Description
(i)液体試料塗布部位;
(ii)試料塗布部位と液体連通する少なくとも1つの試験流路;
(iii)試料塗布部位と流体連通する参照通路、および
(iv)試験流路と参照流路が互いに接触する連結部位であって、典型的には出口、導管、チャンバ、又は、液体の前方への流動を可能にする他の部分を備える連結部位、
を備え、
ここで、参照流路に沿って流動する液体は、連結部位に到達した時、試験流路に沿った液体の流動を防止する効果を有する。
逆に、試験流路に沿って流動する液体が、参照流路の液体より前に連結部位に到達する場合、少なくとも幾つかの実施形態では、参照流路に沿った液体の流動が防止され得る可能性がある。参照流路又は試験流路に沿った液体の流動の防止は必ずしも永久的ではない:検査が実施され、読み取られる時間内で液体の流動が防止されれば十分である。
図1は、本発明によるデバイスを示す。デバイスは、共にキャピラリチャネルを備える試験流路4と参照流路6に流体連通した試料塗布部位2を有する。フィルタ8は、任意に、流路の一方又は両方に設けられてもよい。流路は、下流で、共通チャネル12に繋がる連結部位10で集束する。インジケータ部位14は、連結部位10の下流に設けられてもよい。
アガロースでコーティングされた200μmのポリエステル(ゲルボンド(GelBond)、BMA)からベース層を調製した。適切なマイクロ流体の特徴を、厚さ75μmのヒートシール接着PE層からグラフトテック(GraftTeC)カッターを使用して切り取り、2層を一緒に積層した。最後に第3の層を中間層に積層し、75μmのマイクロ流体チャネルを得た。
本発明による検査デバイスの代替の実施形態を図2に示す。図1に示されている実施形態のものと機能的に同等の構成要素は共通の参照番号で示す。
本発明の実現可能性を実際に実証するために、15μmのポリスチレンビーズ(ポリサイエンス(Polyscience))をアミノデキストラン500,000RMM、次いでNHS−LCLC−ビオチンでコーティングし、ビオチン化ラテックスビーズを調製した(NHS=N−ヒドロキシスクシンイミジル、LCLC=「長鎖」、即ち、炭素数12のスペーサー)。200μg/mlのBSA溶液50μlに(非特異的結合部位をブロックするため)、15μmビオチン粒子の5%溶液50μlを添加し、ボルテックスで混合した。試験流体を調製するため、ボルテックスで混合しつつ、ビオチン化された粒子のBSA溶液にストレプトアビジン1μm磁性粒子の溶液5μlを添加した。調製後直ぐに、後述のマイクロ流体デバイスに流体を添加した。
Claims (19)
- 液体試料の少なくとも1つの性質を検査する流体検査デバイスであって、
(i)液体試料塗布部位、
(ii)前記液体試料塗布部位と液体連通する少なくとも1つの試験流路、
(iii)前記液体試料塗布部位と液体連通する参照流路、
(iv)前記試験流路と前記参照流路が互いに接触する連結部位であって、出口、導管、チャンバ、又は、液体試料の前方への流動を可能にする他の部分を備える連結部位、および
(v)前記連結部位の下流に設けられた1つ以上の通気孔であって、液体試料を前記試験流路と前記参照流路に沿って前進させるのに、空気又は他の流体の当該通気孔を介する排出を必要とする通気孔
を備えるデバイスであり、
前記試験流路又は参照流路の一方を流れる液体試料が先に前記連結部位に到達した場合に、前記通気孔の上流に空気ブロック又は他のガスブロックを生じさせて、前記試験流路又は参照流路の他方に沿った液体試料の流れの更なる前進を阻害するように構成され、
前記試験流路および/又は参照流路が、マイクロ流体チャネル、多孔質キャリヤ、又は両方の組み合わせを備え、
前記マイクロ流体チャネルの少なくとも一部が、毛細管の寸法である、
流体検査デバイス。 - 前記試験流路および/又は参照流路が、フィルタ、インキュベーション部位、チャンバ、流動制限の1つ以上を備える、請求項1に記載の流体検査デバイス。
- 前記試験流路および/又は参照流路が、標識又はインジケータを備える、請求項1又は2に記載の流体検査デバイス。
- 前記標識又はインジケータが、前記液体試料と接触した時、移動可能である、請求項3に記載の流体検査デバイス。
- 前記標識又はインジケータが、酵素、蛍光色素、着色色素、および金コロイド粒子又は他の金属コロイド粒子からなる群から選択される、請求項3又は4に記載の流体検査デバイス。
- 前記液体試料中に存在する目的の検体と反応する試薬、又は前記液体試料中に存在する目的の検体と結合する結合相手を含む、請求項1〜5のいずれか一項に記載の流体検査デバイス。
- 前記試薬又は結合相手が、前記試験流路内に配置されている、請求項6に記載の流体検査デバイス。
- 前記試薬の前記検体との反応又は前記結合相手の前記検体との結合が、前記試験流路を流れる液体試料の流速を低下させる、請求項7に記載の流体検査デバイス。
- 前記結合相手が、抗体、抗体の抗原結合フラグメント、又は、抗体若しくはその抗原結合フラグメントの多量体を含む、請求項6〜8のいずれか一項に記載の流体検査デバイス。
- 前記試薬又は結合相手が、粒子状であるか又は粒子と結合している、請求項6〜9のいずれか一項に記載の流体検査デバイス。
- 前記試薬又は結合相手は、目的の検体が存在するとき、前記試験流路に沿った液体試料の流動を妨げる又は遅延させるのに十分な凝集を引き起こす、請求項6〜10のいずれか一項に記載の流体検査デバイス。
- 前記検査の結果を示すインジケータ部位を更に備える、請求項1〜11のいずれか一項に記載の流体検査デバイス。
- 前記インジケータ部位が前記連結部位の下流に設けられている、請求項12に記載の流体検査デバイス。
- インジケータ部位が前記連結部位の上流に設けられている、請求項12に記載の流体検査デバイス。
- インジケータ部位が前記試験流路と前記参照流路の両方に設けられている、請求項14に記載の流体検査デバイス。
- 前記検査結果に対して二者択一の結果を提供する、請求項1〜15のいずれか一項に記載の流体検査デバイス。
- 非デジタル検査デバイスである、請求項16に記載の流体検査デバイス。
- 妊娠試験デバイスであり、前記目的の検体がhCGを含む、請求項8〜17のいずれか一項に記載の流体検査デバイス。
- 液体試料中の目的の検体の存在および/又は量を検出する方法であって、前記液体試料を請求項1〜18のいずれか一項に記載の流体検査デバイスの前記液体試料塗布部位に塗布する工程、および、前記検査結果を示す又は記録する工程を含む方法。
