Description
本発明は、医学上有効な化合物及び/又は他の化合物の経口投与のためのナノ粒子キャリアに関する。
(発明の背景)
噴霧乾燥技術が、主に肺疾患の薬物送達のために特定の特性、例えば粒径、密度及び形状を有する医薬粉末の調製における広い用途を見込んできた。それは、懸濁液又は溶液を液滴に霧状にし次いで熱気流中の乾燥処理による固体粉末の製造に精通した方法である。
乾燥方法として主にしばしば考えられるが、噴霧乾燥は、封入方法であって、活性物質がポリマーマトリックス又はシェルに閉じこめられる方法としても使用され得る。いくつかのコロイド系、例えばエマルジョン又はリポソームが、成功裡に乾燥補助剤、特に糖類、例えばラクトース、ソルビトール及びトレハロースを使用してその構造を保護しながら噴霧乾燥されたことが報告される。
噴霧乾燥技術の利点の一つは、費用有効性であること及び広範囲の医薬製品に適用できる速乾性方法であり及び流動性粉末の製造に繋がることであり、非常に低い水含有量、ゆえに活性の劣化を防ぐことを特徴とする。これは、主にこれらのキャリアが関心の部位において活性化合物の送達のために具体的に設計され、ナノスケールの範囲にある場合、長期安定のキャリアの開発に有意義である。
最近、薬物送達分野の液体コロイド系の噴霧乾燥であって、乳酸-グリコール酸共重合体(poly(lactic-co-glycolic acid))(PLGA)に封入されたDNAを含む単独エマルジョン(油中水型エマルジョン)が成功裡に噴霧乾燥されたことが報告された。別の報告は、オレンジオイルを保存する目的で、かつラクトースの存在下、二重エマルジョン(油中水中油又はO/W/O)の噴霧乾燥においてなされ、また両方の場合において製造された粒子がマイクロサイズの範囲にあった。
狭いサイズ分布範囲、典型的に180〜250nmを有する球状ナノ粒子の必要性が認められてきた。理想的にそのような粒子は、実質的になめらかな表面を有しかつ流動性(free flowing)であるべきである。
本発明は、薬物送達のためのナノ粒子キャリアの製造方法であって、該ナノ粒子は以下の工程、(1)該ナノ粒子キャリアの基剤を形成する1種以上のポリマーを含む水-油-水二重エマルジョンを調製する工程、(2)送達される薬物を該エマルジョン相の一つへ配合する工程、(3)油相又は外側の水相を炭水化物でドープする工程、及び(4)該エマルジョンを噴霧乾燥して100nm〜1000nmの狭い粒径分布のナノ粒子を形成する工程、によって製造される方法を提供する。
このように製造されるナノ粒子は、多機能ナノ粒子であり得る。
該炭水化物は、糖類でよい。
該糖類は、二糖類でよい。
該二糖類は、ラクトース、マルトース、イソマルトース、マンノビオース、トレハロース、セロビオースなどでよい。
該糖類は、陽イオン生物分解性の粘膜粘着性多糖類(muco-adhesive polysaccharide)と混合され得る。
該エマルジョンの該油相は、界面活性剤でドープされ得る。
該エマルジョンの該水相は、界面活性剤でドープされ得る。
該界面活性剤は、非イオン性界面活性剤でよい。
該界面活性剤は、アセチレン型ジオール化学をベースにし得る。
該界面活性剤は、ポリマー非イオン性界面活性剤であり得る。
該水相中のポリマー非イオン性界面活性剤は、部分的に加水分解されたポリビニルアルコール(PVA)であり得る。
該ポリマーは、エマルジョンの油相中にあり得る。
該油相中のポリマーは、PLGA(乳酸-グリコール酸共重合体)であり得る。
油相及び水相両方のポリマーが存在し得る。
該薬物は、油相に添加され得る。
該薬物は、内部の水相に添加される親水性薬物であり得る。
該薬物は、疎水性であり得、かつ任意に油相へ添加され得る。
該薬物は、リファンピシン、イソニアジド、エタンブトール、又はピラジンアミドであり得る。
該油相は、ステアリン酸を含み得る。
このように形成されたナノ粒子は、実質的に球状であり得る。
ナノ粒子の粒径分布は、180nm〜250nm直径であり得る。
後に続く態様の記載は、広く解釈されるべきでありまた本発明の範囲を限定するものでない。
この実験に対して、イソニアジド(INH)エタンブトール(ETH)、ピラジンアミド(PZA)及びリファンピシンを含む抗結核抗生物質が、広範囲にキャリアとして使用されるポリDL、乳酸-グリコール酸共重合体(PLGA50:50)、生物分解性及び生物学的適合性ポリマーのポリマーコアシェルナノ粒子に成功裡に取り込まれた。INH(又はEth又はPZA又はRIF)を組み込むPLGAのサブミクロン固体粒子が、典型的な二重エマルジョン水中油中水(W/O/W)を簡単な噴霧乾燥により得られた。
配合において、ラクトース一水和物が噴霧乾燥助剤として使用された一方で、キトサン、陽イオン生物分解性の粘膜粘着性多糖類が吸収促進剤として使用された。PEGがキャリアの生物循環(bio-circulation)を増加させるために混和された一方で、PVAは、二重エマルジョンの主な安定剤成分として考えられた。
サーフィノール104PG-50(商標)は、サイズ分布を著しく狭くさせながら、共界面活性剤(co-surfactant)として粒径をナノサイズ範囲に減少させる大役を果たした。
2.1材料
現場で使用されている抗結核薬をシグマ社から購入した。ポリDL,乳酸-グリコール酸共重合体(PLGA)50:50(Mw:45000-75000)及び85%脱アセチル化した低分子量のキトサンが両方ともシグマ社から供給された。部分的に加水分解された(87-89%)ポリビニルアルコール(PVA)(Mw:13000-23000)もシグマ社から得られた。メルク社から供給されたステアリン酸、ジェミニジオール型界面活性剤、サーフィノール104PG-50(商標)がエアープロダクツ社から供給された。ポリエチレングリコール(PEG)(Mw9000)をBASF化学から購入した。メルク社から供給されたラクトース一水和物を賦形剤として使用した。
ジクロロメタン、酢酸エチル及びアセトニトリルの分析的かつHPLC等級もメルク社から供給された。
2.2.1配合
ナノ粒子の調製を有機溶媒の蒸発に次ぐ二重エマルジョンW/O/Wからの界面ポリマー沈殿反応に基づく方法から達成した。本発明において、溶媒蒸発及び乾燥の工程が、噴霧乾燥技術の適用により一つの工程に組み合わされた。
短く言えば、50mgのINHを2mlのリン酸緩衝溶液(pH7.4)に溶解し、これを8mlの有機溶媒(DCM又は酢酸エチル)に溶解された100mgのPLGA(50:50)の溶液に添加した。任意に2mlの0.2%(w/v)のステアリン酸も同一溶媒(DCM又は酢酸エチル)に溶解され得る。サーフィノール104PG-50(商標)の滴をPLGA油相又はPVAを含む外側の水相に意図的に添加した。
混合物を高速ホモジナイザー(シルバーソン社製L4R)を5000rpmで3分間使用する乳化に被してW/Oエマルジョンを生成した。得られた第一エマルジョンを次いで直ぐに、既知濃度のPVA(1又は2%w/v)、PEG0.5%w/v、キトサン及びラクトース水溶液の水性相容積(aqueous phase volume)へ明示された容積比で注ぎ、高速ホモジナイザー(シルバーソン社製L4R)を8000rpmで5分間用いて乳化し二重エマルジョンW/O/Wを形成した。得られた最終エマルジョンを直接噴霧乾燥器を通じて送り込み、表1に特定される条件を使用してナノ粒子を製造した。
標準ノズル(直径0.7mm)を有するビュッヒミニ噴霧乾燥器モデルB-290(ビュッヒラボ、スイス)を使用して様々な処方の乾燥粉末を備えた。
使用された条件を表1に集めた。
粒径及び粒径分布をマルバーンZetasizer Nano ZS(マルバーンインスツルメント社、英国)を使用する動的レーザー散乱又は光子相関分光法により測定された。各サンプルに対して、必要に応じて、3-5mgの噴霧乾燥された粉末を濾過水中(0.2μmフィルター)に粒子を懸濁し、ボルテックスし及び/又は2分超音波分解することにより調製された。各サンプルを3通りに測定した。
2.2.3ゼータ電位
粒子のゼータ電位をZetasizer Nano ZS(マルバーンインスツルメント社、英国)を使用して測定した。
3mgのサンプルの噴霧乾燥したナノ粒子を1-2mlの脱イオン水に懸濁し及び次いで測定前にボルテックス又は超音波分解した。それぞれの測定を3通り行った。
2.2.4走査型電子顕微鏡
噴霧乾燥したナノ粒子の表面形態学を走査型電子顕微鏡(LEO1525フィールドエミッションSEM)により視覚化した。少量のナノ粒子粉末を真鍮のスタブ(stub)上に両面接着テープを用いて取り付け、またスパッターにより金の薄層を真空塗布した。
2.2.5薬物の組込み
ナノ封入処理の後、粒子粉末中に封入された親水性薬物イソニアジドの量を分光学的方法(UV-Vis、熱スペクトルヘリオス(Thermo Spectronic Helios)α)を使用して3通り測定した。ナノ粒子中のINHの封入有効性を取り込まれたINHと調製に使用されたINHの理論量の質量比として決定された。封入されていない薬物を除去するために50mgの沈殿粒子を20mlの脱イオン水に再懸濁させ、遠心分離し(10000rpm/10C/5分)、上清をUV-Vis分光高度計に被り、INH評価のためにλ=262nmで読み取った。INHの封入量を全初期INH量から上清中のINHを減じることにより決定された。
INH噴霧乾燥した粉末の安定性を光ダイオードアレイ(PDA)検出器を備えるシマヅ社製機械を使用する逆相-高速液体クロマトグラフィー(RP-HPLC)により評価した。
以下の特性を適用した:カラム フェノメネックス(商標)[C18(5μm)、(250×4.6mmID)]、95%(v/v)緩衝液NaH2PO4(pH6.8)を有する5%(v/v)アセトニトリルの移動相、1ml/分の流速及び30℃の温度。この検出は、PDAを使用してλ=259nm、全注入容積20μlにおいて実施された。
3.結果と議論
全ての噴霧乾燥が約220-800nmのサイズ範囲の製造されたナノ粒子に行われた。PVA濃度が1-2%に変化したサンプルが示すように、液体供給の濃度が粒径にいかなる影響を示さなかった。ラクトース及びサーフィノール104PG-50(商標)の添加のみがナノ粒子のサイズ及び形態学に有意な影響を示した。面白いことには、油相に添加されたジェミニ界面活性剤のただ1滴は、有機溶媒の型又はPVAの濃度に関係なく急激に製品のサイズ及びサイズ分布を減じた。
全ての実験セットの中に、温度に加えて噴霧乾燥の全ての他のパラメータが一定に保持された。質量比PLGA:INH(2:1)も変化しなかった。ラクトースの添加はナノ粒子の形状を著しく改良した。この効果は、ジクロロメタンが有機溶媒として使用された場合に目立っていた。
研究された全ての配合に対するこの粉末の収量は、40-70%の範囲であった。
熱分析により決定された選択された試料の残りの水含有量は、非常に低レベルの湿気を示した(〜3%)。
HPLCから得られた結果がもしかするとラクトースとの相互作用のためにINHの減成を示した。この課題が、配合にこれらを組み込む前にラクトースの官能基をキトサンでキャップすることにより克服された。
INHの封入有効性は、約60%である。
二重エマルジョン技術においてほとんど一般的に使用される有機溶媒は、ジクロロメタン(DCM)及び酢酸エチル(EA)である。
従って、我々はナノ粒子のサイズ及び形態学を有機溶媒を変化させることによって観察することにした。すべての場合、酢酸エチルが有機溶媒として使用された場合、得られた第一エマルジョンは一時的に安定なエマルジョンの性状を示し、この観察は、DMCを伴って得たものと比較した場合、エマルジョンのより少ない乳状の外見に基づく。
EA試料は、DCMで調製された試料と比較して非常に不規則な表面形態学を形成した。EAに由来の粒子は、ラクトースの添加前非常にくぼみかつ皺がよっていた。小さいドーナッツ形状の粒子も観察された。
3.2添加剤の効果
3.2.1粒径及び形態学におけるラクトースの効果
ナノ粒子のサイズ及び形状及び表面形態学が、相の組成に強く影響された。ラクトースの初期濃度が5-10%w/v増加したので、これらの粒子は、非常に皺のよったものからほとんど滑面球に変わった。図1C及びDのSEM図に描写されるようにドーナッツ型粒子のフラクションが、使用された溶媒の型にかかわらず目立って減少した。しかし、さらに多くの表面のなめらかさが、観察の目盛りにおいてDCMを伴って観察された。
ラクトースを添加しない配合と比較した時、使用された有機溶媒の型にかかわらず粒径が減少した。この減少は、図2に示される結果に例証されるようにDCMの場合非常に
大きく目立ち、ラクトースが配合に添加された場合、粒子のz-平均サイズは、1200nmより大〜450nmに落ちた。
ゼータ電位が、配合中キトサンの存在のために正の範囲であった。その初期濃度が、0.05、0.1及び0.3%(w/v)の間に変化し、この配合の最適化が、キトサン0.3%を伴ってなされ、これは高い正のゼータ電位〜+45mVとなった。
アセチレンジオール化学に基づく非イオン性界面活性剤が、低表面張力及び良好な脱発泡(de-foaming)及び表面湿潤特性を提供する界面活性剤の独自の類を代表する。
水/空気表面に垂直に延伸する(orient)殆どの界面活性剤と反対に、アセチレンジオール界面活性剤は、その分子構造に起因して水平に延伸する。この界面活性剤の小型分子が、動的表面張力(DST)の低値を提供する界面領域に非常に素早く移動し得る。サーフィノール104PG-50(商標)のバルク濃度2.10-6モル.cm-3に対して、DSTは、約0.00035N.cm-1 (約35ダイン.cm-1)落下したと報告された。実際には、それが、我々の配合に共界面活性剤としてそれを選択するように我々を動機つけたこの表面張力を著しく減少させる特性である。サーフィノール104PG-50(商標)が、薬物の水性相への導入前に内部油相に添加された。得られた生成物は、約230nmの非常に小さい粒径を特徴とし、この実験結果は、再現可能である。
このサイズ分布は、凝集を阻止するためのサーフィノール104PG-50(商標)の能力におそらく起因して均一に非常に狭い(多分散性指数(PDI)〜0.1)。
3.2.3形態学におけるPEG及びステアリン酸の効果
マクロファージに容易に認識されにくい実体(entities)を生成しかつ従って血液中の持続性循環時間を示すために、ポリエチレングリコール(PEG)が薬物送達方法に広範囲に使用されていることがよく確立されている。生物レベルにおいて、ナノ粒子をPEGで被覆することは、立体的に血液成分とその表面との相互作用を妨げ、血漿タンパク質とポリエチレングリコール化されたナノ粒子との結合を減少させる。これは、オプソニンとの薬物キャリア相互作用を阻害し細網内皮系(RES)による捕捉を示した。
脱イオン水に溶解されたPEGを外側相に0.5%w/vの初期濃度でPVAとともに導入した。
水性外側相の5mlのPEG(0.5% w/v)の存在とポリマーの油相へ添加された2mlのステアリン酸(0.2% w/v)をサーフィノール104PG-50(商標)との共界面活性剤として組み合わせたとき、図3に表されるような表面形態学の顕著な改良が観察された。ゼータサイザーの読みは、約270nmのより小さい粒径及び非常に狭い分布(PDI〜0.2)を提供した。
また、本発明は以下の態様であり得る。
〔1〕薬物送達のためのナノ粒子キャリアの製造方法であって、前記ナノ粒子が以下の工程、(1)前記ナノ粒子キャリアの基剤を形成する1種以上のポリマーを含む水-油-水の二重エマルジョンを調製する工程、
(2)送達される薬物を前記エマルジョン相の一つへ配合する工程、
(3)油相又は外側の水相を炭水化物でドープする工程、及び
(4)前記エマルジョンを噴霧乾燥して100nm〜1000nmの狭い粒径分布のナノ粒子を形成する工程、
により製造されることを特徴とする方法。
〔2〕このように製造された前記ナノ粒子が、多機能ナノ粒子である前記〔1〕記載の方法。
〔3〕前記炭水化物が、糖類である前記〔1〕又は〔2〕記載の方法。
〔4〕前記糖類が、二糖類である前記〔3〕記載の方法。

Claims (18)
- 薬物送達のためのナノ粒子キャリアの製造方法であって、前記ナノ粒子が以下の工程、(1)前記ナノ粒子キャリアの基剤を形成する1種以上のポリマーを油相に含む水-油-水の二重エマルジョンを調製する工程、
(2)送達される薬物を前記エマルジョン相の一つへ配合する工程、
(3)油相又は外側の水相を炭水化物でドープする工程、
(4)(i)前記油相、又は(ii)前記油相及び前記外側の水相のいずれかを、非イオン性界面活性剤でドープする工程であって、前記油相は、アセチレンジオール構造を有する非イオン性界面活性剤でドープされる工程、及び
(5)前記エマルジョンを噴霧乾燥して100nm〜1000nmの狭い粒径分布のナノ粒子を形成する工程、
により製造されることを特徴とする方法。 - このように製造された前記ナノ粒子が、多機能ナノ粒子である請求項1記載の方法。
- 前記炭水化物が、糖類である請求項1又は2記載の方法。
- 前記糖類が、二糖類である請求項3記載の方法。
- 前記二糖類が、ラクトース、マルトース、イソマルトース、マンノビオース、トレハロース、及びセロビオースを含む群から選択される請求項4記載の方法。
- 前記糖類が、陽イオン生物分解性の粘膜粘着性多糖類と混合される請求項3〜5のいずれか1項記載の方法。
- 前記多糖類が、キトサン及び/又はその誘導体である請求項6記載の方法。
- 前記工程(4)の前記外側の水相をドープする非イオン性界面活性剤が、ポリマーの非イオン性界面活性剤である請求項1記載の方法。
- 前記外側の水相中の前記ポリマーの非イオン性界面活性剤が、PVAである請求項8記載の方法。
- 前記油相中の前記ナノ粒子キャリアの基剤を形成するポリマーがPLGA(乳酸-グリコール酸共重合体)である請求項1〜9のいずれか1項記載の方法。
- 前記薬物が、前記油相に添加される請求項1〜10のいずれか1項記載の方法。
- 前記薬物が、内部の水相に添加される親水性薬物である請求項11記載の方法。
- 前記薬物が、疎水性である請求項11又は12記載の方法。
- 前記薬物が、リファンピシン、イソニアジド、エタンブトール、又はピラジンアミドから選択される請求項1〜13のいずれか1項記載の方法。
- 前記エマルジョンの外側の水相が、ポリエチレングリコール(PEG)を含む請求項1〜14のいずれか1項記載の方法。
- 前記油相が、ステアリン酸を含む請求項1〜15のいずれか1項記載の方法。
- このように形成された前記ナノ粒子が、実質的に球状である請求項1〜16のいずれか1項記載の方法。
- 前記ナノ粒子の粒径分布が、180nm〜250nm直径である請求項17記載の方法。
