Description
本願明細書において用いられる以下の用語は、以下のとおり定義される。
一連のテストを、生物学的サンプルの試薬との反応からの計測値の正確性および精度への、分取の影響を実証するために実施した。マルチスティックス(MULTISTIX)(登録商標)製品(バイエル(Bayer))製の試薬を、検出すべき分析物を含有する、および含有しないケクスティックス(CHEKSTIX)液体(バイエル(Bayer))または他の標準化テスト溶液でテストした。試薬表面のいくつかは疎水性であったが、他のものは親水性であった。テストした試薬のいくつかは水溶性であったが、他のものは不水溶性であった(表1を参照のこと)。すべての場合において、試薬は、基材としての非多孔性ポリスチレンフィルムの上部に置いた吸収性材料(すなわち吸収剤)の多孔性パッドに含有させた。
試薬含有パッドは、浸漬したときに、組成物に応じて異なる量の液体を吸収したことは明らかであった(表1)。普通、きわめて疎水性の試薬表面は微量の液体を吸収する。70ダイン/cm2の表面張力を有するきわめて疎水性のポリスチレン表面はわずかに0.2μLのフィルムを保持した。含浸された液体の量における顕著な偏差は、類似の表面エネルギーの乾燥試薬の間に見いだされ、および試薬要素によって影響されていた。
サンプル液体を試薬パッド上に多量の液体でピペットするとき、得た結果の性能を補正する必要性に追加して、吐出した体積がどのように液体の拡散に影響するのかを判定することも重要である。研究を実施して、液体小滴の量の増加の、親水性表面およびきわめて疎水性の試薬表面上での液体の拡散への影響を評価した。小滴サイズを増加するに伴って、親水性試薬パッド上の液体の直径が増加する(図5を参照のこと)ことを、予期したとおり見出した。しかしながら、きわめて疎水性の試薬パッド(グルコース)の場合には、大きな小滴を用いたときに少量の吸収および液体の膨張が生じた(図4を参照のこと)。しかしながら、意外なことに、小滴サイズが100nL未満であるとき、液体は瞬間的に吸収され、膨張が生じる(小滴のサイズと相対的に)ことを見出した。すなわち、小滴が微小であるとき、疎水性試薬パッドは親水性であるかのような挙動を示した。複数のより微小な小滴の吐出が、均一な被覆率をもたらすために好ましいであろうと結論づけた。また、サンプル体積の制御により、利用可能な試薬パッドのより効率的な使用が可能であるであろう。
これらのテストで用いた例えばグルコースパッドといった試薬パッドがきわめて疎水性である場合、液体サンプルにとって、大きな小滴ではパッドに浸透することは困難である。(図4の液体吸収についてのテストの結果に注意)。グルコースパッドにおいて、これは、サンプル液体の進入を制限することにより意図的になし得る。サンプル中のグルコースの試薬との反応からの変色は、表面上に位置する液滴の基部下で液体が表面に接触する場合にのみ生じる。従って、大きな小滴をピペットするのではなく、多くの微小な小滴を既定のエリア上に吐出することが、最良の結果を得るために好ましいであろう。これがなされるとき、浸漬のものと同等のまたはそれを超える性能を、達成することが可能である(表2中の微小な小滴で達成したFOMに注意)。
図1は、ノズル12aが液体チャンバ10aから提供され、および複数のノズル開口が小滴14aを、疎水性表面16a上に支持されている試薬含有エリア18a上に分取する本発明の第1の実施形態を示す。
水溶性試薬が試薬表面中にあるとき、サンプル液体、例えば尿がそれらを吸収する。次いで、液体が試薬エリアに拡散するに伴って、水溶性要素は、溶剤面で液体と共に移動する。この移行は、不均一な発色を生成する。水溶性要素とのテストを用いて移行を計測した。大きな液滴とテストした白血球試薬(水溶性要素を含有する)は、サンプル体積に関係なく、およびサンプルが白血球を含有していたか否かに関わらず、実質的に同一の結果を示した(<3のFOM、表2)。しかしながら、テストパッドをサンプル液体中に浸漬したとき、または液体を微小な小滴で適用したとき、白血球の存在または不存在は容易に判定された(>10のFOM、表1または2)。水溶性要素の拡散は、浸漬または微小な小滴適用のいずれにおいても明らかであった。微小な小滴の反復適用での試薬表面の同時被覆は移行を制限し、および生じたいずれの移行も予測可能なパターンとなった。大きな小滴(>3μL)の場合には、不作為な凝集が移行に不均一さを生じさせていた。きわめて微小な小滴(<10nl)の場合には、不作為な凝集がまた蒸発に伴って生じた。これは、不均一な移行だけではなく、吐出液体に欠く開放空隙も生成した。
典型的な実施形態において、ノズルは、試薬表面上約5mm以下の間隔とされるであろう。図1〜3においては、ノズルプレートは、小滴がテストエリア上に垂直に落下するようホールで穿孔されている。プレートはまた、小滴が角度をもって落下するよう湾曲していてもよい。交差汚染を最小限とするために、ノズルおよびテストエリア間の距離はできるだけ短く、小滴のサイズおよびそれらの軌道によって決定されるとおり、正確な小滴の配置のため典型的には、1mmを超え、および5mm未満であろう。
微小な小滴を試薬エリア上に分取するこの方法を利用するとき、試薬のサンプルに対する光学的応答を読み取る方法を向上することが可能である。従来の技術においては、例えば発色といった試薬の応答は、所望の波長を有する光をエリアに供給し、および光検出器への反射または透過光を計測することにより全試薬エリアについて測定される。計測される光の量は、反応したサンプル中の分析物の量と相関する。実質的には、試薬エリア全体での発色の平均が読み取られる。より正確な計測は、サンプル液体の吐出に応じたパターンおよび方策で試薬エリアを照射する。すなわち、光線は反応エリアをトラバース(スキャン)することができ、および反射または透過光が、1回に1スポット(ピクセル)で、受光され、および計測されることができる。このようなプロセスは、従来のテレビセットにおける陰極線管のスクリーン上でのパターントレースに類似するとして考えることができる。
100μm未満のノズルホールの一定サイズの小滴を送達する能力に影響する微粒子(例えば細胞、キャスト、結晶、生分子等)を含む尿をサンプル液体として適応させるために、100nLの小滴サイズ限界を選択した。100μmを超えるホール直径は、微粒子を洗い流すため、および交差汚染のキャリーオーバーを回避するために、十分な分離を許容することがさらに示された。総サンプル液体は単一の小滴としてピペットによって送達されるものと同等であり、約1〜1000の微小な小滴が約0.2〜5秒の短時間で送達されるため、ノズルホールは反復的に用いられると共に閉塞してはならない。所望の結果は、100μmを超えるホール直径が、試薬からの均一な応答を提供するために、反応エリア上へ何千もの微小な小滴が均一に吐出されることを許容したことであった。100μm未満のホール直径でみられた追加の問題は、吸収が完了する前の液体の蒸発およびエアロゾルの形成であった。
ノズルは、ソースからのサンプル流体または加圧下の試薬で供給され得、これは、例えば、逆止弁、スプリッタ、空気供給および通気を伴うまたは伴わない、シリンジポンプ、ダイヤフラムポンプ、加圧コンテナ、ピエゾアクチュエータ、ダイヤフラム、蠕動、ボルテックス、吸引、遠心分離等であり得る。
さらなる従来の技術においては、例えば吸収剤パッドといった吸収性表面に試薬が追加され、次いで乾燥される。生物学的流体サンプルが追加され、および発色または他の光学的応答の発生に十分な時間が経過した後、試薬含有表面が読み取られ、およびサンプル中の分析物の量と相関される。本発明において、生物学的サンプルの精確な吐出および試薬エリアの光学的画像が、所望の場合に、生物学的サンプルがテストされているときでの追加の試薬の吐出を可能とする。
液体は、サンプルが追加された後に試薬エリア上に分取されることができる。例えば、3μLの尿をグルコース試薬に添加し、続いて、3μLのリン酸緩衝剤pH6.5を添加した。尿サンプルは、0秒で試薬と液体接触をなし、および色反応が開始される。液体緩衝剤は、10秒後に試薬と液体接触をなし、および色反応は継続する。FOMは、グルコース試薬結果がサンプル中の高塩化物含有量によって阻害されるため、高比重尿がサンプルとして用いられるときは低減される。サンプル後の第2の液体試薬の適用では、表3に示すとおり、希釈サンプル中の低下した溶解塩により、低および高サンプル間の偏差は低減され、およびFOMが向上される。
Claims (33)
- 生物学的流体のサンプルを、前記生物学的流体中の分析物と反応するための試薬を含有するエリアを有する表面上に吐出する方法であって、
(a)前記生物学的流体を、ノズルと液体的に連通するリザーバ中に入れるステップと、
(b)前記生物学的流体を前記ノズルから、0.1〜2mmの範囲の直径を有する小滴で、前記試薬含有エリア上に、または前記試薬含有エリアに隣接して分取するステップと、
(c)前記生物学的流体と前記試薬との間の反応の結果を試薬含有エリアの分光写真画像から読み取るステップと
を含み、
前記生物学的流体サンプルが、尿、血液、唾液、または粘液あるいは組織の抽出物からなる群より選択され、
前記ノズルが、インクジェットノズルであり、前記試薬エリアから1〜5mmに配置される、
ことを特徴とする方法。 - 試薬を含有するエリアを同時に被覆するに十分な小滴を分取することを特徴とする請求項1に記載の方法。
- 小滴が線の配列で分取されて試薬を含有するエリアが被覆されることを特徴とする請求項1に記載の方法。
- 前記生物学的流体と前記試薬との間の反応の結果が、前記分光画像の所定の部分に光を指向化させると共に、前記分光画像の各部分から戻る光を計測することにより読み取られることを特徴とする請求項1に記載の方法。
- ノズルが前記小滴を分取する開口を有し、前記開口は100μmから1000μmの間の直径を有することを特徴とする請求項1に記載の方法。
- 前記分光画像の前記所定の部分が全試薬エリアから構成されることを特徴とする請求項4に記載の方法。
- 前記分光画像の前記所定の部分が1滴の小滴によって接触されるエリアから構成されることを特徴とする請求項4に記載の方法。
- 前記生物学的流体と前記試薬との間の反応の結果が、小滴の配列によって接触されるエリアの配列において読み取られることを特徴とする請求項7に記載の方法。
- 前記生物学的流体が前記試薬エリアに隣接して吐出されることを特徴とする請求項1に記載の方法。
- 生物学的流体のサンプルを、前記生物学的流体中の分析物と反応するための試薬を含有するエリアを有する表面上に吐出する方法であって、
(a)前記生物学的流体を、ノズルと液体的に連通するリザーバ中に入れるステップと、
(b)1種または複数種の試薬液体を、(a)の前記ノズルまたは第2のノズルと液体的に連通する1つまたは複数のリザーバ中に入れるステップと、
(c)前記生物学的流体を(a)の前記ノズルから、0.1〜2mmの範囲の直径を有する小滴で、前記試薬含有エリア上に、または前記試薬含有エリアに隣接して分取するステップと、
(d)(a)の前記ノズルまたは第2のノズルから(b)の1種または複数種の試薬液体を、0.1〜2mmの範囲の直径を有する小滴で、前記試薬含有エリア上に、または前記試薬含有エリアに隣接して分取するステップであって、(c)の前記生物学的流体の前記分取の前、最中または後になされる前記分取するステップと、
(e)前記生物学的流体と前記試薬との間の反応の結果を試薬含有エリアの分光写真画像から読み取るステップと
を含み、
前記生物学的流体サンプルが、尿、血液、唾液、または粘液あるいは組織の抽出物からなる群より選択され、
前記ノズルが、インクジェットノズルであり、前記試薬エリアから1〜5mmに配置される、
ことを特徴とする方法。 - (b)の1種または複数種の試薬液体を、前記生物学的流体の分取前に前記試薬エリア上に分取することを特徴とする請求項10に記載の方法。
- (b)の1種または複数種の試薬液体を、前記生物学的流体の分取後に前記試薬エリア上に分取することを特徴とする請求項10に記載の方法。
- (b)の前記1種または複数種の試薬液体を、前記生物学的流体の分取の最中に前記試薬エリア上に分取することを特徴とする請求項10に記載の方法。
- 試薬を含有するエリアを同時に被覆するに十分な小滴を分取することを特徴とする請求項10に記載の方法。
- 小滴が線の配列で分取されて、試薬を含有するエリアが被覆されることを特徴とする請求項10に記載の方法。
- 前記生物学的流体と前記試薬との間の反応の結果が、前記分光画像の所定の部分に光を指向化させると共に、前記分光画像の各部分から戻る光を計測することにより読み取られることを特徴とする請求項10に記載の方法。
- ノズルが前記小滴を分取する開口を有し、前記開口は100μmから1000μmの間の直径を有することを特徴とする請求項10に記載の方法。
- 前記試薬エリアの前記所定の部分が全試薬エリアから構成されることを特徴とする請求項16に記載の方法。
- 前記試薬エリアの前記所定の部分が1滴の小滴によって接触されるエリアから構成されることを特徴とする請求項16に記載の方法。
- 前記生物学的流体と前記試薬との間の反応の結果が、小滴の配列によって接触されるエリアの配列において読み取られることを特徴とする請求項19に記載の方法。
- 前記生物学的流体または前記試薬液体が前記試薬エリアに隣接して吐出されることを特徴とする請求項10に記載の方法。
- 前記分析物と反応する試薬を含有する表面上に分取された生物学的流体のサンプルの分析物含有量を計測するための装置であって、
(a)生物学的流体の前記サンプルを分取するノズルであって、該ノズルがインクジェットノズルであり、100μmから1000μmの間の直径を有する1つまたは複数の開口を含有すると共に、前記試薬含有表面上1〜5mmに配置されたノズルと、
(b)前記生物学的サンプルを(a)の前記ノズルに供給するためのリザーバと、
(c)前記ノズル開口が、生物学的流体の前記サンプルを、前記試薬を含有する表面の所定の領域上に分取するよう、前記ノズルを前記表面に対して配置する、または前記表面を前記ノズルに対して配置する手段と
を含み、
前記生物学的流体サンプルが、尿、血液、唾液、または粘液あるいは組織の抽出物からなる群より選択される、
ことを特徴とする装置。 - 前記表面の前記所定の領域が全試薬含有領域であることを特徴とする請求項22に記載の装置。
- 前記表面の前記所定の領域が、前記ノズル開口によって分取される1滴の小滴によって接触されるエリアであることを特徴とする請求項22に記載の装置。
- 前記表面の前記所定の領域が、前記試薬を含有する表面の一部分であることを特徴とする請求項22に記載の装置。
- 前記生物学的流体と前記試薬との間の反応の結果を、試薬含有表面の分光写真画像から読み取る手段をさらに含むことを特徴とする請求項22に記載の装置。
- 前記生物学的流体と前記試薬との間の反応の結果を読み取る前記手段が、前記分光画像の所定の部分に指向化される光の光源と、前記分光画像の前記部分から戻る光を計測する検出器とを含むことを特徴とする請求項26に記載の装置。
- 前記分光画像の前記所定の領域が、全試薬含有表面から構成されることを特徴とする請求項27に記載の装置。
- 前記分光画像の前記所定の領域が、1滴の小滴によって接触されるエリアから構成されることを特徴とする請求項27に記載の装置。
- 前記分光画像の前記所定の領域が、前記試薬含有表面の一部分から構成されることを特徴とする請求項27に記載の装置。
- 試薬液体を前記試薬含有表面上に分取する手段をさらに含むことを特徴とする請求項22に記載の装置。
- 試薬液体を分取する前記手段が、100μmから1000μmの間の直径を有する1つまたは複数の開口を含有すると共に、前記試薬含有表面上1〜5mmに配置されたノズルを含むことを特徴とする請求項31に記載の装置。
- 前記ノズル開口が、前記試薬液体を前記試薬含有表面の所定の部分上に分取するよう、前記ノズルを前記試薬含有表面に対して配置する、または前記表面を前記ノズルに対して配置する手段をさらに含むことを特徴とする請求項32に記載の装置。
