Description
1.ガランタミンと比較して顕著に減少したChE-Iとしての活性。
2.ガランタミンと比較して減少したニコチン性APLとしての活性。
3.ガランタミンと比較して減少した胃腸の副作用。
これらのアプローチ全てが研究された。また、実施例において説明され、図に示される。
「アリール」という用語は、アルキル基、アルコキシ基、アルキルカルボニル基、ハロ−又はトリハロメチル基から成る群から独立に選択される0、1、2、 3、4、又は5個の置換基を有するフェニル基を意味するものとする。
記載されている誘導体の立体化学はガランタミンのものと同一である。
脳血液関門を通過しての脳への選択されたR1プロドラッグの輸送が増加し、中枢神経系の標的部位の近傍でガランタミンに酵素的に変換し、かつ、このような部位で相互作用する効果で、3つのプロ−ガランタミン(および比較としてガランタミン)につき、図6で示すように、マウスにおける薬剤性の記憶喪失を大幅に改善させている。

20%のマウス脳ホモジネートを基質としてのアセチルチオコリン200μMを補って使用し、初期の反応動力学はRiddles PW, Blakeley RL, Zerner Bの 「エルマン試薬の再評価(Reassessment of Ellman's reagent)Methods Enzymol. 1983;91:49-60に従って測定した。図に示すように、それぞれ50μMのプロ−ガランタミンでさえ、1μMのガランタミンのサイズに相当する脳のコリンエステラーゼ阻害レベルに達することができなかった。開裂しないガランタミン誘導体(Gln 1063)はマイナスの値となった。誘導体Gln 1063においては式IのR1は−O−Si(CH3)2−C(CH3)2−C(CH3)2Hである。
ブチリルコリンエステラーゼ、25単位/mlが使用された。反応温度は37℃であった。蛍光性の反応生成物ガランタミンの生成は蛍光検出により決定された。
ガランタミンおよびGln 1062それぞれの存在下でのアセチルコリンの反応の増加を全細胞パッチクランプ記録により決定した。ガランタミンは40%までの反応の最大増加を示し、プロ−ガランタミンは17%までの最大増加しか示さなかった。
図4aは、脳と、脳における、適用されたプロ−ガランタミンの測定可能な濃度とプロ−ガランタミンの開裂により得られたガランタミンの濃度とを示す。最も高濃度で始まる曲線は、脳での、ベンゾイルガランタミンである誘導体GLN-1062である。図4bは、図4aの抜粋(拡大表示)であり、0.00と1.00μg/g(基質の重さ/体重)との間の濃度範囲をより詳細に示す。図4bでは上の曲線は脳で得られたガランタミンの濃度を示し、中間の曲線は血液中のガランタミンの濃度を示し、約0.3μg/gの濃度で始まり減少する曲線は血液中のGLN-1062の濃度を示す。

スコポラミンはT迷路試験で変更を増やすことにより測定できる記憶障害を誘導する。認識強化薬剤はT迷路試験の20分前にスコポラミンと一緒に様々な用量で腹腔内投与された。回復は用量の関数として測定され、それぞれの薬剤についてEC50が決定された。

(−)−ガランタミン臭化水素酸塩 (1.0 mol)および トリエチルアミン (4.0 mol) のDCM (30 mL)溶液に, DMAP (0.5 mol) を加え、さらにそれぞれの酸塩化物または酸無水物(1.2 mol)を加えた。混合物をアルゴン下、室温で一晩攪拌した。 反応混合物を10% NaHCO3および食塩水で洗浄し乾燥させ(Na2SO4)、濃縮した。得られた粗化合物をカラムクロマトグラフィーまたは再結晶で精製し、純粋な生成物を得た。

対応する酸 (13.87 g, 135.8 mmol) のDCM (250 mL )溶液にDCC (33.62 g, 162.9 mmol) を加え、さらにDMAP (3.32 g, 27.15 mmol)を加えた。反応混合物をさらに30分間室温で攪拌した。これに、(−)−ガランタミン臭化水素酸塩 (10.0 g, 27.15 mmol)およびトリエチルアミン (4.6 mL, 32.59 mmol) を加え、混合物をアルゴン下、室温で一晩攪拌した。析出したDCHUをろ過で除き、ろ液を留去した。さらにDCHUを続いての冷酢酸エチルでの粉砕とろ過で除いた。酢酸エチル溶液をロータリーエバポレータで留去し得られた粗生成物をカラムクロマトグラフィーで精製し所望の生成物を得た。
基本手順2を用いて、2−メチル−ブタノイル−ガランタミン(=(4aS,6R,8aS)−4a,5,9,10,11,12−ヘキサヒドロ−3−メトキシ−11−メチル−6H−ベンゾフロ[3a,3,2−ef][2]ベンザゼピン−6−オール,2−メチル-酪酸(エステル))を収率53%で固体で得た。
基本手順1を用いても、全ての観点(HPLC、融点、1H-NMR)において同一の、同じ生成物が収率58%で得られた。
例9:
基本手順1を用いて、2−メチル−プロパノイル−ガランタミン(=(4aS,6R,8aS)−4a,5,9,10,11,12−ヘキサヒドロ−3−メトキシ−11−メチル−6H−ベンゾフロ[3a,3,2−ef][2]ベンザゼピン−6−オール, 2−メチル-プロピオン酸(エステル))を固体で収率63%で得た。
2−メチル−プロパノイル−ガランタミン塩酸塩
2−メチル-プロパノイル-ガランタミン (150 mg, 0.43 mmol)の酢酸エチル (5 mL)溶液に、塩酸で飽和させた酢酸エチル(5 mL)を0℃で攪拌しながらゆっくり加えた。反応混合物を室温で1時間攪拌した。溶媒を留去し、得られた残渣を乾燥エーテルで洗浄し、高真空で乾燥させて所望の生成物を黄白色固体で164 mg(97%)得た。
C21H27NO4 (1.5 HCl)としての計算値: C, 61.2; H, 6.97; N, 3.40.実測値: C, 61.62; H, 6.95; N, 3.91.
2-メチル-プロパノイル-ガランタミン クエン酸塩
2-メチル-プロパノイル-ガランタミン (150 mg, 0.43 mmol) のメタノール (5 mL)溶液に、クエン酸のメタノール溶液(5 mL)を室温で攪拌しながらゆっくり加えた。反応混合物を室温で1時間攪拌した。溶媒を留去し、得られた残渣をメタノール−ジエチルエーテルから析出させて所望の生成物を黄白色固体で187 mg (81%) 得た。
C27H35NO11 (1.0 H2O) としての計算値: C, 57.14; H, 6.57; N, 2.47. 実測値: C, 57.43; H, 6.48; N, 2.53.
攪拌している(−)−ガランタミン臭化水素酸塩 (1.10 mmol)のピリジン(6 mL)溶液に、0℃窒素下で対応する酸塩化物(2.2 mmol)を加え、混合物をTLCが反応が終了したことを示すまで攪拌した。その後、CH2CL2 (10 mL) および水(10 mL) を加え、攪拌を30分続けた。有機層を分離し、水(2×10 mL)で洗浄し、無水MgSO4で乾燥させ溶媒を除去した。残渣をフラッシュクロマトグラフィーで精製し、例1の生成物と全ての観点で同一の生成物を得た。
例12:
R1−ピリジノイルガランタミンの合成
上記で提供した例に加えて、下記の化合物を上記の基本手順で製造した。
マウス脳ホモジネートの調製
脳は動物(マウス)から取り出され、液体窒素で急速冷凍され、使用するまでマイナス80℃で保存される。抽出物を調製する前に、冷凍脳は氷の上で溶かされ、重量が測定される。氷冷した緩衝液(130 mM NaCl, 5 mM KCl, 2.5 mM CaCl2, 1 mM MgCl2, 5mMグルコース, 5 mM HEPES, pH 7.4)を溶かしたマウス脳に加えた(1:4、重量対体積、20%脳ホモジネートが得られる)。その後、組織は氷上の陶器のホモジナイザーで、240rpmで11回ピストンを上下に動かして、ホモジナイズされる。新鮮に調製されたマウスのホモジネートはアリコートに分けられる。
脳エステラーゼ阻害の測定方法。結果は図1に示される。
修正エルマンエステラーゼ試験が用いられる。簡単に言えば、この方法は基質であるアセチルチオコリンのアセテートとチオコリンへの開裂を利用するものである。後者はDTNB(5,5´-ジチオビス−(2−ニトロ−安息香酸))と反応して黄色い化合物となり、これは分光学的に定量できる。インキュベーションのための緩衝液はpH 7.2で51 mmol/lのリン酸ナトリウム緩衝液と0.05% のトゥイーン(Tween)20を含み、100 mg/l のDTNBと0.2 %のマウス脳ホモジネート(例13に記載のように調整されたもの)を添加される。調べられる化合物は所望の濃度で加えられる。混合物は37℃にされ、反応は200μMのアセチルチオコリンを加えることにより開始される。1秒間隔で、A405がマイクロプレートリーダーで40秒測定される。吸光度−時間曲線の直線状の部分は酵素反応の開始速度を表し、反応速度の計算に用いられる。曲線の傾きは反応速度に対応する。阻害は以下の等式に従って、阻害されていない反応に対するパーセントで示される。

マウス脳ホモジネートのプロドラッグ開裂の測定法
例13に従って調製されたマウス脳ホモジネートのアリコートにプロガランタミン誘導体を加え、プロドラッグの最終濃度を10μMに調整する。インキュベーション時間の終わりに12μl の0.1M NaOH、および 100μlの飽和KClを0.1mlの反応混合物に加え、よく混合する。残りのプロドラッグと、遊離したガランタミンを200μlのトルエンで抽出する。トルエン抽出段階は150μlのトルエンを用いて2回繰り返し、得られた抽出物は蓄えられ、乾燥され、50%メタノールに溶解され、続くHPLC分析に用いられる。
医薬候補の、HEK-293細胞に発現したニコチン性アセチルコリン受容体(nAChRs)へのアロステリック調節効果の電気生理学による研究。結果は図3に示される。
ヒトα4β2、ヒトα3β4、またはキメラ鶏α7 (マウス5HT3とともに) nAChRを発現しているHEK-293細胞を、フィブロネクチンでコートされたカバースリップに測定前3日間固定した。細胞を含む選択されたnAChRを細胞外緩衝液(145 mM NaCl, 5 mM KCl, 1 mM MgCl2, 2 mM CaCl2, 10mM D-グルコース, 10 mM HEPES, pH 7.3, 約300 mOsm)で満たした記録用の浴に入れた。パッチクランプシステムは倒立顕微鏡(ツァイス(Zeiss)、ドイツ)、PatchMasterソフトウェア付のコンピュータ制御パッチクランプ増幅器(HEKA, ドイツ)、チューブかん流システム(ALA, USA)とU環アプリケーター(IMM, ドイツ)、およびデュアルマイクロマニピュレータからなっていた。パッチ用のピペットは火で研磨された10mm長さで1.5mm幅のホウケイ酸ガラスキャピラリー(WPI, ドイツ)から引いた。プログラムで制御できる引くための道具(Sutter, USA)を使用して、すぐ使える2対のピペットを調製した。パッチ用のピペット(抵抗4-8 MΩ)それぞれは1回のみの使用とした。ピペットを内部緩衝液(140 mM CsCl, 11 mM EGTA, 10mM HEPES, 2 mM MgCl2, pH 7.3, 約300 mOsm)で満たし、作動している電極に接続した。実験のための作動している電極、および参照の電極は毎日新しくし、塩素化されたばかりの銀ワイヤ(40 mm x 0.4 mm)で作製し、パッチクランプ増幅器のヘッドステージ回路に接続した。パッチは振幅-1 mV、持続時間20 msの方形テストパルスを使って行った。ギガシール(gigaseal)の形成後すぐ、保持電位-70 mVをパッチ電極に付与し全セルの記録は陰圧パルスを使って達成させた。早くて遅い膜キャパシタンスと連続した抵抗過渡信号に対して必要な補償は全てPatchMasterソフトウェア内で自動で行った。全細胞の電流は、適当なnAChR サブタイプ(α4β2および α3β4 EC50 = 30μM, キメラ α7 EC50 = 3μM)それぞれに対してのEC50でニコチンを投与することにより作動させた。nAChRのサブタイプそれぞれに対する選択された化合物のアロステリックを促進するリガンドの効果を評価するために、これらの化合物は、刺激するニコチン溶液に次の濃度で加えられた:1, 5, 10, 50, 100, 500, 1,000, 5,000 および10,000 nM、そして、500 msパルスの間、溶液を細胞表面にU管を介して加え、10kHzにデジタル化された対応する電流を10秒間記録した。連続的な電流刺激を2分間隔で行い、nAChRの脱感作を避け、刺激のための溶液の全交換を確実にした。電流の平均ピーク増幅は選択された化合物の濃度の存在下で測定され、その化合物がない状態で測定された値(参照)と比較され、参照の%として計算された。特定の化合物のAPL効果の測定は5つの細胞の最低値について繰り返され平均値+/- SDを得た。15%を超えなかった観測されたAPL効果の平均値は顕著でないとして処理した。特定の化合物の濃度依存性のAPL効果を示すために、参照値+/- SDの対応する%が使用された濃度に対してプロットされた。
プロガランタミンGln-1062 (3mg/kg)のマウスでの薬物動態、結果を図4に示す。
研究の目的
Gln-1062およびその開裂産物ガランタミンの血液および脳での薬物動態プロファイルの決定。Gln-1062およびその開裂産物ガランタミンの脳対血液の濃度比を決定し、血液脳関門の浸透能力を評価する。
生物分析
Gln-1062評価の分析方法を、直線性、正確性&精度、およびSAM血液および脳ホモジネートにおける回復について、LC/MS/MSを用いて評価した。

この方法を、直線性、正確性&精度、およびSAM血液および脳ホモジネートにおけるGln-1062回復について評価した。
A 直線性、精度&正確性
単一の標準曲線および3つの品質管理レベル(QC)につき6つの反復(全部で18のQC)を抽出して分析した。較正曲線の直線性を重み付き最小二乗回帰分析で決定した。
許容基準
i.9つの標準のうち少なくとも6つが名目値から±15%の精度でなくてはいけない、ただし、評価法の下限(LLOQ)においては、±20%は許容される。
ii.バッチQCの3分の2およびそれぞれのレベルの少なくとも半分のQCは名目値から±15%の計算精度でなくてはいけない。
iii.内部アッセイの平均の正確性および精度
1.精度および正確性を決定するために6つのQCのうちの4つが得られなければならない。
2.それぞれのQCレベルの変数の内部アッセイ係数(%CV)は15%および許容されるそれぞれの検証のための平均値の精度を超えてはならない。
血液および脳ホモジネートマトリックスのGln-1062の直線性、正確性および精度をそれぞれ表1および表2に示す。
血液および脳ホモジネートマトリックスからのGln-1062の回収もまた評価した。
回収は抽出したマトリクスQCサンプル中の分析物の濃度をポスト−抽出物(ブランク抽出物)サンプルマトリックスにおいて調製された標準曲線で5.3Bに記載されているように定量することにより決定した。
血液および脳ホモジネートマトリックスからのGln-1062の回収はそれぞれ表3および4に示す。
血液収集:
血液サンプルを眼窩後の神経叢抗凝固剤抗凝固剤とやさしく混合し、セクション5.3Aに記載されているように血液サンプルの50μlアリコートがすぐに沈殿した。それぞれの時点での残った血液サンプルの量をドライアイス上で冷凍した。
脳の収集:
血液を引いた後すぐ、脳はリン酸緩衝食塩水(pH 7.4)でかん流させ、取り出してドライアイス上で冷凍した。
全ての脳サンプルを分析部に移送し、分析まで、−80±10℃で冷凍しておいた。
脳サンプルを氷で溶かし、重さを測った。適切な量の氷冷ホモジネート用媒体(メタノール:水:20:80、v/v)を加えた。氷上、ポリトロン(polytron)ホモジナイザーで脳サンプルをホモジナイズし、ホモジネート用媒体で量4mlのホモジネートにつき、脳1gmを得られるように調製した。ホモジネートのあと、すぐに脳ホモジネートサンプルを分析まで−80℃で冷凍した。
ガランタミンおよびいくつかのR1プロ−ガランタミンそれぞれの投与後の、フェレットにおける胃腸の副作用の行動インデックス。結果を図5に示す。
試験システム
実験の日に750-1000グラムの重さの14匹の大人のオスのPutoris furo フェレット(Marshall BioResources (North Rose, USA))を本実験で使用した。スポンサーの同意を得て、14匹の動物のうちの4匹を2つの実験群に含めた。(表3および表4、フェレット番号1,2,3、および4)
動物の馴化は5日間続けた。受け取りの際、動物はSyncrosome の前提でケージに集めて入れられた。動物は好きなときに餌と飲料水をとれるようにした。
試験アイテムと参照化合物
この研究のあいだ、参照化合物(ガランタミン)1つと、ガラントス(Galantos)の候補化合物(GLN979)をそれぞれ2つの投与量で試験した。いずれの化合物も表1に詳述されている投与量と濃度とで腹腔内投与した。
異なる化合物の発送の詳細を表2に示す。

4つの実験群につき、投与は麻酔されていない動物に腹腔経路でT0(図1)で行った。それぞれの群の投与量は表1に詳細に示す。
嘔吐試験
化合物溶液の腹腔投与の後、動物は熟練技術者が4時間連続して観察した。この間、嘔吐の機会数(胃腸の中身の一部を排出するに至る一連の吐き気)を記録した。
4時間の観察の間、いくつかの副作用(唾液分泌, 震え, 呼吸困難、および下痢)もまた観察した。これらの行動それぞれにおいて、採点方法はスポンサーと決定した。重篤度にしたがい、それぞれのパラメータは以下のように定量化した。
−なし(None);行動は観測されない
−おだやか(Mod,):行動は低い頻度および/または低い強度で観測される
−強い(Int.):行動は頻繁におよび/または継続的におよび/または高い強度で観測される
参照または試験化合物の腹腔内投与を受けた全ての動物を嘔吐反応および行動の両方の形式にかかわらず、この試験に含めた。
マウスにおけるスコポラミンにより誘導された記憶喪失からの回復、結果を図6に示す。
薬剤調製
Gln 1062、Gln 0979およびガランタミンをスポンサーから提供された 96mM NaCl (等浸透圧)中の15% -ヒドロキシプロピル−β−シクロデキストリンに溶解した。Gln 1062および Gln 0979は、濃度0.01, 0.03, 0.1 and 0.2mg/ml で使用し、これは10ml/kg で与えられるとそれぞれ0.1, 0.3, 1 and 2mg/kg i.pの投与量になる。ガランタミンは濃度0.03, 0.1, 0.2 and 0.5mg/mlで用いられ、これは10ml/kg で与えられるとそれぞれ0.3, 1, 2 and 5mg/kg i.p., の投与量になる。参照動物は96mM NaCl 中の15% -ヒドロキシプロピル−β−シクロデキストリンの注射を媒体として受けた。
ニコチン((−)ニコチン酒石酸水素塩、シグマ(Sigma)、フランス), スコポラミン(−(−)スコポラミン塩酸塩, シグマ、フランス)を食塩水(0.9% NaCl, Aguettant, フランス)にそれぞれ濃度0.04および 0.1mg/mlで溶解した。10ml/kgの量で投与され、それぞれ投与量0.4および 1mg/kgに達した。
4〜5週間、老齢のオスのCD−1マウス((Janvier; Le Genest St Isle −フランス)を研究に用いた。マウスはグループに分けてケージにいれ(1ケージ10マウス)調製された温度(21-22℃)の部屋で、反対の明暗サイクル(12時間/12時間;光オン:17:30−05:30; 光オフ: 05:30−17:30)を保ち、好きなときに餌と水をとれるようにした。
Gln 1062、Gln 0979およびガランタミンの認識を強化させる性質は同じ実験条件下でT迷路交代モデルでスコポラミン処理したマウスで評価した。Gln 1062およびGln 0979の両方を0.1、0.3、 1、および 2mg/kg i.pの投与量でテストした。ガランタミンは0.1、0.3、1、2、および5mg/kg i.pの投与量でテストした。ニコチンは0.4mg/kg i.pの投与量でテストした。これらの化合物はすべて、記憶障害を誘導するために用いられた1mg/kg i.pスコポラミン(T迷路試験前20分)の注射を受けたすぐ後に投与した。
記憶能力はT迷路での自発的交代のパーセンテージにより評価した。食塩水を注射されたマウスの交代の数を交代しなかった記憶能力の基礎レベルとして用いた。
Gln 1062、ln 0979およびガランタミンは異なる動物を用いた3セットの実験で別々にテストした。それぞれの実験のセットは、少なくとも1つのそれぞれの食塩水/媒体、スコポラミン/媒体、およびスコポラミン/ニコチン(0.4mg/kg)群を常に含む、一連の毎日の実験に分けた。
T迷路装置は本線(55cm長さ( 10cm幅 (20 cm 高さ)および2つのアーム(30cm長さ (10 cm 幅 ( 20cm高さ)が、本線の右と左との関係で90度で位置している灰色のフレキシガラスからなるものであった。はじまりの箱(15cm長さ(10 cm 幅)を本線からギロチンドアで離した。水平のドアは、交代作業を選択させられている間、特定のアームを閉め出すためにあった。
この実験プロトコルは1つのセッションからなり、これは1「強いられた選択」試験で始まり、14の「自由選択」試験に続く。はじめの「強いられた選択」試験において、動物はスタートのアームに5秒閉じ込め、その後、左のゴールか、または右のゴールを水平のドアでブロックしたまま動物を放す。マウスは、離された後、迷路をうまく通り抜け、最終的に開いているゴールのアームに入り、スタートの地点に戻る。動物がスタート地点に戻るとすぐ、閉まっているゴールのドアは開放され、この動物は左と右のゴールのアームを自由に選ぶことができるようになる(「自由選択試験」)。4本の足をアームに置いたとき、入ったと考えることとする。14個の自由選択試験が行われるか、10分間が過ぎるときのいずれか早い方と同時に1つのセッションが終わり、動物が迷路から出される。試験の平均持続時間は6分である。
装置は動物ごとにアルコール(70°)を使って洗浄する。尿および糞は迷路から取り除く。
試験の間、動物の取り扱いとオペレータの視界はできるだけ小さくする。
4個の自由選択試験の交代のパーセンテージはマウスごとに決定され、作動記憶能力のインデックスとして用いた。このパーセンテージは連続的な試験(すなわち、左−右−左−右、など)の間のT迷路の異なるアームへの進入として規定された。
差異分析(ANOVA)を結果のデータについて行った。フィッシャー(Fisher)の保護された最小有意差を一対比較に用いた、p値≦0.05を有意とした。スコポラミンにより誘導された記憶障害の薬剤による回復は、食塩水/媒体群の反応を100%、スコポラミン/媒体群の反応を0%として計算した。
それぞれの薬剤について、ED50を決定するために回復の状態をS字状の投与量反応モデル(グラフパッド(graphpad)ソフトウェア)に続いてプロットした。ED50は、曲線適合表から読み取ったが、これは50%の反応と関連する有効投与量を示す。
Claims (3)
- 下記化合物を含む、コリン作用性障害に関連する神経変性疾患、精神疾患、または神経疾患の治療のための、医薬組成物。
- 前記疾患が、アルツハイマー病、パーキンソン病、他のタイプの痴呆、統合失調症、てんかん、低酸素状態の後の脳中の酸素および栄養欠乏、無酸素症、仮死、心停止、様々なタイプの中毒、知覚麻痺、自閉症、手術後の精神錯乱および/又は亜症候群性の手術後の精神錯乱、アルコール依存、ならびにニコチン渇望からなる群より選択される請求項1に記載の医薬組成物。
- 神経変性疾患がアルツハイマー病およびパーキンソン病から選択される請求項2に記載の医薬組成物。
