脳卒中からの脳の回復は、患部への血液供給の回復(血管新生)およびニューロンの回復(神経新生)を含む複雑なシステムである。血管新生のために、内皮細胞の増殖が、早くも12〜24時間で起こり得る。その後、これらの新たに生成された細胞は、血管内皮細胞成長因子(VEGF)および血小板由来成長因子(PDGF)などの多くの内皮細胞マイトジェンに応答して虚血脳領域の方へと遊走し、そして3〜7日後には梗塞周辺皮質において新たな血管を形成する。その後、血管新生は、少なくとも21日間続く。重要なことには、げっ歯類において脳卒中後の血管新生を増加させる薬物であるナイアスパンは、脳卒中からの機能的な回復を改善し、このことは、脳卒中後の脳の血管新生の増強により、脳卒中の予後は改善され得ることを示唆する。さらに、脳の血管新生は、血液脳関門の形成、ニューロングリア環境への包埋、異なるマトリックスレセプター発現、および発生後の血管新生が比較的に難しいことを含む、脳の脈管構造の独特な特性に因り、脳以外の血管新生とは異なっている。

内皮細胞

ドメインVおよびエンドスタチン
ヒトエンドスタチンを、Cell Sciences, Inc.(Canton, MA)およびSigma Chemical(St. Louis, MO)から購入した。ドメインV(DV)を、ベクターpSecTag2A(Invitrogen, Carlsbad, CA)にクローニングし、そしてトランスフェクションされた293FT(ATCC, Manassas, VA)細胞から、そのC末端6×HisタグおよびNi−ATAアガロースビーズ(Qiagen, Valencia, CA)カラムクロマトグラフィーを介して精製した。透析したDV(PBSに対して)の純度を、抗DV抗体(R&D systems, Minneapolis, MN)および抗his抗体(EMD Chemicals, Gibbstown, NJ)を用いてのSDS−PAGEおよびウェスタンブロットを介して確認した。以前に実証された抗血管新生性濃度のDV(250nM)およびエンドスタチン(600ng/ml)および熱で失活させた対照(100℃で30分間)を用いての30分間の前処置を、全ての実験について使用した。全ての記載した実験について、N=15である(5つの別々の実験、各々の条件は3重に実施した)。Sigmastatソフトウェアパッケージを用いてのスチューデント独立t検定によって、全ての実験についての統計学的有意性(p<0.05)を決定した。
エンドセリン−1による中大脳動脈in vivo脳卒中モデル
Harlan Sprague Dawleyラット(n=10)に、エンドセリン−1(American Peptide Company, Sunnyvale, CA)またはPBS注入を用いて中大脳動脈閉塞の定位的手術を行なった。ラットを、手術から3日後または7日後に絶ち、そして脳組織をドメインVおよびGAPDHウェスタンイムノブロット分析のために処理した。
In vitroにおける血管新生アッセイ
マトリゲル実験を、記載(7)の通りに実施した。12〜18時間後、細胞を撮影し、そして細管形成をAdobe Photoshop CSを用いて定量した(細管のピクセル/高倍率視野、1つの条件につき10箇所の領域)。細胞の遊走を、製造業者の指示に従って、改変ボイデンチャンバー(NeuroProbe, Gaithersburg, MD)を用いて評価した。I型コラーゲンでコーティングされたポリカーボネート膜を横切って、無血清培地+/−3%ウシ胎児血清の方へと向かう遊走を、6〜8時間後に評価した。MTS溶液と共にVEGF20ng/mlを含む無血清培地(Promega, Madison, WI)中で製造業者の指示に従って48時間後に増殖を評価した。
ニューロン−脳内皮細胞の共培養細管形成アッセイ
生後8日目のラットから単離した大脳顆粒ニューロンを、ラミニン(Invitrogen)でプレコーティングしたチャンバースライドに加え、そして一晩増殖させた。その後、脳内皮細胞を一晩かけて加え、その後、無血清培地中で2〜6時間かけて実験的な処置を行なった。固定した細胞を、共焦点顕微鏡(Zeiss, New York, NY)のために抗フォン・ヴィルブランド因子抗体(Dako, Denmark)または抗TUJ1抗体(Neuromics, Edina, MN)および適切な蛍光色素でタグ化した二次抗体(Jackson ImmunoResearch, West Grove, PA)を用いて免疫染色した。
結果
ピークの梗塞周囲の血管新生は、3〜7日後に始まるので、同じ動物に由来する、脳卒中を起こしたおよび脳卒中を起こしていない対照の大脳半球を、ドメインVレベルについて、脳卒中から3日後および7日後にウェスタンイムノブロットによって調べた(各時点についてn=5)。脳卒中の誘発から3日後に、脳卒中を起こした大脳半球におけるドメインVの530%±20%(標準偏差)の増加を、GAPDHローディング対照に対して正規化しそして光学密度測定法(ImageJ, NIHソフトウェア)によって定量して実証した(スチューデント独立t検定によってp<0.001)(図1A〜1B)。
脳血管新生および神経血管微小環境の形成に対するDVの効果
in vitroにおける脳血管新生におけるドメインVの役割を実証するために、マトリゲル毛細血管アッセイ、細胞遊走アッセイ、および細胞増殖アッセイを含む血管新生アッセイを、微小血管脳内皮細胞およびヒト組換えドメインVを用いて実施する。ニューロン−脳内皮細胞の共培養毛細血管アッセイを使用して、より脳に似た環境における毛細血管形成に対するドメインVの効果を研究し、並びに、神経血管微小環境の形成を研究する。前記の研究は、in vitroにおける焦点性虚血をモデリングするために、酸素およびグルコースを欠乏させた環境(OGD)中で実施する。DVは、有意に、そして用量依存的に、正常な条件およびOGD条件の両方において、脳血管新生、並びにニューロンと内皮細胞との相互作用を増強するだろう。
DVのクローニングおよび精製

内皮細胞増殖に対するDVの効果
内皮細胞の増殖は、血管新生における重要な初期の工程である。内皮細胞に対するドメインVの増殖効果を評価するために、マウスおよびラットの両方に由来する微小血管脳内皮細胞を、種特異的な差異を除外するためと、in vivoにおける脳卒中モデルにおけるラットの使用に対応するための両方のために使用する。ヒト臍静脈内皮細胞(HUVEC、Lonza, Basel, Switzerland)を対照として使用し、そしてヒト脳微小血管内皮細胞(Cell Systems, Kirkland, WA)を使用して、ドメインVの効果が、ヒトにも適用可能であり得るかどうかを決定する。増殖アッセイについて、細胞を、トリプシン0.25%(Invitrogen)を用いて収集し、20ng/mlのVEGFを含む無血清培地中のビヒクル対照または種々の濃度のDV(1〜450nMの濃度を、全てのin vitroにおける血管新生実験について使用)懸濁液を用いて30分間かけて前処置し、そしてその後、5×103個の細胞/ウェルの密度で96ウェルプレートに加える。その後、細胞を、正常な増殖条件下において48時間かけてインキュベーションする。その後、MTS溶液を、1時間かけて加え(20ml/ウェル)、その後、プレート解読機で490nmにおける吸光度を解読する。
内皮細胞遊走に対するDVの効果の決定
血管新生性の内皮細胞が増殖するにつれて、それらは、血管を要求する組織によって放出されるケモアトラクタントに応答して、新たな血液の供給を必要とする組織領域へと向かって遊走し始める。DVが内皮細胞の遊走に影響を及ぼし得るかどうかを評価するために、細胞を収集し、そして改変ボイデンチャンバー(NeuroProbe, Gaithersburg, MD)の上のチャンバーにローディングし(37℃で種々のDV懸濁液濃度を有する適切な無血清培地中での30分間のプレインキュベーションを含むまたは含まない)、そしてI型コラーゲン、ラミニンまたはフィブロネクチンでコーティングされたポリカーボネート膜を横切って、37℃の下のチャンバー中の無血清培地+/−3%FBSに向けて6〜8時間かけて遊走させる。重要なことには、これらの細胞外マトリックスは、それぞれ、脳内皮細胞の中性の、抗血管新生性のおよび血管新生促進性の基質を示し(11)、そしてその使用により、DVの作用がECM特異的であるかどうかを決定することが可能となる。その後、膜(遊走細胞を含む)をアセトンで固定し、そして細胞をクリスタルバイオレット(Sigma)で染色し、そして顕微鏡下で計測する。さらに、DVが、VEGF、脳由来神経栄養因子(BDNF)、神経成長因子(NGF)および塩基性FGF(bFGF)を含む種々のケモアトラクタントへの方への遊走に異なって影響を及ぼすのうかどうかを決定する。
内皮細胞のマトリゲルによって誘発される細管形成に対するDVの効果の決定
一旦、内皮細胞が、新たな血液供給を要求する目的の組織ターゲットに到達すると、細胞は毛細血管形態形成を受けて、新たな血管を作る。マトリゲル細管形成アッセイは、in vitroにおいて内皮細胞毛細血管形態形成を研究するための十分に確立された技術である(12)。この目的を達成するために、これらのアッセイを前記したように実施(種々のDV濃度と内皮細胞とのプレインキュベーションを含むまたは含まない)する。細胞を種々の時点で観察し、その後、12〜18時間後に4%パラホルムアルデヒド中で固定する。標準的な画像取得ソフトウェアを使用して、倒立顕微鏡に取り付けたCCDカメラを用いて画像を獲得する。細管形成を、Adobe Photoshop CSを用いて定量する(細管のピクセル/顕微鏡視野、1つの条件あたり10箇所の視野)。
共培養液中における内皮細胞の細管形成およびニューロンとの相互作用に対するDVの効果の決定
マトリゲルよりも脳に似た環境において脳内皮細胞毛細血管形成をドメインVがモデュレーションする能力を調査するために、大脳顆粒ニューロンと脳内皮細胞からなる共培養アッセイシステムを開発した。生後8日目のラット大脳を、それが脳内皮細胞と相互作用する新生ニューロンの容易に入手可能でかつ豊富な入手源であることからニューロン入手源として選択した(13)。ラミニンに加えて、ニューロンを、コラーゲンIまたはフィブロネクチンでプレコーティングしたウェルに加えて、内皮細胞細管形成に対する各々の基底にある基質の相対的な重要性を決定する。内皮細胞細管形成の程度に対する種々の濃度のドメインVの効果を、種々のインキュベーション期間(1、3、6、9時間)について決定する。細管形成に対するドメインVの用量依存的な増強作用を、実証する。
DVのin vitroアッセイにおける酸素−グルコース欠乏(OGD)の効果
in vitro虚血モデル(15)におけるドメインVの効果を、低酸素チャンバー中の低グルコース培地(1g/L)中の前記の血管新生アッセイを使用して示す。各アッセイについて、低グルコース培地±DV中の培養液を、95%N2/5%CO2を1時間流した低酸素チャンバー中に入れ、その後、各血管新生アッセイについて前記した実験持続時間かけてチャンバーに封をする。OGDのみで、脳内皮細胞血管新生が増加し、そしてこれはさらにDVによって増強される。
脳内皮細胞におけるa5b1インテグリンおよびDVの血管新生促進作用
DVの血管新生促進作用にα5β1が必要であることは、α5β1の遮断および/またはα5β1の減少が、ドメインVの血管新生促進作用を軽減または阻害することを意味する。それ故、遊走、増殖および細管形成アッセイにおける、DVで処置および非処置の脳内皮細胞に対する、それぞれ、機能的遮断抗体およびIgG対照(Millipore、10〜20mg/ml、細胞と共にプレインキュベーション)を用いてのa5b1インテグリンの機能的阻害の効果、並びに、siRNA(Accellの予め設計されたsiRNA、Dharmacon, Chicago, IL)を用いてのα5β1レベルの減少の効果を示す。a5b1ノックダウンは、ウェスタンブロットを介して確認される。
内皮細胞のa5b1の表面局在化に対するDVの効果の決定
DVへの曝露に応答した脳内皮細胞のα5β1インテグリンの表面局在化を直接測定するために、Milner et al. (15)の方法を使用して、CellQuest(Becton-Dickinson)ModFit LT(Verity)ソフトウェアを含むBecton-Dickinson FACSCalibur機器を使用したフローサイトメトリーのために、プラスチックまたは種々の基質(ラミニン、コラーゲン、フィブロネクチンまたは固定したDV、4℃で一晩かけてウェルに40mg/mlで加える)(DVへの曝露を含むまたは含まない)上で増殖させた細胞を調製する。
DVとα5β1インテグリンとの相互作用およびDVによるα5β1インテグリンの活性化の決定
DVがa5b1インテグリンと結合しそして活性化し、その結果、RGDフィブロネクチン/α5β1結合モチーフを介してフィブロネクチンに対する脳内皮細胞の親和性が増加する。固定したフィブロネクチンへの脳内皮細胞の接着増加に対するDVの効果、および、RGD配列GRGDS(配列番号1)vs対照ペプチドSDGRG(配列番号2)を示す。
脳卒中によって生じるDV
DVを脳卒中後に投与し、そして脳卒中の病態および機能的回復に対するその効果を示す。脳卒中後の血管新生は、神経血管微小環境の形成のようにin vivoにおける脳卒中においてDV投与後に増加し、そしてラットにおける病的および機能的脳卒中の予後が、改善する。DV療法は、動物において十分に耐容性がある。
エンドセリン−1によるin vivoにおける脳卒中モデルおよびDV療法
Harlan Sprague Dawleyラットに、前記したようにエンドセリン−1(またはPBS偽対照)によって誘発される中大脳動脈閉塞(MCAo)を行なう。血流中断を、レーザードプラ流速測定計(BPM2モデル, Vasamedics Inc, St. Paul, MN)を使用して確認する(16)(49)。MCAoから24時間後、動物を無作為に2つの群に分け、一方には、腹腔内(I.P.)注入によって2〜6mg/kgのろ過したDV(公開(7)されているように腫瘍血管新生を阻害するに十分な量)の注入を施し(処置群)、そして一方には2週間におよび2日間毎にビヒクル対照を施す。また、MCAoから24時間後に開始して、ラットに、毎日(14日間)、新たに合成されたDNAを標識するためのブロモデオキシウリジン(BrdU;100mg/kg;Sigma)の腹腔内注入を施し、回復中のラット脳における新生ニューロンを同定する。
脳卒中組織の免疫組織化学およびウェスタンイムノブロット
ラットにおける脳卒中後の血管新生および神経血管微小環境の形成±DV療法を評価するために、脳卒中を起こした動物および偽の脳卒中を起こした動物からの新鮮な脳組織を液体窒素中で凍結する。血管新生の免疫組織化学について、Microm HM 550クライオスタット(Walldorf, Germany)を用いて得えた脳卒中を起こした大脳半球の凍結切片を、血管抗原すなわちCD31およびフォン・ヴィルブランド因子に対する抗体(Dako)を用いてプローブする。BrdU+ニューロンと血管との相互作用の程度を評価し、そしてAdobe Photoshop CSを用いて定量する。さらに、脳卒中の組織による注入DVの取り込みおよび分布を、DVの6×HISタグに対する抗体(EMD)を用いて示す。非処置の脳卒中を起こした動物と比較して、DV療法により、脳卒中を起こした脳組織周辺において、血管が増加、すなわちCD31および/またはフォン・ヴィルブランド陽性細胞が増加し、これはまた、α5β1インテグリン発現が増加し、およびこれらの血管とBrdU+ニューロンとの間の相互作用が増加しているであろう。投与したDVは脳卒中部位に局在し、そしてその結果、梗塞サイズが縮小する。
脳卒中によって誘発される脳性麻痺のためのパールカンドメインV療法
胎児期または新生児期の間に起こる脳血管事象である、周産期の虚血性脳卒中(PAS)は、乳児脳性麻痺の主要な原因である。残念なことに、PASは、典型的には遡及的に診断され、すなわち傷害が起こった後に、そして組織プラスミノーゲンアクチベーターなどの伝統的な脳卒中の療法には遅すぎる時期に診断される。それ故、成功裏の脳性麻痺療法は、脳の自己修復機序を活用すべきである。残念なことに、脳の自己修復は十分に解明されていないが、血管再建(血管新生)および神経再増殖(神経新生)の神経血管微小環境において起こるようである。これらの両方の過程には、結果が未知である、プロテアーゼによって駆動される細胞外マトリックス(ECM)の再構築が関与する。脳卒中後の脳血管新生および神経血管微小環境の形成は、一部には、ECMであるパールカンの生物活性フラグメントの生成によって刺激され、そして、この自己修復機序を、PASによって誘発される脳性麻痺療法のために活用する。脳卒中は、急速にパールカン(これは、研究されている最もプロテアーゼに対して感受性の高いECM成分である)の生物活性フラグメントを生成し、そしてパールカンは、血管新生および神経新生の両方に必要とされる。さらに、パールカンフラグメントであるドメインV(DV、またエンドレペリンとしても知られる)は、α2β1インテグリンを通して脳以外の系においては血管新生を阻害することができる。
投与したDVは、脳卒中および梗塞周辺組織に局在化する
脳組織を免疫組織化学によって分析して、投与したDV(組換えDVのc末端上のHISエピトープタグ)並びに血管(フォン・ヴィルブランド因子)を検出して、投与したDVが、脳卒中を起こした脳組織において検出され得るかどうか、そしてそれがどこに局在化し得るかを決定した。図7は、投与したDVが、脳卒中を起こした脳組織に豊富に見られ、そして梗塞周辺皮質においてはより少ない程度で見られたが、同じ動物の反対側の脳卒中を起こしていない皮質には見られなかったことを実証する。さらに、投与したDVの実質的に全てが、梗塞周辺皮質の血管周辺分布域に蓄積し、そしてより少ない程度で脳卒中組織に蓄積し、これは、in vivoにおけるその脳血管促進効果および活性化固形腫瘍の血管周囲にターゲティングするその能力と一致する。重要なことには、このことは、DVが、少なくとも反管腔側の血管周囲領域に限り、以前として不明である機序を介して血液脳関門を通過することができ、そこでアストロサイトおよびニューロンなどの他の細胞型に曝されそして効果を発揮することができることを示唆する。図8は、それぞれPBSおよびDVで処置した脳卒中を起こした動物の、脳卒中を起こした皮質および反対側の皮質から得た、溶解液のウェスタンブロット分析による類似の結果を実証する。
投与したDVは、脳卒中および梗塞周辺組織に局在する
DV療法が、梗塞脳組織のすぐ近くに隣接する脳領域として定義された、梗塞周辺脳組織中を遊走する神経芽細胞の程度に影響を及ぼし得るか、そしてこれにより新たなニューロンを用いての、脳卒中を起こした脳組織の再増殖に影響を及ぼし得るかどうかを決定するために、PBSおよびDVで処置した脳卒中を起こした動物からの脳切片のダブルコルチン免疫組織化学を使用し(図9A)、そしてそれぞれの脳卒中梗塞周辺組織における高倍率視野(HPF、1匹の動物あたりn=20の画像)あたりのダブルコルチンのピクセル平均数を定量した。DV処置動物におけるダブルコルチン陽性ピクセルの有意な増加を認めた。さらに、DVは神経血管微小環境の形成を育成するという仮定に一致するように思われることに、有意に(p<0.001)より高い比率の同定されたダブルコルチン陽性細胞を、梗塞周辺脳組織におけるPBS処置対照(60%+/−3%)と比較してDV処置動物(90%+/−5%、1匹の動物あたりn=20の画像)において、フォン・ヴィルブランド抗体またはHIS抗体に対する抗体によって同定したところ(図9B)、血管のすぐ近くに認めた。
DVは、in vitroにおいて皮質ニューロンの遊走を増強する
前記の結果によって示唆されるように、脳内皮細胞に対する効果を通して神経血管微小環境の形成を育成することによって、DVは、神経保護性であり得、神経新生および神経遊走を増強し得る。さらに、DVが、血液脳関門を通過するならば、それはまたニューロンに対して直接的な作用を有し得る。DVが、脳卒中に対するその治療能力と一致し得る、直接的な神経作用を有し得るかどうかを決定した。この目的を達成するために、胎生15日齢のC57Bl6マウスからのマウス一次皮質ニューロンを、特に髄膜を除去することに注意を払いながら一次細胞単離のための慣用的な無菌的な解剖技術を使用して単離して、そしてin vitroにおける遊走アッセイを、これらの細胞を用いて実施した。
図11は、脳卒中後の3日目以降に、偽対照とドメインVで処置した脳卒中を起こした動物との間に統計学的な差異が全くなかったことを示す。ドメインV処置動物を、脳卒中後1、3、5、7日目に0.5mg/kgを用いて処置した。
ドメインVは、BDNFの遊離を誘発する
本明細書におけるin vivoでの結果は、DVによる処置が、神経血管脳修復を助け、これは、一部には、脳由来神経栄養因子(BDNF)などの神経血管促進因子のDVにより誘発される遊離に起因し得ることを示唆する。図12Aは、24時間後、DVにより、有意に(**p=0.0025)脳微小血管内皮細胞によるBDNFの遊離が増強することを実証する。同様に、脳卒中後のDVによる処置により、PBS処置対照と比較して、脳卒中から3日後および7日後に脳卒中を起こした脳のBDNFレベルが有意に増加した(図12B〜12C)。さらに、一次単離ラットアストロサイトまたは皮質ニューロンへのDVの添加によるBDNFレベルに対する効果は全く見られず、このことは、in vivoにおける脳BDNFレベルに対するDVの効果は、主に、脳内皮細胞により媒介される効果であり得ることを示唆する。

脳微小血管内皮細胞は、以前に同定されたDVレセプターα2β1インテグリンを発現しないので、脳内皮細胞に対するDVの血管新生促進作用は、α2β1の不在および別個の血管新生促進性DVレセプターの存在の両方に起因し得る。α5β1は、血管の発達にとって非常に重要であり、そして脳卒中後の脳の血管新生を促進するが、それ以外では、低酸素症後に脳内皮細胞において再発現されるまでは成熟脳においてはダウンレギュレーションされている。血管新生促進成長因子のアンジオポエチン1の単量体変異体は、α5β1インテグリンに結合し、そしてα5β1インテグリンを介して血管の安定化を促進し、このことは、血管新生および血管再構築におけるこのレセプターの重要性をさらに強調する。
C57マウス脳内皮細胞へのα2β1インテグリンの添加は、ドメインVの細胞増殖活性を逆転させる。DVは、α2β1インテグリン(抗血管新生性レセプター)およびα5β1インテグリン(血管新生促進性レセプター)の両方を発現する大半の内皮細胞において抗血管新生性であるので、DVは、α5β1インテグリン(Kdは160nM)を超えるα2β1インテグリン(Kdは約30nM)に対するその増加した親和性に因りこれらの細胞において抗血管新生性であると考えられた。α2β1インテグリンを、通常はこのレセプターを発現しないマウス脳微小血管内皮細胞にトランスフェクションした。これは、C末端RFP融合タンパク質(Texas A&M University Biomedical Engineering)を含むα2−サブユニットインテグリンをコードする配列を含むプラスミドベクター(pEGFP−N2, Clontech)を用いてのトランスフェクションによって達成され、空のベクターを、対照として使用した。細胞を、抗生物質を全く含まない培地中で24時間の間に回復させた。倒立蛍光顕微鏡を使用して24時間後にトランスフェクション効率を評価した。細胞増殖を、製造業者の指示に従ってMTS溶液(Promega, Madison, WI)を含む無血清培地中で48時間後に評価した。DVにより、細胞培地対照と比較して、マウス脳内皮細胞の増殖は増加し、これは、VEGF中和抗体またはα5β1インテグリン機能遮断抗体の添加によって遮断され得る(図14A)。α2β1インテグリンの細胞への添加により、細胞の増殖は増加するよりもむしろ抑制され、このことは、α2β1インテグリンのマウス脳内皮細胞への添加は、DVを、増殖性から抗増殖性へと変換することを実証する。バーは、平均値+/−標準偏差である(図14B)。**p<0.01。##p<0.01。
結合アッセイを、記載(17)のように光学バイオセンサー(IAsys; Affinity Sensors, UK)を使用して実施した。簡潔に言うと、センサーの表面上の、アクセプターと称されるA5ドメインに共有結合させるために、表面上に存在するカルボン酸基を、0.1MのN−ヒドロキシスクシンイミドと0.4MのN−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(Pierce)の1:1混合物の注入によって活性化した。その後、リン酸緩衝食塩水(PBS)中に溶解したアクセプターを、応答プラトーに到達するまで活性化表面に結合させた。残留活性基を、100μLの1Mのトリス−HCl(pH8.5)の注入によって遮断した。
DVは、in vivoにおいて血液脳関門の透過性に有意に影響を及ぼさない
DVは、in vitroにおいて脳内皮細胞におけるTEERを減少させ得ることが実証されたので(血液脳関門の透過性がすでにマイナスに影響を受けている場合の脳卒中後の潜在的に懸念される作用である)、DVが、in vivoにおいて血液脳関門の透過性に対して任意の作用を及ぼしたかどうかを決定した。
[3H]マンニトール(14.2Ci/mmol)および[14C]スクロース(412mCi/mmol)をMoravek Biochemicals(USA)から購入した。残りの化学品は、Sigma Chemical Company(UK)から購入した。マウスは、Harlan, UKから購入した。
BALB/c系においては、[14C]スクロースおよび[3H]マンニトールによって測定した全ての脳領域における血管空間は、DV(1mg/kg、図15A)の腹腔内注入の8時間後または24時間後に有意に影響を受けなかった。さらに、脳の水分含量は0時間目において73.3±1.3%であり、そしてDV(1mg/kg)の腹腔内注入によって8時間後(64.6±3.4%)または24時間後(72.8±2.9%)に有意に影響を受けなかった(1要因の分散分析)。C57Bl6系においては、[14C]スクロースおよび[3H]マンニトールによって測定した全ての脳領域における血管空間は、DV(1mg/kg、図15B)の腹腔内注入から2時間後に有意に影響を受けなかった。
Claims (8)
- パールカンのドメインVのアミノ酸配列を有するエンドレペリンタンパク質を含む、脳血管の生成を刺激または増強することによる患者の治療のための医薬組成物であって、経口、皮下、静脈内または腹腔内投与される、医薬組成物。
- 医薬組成物が、経口投与される、請求項1記載の医薬組成物。
- 医薬組成物が、患者の体重あたり0.1mg/kg〜約10mg/kgの量で投与される、請求項1記載の医薬組成物。
- 医薬組成物が、腹腔内投与される、請求項1記載の医薬組成物。
- 医薬組成物が、静脈内投与される、請求項1記載の医薬組成物。
- 医薬組成物が、皮下投与される、請求項1記載の医薬組成物。
- 脳卒中回復を改善するための使用のための、請求項1記載の医薬組成物。
- 脳卒中後の血管新生を刺激するための、そして、脳卒中後の神経新生に必要で神経芽細胞遊走に向けられた神経血管微小環境の形成を成長させるための使用のための、請求項1記載の医薬組成物。
