Description
本発明は、HSV感染症の治療および予防のために用いることができる分子、組成物、および方法に関する。より詳しく述べると、本発明は、HSV特異的T細胞、特にCD8+ T細胞の抗原特異性を有する方法、分子、および組成物を開発するために用いることができるHSVタンパク質のエピトープを同定する。
細胞性免疫応答は、ヒトにおける重度の再発性HSV感染症を制限するために必要である。初回の性器HSV-2感染症は持続して重度であるが、再発は重症度が低く、よりしばしば無症候性である。原発性HSV-2感染症の緩解は、CD8+ 細胞毒性Tリンパ球(CTLs)を含む抗原特異的T細胞の浸潤に関連している。ヒトにおける再発性HSV-2感染症の連続的な病変部の生検研究から、病変が成熟するにつれてCD8+優勢へとシフトすること、および局所的CTL活性がウイルス排泄と相関することが示されている(コエル(Koelle, DM)ら、J. Clin. Invest. 1998、101:1500〜1508(非特許文献1);カニンガム(Cunningham, AL)ら、J. Clin. Invest. 1985、75:226〜233(非特許文献2))。このように、CD8+ CTLによって認識されるHSV抗原は、新規治療およびワクチンのために用いることができる。
本発明は、HSV抗原、HSV抗原を含むポリペプチド、該ポリペプチドをコードするポリヌクレオチド、ベクター、および該ポリヌクレオチドを含む組換え型ウイルス、該ポリペプチドを提示する抗原提示細胞(APC)、HSVに対して産生された免疫細胞、ならびに薬学的組成物を提供する。薬学的組成物は、予防的および治療的のいずれにも用いることができる。本発明の抗原はヘルペス病変から採取したT細胞によって認識される。本発明はさらに、HSV感染症を予防および治療する方法、抗ウイルスおよび/または免疫調節リンフォカインの分泌を増強する方法、ならびにHSV特異的抗体産生を増強する方法を含む方法を提供する。HSV感染症を予防および治療するため、抗ウイルスおよび/または免疫調節リンフォカインの分泌を増強するため、HSV特異的抗体産生を増強するため、および一般的にHSV-特異的免疫を刺激および/または増強するために、本方法は、本発明のポリペプチド、ポリヌクレオチド、組換え型ウイルス、APC、免疫細胞または組成物を被験者に投与することを含む。HSV感染細胞を死滅させる方法、およびウイルス複製を阻害する方法は、HSV感染細胞を本発明の免疫細胞に接触させる段階を含む。本発明の免疫細胞は、本発明の抗原、または本発明の抗原を提示するAPCによって刺激される細胞である。そのような免疫細胞を産生する方法も、本発明によって提供される。本方法は、本発明の抗原を提示するように改変されたAPC、特に樹状細胞に免疫細胞を接触させる段階を含む。好ましい態様において、免疫細胞はCD4+またはCD8+ T細胞のようなT細胞である。
本発明は、HSV感染症の予防および治療のために有用であるHSV抗原を提供する。ヘルペス領域に由来するT細胞によって認識されることが確認された抗原および/またはその構成エピトープを本明細書において開示する。幾つかの態様において、本発明の抗原に対する特異性を有するT細胞は、ウイルス感染細胞に対して細胞障害活性を示した。T細胞の特異性に関与する免疫原性抗原の同定によって、改善された抗ウイルス療法および予防方法の開発が容易となる。抗原または本発明の抗原をコードするポリヌクレオチドを含む組成物により、HSV感染症を予防および治療するために有効にターゲティングされるワクチンが提供される。
本出願において用いられた全ての科学技術用語は、特に明記していなければ当技術分野で一般的に用いられている意味を有する。本出願において用いられるように、以下の用語または句は、明記された意味を有する。
一つの態様において、本発明はポリペプチドがICP0もしくはUL47タンパク質またはその断片を含む、単離された単純ヘルペスウイルス(HSV)ポリペプチドを提供する。一つの態様において、断片はICP0の92位〜101位のアミノ酸またはその置換変異体を含む。別の態様において、断片はUL47の289位〜298位のアミノ酸、548位〜557位のアミノ酸、550位〜559位のアミノ酸、551位〜559位のアミノ酸、および/もしくは551位〜561位のアミノ酸またはその置換変異体を含む。アミノ酸残基に関しては、ドラン(A. Dolan)ら(1998、J. Virol. 72(3):2010〜2021)に記述のHSV-2ゲノムのタンパク質を参考とする。
本発明は、本発明の1つまたは複数のポリペプチドをコードするポリヌクレオチドを提供する。ポリヌクレオチドはベクターに含めることができる。ベクターはさらに、本発明のポリヌクレオチドに機能的に結合した発現調節配列を含む。幾つかの態様において、ベクターは、関係する他の分子をコードする1つまたは複数のポリヌクレオチドを含む。一つの態様において、本発明のポリヌクレオチドとさらなるポリヌクレオチドとを、融合タンパク質をコードするように結合することができる。
本発明は、HSV感染症を治療および予防するために有用である組成物を提供する。組成物はウイルス複製を阻害するために、そしてウイルス感染細胞を死滅させるために用いることができる。一つの態様において、組成物は薬学的組成物である。組成物は、本発明のポリペプチド、ポリヌクレオチド、組換え型ウイルス、APC、または免疫細胞の治療的または予防的有効量を含みうる。有効量は、例えばT細胞を活性化することによって、免疫応答を誘起または増強するために十分な量である。T細胞の活性化の一つの測定法は、D.M. コエル(Koelle)ら、1997、Human Immunol. 53:195-205)の記述のような細胞毒性アッセイ法である。幾つかの態様において、組成物はワクチンである。
治療には予防と治療法が含まれる。予防または治療は単回直接注射を1回または多数回行うことによって達成される。投与はまた、複数の部位にほぼ同時に行うことができる。患者または被験者には、ヒト、ウシ、ウマ、イヌ、ネコ、ブタ、およびヒツジ動物のような哺乳類が含まれる。好ましくは、患者または被験者はヒトである。
従来の技術を用いて、同定されたHSV抗原のインビボ有効性を確認することができる。例えば、一つの技術はマウスチャレンジモデルを利用する。しかし、当業者はこれらの方法が定常的なものであり、他のモデルを用いることができることを認識すると思われる。
本発明は、被験体におけるHSV感染症を治療および/または予防する方法を提供する。本方法は、本発明の組成物を被験体に投与することを含む。組成物は治療的または予防的ワクチンとして用いることができる。一つの態様において、HSVはHSV-2である。または、HSVはHSV-1である。本発明はさらに、HSV複製を阻害する、HSV感染細胞を死滅させる、抗ウイルスおよび/または免疫調節活性を有するリンフォカインの分泌を増加させる、ならびにヘルペス特異的抗体の産生を増強する方法を提供する。本方法は、例えば、本明細書に提示した実施例に記述のように、HSV感染細胞を、本発明の抗原に対して産生された免疫細胞に接触させる段階を含む。接触させる段階は、インビトロで行うこともインビボで行うこともできる。好ましい態様において、免疫細胞はT細胞である。T細胞には、CD4およびCD8 T細胞が含まれる。本発明の組成物は、免疫病理疾患に対して認容される物質として用いることができる。
以下の実施例は、本発明を説明するため、そして当業者が本発明を実施および用いる際に役立つように提供する。実施例はいずれにせよ本発明の範囲を制限するものではない。
本実施例は、特異的CD8 CTLが性器HSV-2病変に局在することを証明する。これは、インサイチューで抗原/APCに遭遇しているが、読みとりアッセイ法の前にインビトロで抗原によって再刺激されていない細胞を用いた、再発性の性器HSV-2病変の連続病変生検試験によって示される。
病変浸潤リンパ球(LIL)を、50 μMアシクロビル(ACV)の存在下で植物性赤血球凝集素(PHA)およびIL-2によって1サイクル増殖させた。典型的に、細胞5×106個〜5×107個を2週間後に得た。これらのバルク集団の表現型は、既に記述されている(コエルら(Koelle, DM)、J. Clin. Invest. 1998、101:1500〜1508)。TCR αβ、CD3+細胞では、病変部が成熟して培養が陰性となるに従って、CD8優勢となるように徐々にシフトする。
局所反応は、症状の2日目という早期での、K562および同種異系HSV-2感染リンパ球継続株(LCL)の溶解によって決定されるように、高レベルのNK-細胞活性を有した。NK細胞は、正常な皮膚と比較して病変から増殖させた細胞において選択的に濃縮された。HSV特異的CD4細胞も同様に早期に濃縮した。病変部は「Th1」(インターフェロン-γ(IFN-γ)、IL-12 p40、IL2)および「Th2」(IL-4、IL-5、IL-10、IL-13)mRNAの双方において濃縮された(ファンフーリス(Van Voorhis, WC)ら、J. Infect. Dis. 1996、173:491〜95)。病変浸潤HSV-2特異的CD8 CTLのサイトカインパターンにはインターフェロン-γが含まれる。
溶解は、エフェクター:標的(E:T)比が20:1またはそれより低い割合での特異的放出百分率である。
1HSV-2ゲノムの標準マップ内のエピトープの位置(ドラン(Dolan, A.)ら、J. Virol. 1998、72:2010〜21);HSV-2型特異的TCCのエピトープマッピングは、記載のように(コエル(Koelle, DM)ら、J. Virol. 1994、68:2803〜10;コエル(Koelle, DM)ら、J. Virol. 1998、72:7476〜83)HSV-1×HSV-2中間型組換え型ウイルス(IRV)を用いる(プレストン(Preston, VG)ら、J. Virol. 1978、28:499〜517)。境界はおおよそである。
2記載のように(コエル(Koelle, DM)ら、J. Infect. Dis. 1994、169:956〜61)、HSV-2に感染した部分的一致LCLのHLA対立遺伝子制限死滅;タイピング方法によって認められる血清学的またはDNA定義。
3dkRW22。RW22およびRW 1991.22は、1991年に被験者RWに由来するT細胞クローンを意味する。「22」という名称が与えられた2つのクローンを1991年にRWから個別に得た。本出願を通じて、個別に由来する2つのクローンはdkRW22およびcpRW22によって区別される。
粘膜免疫応答をPBMCから分離して、HSV特異的CTLがマウス粘膜に局在することは、膣接種に対する保護に関連する。本実施例は、CD8+細胞を含むHSV特異的T細胞が子宮頚部リンパ球において検出されうることを証明する。
本実施例において、原発性性器HSV-2感染症に一致した症状を呈する患者から生検標本を採取した。採取した細胞の表現型を決定し、標本からのLILおよびPBMCに増殖および細胞毒性アッセイを行った。結果は、原発性性器HSV-2病変に存在するCTLのHSV特異的増殖および細胞障害反応が、再発性疾患の際に検出される反応に典型的であることを示している。
1バルク細胞は、APCとしての放射線照射自己PBMC(105個/ウェル)と共に104個/ウェルで用いた。結果は4日目での3Hチミジン取り込みのcpm平均値を表す。15日目PBMCは、生存細胞105個/ウェルで用いた。
2バルク細胞は、表示のようにMOI 10で18時間感染した自己由来(au)またはHLA不一致(al)LCLに対して、51Cr放出においてエフェクター:標的比 20:1で用いた。CD8+細胞はミディマクス(MidiMacs)(登録商標)(ミルテニ(Miltenyi))によって濃縮した。結果は特異的放出百分率であり;自然放出は<22%であった。
本実施例は、発現クローニングによって、ICP0の抗原性を証明する。特に、ICP0のアミノ酸92〜101位内のエピトープを同定する。さらに、ICP0の抗原性をワクシニアを用いて確認する。アミノ酸の番号付けは、ドランら(Dolan、J. Virol. 1998、72:2010〜21)の命名および番号付けを用いる。
ブーン(Boon)らのCos-7発現クローニング法を、発現クローニングのために用いた(デプラエン(De Plaen E)ら、レフコビッツ(Lefkowits, I.)編、「免疫学方法マニュアル(Immunology Methods Manual)」第2巻、ニューヨーク、アカデミックプレス、1997:691〜718)。インターフェロン-γ分泌は、洗浄した自家LCL刺激体(MOI 10で18時間の偽感染またはHSV-2感染)1×104個および反応TCC5×104個をTCM 200 μlにおいて1試料あたり3本ずつ24時間播種することによってCD8 T細胞読み出しとして試験した(ティゲス(Tigges, MA)ら、J. Virol. 1992、66:1622〜34)。
;KpnIおよびXba I部位に印をつけた:それぞれ、配列番号:4および5)を消化して、pcDNA 3.0にクローニングして、配列決定した。これはB* 4501に関してゲンバンク61710と同一であった。発現は、FITC標識、対立遺伝子特異的mAb B12(ワンラムダインク)によってチェックした。48時間目に、フローサイトメトリーによって、トランスフェクトしたCos-7の40%が表面HLA B45を発現したのに対し、ベクターに関しては<1%であった。CD8 TCC RW3およびdkRW22の制限対立遺伝子(表3)であるHLA A*0201を、同様にクローニングして、発現をmAb MA2.1によって証明した(マクミカエル(McMichael, AJ)ら、Human Immunol. 1980、1:121〜29)。
HSV特異的CD8クローンは全て、特異的にIFN-γを放出した(表3)。さらに、HLA制限に関する確認試験としてのインターフェロン-γアッセイ法の用途を調べた。クローンRW51は、HLA B*4501をトランスフェクトしたCos-7細胞に曝露した後HSV-2を感染させた細胞では、インターフェロン-γを特異的に放出したが、A*0201をトランスフェクトした細胞では放出しなかった(表3)。
Cos-7細胞7×103個に対するTCC5×104 個の反応を24時間でチェックした。
;配列番号:19)はCTL(図4)およびインターフェロン-γ(表4)アッセイ法において活性であった。その他の候補エキソン2ペプチドは活性ではなかった。高いEC50値(〜1μM)は、ペプチド結合溝の外側に存在すると予想されるカルボキシ末端テールによる可能性があり、HLA B*4501に対する結合を減少させる可能性がある。ラウス(B. Rouse)のワクシニア-ICP0(マニカン(Manickan, E)ら、J. Virol. 1995、69:4711〜16)を増殖させて、力価を測定した(コエル(Koelle, DM)ら、J. Virol. 1994、68:2803〜10)。クローンRW51は、vac-ICP0標的を特異的に溶解した(図4)。ワクシニアを利用できることは偶然であり、発現クローニングの結果を確認する必要がない。エピトープの範囲を狭めるために、ICP0(AERQGSPTPA;配列番号:6)のアミノ酸92〜101位を含むペプチドを合成した。このペプチドのIC50は、1〜10ナノモルのあいだである(図5)。
本実施例は、発現クローニングによってタンパク質UL47のコード領域内に含まれるDNAによってコードされるHSVポリペプチドの抗原性を証明する。発現クローニングおよびライブラリー調製は実施例4に記載した通りであった。
抗原タンパク質に関してライブラリーをスクリーニングするために、平底マイクロタイタープレートに播種した(9,000個/ウェル)Cos-7細胞に、24時間後にライブラリーのプールおよびA*0201 DNA(50および25 ng/ウェル)を同時トランスフェクトした。クローニングしたT細胞を48時間後に加え、インターフェロン-γアッセイ法を実施例4に記載したように、24時間上清について実施した。pCNA 3.1-his Cからのライブラリーに1つの陽性プールを認めた。このプールからの微生物を播種して、次のラウンドのアッセイのためにコロニー96個からDNAを調製した。C1F1C7と呼ばれる一つの陽性クローンを、インターフェロン-γ放出アッセイ法の追跡ラウンドにおいて認めた。ウイルス挿入断片の配列決定によって、それがHSV-2ゲノムのヌクレオチド102875〜101383位からの1.4 kbのSau3aI断片であったことが判明した。配列は、アミノ酸292〜696位までのHSV-2 UL47のC-末端領域、短い介在領域、さらにHSV-2 UL46のN-末端アミノ酸70個をコードする。

(3'プライマー;配列番号:10)であった。それぞれの場合において、クローニングを容易にするために、5'プライマーは、組み込まれたBamH I部位(下線)を含み、3'プライマーは組み込まれたXba I部位(下線)を含んだ。
材料と方法
細胞株とウイルス:
EBV-LCLを施設内でPBMCから調製し;ARENT、PITOUT、HERLUF、およびKAS011はネポム(G. Nepom)から得た。HSV-1 E115、HSV-2 333、HG52、組換え型vac-ICP0-HSV-2(ロウズ(B. Rouse)提供)および野生型ワクシニアNYを産生させ、Vero細胞またはBSC-40細胞において力価を測定した。
HSV-2 ゲノムDNAをSau3A Iによって消化して、再抽出し、かつクレノウ断片、dTTP、およびdCTPによって部分的に埋めた。プラスミドpcDNA3.1(+)myc-his A、B、およびC(インビトロゲン社)をXhoIによって消化して、dATPおよびdGTPによって部分的に埋めた。ライゲーションの後、DNAを大腸菌(E. coli)株DH10Bに電気穿孔した。各ライブラリーは、一次形質転換体数千個を有した。各ライブラリーの大多数はバルクで直ちに増幅させて(4ml LB-amp、一晩)少量ずつ小分けした。ランダムクローン20個はそれぞれ、単一のHSV-2 Sau3A I断片を含んだ。トランスフェクションのためにDNAを作製するために、深い96ウェルプレートに、〜15コロニー/ウェルでライブラリーを、または選択された個々のクローンのいずれかを接種した。一晩増殖させた後、DNAを96ウェルフィルターによって調製した。
Xba I部位を下線で示す;配列番号:11)は、ICP0ゲノムクローンにおけるHSV-2 DNAの3'末端の配列に由来するものであった。MMLV逆転写酵素を用いた。スプライシングを調べるため、PCRは、pfuポリメラーゼ、cDNA、上記の3'プライマー、および5'プライマー
(KpnI部位;配列番号:12)を用いた。ICP0のエキソン1を単離するために、PCRは同じ5'プライマーと3'プライマー
(Xba I部位;配列番号:13)を用いた。産物はpCDNA3.1-his-Bにクローニングした。
CTLアッセイ法は、標準的な4時間の51Cr放出によって行った。標的EBV-LCLをMOI 10でHSVに18時間感染させ;エフェクター:標的比は20:1であった。抗クラスI mAb W6/32を10 μg/mlで用いた。ウイルスRNA発現の阻害作用を調べるために、アクチノマイシンDを5μg/mlで、感染前、90分間感染時、洗浄時、およびアッセイ期間にわたって30分間使用した。
リンパ球を、標準的な方法によってCD3、CD4、CD8、CD16/56、TCRαβ、またはTCRγδに対する標識mAbによって染色した。トランスフェクトしたCos-7細胞におけるHLA発現を測定するために、トリプシン処理した細胞をHLA B*4501に対して反応性のFITC-標識mAb B12(ワンラムダインク(One Lambda, Inc))1μg、またはHLA A*0201に対して反応性のmAb MA2.1細胞上清と共に混合した後、FITC-標識ヤギ抗マウスIgGと混合した。
HLA B44対立遺伝子を定義するため、多様なエキソンの直接配列決定を行った。
γ-インターフェロンは、エンドゲン社の試薬を用いてELISAによって測定した。プレートを0.25 μg/ml捕獲mAb M700A-E 100 μlによってコーティングして、1%BSAの0.2 M NaCl、3mM KCl、0.05 Mトリス、pH 9(TBS)溶液によって1時間ブロッキングした。後のインキュベーションは各100 μlであり、その前にPBS/0.2%ツイーン-20によって3〜5回洗浄し、室温で回転させて行った。0.1%BSA、0.05%ツイーン20、および4μg/ml免疫グロブリン阻害試薬#6LD1068(バイオレクラメーションインク、イーストメドウ、ニューヨーク州)を含むTBS(試料緩衝液)によって希釈した試料および標準物質を2時間加えた。試料緩衝液において100 ng/mlとなるように希釈したビオチン結合検出mAb(M701B)を1時間加えた。1%BSA、0.05%ツイーン20を含むTBS中で100 ng/mlとなるように希釈したアビジンD:HRP(A-2004)を1時間加えた。TMB基質を10分間加えた。検出下限は2〜10 pg/mlの範囲であった。
CD8+ T細胞クローンcpRW22(dkRW22と同じ起源に個別に由来する)を、HLA-A2に結合すると予想され、HSV-2遺伝子UL47に由来する一連の合成ペプチドに対して調べた。これらのペプチドの一つはIFNγELISPOTアッセイ法においてcpRW22によって陽性に認識された。陽性と採点されたUL47ペプチドの配列は、
(配列番号:18)であり、このペプチドはUL47のアミノ酸548〜557位を含む。

天然に加工されるUL47ペプチドを決定するために、C1R-A2/3D9.6H7細胞1.5×1010個からA2分子を精製して、結合したペプチドを酸溶出によって剥離させた。これらのペプチドを以下の条件下でHPLCカラム上で分画した:TFAイオン対形成剤;0〜10%アセトニトリル(ACN)を5分、10〜45%ACNを50分、45〜60%ACNを5分。これらの画分が、IFN-γELISPOTアッセイ法においてcpRW22T細胞による認識に関してT2標的を感作できるか否かを調べた。標的は、血清不含培地+HuB2M3μg/ml中で各画分の5%によって32℃で4時間パルスしたT2細胞(20,000個)であった。次に、標的を2回洗浄し、ELISPOTプレートのウェル2個ずつ(10,000個/ウェル)に移した。反応体はCTLクローンcpRW22(20,000個/ウェル)であった。
(配列番号:1)である。UL47/550〜559ペプチドをコードしうるHSV-2の遺伝子断片はC1R-A2/3D9.6H7細胞に含まれることがその後確認された。これは、pBIBレトロウイルスベクターのクローニング部位の隣接領域に対して、およびUL47/550〜559をコードするDNA配列(図17)に対して作製したプライマーを用いたPCRを行うことによって実施した。これらのPCRプライマーおよび多様なPCR条件を用いて、C1R-A2/3D9.6H7細胞は、HSV-2に由来する少なくとも2個のレトロウイルス挿入断片を含むことが証明された(図18A〜C)。一つの挿入断片はUL52遺伝子の2つの断片をコードする。第二の挿入断片は、UL47/550〜559ペプチドをコードする部分を含む、UL47遺伝子の大きい部分をコードする。
本実施例は、病変由来材料を用いてHSV特異的CD8 CTLによって認識されるさらなるタンパク質を同定することができる方法を示す。
直腸周囲、臀部および/または大腿部皮膚を清浄して麻酔後に、それらから穿孔生検(3〜4mm)を採取した。疑われる原発性疱疹からの病変を可能な限り速やかに生検を採取して、初回感染時に少なくとも2回の連続生検を行うことが好ましい。治癒段階での再発性の性器HSV-2病変(後期潰瘍/痂皮)は、そのような病変からのLILは高いCTL活性を有することから、抗原/エピトープ発見にとって好ましい。病変の一部は、免疫組織学のためにOCT培地においてイソペンタン/液体窒素中で瞬間的に凍結することができる。生検の一部を解離させて、細胞を限界希釈によって増殖させ、その一部をバルク培養に用いた(コエル(Koelle, DM)ら、J. Clin. Invest. 1998、101:1500〜1508)。LILは、組織を細切することによってバルクで増殖させ、48ウェルプレートにおいてアシクロビル(50 μM)を含むT細胞培地中で0.8 μg/ml PHAおよびフィーダーPBMC 7.5×105個/ウェルによって刺激した。増殖はIL-2(50 U/ml、ヘマゲン社(Hemagen))によって補助し、通常14〜21日で細胞1〜5×107個を生成する。CD8選択細胞のCTL活性を、エフェクター:標的比20:1の4時間の51Cr放出アッセイ法において自己由来および同種異系感染LCL偽ならびにHSV-2感染LCLに対して調べる(ティゲス(Tigges, MA)ら、J. Virol. 1992、66:1622〜34)。この段階で溶解活性があれば、HSV特異的CD8 CTLクローンの回収が予測される。
候補培養物の好ましいスクリーニングは、HSV-2に感染した(18時間、MOI 10)、または感染していない自己由来LCLに対する分割ウェルCTLアッセイ法である。LCLは、EBV形質転換B細胞株であり(コエル(Koelle, DM)ら、J. Clin. Invest. 1993、91:961〜68;ミラー(Miller, G.)ら、Proc. Natl. Acad. Sci. USA 1972、69:383〜87)、これはPBMCから確立するために約6週間を要する。LCLはHSV感染に対して許容的であるが、皮膚線維芽細胞と比較すると、HSV媒介HLAクラスIのダウンレギュレーションに対して比較的抵抗性である。ほとんどの被験者は、登録してLCLを調製してから生検を行う。したがって、LCLは、TCCをスクリーニングに準備できる際には使用可能であると思われる。好ましくは、自己由来HSV-2はVero細胞上で単離、増殖、および力価を測定される(コエル(Koelle)ら、1993、上記)。
スクリーニングアッセイ法において陽性と採点されたT細胞は、リッデルら(Riddel、Nature Medicine 1996、2:216〜23;米国特許第5,827,642号)の方法によって増殖させる。当初の微量培養ウェルにおける細胞の「残りの」半数(〜5×104個)を25 ml T細胞培地において放射線照射(3300 ラド)混合同種異系PBMC 2.5×107個、放射線照射(8000 ラド)LCL 5×106個、および30 ng/ml mAb OKT3(抗CD3)と共に混合する。24時間目と、その後週に2回、rhIL-2(50 U/ml、カイロン社、エメリービル、カリフォルニア州)を加える。OKT3は4日目に洗浄して除去する。典型的に、T細胞は最初のサイクルの終了時に細胞約1〜5×107個まで増殖する。増殖が約12日で目に見えて遅くなれば確認CTLアッセイを行うことができる。同一の第二のサイクル後に保存された細胞数は、さらに200〜1000倍の増殖が通常起こるため、本質的に無制限である。増殖した細胞の融解した少量をCTL、増殖、およびサイトカインアッセイ法に用いる。クローンの約10〜20%が増殖できず;抗原特異性の喪失はまれであるが、複製能の喪失は起こる可能性がある。
エピトープマッピングは、分子、生体情報、および合成方法によって行うことができる。ゲノムライブラリースクリーニング(上記)は、アミノ酸25〜300個の範囲である遺伝子断片を初回「陽性」として生じる。陽性分子クローンにおけるHSV-2コード配列は、HSV-2挿入断片の入れ子状態の切断、またはHSV-2 DNAの内部制限部位での切断、およびプラスミドの再構築を行うために、エキソヌクレアーゼIII消化のような標準的な方法を用いて短くすることができる(ガビン(Gavin, MA)ら、J. Immunol. 1993、151:3971〜80)。陽性構築物の一部を再増幅するために校正機能を有するポリメラーゼによるPCRを用いることが好ましい。切断は、長さアミノ酸50〜100個の陽性断片のために設計される。モチーフマッチングペプチドに関して、「Pマイナス1」の位置でのN-末端ペプチドがしばしばTCRの「上部」に面し、T細胞誘発を必要とするため、P1「アンカー」は合成ペプチドの残基2に存在する。モチーフが不明の場合、5個重なり合う15量体を作製する。ペプチドはCTLおよび/またはインターフェロン-γアッセイ法において1 μMおよび10 μMで調べる。
本実施例は、他のHLA-B45陽性供与体が先に定義されたICP0 92-101ペプチドに対して検出可能なCD8+ T細胞反応を有することを証明する。
結果は、エフェクター:標的比10:1〜20:1での4時間CTLアッセイ法における特異的放出百分率である。
11μg/ml ICP0 92-101を90分ローディングした、またはMOI 5でHSV-2に18時間感染した標的LCL(RW=B*4501、HV=B*4501以外)。

本実施例は、他のHLA-A2陽性供与体がこれまでに定義されたUL47ペプチド550〜559に対して検出可能なCD8+ T細胞反応を有することを証明する。
;配列番号:20)特異的A2制限クローンHV2は、供与体HV5101に由来した。このように、RW1874とHV5101のPBMCにおいてUL47特異的CD8+ T細胞が検出されることが予想された。PBMCをインビトロで、3つのA2制限エピトープの一つの1μg/mlによって2回刺激した:インフルエンザM1/58-66、UL47/289-298、またはUL47/550-559。次に、T細胞を、ペプチドなし、刺激ペプチド、またはHIVに由来する対照ペプチド(RT)のいずれかをパルスした標的に対する51Cr放出アッセイ法において調べた。調べた供与体 は全てHIV陰性であることが知られていた。
