Description
ハクサイ軟腐病を防除する乳酸菌の探索を目的として、発酵乳や発酵食品などから乳酸菌を分離した。乳酸菌の分離方法は次の通りである。すなわち、サンプル(例えば、イカの塩辛)を10倍量の滅菌水とともに磨砕し、その磨砕液を滅菌水で適宜希釈した。その希釈液をシャーレに入れ、約50℃に冷ましたMRS寒天培地(DifoTMLactobacilli MRS Broth 55g、炭酸カルシウム5g、寒天12g/L)と混釈したのち固化し、30℃で2,3日間培養した。培養後、周りにクリアゾーンを形成しているコロニーを乳酸菌として分離した。

SOK04SW株の細菌学的性質を分析したところ、次の通りであった。
MRS寒天培地(Oxoid製、Hampshire、英国)を用い、培養温度30℃、培養時間24時間、嫌気条件で培養した菌株を供試菌体とした。
DNA抽出はInstaGene Matrix(BIO RAD製、CA、米国)により、PCRはPrimeSTAR HS DNA Polymerase(タカラバイオ製)により、サイクルシークエンスはBigDye Terminator v3.1 Cycle Sequencing Kit(Applied Biosystems製、CA、米国)により、それぞれ実施した。使用プライマー(中川恭好他:遺伝子解析法 16S rRNA遺伝子の塩基配列決定法、日本放線菌学会編、放線菌の分類と同定、88-117pp.日本学会事務センター、2001)は、9F, 339F, 785F, 1099F, 536R, 802R, 1242R, 1541Rである。シークエンスはABI PRISM 3100 Genetic Analyzer System(Applied Biosystems製、CA、米国)により行い、解析ソフトウェアはAuto Assembler(Applied Biosystems製、CA、米国)及びアポロン(テクノスルガ製)を用いて、アポロンDB細菌基準株データベース(テクノスルガ製)及び国際塩基配列データベース(GenBank/DDBJ/EMBL)に対する相同性検索を実施した。
上記で得られた16S rDNAの塩基配列約1500bpを用い、推定される近縁菌群の基準株由来の16S rDNAを取得して分子系統解析を実施した。詳細には、データベースとしてアポロンDB−BA3.0(テクノスルガ製)及び国際塩基配列データベース(GenBank/DDBJ/EMBL)を用い、またソフトウェアとしてCLUSTAL W(THOMPSON (J.D.)他: CLUSTAL W: improving the sensitivity of progressive multiple sequence alignment through sequence weighting positions-specific gap and weight matrix choice. Nucleic Acids Research, 1994, 22:4673-4680)及びMEGA ver3.1(KUMAR,(S.)他: MEGA3: Integrated Software for Molecular Evolutionary Genetics Analysis and Sequence Alignment. Briefings in Bioinformatics, 2004,5,150-163)を用いた。

株名の末尾のTはその種の基準株を示す。括弧内はアクセッション番号であり、同番号にTecsrg標記のある株は(株)テクノスルガ・ラボのシークエンス株であることを示す。
MRSブロス(Oxoid製、Hampshire、英国)を用い、培養温度30℃、培養時間24時間、嫌気条件で培養した培養菌株から河本らの方法(河本好章、江崎孝行「細菌の系統分類と同定方法」第18回日本細菌学会技術講習会テキスト、日本細菌学雑誌、2000, 55, 545-584)によりDNAを抽出・精製した。
上記4と同条件で培養した培養菌株から上記河本らの方法によりDNAを抽出し、得られた粗抽出DNAを浜本の方法(浜本牧子「細胞外多糖を生成する担子菌系酵母のDNA抽出・精製法」、Microbiol. Cult. Coll. 1994, 10, 116-118)によりセシウムクロライドを用いた超遠心分離によって精製した。
上記4と同条件で培養した培養液を遠心分離して上清を回収し、これより有機酸を抽出した。抽出した有機酸からHPLCを用いて乳酸を測定した。標準試料としては、L−乳酸(和光純薬工業製)とD−乳酸(武蔵化学研究所製)を用いた。また、HPLC分析の条件は、カラム:SUMICHIRAL OA-5000 4.6φ×150mm、移動相:2mM硫酸銅2%イソプロパノール、検出:UV254nm、流速:0.8mm/分、温度:40℃、注入量:5μLとした。
上記2の16S rDNAによる相同性検索の結果より、SOK04SW株は、ラクトバチルス属に含まれ、既知種ではラクトバチルス・ナンテンシス(L. nantensis)に最も近い近縁であると考えられる。

SOK04SW株は、京都府の一機関である京都府農業資源研究センター(所長:並木 隆和、あて名:郵便番号619−0244 日本国京都府相楽郡精華町大字北稲八間小字大路74番地)の名称で、次の通り寄託されている。

実施例2の上記1及び2と同様の方法により、SOK04BY株の細菌学的性質を分析した。

植物病害防除能を有する乳酸菌として、上記SOK04SW株とSOK04BY株を用いて、乳酸菌製剤を調製した。調製は、菌株10重量%、ラウリル硫酸ナトリウム0.5重量%、リグニンスルホン酸ナトリウム4.5重量%、ホワイトカーボン2.5重量%、クレー82.5重量%を均一に混合粉砕することにより水和剤とした。
軟腐病菌に汚染された圃場にハクサイ苗(品種:無双)を定植して、慣行栽培を行った。その後、結球開始期頃から下記表5に示す乳酸菌製剤の1000倍液(製剤1gを1Lの水に希釈した液)を3〜5回散布し、収穫期に発病調査を実施した。なお、乳酸菌製剤を散布しない対照区も設けた。
防除価=(1−処理区の発病度/対照区の発病度)×100
実施例5とは異なる軟腐病汚染圃場において、実施例5と同様にして病害防除効果試験を行った。但し、本実施例では、下記表6に示す乳酸菌製剤の500倍液と1000倍液をそれぞれ用いた。また、比較のために、市販の微生物農薬(非病原性エルヴィニア細菌剤)であるバイオキーパー(セントラル硝子株式会社製)も用いた。
実施例5及び6とは異なる軟腐病汚染圃場において、実施例5と同様にして病害防除効果試験を行った。また、比較のために、上記市販のバイオキーパーも用いた。
実施例5〜7とは異なる軟腐病汚染圃場において、実施例5と同様にして病害防除効果試験を行った。その結果、表8に示すように、SOK04SW株とSOK04BY株を併用したSW・BY混合製剤は、単独使用のSOK04SW株よりも優れた病害防除効果を有していた。
ミズナ種子(品種:城南千筋)270mgをSOK04SW株の懸濁液(107cfu/ml、800ml)に20℃、24時間浸種処理した後、市販の培土を詰めた硬質ポット(直径15cm)に14粒ずつ播種した(8反復/区)。イタリアンライグラス培地(種子粉末1g、蒸留水5ml)で20℃、一週間培養した立枯病菌(ピシウム属、Pythium aphanidermatun OPU693株)を滅菌培土50gと一緒に磨砕し、それを滅菌培土で300倍に希釈して汚染土を作製し、該汚染土で上記の播種したミズナ種子を覆土した。温室内で栽培し、播種7日後及び14日後に、それぞれ不発芽株と立枯株を発病株として調査し、併せて発病株とした。なお、対照区では、滅菌水にミズナ種子を20℃、24時間浸種処理し、その他は同様に行った。

- 請求項1又は2に記載の菌株で植物及び/又は土壌を処理することを特徴とする植物病害防除方法。
