Description
細胞培養物の調製
Caco−2細胞(ATCC番号:HTB−37、Manassas,VA,USA)を、(Thwaitesら、1993a;1993b)に記載のとおりに培養した。細胞単層を、高密度(4.4〜5.0°−105細胞/cm2)で直径12mm又は24.5mmの組織培養インサート[Transwellポリカーボネートフィルター(Costar)]上に播種することにより調製した。細胞単層を、空気中5%のCO2の加湿雰囲気中で37℃で維持した。細胞のコンフルエンスを顕微鏡及び経上皮抵抗の決定によって推定した。リシンの放射標識流動を、播種後18〜25日及び給餌後18〜24時間に実施した。
腸リシン取り込みについてL.ロイテリ(L.reuteri)株を選択する方法
Caco−2細胞単層を洗浄し(500mlの改変Krebs緩衝液(組成は以下のとおり(全てmmol/l)、NaCl 140、KCl 5.4、CaCl2 2.8、MgSO4 1.2、NaH2PO4 0.3、KH2PO4 0.3、HEPES 10、グルコース10(Tris塩基でpHを7.4にして37℃で))中で4回)又は必要に応じてNa+不含Krebs緩衝液(上記と同様であるが、NaCl及びNaH2PO4の代わりにコリンCl−)で、予め温めた(37℃)Na+不含Krebbs緩衝液2mlを含む6ウェルプレート(Life Technologies)中に配置した。新たなKrebs緩衝液又はNa+不含Krebbs緩衝液のアリコートを、チャンバー中に配置した。放射標識リシン((3H)Amersham)を、トレーサー濃度(0.2μCi/ml)で使用した。次いで上皮層を、7.0×107cfu/mlの濃度の試験すべき種々のLAB(ラクトバシラス・ロイテリATCC PTA−6127、ラクトバシラス・ロイテリ1068、ラクトバシラス・ロイテリSD2112及びバシラス・コアグランス・ラクリス−S株)と共に37℃で6時間インキュベートした。200μLのサンプルを、経上皮輸送の決定のために基礎溶液から60分毎に採取した。放射標識を、シンチレーションカウントによって決定した。結果については図1を参照のこと。
選択された株を含む飼料製品の製造
本実施例では、L.ロイテリ「Shiny」(ATCC PTA−6127)を、市販製品にこの株を添加するために、一般的に良好な成長特徴及び実施例2で既に言及した選択における好ましい結果に基づいて選択する。このL.ロイテリShiny株は、業界でラクトバシラスを増殖させるための標準的な方法を使用して、増殖及び凍結乾燥させる。
イヌ試験
45匹のビーグル犬を使用して試験を実施する。イヌに、実施例3の食餌に対応する標準的な乾燥食餌を1週間給餌し、その後試験を開始する。試験開始の直前に、参加するイヌの毛の状態を、実施例5に記載したように評価パネルによって評価する。イヌを、15匹ずつ3グループに分ける。1グループのイヌに、L.ロイテリ「Shiny」(ATCC PTA−6127)でコーティングした乾燥ペレットを給餌し、他のグループのイヌに、L.ロイテリ株1068でコーティングした乾燥ペレットを給餌し、3番目のグループには標準的な食餌を続けることにより、コントロール食餌を提供する。全てのグループを、飼料及び水に自由にアクセスできるようにする。12週間後、各イヌの毛の状態を再度評価する。L.ロイテリ「Shiny」コーティングを有するペレットを給餌したイヌは、顕著に艶が増した外観を有し、L.ロイテリ1068株を有するペレットを給餌したイヌ及びコントロール群のイヌと比較して、顕著なフケを示さない。
爪の成長及び毛皮の特徴
実施例4のイヌの爪は、熟練のネイルグルーマーがスコア付けし、毛皮は、ビーグルの繁殖を専門とする2人の経験のあるドッグショー審判がスコア付けする。試験前に、イヌを、毛皮及び爪の品質によって、グループ間に均等に分配する。12週間後、爪を、カットしたときの割れ及び成長に関して品質を評価し、1を悪いとし、5を優れているとして、5つのレベルの尺度でスコア付けする。毛皮を、全体的な品質、フケ、毛皮の長さ及び艶について、1(最も悪い)から5(最も良い)の範囲の尺度で評価する。
Claims (14)
- ラクトバシラス・ロイテリ(Lactobacillus reuteri)の非病原性のリシン取り込み刺激乳酸菌株を選択するステップと、選択した株の生物学的に純粋な培養物を含む製品を動物に提供するステップとを含む、コンパニオン動物における毛皮及び爪の品質を改善する方法。
- 選択するステップが、乳酸菌株が動物におけるリシンの吸収を直接促進又は改善する能力を分析することを含む、請求項1に記載の方法。
- 選択するステップが、腸上皮透過性研究のためのCaco−2細胞系の使用を含む、請求項1に記載の方法。
- 選択が、リシンの放射標識流動の使用を含む、請求項1に記載の方法。
- 株が、ラクトバシラス・ロイテリ(Lactobacillus reuteri)株ATCC PTA−6127の生物学的に純粋な培養物である、請求項1に記載の方法。
- ラクトバシラス・ロイテリ(Lactobacillus reuteri)ATCC PTA−6127の生物学的に純粋な培養物。

b)コーティング基材でコーティングされ、次いで選択された株の生物学的に純粋な培養物の乾式噴霧でコーティングされた飼料混合物
を含む、ペット動物における毛皮及び爪の品質改善剤。 - 飼料混合物がペレット化されている、請求項8に記載の改善剤。
- ラクトバシラス・ロイテリ(Lactobacillus reuteri)株がATCC PTA−6127である、請求項8に記載の改善剤。
- 飼料混合物が、トウモロコシ、コーングルテン、鶏肉及び魚粉、塩類、ビタミン並びにミネラルを含む、請求項8に記載の改善剤。
- コーティング基材が獣脂を含む、請求項8に記載の改善剤。
- 生物学的に純粋な培養物が、製品1グラム当たり107CFUのレベルで乾式噴霧により適用され、その後獣脂が、脂肪層に付着するか又は脂肪層中に部分的に浸透するように配置される、請求項12に記載の改善剤。
- 飼料、錠剤及びカプセルからなる群より選択される形態である、請求項8に記載の改善剤。
