1ウェルあたり100μlの、2×TY培地中の0.2mMのIPTGを添加した。その後、プレートを30℃で一晩、250prmで震盪させながらインキュベートした。深型ウェルプレートを2,500rpmで10分間遠心分離し、上清を注意深く除去した。ペレットを、150μlのTES緩衝液(50mMのTris/HCl(pH8)、1mMのEDTA(pH8)、20%のスクロース、Complete protease inhibitor,Rocheを補充した)中に再懸濁させた。150μlの希釈したTES緩衝液(1:5のTES:水の希釈)を添加し、そして氷上で30分間インキュベーションすることにより、低張性ショックを生じさせた。その後、プレートを4000rpmで10分間遠心分離し、細胞と破片を除去した。上清を注意深く別のマイクロタイタープレートに移し、機能性アッセイまたはELISAにおける即時の試験のために氷上で維持した。
1mlの2×TYAGの出発培地に、新たに画線した2×TYAG寒天プレート由来の単一コロニーを接種し、震盪させながら(240rpm)37℃で5時間インキュベートした。この培養物のうちの0.9mlを使用して400mlの同じ培養液に接種し、30℃で一晩、激しく震盪させながら(300rpm)増殖させた。
huRANTESに誘導されるカルシウムの流入の、scFv抽出物を使用する阻害
様々なhuRANTES scFvのペリプラズム抽出物を上記のように生産した。hCCR5を発現しているL1.2細胞を、10%のFCSを補充したRPMI培地中で培養した。scFvを含む抽出物を、2nM〜10nMのhuRANTES(Peprotech,Rocky Hill NJ)とともに、室温で30分間インキュベートした。細胞をPBS中で洗浄し、2μMのFura 2/AMとともに充填した。充填した細胞のうちの100μlを、96ウェルの黒色の透明平底プレート(96−well black,transparent flat−bottom plate)のそれぞれのウェルに添加し、カルシウムの流入速度を、ケモカインscFv混合物を添加した際に、Flex station II機器(Molecular Devices)上で340nmまたは380nmでの励起させたときの514nmの蛍光を測定することによって記録した。個々のscFv抽出物の阻害活性を、無関係なscFvを含む抽出物に対する比較によって評価した。
huRANTESに誘導される細胞の化学走査のscFvによる阻害
野生型L1.2細胞と、hCCR5を発現しているLl.2細胞を、10%のFCSを補充したRPMI培地中で培養した。実験の前日に、細胞を、0.6mg/mlの酪酸とともにインキュベートした。様々な濃度の精製したscFvを、0.2nM〜10nMのhuRANTESとともにインキュベートし、化学走査用の96ウェルプレート(chemotaxis 96−well plate)(Neuroprobe)の底部チャンバーに入れた。フィルタープレートを、化学走査用のプレートの上に置き、それぞれのウェルに、20μlの106細胞/mlの懸濁液を重層した。プレートを37℃で2時間インキュベートした。フィルターを通り抜けて移動した細胞をDRAQ5(Alexis Corporation)で染色し、FMAT 8200リーダー(Applied Biosystems,Foster City CA)上でカウントした。それぞれの候補抗体についてのIC50(ここでは、huRANTESに誘導される細胞の移動の50%が阻害される、すなわち、50%阻害濃度)を決定した(表4)。
scFvのIgG形式への再フォーマット
選択したscFvのVH配列とVL配列を、5’末端にリーダー配列とHindIII制限酵素部位を導入する特異的オリゴヌクレオチドを用いて増幅させた。重鎖配列と軽鎖配列の3’末端に、それぞれ、ApaI部位またはAvrII部位を導入した。この増幅させたVH配列をHindIII/ApaIで消化し、pCon_gamma1発現ベクター(LONZA,Basel,Switzerland)にクローニングした。増幅させたVL配列をHindIII/AvrIIで消化し、pCon_lambda2発現ベクター(LONZA)にクローニングした。この構築物を配列決定により確認し、その後、哺乳類細胞にトランスフェクションした。
天然のヒトhuRANTESの生産
THP1細胞を、10μg/mLのLPSとともに、1×106/mlの濃度で、10mlの培地中で培養した。37℃で一晩のインキュベーションの後、細胞を遠心分離し、上清を回収し、そして天然のhuRANTESの濃度を、実施例4に記載したような化学走査アッセイにおいて概算した。上記に記載したようにLPSで刺激した場合には、天然のhuRANTESだけではなく、CCR5の他のリガンドもまた、THP1細胞によって生産される。したがって、抗huRANTES抗体の中和能力を決定するための化学走査アッセイにおいてこれらの上清を使用した場合は、CCR5の他のリガンドを、それぞれ5μg/mlの濃度の、hMIP−1α、hMIP−1β、hMCP−2、hMIP−1δに対する抗体(R&D Systems)の混合物で中和した。
IgG1形式に再フォーマットしたscFvを使用した、huRANTESに誘導されるカルシウムの流入または細胞の化学走査の阻害
scFvを、実施例5において上記に記載したようにIgG形式に再フォーマットした。huRANTESに誘導されるカルシウムの流入または細胞の化学走査に対するIgGの中和能力を、実施例3および実施例4に記載した細胞をベースとするアッセイを使用して評価した。図1に例として示すように、IgGs C8、1D9、および1E4は、用量依存性の様式で組み換えhuRANTESと天然のhuRANTESの両方の活性を阻害する。全ての抗体についてのこれらのアッセイにおけるIC50を、表5および6にまとめる。
グリコサミノグリカンに固定化したhuRNATESに対する抗体の結合
多くのケモカインを用いた場合には、huRANTESは、内皮細胞のような細胞の表面で発現されるグリコサミノグリカン(GAG)に対してオリゴマー化し、結合することができる。この状況で抗体が確実にhuRANTESに結合できるようにするために、これらを以下のアッセイにおいて試験した。ストレプトアビジンでコーティングした96ウェルプレート(Roche,Basel,Switzerland)を、原型のGAG(Sigma,St.Louis,MO)としてのビオチン標識ヘパリンでコーティングした。洗浄後、過剰量のヘパリンhuRANTESを、GAGの固定化のためにウェルに添加した。試験される抗体とのインキュベーションの後、ウェルを洗浄し、結合を、HRPに結合させた抗ヒトIgG Fcg特異的抗体(Jackson,West Grove,PA)によって明らかにした。図2に示すように、いくつかの抗体は、GAGに結合されている場合には、huRANTESに結合できたが、他のものは、おそらく、huRANTES上にあるそれらのエピトープがオリゴマー構造の中にありもはや接近できないために結合できなかった。GAGの状況でhuRANTESに結合する抗体の能力を表7にまとめる。
抗体2D1の親和性成熟
選択したリード候補(2D1)について、huRANTESに対するその親和性と、huRANTES中和アッセイにおけるその効力を高めるための親和性成熟を行った。重鎖または軽鎖のCDR3の中の5残基のストレッチを、6つのライブラリー(ライブラリーの大きさは5×107から109までの範囲である)を作製するために無作為化した。3つの高ストリンジェンシーの選択ラウンドを実施例2に記載したように行った。改良された変異体のスクリーニングを、エピトープ競合アッセイにおいてscFvペリプラズム抽出物を使用して行った。簡単に説明すると、もとの抗体(2D1)をプレート上にコーティングし、希釈したペリプラズムscFv抽出物をそれぞれのウェルに添加した。その後、ビオチニル化huRANTESを添加し、室温で2時間インキュベートした。洗浄後、コーティングしたもとの抗体に結合したままであるhuRANTESを、ストレプトアビジン結合HRP(Jackson,West Grove PA)を使用して明らかにした。改良された変異体2D1を同定するための参照として、scFvを、IgG形式のコーティングされた2D1に競合させるために使用した。
1E4を発現する安定なCHOK1SV細胞株の作製
CHOK1SV細胞株(Lonza Biologics,plcに所有権がある)を使用して、抗体1E4の生産のためにの半安定的トランスフェクションによってプールを作製した。簡単に説明すると、6mMのL−グルタミンを補充した培地CD−CHO(Invitrogen)中の指数関数的に増殖している細胞を、以下の条件下でエレクトロポレーションした:0.4cmのキュベット中、700μLの新しいCD−CHO中の1.0×107個の生存している細胞を、100μLのTris EDTA緩衝溶液(pH7.4)中の40μgのDNAと穏やかに混合し、続いてすぐに、300ボルト、900μFの1回のパルスを送った。2つのキュベットの内容物を、200mLの新しい予め温めておいたCD−CHO培地中にすぐに移した。続いて、この細胞懸濁液を、4つの組織培養−処理T75フラスコに分け、空気中の10%のCO2および37℃の温度に設定した加湿したインキュベーターの中に入れて、半安定プールを作製した。トランスフェクションのおよそ24時間後、選択圧(50μMのMSXの補充による)をかけた。その後、個々の安定にトランスフェクトされたクローンをClonePix技術(Genetix)を使用して選択し、1E4の生産性についてスクリーニングした。
CHO上清からの1E4の大規模の精製
このプロセスには、MabSelect SuReアフィニティクロマトグラフィー(GE Healthcare)、低pH処理によるレトロウイルスの不活化、SPセファロース陽イオン交換クロマトグラフィーのためのpHの調整、Capto Q(GE Healthcare)陰イオン交換クロマトグラフィーの前の濃縮/膜分離、および最終的な処方緩衝液への濃縮/膜分離が含まれる。
CHO上清から精製した抗体1E4の機能的特性
RANTESは、受容体CCR1、CCR3、およびCCR5のリガンドである。これらの受容体のうちのそれぞれ1つとの相互作用をブロックする、CHO上清から精製した1E4の能力を、化学走査アッセイおよびカルシウム流入アッセイにおいて評価した。
hCCR1、hCCR3、またはhCCR5のいずれかを発現しているL1.2細胞を、10%のFCSを補充したRPMI培地中で培養した。最適な結果のために、hCCR1を発現している細胞を、1%のFCSを含む培地中で一晩飢餓状態にした。実験の前日に、全ての細胞を0.3mg/mlの酪酸とともにインキュベートした。様々な濃度の1E4を、4nM〜25nMのhuRANTES(Peprotech,Rocky Hill NJ)とともに、室温で30分間インキュベートした。細胞をPBS中で洗浄し、2μMのFura 2/AMとともに充填した。充填した細胞のうちの100μlを、96ウェルの黒色の透明平底プレートのそれぞれのウェルに添加し、カルシウムの流入速度を、ケモカイン−抗体混合物の添加後に、Flex station II機器(Molecular Devices)上での340nmまたは380nmで励起させたときの514nmの蛍光を測定することによって記録した。図3に示すように、1E4は、用量依存性の様式で、hCCR1、hCCR3、またはhCCR5のいずれかを発現する細胞中へのカルシウムの流入を阻害することができた。IC50((ここでは、huRANTESに誘導されるカルシウムの流入の50%が阻害される、すなわち、50%阻害濃度)を決定した(表8)。
野生型L1.2細胞と、hCCR1、hCCR3、またはhCCR5のいずれかを発現しているL1.2細胞を、10%のFCSを補充したRPMI培地中で培養した。最適な結果のために、hCCR1を発現している細胞を、1%のFCSを含む培地中で一晩、飢餓状態にした。実験の前日に、全ての細胞を0.3mg/mlの酪酸とともにインキュベートした。最適な結果のために、hCCR1を発現している細胞を、1%のFCSを含む培地中で一晩、飢餓状態にした。様々な濃度の1E4を、1nM〜10nMの組み換えhuRANTES、または1nMの天然のhuRANTES(実施例6に記載したように作製した)とともにインキュベートし、化学走査用96ウェルプレート(Neuroprobe)の底部チャンバーの中においた。フィルタープレートを化学走査用のプレートの上に置き、それぞれのウェルに、20μlの106細胞/mlの懸濁液を重層した。プレートを37℃で2時間インキュベートした。フィルターを通り抜けて移動した細胞をDRAQ5(Alexis Corporation)で染色し、FMAT 8200リーダー(Applied Biosystems,Foster City CA)上でカウントした。図4に示すように、1E4は、用量依存性の様式で、hCCR1、hCCR3、またはhCCR5のいずれかを発現する細胞中へのカルシウムの流入を阻害することができた。IC50((ここでは、huRANTESに誘導される細胞の移動の50%が阻害される、すなわち、50%阻害濃度)を決定した(表9)。
1E4抗体の交差反応性
1E4を、ELISAにおいて、様々な種に由来するケモカインのパネルに結合するその能力について試験した。このパネルには以下のケモカインを含めた:ヒトRANTES、カニクイザルRANTES、ラットRANTES、マウスRANTES、ヒトITAC、ヒトIP−10、カニクイザルIP−10、ヒトMIG、カニクイザルMIG、ヒトMIP1α、ヒトMIP1β、ヒトMCP−1、ヒトMCP−2。簡単に説明すると、ヒト、マウス、ラット、およびカニクイザルから単離したcDNAからクローニングしたケモカインを融合タンパク質として発現させ、実施例1に記載したように精製した。ケモカインをmaxisopbプレート(Nunc,Denmark)中に5μg/mlでコーティングし、一定の濃度範囲の1E4とともにインキュベートした。結合のレベルは、西洋ワサビペルオキシダーゼ(Jackson)に結合させた抗ヒトFc−γ特異的抗体と蛍光基質を使用して明らかにした。図5に示すように、抗体1E4は、ヒトRANTESとカニクイザルRANTESに対してのみ結合し、他の種由来のRANTESにも、試験した他のヒトケモカインのいずれにも結合しなかった。全てのケモカインの適切なコーティングは、個々のケモカインに特異的なモノクローナル抗体を使用して制御し、試験したケモカインは全てこの形式で検出することができた。
1E4抗体のエピトープマッピング
ELISAにおいて、抗体1E4は、ヒトRANTESとカニクイザルRANTESの両方に対して同等と見られる親和性で結合する(図5)。1E4に対する結合についてhuRANTES上におそらく必要とされる残基を同定するために、いくつかの種に由来するRANTESタンパク質配列を図6に示すようにアラインメントした。このアラインメントにおいては、ヒト配列とカニクイザル配列との間で保存されており、それに対して1E4が結合できないマウスRANTESとラットRANTESにおいては異なる複数の残基を分析して、以下のアミノ酸を同定した:A16、R17、P18、G32、P37、R59、およびS64。マウスRANTESの3つの突然変異体を、そのような位置にヒト残基を導入するために、部位特異的突然変異誘発によって作製した:[S16A/L17R/A18P];[S32G/L37P]、および[Q59R/Y64S]。マウスRANTESのこれらの突然変異体形態を発現させ、実施例1に記載したようにインビボでビオチニル化させた。マウスRANTESのこれらの変異体を、ストレプトアビジンでコーティングしたプレート(Streptawell,Roche)の中に捕捉した。ビオチニル化ケモカインのコーティングを、抗マウスRANTESポリクローナル抗体(R&D Systems)を使用して確認した。その後、これらの残基の導入によりマウスRANTESに対する1E4の結合を回復できたかどうかを試験した。簡単に説明すると、マウスRANTES、ヒトRANTES、ならびにマウスRANTESの3種類の突然変異体形態と、対照上清を、室温で30分間、Streptawellプレート(Roche)中に補捉した。洗浄後、抗体1E4を1%のBSA−PBS中の1μg/mlの濃度で添加し、室温で1時間インキュベートした。プレートを洗浄し、西洋ワサビペルオキシダーゼに結合させられたヤギ抗ヒトIgG Fcγ特異的抗体(Jackson)とともにインキュベートした。洗浄後、シグナルをTMB(Roche)を用いて明らかにし、H2SO4で停止させた。プレートを450nmで読み取った。図7に示すように、[S16A/L17R/A18P]突然変異体は、マウスRANTESに対する1E4の結合を回復し、このことは、A16、R17、およびP18がヒトRANTESの1E4エピトープの完全性に重要であることを示している。
1E4の親和性と結合速度
ヒトRANTESとカニクイザルRANTESに対する1E4の親和性と結合速度を、Biacore 2000機器(Biacore AB,Uppsala,Sweden)上で特性決定した。433RU(反応単位)のロバ抗ヒトIgGポリクローナル抗体を、C1 Biacoreチップ上にEDC/NHS化学反応によって固定化した。この表面を、抗体1E4を捕捉するために使用した。この表面を、10mMのグリシン(pH=2)の30μL/分での注入、その後の、HBS−EP緩衝液中で1分の安定化時間によって、それぞれのサイクルの後に再生させた。
虚血の動物モデル
材料および方法
動物:8週〜12週齢のC57BL/6マウスを実験に使用した。全ての動物実験は倫理審査委員会(local ethical Committee)によって承認された。
外科手術:マウスを、最初に4%のイソフルオランで麻酔し、その後、挿管した。人工呼吸を、人工呼吸器(rodent respirator)(モデル683;Harvard Apparatus)を使用して、1分あたり120回の呼吸(120 breaths)で300μLの1回換気量で行った。.麻酔は、酸素吸入器(ventilator)によって100%送達される2%のイソフルランで維持した。開胸術を第4肋間胸骨で行い、その後、心膜を取り除いた。8−0 Proleneでの縫合は、左心房の内縁で左冠動脈の下を通し、閉塞を生じるようにスリップノット(slipknot)で結んだ。
虚血再潅流モデルにおける阻害性RANTESの影響
モデル1:虚血再潅流
マウスの虚血再潅流モデルのプロトコールを説明している図を、図8に示す。このプロトコールでは、B6マウスを3つのグループに分け、媒体対照(PBS)、アイソタイプ対照(実施例10に記載したmAb 64)、またはラット抗mRANTESモノクローナル抗体を、以下のスケジュールにしたがって投与した:
グループ1:PBSを、再潅流の5分前に、i.p.またはi.v.で投与した;
グループ2:ラットIgG2a(mAb 64;アイソタイプ対照)を、再潅流の5分前に、i.p.(1mg/マウス)またはi.v.(1.0mg/マウス)で投与した;
グループ3:ラット抗マウスRANTES(mAb 478)を、再潅流の5分前に、i.p.(1mg/マウス)またはi.v.(0.1、0.3、0.5、1.0mg/マウス)で投与した。
