Claims (17)
- 以下のことを含む、組換体クローンの選択方法:
− 毒性蛋白質をコードするヌクレオチド配列をゲノム中に含む宿主細胞を得ること、そして
− DNAインサートおよび前記毒性蛋白質に対する機能的な抗毒性蛋白質をコードするヌクレオチド配列を前記宿主細胞に導入すること、ただし、前記組換体クローンは前記DNAインサートおよび前記毒性蛋白質に対する機能的な抗毒性蛋白質をコードする前記ヌクレオチド配列の導入後に生き残る組換体クローンであり、前記DNAインサートは、抗毒性蛋白質の発現生産を可能にする転写および/または翻訳信号を含む配列を有し、前記抗毒性蛋白質の発現生産を可能にする転写および/または翻訳信号を含む配列により、前記毒性蛋白質に対する機能的な抗毒性蛋白質をコードする前記ヌクレオチド配列が発現される。 - 抗毒性蛋白質および毒性蛋白質が、以下の組み合わせから成る群から選択されることを特徴とする請求項1に記載の方法:CcdA蛋白質/CcdB蛋白質、Kis蛋白質/Kid蛋白質、Phd蛋白質/Doc蛋白質、RelB蛋白質/relE蛋白質、PasB蛋白質/PasA蛋白質、PasC蛋白質/PasA蛋白質、mazE蛋白質/mazF蛋白質、その他のプラスミド由来か或いはプラスミド由来ではない抗毒性蛋白質/毒性蛋白質。
- 毒性蛋白質に対する抗毒性蛋白質をコードするヌクレオチド配列が、DNAインサートの正しい組み込みの後、発現されることを特徴とする請求項1または2に記載の方法。
- 一つ以上の組換体クローンを回収することをさらに含むことを特徴とする請求項1〜3のいずれか一項に記載の方法。
- 毒性蛋白質に対する抗毒性蛋白質をコードするヌクレオチド配列が、プロモータ/オペレータ配列を含むインサートの導入後に発現されることを特徴とする請求項1〜4のいずれか一項に記載の方法。
- 毒性蛋白質に対する抗毒性蛋白質をコードするヌクレオチド配列が、毒性蛋白質に対する抗毒性蛋白質をコードするヌクレオチド配列の転写および/または翻訳信号を含むインサートの導入後に発現されることを特徴とする請求項1〜5のいずれか一項に記載の方法。
- 第1組換え部位と第2組換え部位との間に介在するカセット配列を含む核酸のコンストラクトであって、前記第1組換え部位および前記第2組換え部位がファージλ由来のatt部位であり、前記カセット配列は、毒性蛋白質に対する機能的な抗毒性蛋白質をコードするヌクレオチド配列およびDNAインサートの組み込みのための部位を含み、前記抗毒性蛋白質は、カセット配列中への前記DNAインサートの正しい組み込みの後に発現され、前記DNAインサートは、抗毒性蛋白質の発現生産を可能にする転写および/または翻訳信号を含む配列を有することを特徴とする核酸のコンストラクト。
- 抗毒性蛋白質および毒性蛋白質が、以下の組み合わせから成る群から選択されることを特徴とする請求項7に記載の核酸のコンストラクト:CcdA蛋白質/CcdB蛋白質、Kis蛋白質/Kid蛋白質、Phd蛋白質/Doc蛋白質、RelB蛋白質/relE蛋白質、PasB蛋白質/PasA蛋白質、PasC蛋白質/PasA蛋白質、mazE蛋白質/mazF蛋白質、その他のプラスミド由来か或いはプラスミド由来ではない抗毒性蛋白質/毒性蛋白質。
- インサートを含むヌクレオチド配列がプロモータ/オペレータ配列を含むことを特徴とする請求項7〜8のいずれか一項に記載の核酸のコンストラクト。
- 第1および第2組換え部位がリコンビナーゼによって認識される請求項8〜9のいずれか一項に記載の核酸のコンストラクト。
- 請求項7〜10のいずれか一項に記載の核酸のコンストラクトを含むことを特徴とするベクター。
- 自己複製ベクターであることを特徴とする請求項11に記載のベクター。
- ウィルスまたはプラスミド・ベクターであることを特徴とする請求項12に記載のベクター。
- カセット配列を含むベクターであって、前記カセット配列は、毒性蛋白質に対する機能的な抗毒性蛋白質をコードするヌクレオチド配列およびDNAインサートの組み込みのための部位をコードし、前記抗毒性蛋白質は、カセット配列中への前記DNAインサートを含むヌクレオチド配列の正しい組み込みの後に発現され、前記DNAインサートは、抗毒性蛋白質の発現生産を可能にする転写および/または翻訳信号を含む配列を有することを特徴とするベクター。
- 自己複製ベクターであることを特徴とする請求項14に記載のベクター。
- 以下のものを含むクローニングおよび/または配列決定キット:請求項7〜10のいずれか一項に記載の核酸のコンストラクト、または前記核酸のコンストラクトを含むベクター。
- 請求項11〜15のいずれか一項に記載のベクターを含む組換原核細胞であって、毒性蛋白質をコードする組換ヌクレオチド配列をその染色体DNA中に組込まれている組換体原核細胞。
