Description
(1)乳化能を有する反応生成物から成る乳化素材を製造するための固定化リパーゼを製造する方法であって、
澱粉粒の形態を保持した澱粉粒又はセルロース粉末に、酵素リパーゼを混合し、該澱粉粒又はセルロース粉末担体に、粉末状態を維持して、該酵素リパーゼを結合させて、澱粉粒又はセルロース粉末固定化リパーゼとする工程において、
1)上記澱粉粒に、α−アミラーゼ及び/又はグルコアミラーゼを作用させて、その表面に穴を開けた空洞澱粉粒又は粒表面をそのまま又はそれを一部糊化した澱粉粒を用いて、
2)澱粉粒表面又はセルロース粉末を、酸化処理した状態で、及び/又は、
3)澱粉粒又はセルロース粉末表面に卵白、ゼラチン、カゼインナトリウム、又はツェインをコーテイングして、
4)これらに酵素リパーゼを混合して該澱粉粒又はセルロース粉末担体に、粉末状態を維持して、酵素リパーゼを結合させる処理を行うことで澱粉粒又はセルロース粉末固定化リパーゼとするか、又は、
5)澱粉粒固定化リパーゼと、セルロース粉末固定化リパーゼ又はセルロースを混合した固定化リパーゼとする、
ことを特徴とする澱粉粒及び/又はセルロース粉末固定化リパーゼの製造方法。
(2)上記澱粉粒に、粉末状態を維持できる程度に、水分を加えて、高温処理して、表面を糊化させた澱粉粒とする、前記(1)に記載の固定化リパーゼの製造方法。
(3)上記澱粉粒固定化リパーゼと、上記セルロース粉末固定化リパーゼ又はセルロースを混合した固定化リパーゼとすることにより、該固定化リパーゼをカラムに詰めて使用する際に、固定化リパーゼが油液中での反応の際に固着するのを防止し、かつ、反応液の流速を維持するようにした、前記(1)に記載の固定化リパーゼの製造方法。
(4)前記(1)から(3)のいずれかに記載の方法で製造した固定化リパーゼを使用して、原料として、油脂及び/又はその分解物と、グルコース、又はオリゴ糖の被反応物を用いて、当該固定化リパーゼを上記原料に作用させて、乳化能を有する油脂反応物から成る乳化素材を製造することを特徴とする乳化素材の製造方法。
(5)油脂が、大豆油、ナタネ油、コーン油、米油、ゴマ油、又はオリーブ油であり、油脂の分解物が、脂肪酸、又はグリセリンである、前記(4)に記載の方法。
(6)前記(1)から(3)のいずれかに記載の方法で製造した固定化リパーゼを、油脂及び/又はその分解物と、グルコース、又はオリゴ糖から成る被反応物に作用させて作製した乳化能を有する反応生成物から成る乳化素材を構成要素として含むことを特徴とする乳化剤。
本発明は、澱粉粒又はセルロース粉末に、リパーゼを結合させることを特徴する澱粉粒又はセルロース粉末固定化リパーゼの製造方法であり、固定化するための条件として、澱粉粒に、水又はエタノール水溶液を、澱粉粒重量当たり5〜40%加えて、リパーゼを混合、結合させること、また、固定化を強くするために、α−アミラーゼ及び/又はグルコアミラーゼを作用させて、その表面に穴を開けた澱粉粒又は粒表面を一部糊化した澱粉粒を用いること、また、結合を強化するために、澱粉粒表面又はセルロース粉末を酸化処理した後に、リパーゼを結合させること、更に、澱粉粒表面又はセルロース粉末の表面を、タンパク質素材でコーテイングして、リパーゼを結合すること、などを特徴とするものである。
(1)酵素反応物1mLを、10mLバイアルに秤取り、水5mLを加えて撹拌する。
(2)これに、着色油100μLを加えて、1分間、振盪撹拌し、その後、1時間室温で静置する。
(3)着色油の色の、水層への分散程度を観察して、++:全体によく着色、+:局部的に着色、±:僅かに着色、−:表層又は下層に着色液が残存している、の4段階で評価する。
1.クエン酸モノグリ(理研ビタミン(株)ポエムK−30 HLB 3.0)
2.コハク酸モノグリ(理研ビタミン(株)パンマック200V)
3.ジアセチル酒石酸モノグリ(理研ビタミン(株)ポエムW−90VP)
(1)澱粉粒又はセルロース粉末固定化リパーゼを製造し、提供することができる。
(2)固定化の方法は、例えば、澱粉粒に、リパーゼの水又はエタノール水溶液を、澱粉粒重量当たり5〜40%加えて、混合、結合させるものであり、極めて簡便、低コストである。
(3)澱粉粒に、α−アミラーゼ及び/又はグルコアミラーゼを作用させて、その表面に穴を開けた澱粉粒又は粒表面を一部糊化した澱粉粒を用いることで、固定化を増強することができる。
(4)澱粉粒表面又はセルロース粉末を、酸化処理することで、固定化を増強することができる。
(5)卵白、ゼラチンなどのタンパク質で、澱粉粒又はセルロース粉末表面をコーテイングしてから、酵素を結合させることにより、固定化を増強することができる。
(6)タンパク質素材でコーテイングすることにより、固定化を強化することができる。
(7)本発明の固定化リパーゼにより、油脂及びその分解物と、グルコース、オリゴ糖から成る被反応物を用いて、乳化素材を製造することができる。
Claims (6)
- 乳化能を有する反応生成物から成る乳化素材を製造するための固定化リパーゼを製造する方法であって、
澱粉粒の形態を保持した澱粉粒又はセルロース粉末に、酵素リパーゼを混合し、該澱粉粒又はセルロース粉末担体に、粉末状態を維持して、該酵素リパーゼを結合させて、澱粉粒又はセルロース粉末固定化リパーゼとする工程において、
1)上記澱粉粒に、α−アミラーゼ及び/又はグルコアミラーゼを作用させて、その表面に穴を開けた空洞澱粉粒又は粒表面をそのまま又はそれを一部糊化した澱粉粒を用いて、
2)澱粉粒表面又はセルロース粉末を、酸化処理した状態で、及び/又は、
3)澱粉粒又はセルロース粉末表面に卵白、ゼラチン、カゼインナトリウム、又はツェインをコーテイングして、
4)これらに酵素リパーゼを混合して該澱粉粒又はセルロース粉末担体に、粉末状態を維持して、酵素リパーゼを結合させる処理を行うことで澱粉粒又はセルロース粉末固定化リパーゼとするか、又は、
5)澱粉粒固定化リパーゼと、セルロース粉末固定化リパーゼ又はセルロースを混合した固定化リパーゼとする、
ことを特徴とする澱粉粒及び/又はセルロース粉末固定化リパーゼの製造方法。 - 上記澱粉粒に、粉末状態を維持できる程度に、水分を加えて、高温処理して、表面を糊化させた澱粉粒とする、請求項1に記載の固定化リパーゼの製造方法。
- 上記澱粉粒固定化リパーゼと、上記セルロース粉末固定化リパーゼ又はセルロースを混合した固定化リパーゼとすることにより、該固定化リパーゼをカラムに詰めて使用する際に、固定化リパーゼが油液中での反応の際に固着するのを防止し、かつ、反応液の流速を維持するようにした、請求項1に記載の固定化リパーゼの製造方法。
- 請求項1から3のいずれかに記載の方法で製造した固定化リパーゼを使用して、原料として、油脂及び/又はその分解物と、グルコース、又はオリゴ糖の被反応物を用いて、当該固定化リパーゼを上記原料に作用させて、乳化能を有する油脂反応物から成る乳化素材を製造することを特徴とする乳化素材の製造方法。

- 請求項1から3のいずれかに記載の方法で製造した固定化リパーゼを、油脂及び/又はその分解物と、グルコース、又はオリゴ糖から成る被反応物に作用させて作製した乳化能を有する反応生成物から成る乳化素材を構成要素として含むことを特徴とする乳化剤。
