2−アミノ−5−ブロモ−N,3−ジメチルベンズアミド(式2の化合物)の製造
メカニカルスターラー、熱電対、冷却器およびTeflon(登録商標)フルオロポリマー管(内径1/16インチ(0.16cm)×外径1/8インチ(0.32cm))(管の端部が反応混合物の表面より下に浸水するように配置する)を備えた1000mLフラスコに酢酸(226mL)を添加した。水(85g)中の水酸化ナトリウム水溶液(50%、25g)を15分かけて添加し、次いで2−アミノ−N,3−ジメチルベンズアミド(50g、0.305モル)(製造方法に関しては、PCT公報、国際公開第2006/062978号パンフレットを参照のこと)を添加して、混合物を55℃まで加熱した。一方の口にTeflon(登録商標)管を取り付けた2つ口200mLフラスコに液体臭素(50.1g)を添加し、もう一方の口を1000mLフラスコのTeflon(登録商標)管に連結した。1時間あたり約0.012m3(0.4cu ft)の速度で2.5時間、液体臭素の表面より下でTeflon(登録商標)管を通して窒素気体を流し入れた。この間、蒸発した液体臭素と、窒素中に飛沫同伴した臭素蒸気の全てが2つ口200mLフラスコから流れ出し、1000mLフラスコのTeflon(登録商標)管を通して反応混合物中に入った。臭素蒸気の添加の間とその後30分間は反応温度を約55℃に保持し、次いで45℃まで冷却し、一晩攪拌した。水(88mL)中水酸化ナトリウム水溶液(50%、52g)を0.8mL/分の速度で反応混合物に添加した。水酸化ナトリウム溶液の総体積の約10%が添加されたら、添加を停止し、反応混合物を45℃で1時間攪拌した。1時間後、0.8mL/分の速度で残りの水酸化ナトリウム溶液を添加した。添加の完了後、反応混合物を45℃で30分間攪拌し、次いで10℃まで冷却し、1時間攪拌した。混合物をろ過し、回収した固体をメタノール(130mL)および水(260mL)で洗浄し、次いで45℃の真空オーブン中で一定重量となるまで乾燥させ、133〜135℃で融解する固体として表題の化合物を得た(67g、HPLCによる純度99.4面積%、収率89.7%)。

2−アミノ−5−シアノ−N,3−ジメチルベンズアミド(式1の化合物)の製造
メカニカルスターラー、熱電対、冷却器および水酸化ナトリウム/次亜塩素酸ナトリウムスクラバーを備えた500mL4つ口フラスコに、冷却器に連結された気体注入口を通してアルゴン雰囲気を維持しながら、2−アミノ−5−ブロモ−N,3−ジメチルベンズアミド(実施例1の方法によって製造されたもの)(純度99.1%、24.1g、0.10モル)およびキシレン(100g)を添加した。この混合物を室温で攪拌し、粉末状シアン化ナトリウム(使用直前に粉末化されたもの)(6.2g、0.121モル、純度95%と仮定する)、ヨウ化銅(I)(2.9g、0.015モル)およびN,N’−ジメチルエチレンジアミン(7.6g、0.085モル)を添加した。混合物にアルゴンをパージしながら、さらに15〜20分間攪拌を続け、その後、混合物をアルゴン雰囲気下に維持した。スクラバーを通して排気しながら、混合物を還流下(約140℃)で加熱した。4.5時間後、混合物を25℃まで冷却し、5分間で水(100mL)を添加し、そして攪拌をさらに30分間続けた。混合物をろ過し、そして回収した固体を水(2×50mL)およびキシレン(50mL)で洗浄し、次いで55℃の真空オーブン中で一定重量になるまで乾燥させ、203〜204℃で融解するオフホワイト色固体として表題の化合物を得た(18.2g)。

メカニカルスターラー、温度計および冷却器を備えた100mL3つ口フラスコに、冷却器に連結された気体注入口を通して窒素の流れを保持しながら、2−アミノ−5−ブロモ−N,3−ジメチルベンズアミド(実施例1の方法によって製造されたもの)(純度99.1%、5.0g、0.02モル)およびクロロベンゼン(20g)を添加した。反応混合物を室温で攪拌し、粉末状シアン化ナトリウム(使用直前に粉末化されたもの)(1.25g、0.024モル、純度95%と仮定する)、ヨウ化銅(I)(0.57g、0.003モル)およびN,N’−ジメチルエチレンジアミン(1.51g、0.017モル)を添加した。混合物を還流下(約130℃)で4.5時間加熱し、次いで25℃まで冷却し、水(20ml)を5分間かけて添加し、そして得られた混合物を30分間攪拌した。混合物をろ過し、回収した固体を水(2×10mL)およびクロロベンゼン(10mL)で洗浄し、次いで50℃の真空オーブン中で一定重量になるまで乾燥させ、202〜203℃で融解するオフホワイト色固体として表題の化合物を得た(3.6g)。

メカニカルスターラー、温度計および冷却器を備えた100mL3つ口フラスコに、冷却器に連結された気体注入口を通して窒素の流れを保持しながら、2−アミノ−5−ブロモ−N,3−ジメチルベンズアミド(実施例1の方法によって製造されたもの)(純度99.1%、5.0g、0.02モル)および1,3,5−トリメチルベンゼン(20g)を添加した。反応混合物を室温で攪拌し、粉末状シアン化ナトリウム(使用直前に粉末化されたもの)(1.25g、0.024モル、純度95%と仮定する)、ヨウ化銅(I)(0.57g、0.003モル)およびN,N’−ジメチルエチレンジアミン(1.51g、0.017モル)を添加した。反応混合物を約138℃〜140℃まで3時間加熱し、次いで還流するまで(約155℃)加熱し、次いで23℃まで冷却し、そして水(20mL)を5分間かけて添加した。混合物を30分間攪拌し、次いでろ過した。回収した固体を水(2×10mL)およびクロロベンゼン(10mL)で洗浄し、次いで50℃の真空オーブン中で一定重量になるまで乾燥させ、202〜203℃で融解するオフホワイト色固体として表題の化合物を得た(3.3g)。

メカニカルスターラー、温度計および冷却器を備えた100mL3つ口フラスコに、冷却器に連結された気体注入口を通して窒素の流れを保持しながら、2−アミノ−5−クロロ−N,3−ジメチルベンズアミド(製造方法に関してはPCT公報、国際公開第2006/062978号パンフレットを参照のこと)(4.0g、0.02モル)および1−メチルフタレン(20g)を添加した。反応混合物を室温で攪拌し、粉末状シアン化ナトリウム(使用直前に粉末化されたもの)(1.25g、0.024モル、純度95%と仮定する)、ヨウ化銅(I)(1.15g、0.006モル)およびN,N’−ジメチルエチレンジアミン(1.51g、0.017モル)を添加した。反応混合物を約180℃まで6時間加熱した。反応混合物のHPLC分析では、2−アミノ−5−クロロ−N,3−ジメチルベンズアミドの転化が約95%であって、2−アミノ−5−シアノ−N,3−ジメチルベンズアミドが主生成物であることが示された。
3−ブロモ−1−(3−クロロ−2−ピリジニル)−N−[4−シアノ−2−メチル−6−[(メチルアミノ)カルボニル]フェニル]−1H−ピラゾール−5−カルボキサミド(式5の化合物)の製造
アセトニトリル(24mL)中3−ブロモ−1−(3−クロロ−2−ピリジニル)−1H−ピラゾール−5−カルボン酸(製造方法に関してはPCT特許公報、国際公開第2003/015519号パンフレットを参照のこと)(純度97.6%、3.10g、0.01モル)および2−アミノ−5−シアノ−N,3−ジメチルベンズアミド(すなわち、実施例2の方法によって製造されたもの)(2.00g、0.01モル)の混合物に3−ピコリン(2.92mL、0.03モル)を添加した。混合物を−5〜−10℃まで冷却し、次いで塩化メタンスルホニル(1.08mL、0.014モル)を滴下して添加した。混合物を−10〜−5℃で5分間攪拌し、次いで0〜5℃で3時間攪拌した。3時間後、温度を0〜5℃に保持しながら、水(11mL)を混合物に滴下して添加した。15分後、濃塩酸(1.0mL)を添加し、混合物を0〜5℃で1時間攪拌した。混合物をろ過し、回収した固体をアセトニトリル−水(2:1混合物、2×2mL)およびアセトニトリル(2×2mL)で洗浄し、次いで窒素下で乾燥させて、206〜208℃で融解するオフホワイト色固体として表題の化合物を得た(4.78g、95%の分析に基づく補正収率95.8%)。

アセトニトリル(80mL)中3−ブロモ−1−(3−クロロ−2−ピリジニル)−1H−ピラゾール−5−カルボン酸(製造方法に関してはPCT特許公報、国際公開第2003/015519号パンフレットを参照のこと)(純度97.4%、15g、0.049モル)および2−アミノ−5−シアノ−N,3−ジメチルベンズアミド(製造方法に関してはPCT特許公報、国際公開第2006/62978号パンフレットを参照のこと)(10.0g、0.0525モル)の混合物に3−ピコリン(13.9g、0.148モル)を添加した。混合物を15〜20℃まで冷却し、次いで塩化メタンスルホニル(8.2g、0.071モル)を滴下して添加した。1時間後、温度を15〜20℃に保持しながら、水(37.3g)を反応混合物に滴下して添加した。混合物を45〜50℃で30分間加熱し、次いで15〜25℃まで1時間冷却した。混合物をろ過し、回収した固体をアセトニトリル−水(約5:1混合物、2×10mL)およびアセトニトリル(2×10mL)で洗浄し、次いで窒素下で乾燥させて、オフホワイト色固体として表題の化合物を得た(24.0g、91.6%の分析に基づく補正収率93.6%)。
1H NMR(DMSO−d6)δ10.53(br s,1H)、8.49(dd,1H)、8.36(m,1H)、8.16(dd,1H)、7.87(d,1H)、7.76(d,1H)、7.60(m,1H)、7.41(s,1H)、2.67(d,3H)、2.21(s,3H)。
