Description
〔1〕 被験物質と、アルカリ剤と、TRPA1を発現する細胞とを接触させ、前記アルカリ剤によりTRPA1を介して引き起こされる生理学的事象を測定することを特徴とする、アルカリ剤の刺激を抑制する物質の評価方法、
〔2〕 前記生理学的事象が、細胞内カルシウムイオン濃度の変化である前記〔1〕記載の評価方法、
〔3〕 前記TRPA1を発現する細胞が、TRPA1をコードする核酸が発現可能に宿主細胞に導入されている細胞である前記〔1〕又は〔2〕記載の評価方法、並びに
〔4〕 前記宿主細胞が、HEK293細胞、CHO細胞、COS−7細胞及びNIH3T3細胞からなる群より選ばれた細胞である前記〔3〕記載の評価方法、
に関する。
(A)配列番号:1に示される塩基配列に対して、米国生物工学情報センター(NCBI)のウェブページ〔アドレスhttp://www.ncbi.nlm.nih.gov/BLAST/〕にて利用できるNCBI BLASTにより、Cost to open gap 11、Cost to extend gap 1、expect value 10、wordsize 11の条件(デフォルト値)下、評価対象の配列をアライメントし、算出される値として、少なくとも60%、好ましくは、80%以上、より好ましくは、90%以上の配列相同性を有する塩基配列からなり、かつコードされるポリペプチドが、陽イオンを透過させる機能を少なくとも発現するポリペプチドである核酸、
(B)配列番号:2において、少なくとも1個、好ましくは、1個又は数個のアミノ酸残基の置換、欠失又は付加を有するアミノ酸配列をコードし、コードされるポリペプチドが、陽イオンを透過させる機能を少なくとも発現するポリペプチドである核酸、
(C)配列番号:1に示される塩基配列に完全に相補的な塩基配列からなる核酸とストリンジェントな条件下にハイブリダイズし、コードされるポリペプチドが、陽イオンを透過させる機能を発現するポリペプチドである核酸、
などが挙げられる。前記核酸は、ヒト由来の核酸であってもよく、他の動物に由来する核酸であってもよい。
TRPA1を発現する細胞の作製
ヒトTRPA1をコードするcDNA〔配列番号:1(GenBankアクセッション番号:NM_007332)の63位〜3888位のポリヌクレオチド〕を、哺乳動物細胞用ベクターである商品名:pcDNA3.1(+)(インビトロジェン社製)のKpnI/XbaIサイトに挿入し、ヒトTRPA1発現ベクターを得た。
被験物質を、アルカリ剤としての溶液A〔組成:140mM NaCl、5mM KCl、2mM MgCl2、2mM CaCl2、10mM グルコース、10mMトリス塩酸緩衝液(pH9.0)〕に添加し、試験液を作製した。また、前記溶液Aを標準試験液として用いた。
Claims (4)
- 被験物質と、アルカリ剤と、TRPA1を発現する細胞とを接触させ、前記アルカリ剤によりTRPA1を介して引き起こされる生理学的事象を測定することを特徴とする、アルカリ剤の刺激を抑制する物質の評価方法。
- 前記生理学的事象が、細胞内カルシウムイオン濃度の変化である請求項1記載の評価方法。
- 前記TRPA1を発現する細胞が、TRPA1をコードする核酸が発現可能に宿主細胞に導入されている細胞である請求項1又は請求項2記載の評価方法。
- 前記宿主細胞が、HEK293細胞、CHO細胞、COS−7細胞及びNIH3T3細胞からなる群より選ばれた細胞である請求項3記載の評価方法。
