Description

ySion、およびRB含有内部断片イオンよりなる群から選ばれる1種以上の断片イオンであることを特徴とする。
[化学式2]
酸型MBIT試薬(XMBIT−OH、X=LまたはH)は、標準固相ペプチド合成法または液相有機合成法によって合成された。標準固相ペプチド合成法としては、アミノ酸残基が質量調節基として使用され、該当質量調節基がアラニン(Ala)、セリン(Ser)、ヒスチジン(His)、バリン(Val)、グルタミン(Gln)、フェニルアラニン(Phe)、アルギニン(Arg)またはチロシン(Tyr)などの天然アミノ酸の側鎖であるMBIT試薬と、アシル基またはアミノ酸残基が質量調節基として使用され、該当質量調節基がエチル(C2)、プロピル(C3)、ブチル(C4)、ペンチル(C5)、ヘキシル(C6)、ヘプチル(C7)またはオクチル(C8)である全てのMBIT試薬を合成することができる。液相有機合成法としては、アミノ酸残基が質量調節基として使用され、当該質量調節基がヘキシル(C6)、ヘプチル(C7)またはオクチル(C8)である場合の酸型MBIT試薬を合成することができる。
物質
無水N,N−ジメチルホルムアミド(DMF)、ピペリジン、ジクロロメタン(DCM、HPLCグレード)、トリフルオロ酢酸(TFA、HPLCグレード)、チオアニソール(TA、>99.5%)、エタンジチオール(EDT、>99.5%)、無水酢酸、プロピオン酸、酪酸、ペンタン酸、ヘキサン酸、ヘプタン酸、オクタン酸、ノナン酸、N−Fmoc−アラニンはSigma−Aldrich(St.Louis、MO)から購入した。酢酸−d3、N−Fmoc−アラニン−3,3,3,−d3はCDN isotope(Toronto、カナダ)から購入した。2−塩化トリチルレジンはMerckから購入した。N,N’−ジイソプロピルカルボイミド(DIC)、1−ベンゾトリアゾール、およびN末端がFmocで保護された他の全てのアミノ酸はAdvanced ChemTechから購入した。
1)段階1
N−Fmoc−アラニンまたはN−Fmoc−アラニン−3,3,3−d3(75mg)を、水分が完全に除去されたジクロロメタン溶液1mLに溶かした。ジクロロメタンのみではよく溶けないので、100μLのDMFを入れて完全に溶解させた。準備したN−Fmocアミノ酸溶液と170μLのDIPEAを、火炎乾燥したバイアルに入っている2−塩化トリチルレジン0.1gと混合し、2〜4時間緩やかに攪拌した。レジンをペプチド合成用ポリプロピレン材質のカートリッジ(総量:5mL)に移した後、十分な量のジクロロメタン/メタノール/DIPEA混合溶液(17/2/1、v/v/v)で3回洗浄した。その後、十分な量のジクロロメタンで3回、DMFで2回さらに洗浄した後、ジクロロメタンで2回さらに洗浄し、しかる後に、レジンから溶液を完全に除去し、減圧条件で完全に乾燥させる。
段階1で製造された乾燥レジンに約3mL程度のDMFを入れて2〜3分間十分振とうし、DMFを除去する過程を約5回繰返し行ってレジンをDMFに十分浸漬した。DMFに混ざっている25%ピペリジン溶液を約3mL程度レジンに混ぜ、5分間攪拌した後、溶液を除去した。その後、さらに25%ピペリジン溶液を3mL程度レジンにさらに入れた後、15分間攪拌し、溶液を除去した。しかる後に、レジンを十分な量のDMFで3回、メタノールで3回洗浄し、さらにDMFで3回洗浄した。
アミノ酸側鎖が質量調節基として使用されるMBIT試薬を合成する場合には次のように製造した。
段階3で製造されたレジンに、DMFに混ざっている25%ピペリジン溶液を約3mL程度添加し、5分間攪拌した後、溶液を除去した。その後、さらに25%ピペリジン溶液を3mL程度レジンにさらに加えた後、15分間攪拌し、溶液を除去した。その後、レジンを十分な量のDMFで3回、メタノールで3回洗浄し、さらにDMFで3回洗浄した。
アミノ酸側鎖が質量調節基として使用されるMBIT試薬を合成する場合には次のように製造した。
段階5で製造されたレジンに、TFA/ベンゼン/TA/蒸留水/EDT混合溶液(16.5/1/1/1/0.5、v/v/v/v)2mLを混ぜ、3時間攪拌した。この過程で合成された酸型MBIT試薬がレジンから分離された。レジンは全て濾過し、残った溶液を別途に集め、体積が200μL以下となるように窒素によって乾燥させた。その溶液に、冷たく冷したエーテルを添加して白色粉末状の生成物(酸型MBIT試薬)を沈殿させた。沈殿した生成物は、冷たく冷したエーテルで3回または4回洗浄した後、減圧条件で完全に乾燥させた。
物質
2−アミノ−4−ペンテン酸、無水酢酸(Ac2O−d0)、Boc−l−アラニン−d0、TFA、4−オクテン、5−デケン、1−ヘプテン、およびグラブス触媒(Grubb’s catalyst、2nd generation)はSigma−Aldrich(St.Louis、MO)から購入し、dl−アラニン−d3、および過重水素化無水酢酸(Ac2O−d6)はCDN Isotopes(Quebec、カナダ)から購入した。
1)段階1
2−アミノ−4−ペンテン酸(2mmoL)をpH9〜10の水(4mL)に溶かし、無水酢酸(4.0mmoL)を0℃で投入した。8M NaOHを投入してpH10程度に調節し、反応混合物を4時間0℃で攪拌した。濃い塩酸溶液を投入してpH2以下に作って反応を終了した。生成物はメタノールに溶かし、精製して乾燥させて2−アセトアミド−4−ペンテン酸を固体として回収した。
アラニンベンジルエステルは、Bocで保護されたアラニンに臭化ベンジルを加えてBoc−アラニンベンジルエステルを作った後、TFAを加えてBocを除去する過程によって作られる。ジオキサンと水との混合物(2/1、v/v)に0.33Mで溶けた1−アラニン−d3(1mmoL)に、1.5mLの1M NaOHとジ−3次−ブチルジカルボネート(1.1mmoL)を投入した後、室温で6時間攪拌した。ジオキサンを蒸発させた後、氷を用いて混合物を冷却し、KHSO4飽和水溶液を加えてpHを2〜3程度に低めた。有機生成物は3回にわたって10mLの酢酸エチル(EA)で抽出され、無水Na2SO4の下で乾燥した。シリカゲルクロマトグラフィー精製によって、Boc−dl−アラニン−d3(0.14g、0.74mmoL)が得られた。0.5mmoLのBoc−dl−アラニン−d0またはBoc−dl−アラニン−d3を無水アセトン(5mL)に溶かし、炭酸カリウム(0.75mmoL)と臭化ベンジル(0.55mmoL)を加えた。5時間の還流後、反応生成物は室温に冷却し、濃縮された後、クロロホルム(10mL)に溶解された。有機層を炭酸ナトリウムの飽和水溶液(30mL)で洗浄し、Na2SO4の下で乾燥させた後、シリカゲルクロマトグラフィーで精製してBoc−アラニン−d0ベンジルエステルまたはBoc−アラニン−d3ベンジルエステルを白色固体として得た。Boc−アラニン−d0ベンジルエステルまたはBoc−アラニン−d3ベンジルエステル(0.98mmoL)を無水DCM(10mL)に溶かし、8mmoLのTFAを0℃で加えて1時間攪拌した。減圧の下で溶媒を除去した後、残留物は高真空の下で乾燥させた。油状生成物(アラニン−d0ベンジルエステルまたはアラニン−d3ベンジルエステル)は無水THF(2mL)に保管した。
段階2で製造された、THF(5mL)に溶けたアラニン−d0ベンジルエステルまたはアラニン−d3ベンジルエステル(0.55mmoL)に、BOP試薬(1.01mmoL)を加えた後、30分間室温で攪拌した。DIPEA(3.36mmoL)を0℃で投入し、さらに室温で15分間攪拌した後、段階1で製造された、無水THFに溶けた2−アセト−d3−アミド−4−ペンテン酸または2−アセト−d0−アミド−4−ペンテン酸を加えた後、一晩室温で攪拌した。溶媒を蒸発させた後、残留物はEAに溶解させた。有機層を水で洗浄した。残留油状生成物をシリカゲルフラッシュクロマトグラフィーで精製することにより、ベンジル2−(2−アセトアミド−4−ペンテンアミド)プロパネートを無色固体として得た。
DCMに、段階3で製造されたベンジル2−(2−アセトアミド−4−ペンテンアミド)プロパネート、アルケン(4−オクテン、5−デケン、または1−ヘプテン)、およびグラブス触媒を仕込んで40℃で24時間還流させた。触媒と溶媒を除去した後、シリカゲルクロマトグラフィーで精製した。この反応生成物は、20mol%のPd(OH)2と共に無水エタノールで混合した後、室温で一晩1気圧のH2圧力下で攪拌した。触媒をフィルターリングして濾した後、生成物を真空下で濃縮し、メタノールとエーテルの1:1混合物を用いた再結晶を行うことにより、酸型MBIT試薬を得た。
物質
無水アセトニトリル(ACN、HPLCグレード)、無水DMF、ヒドロキシルアミンヒドロクロライド、トリフルオロ酢酸(TFA、HPLCグレード)、α−シアノ−4−ヒドロキシ桂皮酸(HCCA)、N−ヒドロキシスクシンイミド(NHS)は、Sigma−Aldrich(St.Louis、MO)から購入した。1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC)は、Pierce(Rockford、Il)から購入した。ウシ血清アルブミン(BSA)パウダーはCalbiochemから購入した。

アミノ酸残基が質量調節基として使用され且つ該当質量調節基がアラニン(Ala)、セリン(Ser)、ヒスチジン(His)、バリン(Val)、グルタミン(Gln)、フェニルアラニン(Phe)、アルギニン(Arg)、またはチロシン(Tyr)などの天然アミノ酸側鎖であるMBIT試薬を用いて、BSAのトリプシンペプチドを標識する反応を行った。
エチル(C2)、プロピル(C3)、ブチル(C4)、ペンチル(C5)、ヘキシル(C6)、ヘプチル(C7)またはオクチル(C8)を質量調節基として使用した場合のうち、アミノ酸残基を質量調節基として有し且つ該当質量調節基がヘキシル(C6)、ヘプチル(C7)またはオクチル(C8)であるMBIT試薬を用いて、Hsc82のトリプシンペプチドに標識する反応を行った。
XMBITが連結されたモデルペプチド溶液は、MALDI質量分析のために0.1%TFA溶液に500倍希釈した。LMBITおよびHMBITが連結されたモデルペプチドは、7つの多様な比率で混合した。([L]/[H]=1/1、2.3/1、4/1、6.3/1、9/1、12.3/1、16/1)の7つの混合比で準備された各試料は、マトリクス溶液(5mg/mL HCCA in 50/50/0.1、H2O/ACN/TFA)と1:1の体積比で混合した。試料/マトリクス混合物1μLがMALDIプレートにロードされた。一つの試料スポット当りロードされたモデルペプチドの量はアンジオテンシンIIとロイシンエンケファリンがそれぞれ約250fmolずつであった。
HMBITまたはLMBITに連結されたトリプシンペプチド1:1の比率で混合し、その一分取量(6.4μL)を、PepMapカラム(孔径100Å、粒径3mm、内径75mm、長さ150mm)を取り付けた逆相ナノ液相クロマトグラフィー(Reverse-Phase Nano-Liquid Chromatography、RP−nano−LC)分離機(LC−packings社、Sunnyvale、CA)に注入した。LCは流速0.3μL/分で60分間作動し、2−溶媒勾配を用いて作動した。体積比でH2O/ACN/TFA=95/5/0.1の溶媒AとACN=TFA=100/0.1の溶媒Bが2−溶媒勾配に利用された。[溶媒A]/[溶媒B]勾配は100/0から始まり、0〜20分の間に30/70に変わり、20〜40の間に0/100に変わり、40〜45分まで0/100に維持された。45分となる時点で100/0に急激に落ち、45〜60分の間100/0の比率が維持された。溶出される試料は、マトリクス溶液と共に25秒毎に単一MALDIスポットにProbot microfraction collectorを用いて集められた。60分間合計144個のMALDIスポットにLC溶出してくる試料が分取されて塗布された。
前記方法によってMALDIターゲットに塗布された試料を分析するために、4700 Proteomic Analyzer(Applied Biosystems社、Foster City、CA)が陽イオンモードで質量対電荷比の範囲500〜2500Thで使用された。各塗布されたスポットに対してTime−of−Flight(TOF)スペクトルは1000個の反復レーザーショットに対する結果を蓄積することにより得られた。
(a)質量調節基がアラニン(Ala)、セリン(Ser)、ヒスチジン(His)、バリン(Val)、グルタミン(Gln)、フェニルアラニン(Phe)、アルギニン(Arg)、またはチロシン(Tyr)などの天然アミノ酸の側鎖である場合
−MBIT試薬の検証
製造されたMBIT試薬に正常的に合成されたかを検証するために、それぞれのMBIT試薬をアンジオテンシンII(1046.5Th)に標識して[MAG(1)+H]+イオンの質量を検出し(図8(a)〜図8(h))、タンデム質量分析(図9)を行った。LMBITとHMBITでそれぞれ標識されたアンジオテンシンIIは、同一の質量値で検出された。[MAG(1)+H]+イオンの質量値は、質量調節基がそれぞれアラニン側鎖の場合には1233.6Th、セリン側鎖の場合には1249.6Th、バリン側鎖の場合には1261.7Th、グルタミン側鎖の場合には1290.7Th、ヒスチジン側鎖の場合には1299.7Th、フェニルアラニン側鎖の場合には1309.7Th、アルギニン側鎖の場合には1318.7Th、チロシン側鎖の場合には1325.7Thである。標識シグニチャーと定量信号質量値は、質量調節基がアラニン側鎖の場合には188Th(b0)、114Th(LbS)、および117Th(HbS)、セリン側鎖の場合には204Th(b0)、130Th(LbS)および133Th(HbS)、バリン側鎖の場合には216Th(b0)、142Th(LbS)および145Th(HbS)、グルタミン側鎖の場合には245Th(b0)、171Th(LbS)および174Th(HbS)、ヒスチジン側鎖の場合には254Th(b0)、180Th(LbS)、および183Th(HbS)、フェニルアラニン側鎖の場合には264Th(b0)、190Th(LbS)および193Th(HbS)、アルギニン側鎖の場合には273Th(b0)、199Th(LbS)および202Th(HbS)、そしてチロシン側鎖の場合は280Th(b0)、206Th(LbS)および209Th(HbS)でそれぞれ計測された。前記結果より、本発明で製造された天然アミノ酸残基を用いたMBIT試薬が正常的に合成されたことが分かる。
図8(a)〜図8(h)はアンジオテンシンIIとロイシンエンケファリンとのペプチド混合物を8種のMBIT試薬対で反応させた後、それぞれをMALDI質量分析した結果を概略的に示す図である。図2の(b)に示した8つの相異なる質量調節基RTを有する8種のMBIT試薬対それぞれをモデルペプチドと反応させてMALDI−TOF質量分析した結果を図8(a)〜図8(h)に示す。図8(a)〜図8(h)に示すように、[MXX(n)+H]+のXXはペプチドの種類を示し(AG=アンジオテンシンII、LE=ロシンエンケファリン)、nはペプチドに連結されたMBIT試薬の個数を示す。N末端がアシル化されたジペプチド構造のMBIT試薬(N−アセチル−XXX−AlaまたはAc−XA)において、質量調節基を含むXxx(またはX)がアラニン(図8(a))、セリン(図8(b))、バリン(図8(c))、グルタミン(図8(d))、ヒスチジン(図8(e))、フェニルアラニン(図8(f))、アルギニン(図8(g))、およびチロシン(図8(h))の場合をそれぞれ示した。質量調節基がアラニンの場合には1233.6Th、セリンの場合には1249.6Th、バリンの場合には1261.7Th、グルタミンの場合には1290.7Th、ヒスチジンの場合には1299.7Th、フェニルアラニンの場合には1309.7Th、アルギニンの場合には1318.7Th、チロシンの場合には1325.7ThでアンジオテンシンIIに該当する[MAG(1)+H]+イオンが検出された。また、質量調節基がヒスチジンの場合とアルギニンの場合、809.5Thと828.5Thでロイシンエンケファリンに該当する[MLE(1)+H]+イオンがそれぞれ検出された。モデルペプチドにそれぞれのMBIT試薬で反応が行われた後で増加する質量値が、該当MBIT試薬によって理論的に増加しなければならない質量値と一致するので、各MBIT試薬は成功的に合成された。
−MBIT試薬の検証
製造されたMBIT試薬に正常的に合成されたかを検証するために、それぞれのMBIT試薬をアンジオテンシンII(1046.5Th)に標識して[MAG(1)+H]+イオンの質量を検出し(図17)、タンデム質量分析(図18)を行った。LMBITとHMBITでそれぞれ標識されたアンジオテンシンIIは同一の質量値で検出された。[MAG(1)+H]+イオンの質量値は、質量調節基がそれぞれエチルの場合には1247.7Th、プロピルの場合には1261.7Th、ブチルの場合には1275.7Th、ペンチルの場合には1289.7Th、ヘキシルの場合には1303.7Th、ヘプチルの場合には1317.7Th、オクチルの場合には1331.8Thで分析体がそれぞれ観察された。標識シグニチャーと定量信号質量値は、質量調節基がエチルの場合にはそれぞれ202Th(b0)、128Th(LbS)、および131Th(HbS)で、質量調節基がプロピルの場合にはそれぞれ216Th(b0)、142Th(LbS)、および145Th(HbS)で、質量調節基がブチルの場合にはそれぞれ230Th(b0)、156Th(LbS)、および159Th(HbS)で、質量調節基がペンチルの場合にはそれぞれ244Th(b0)、170Th(LbS)、および173Th(HbS)で、質量調節基がヘキシルの場合にはそれぞれ258Th(b0)、184Th(LbS)、および187Th(HbS)で、質量調節基がヘプチルの場合にはそれぞれ272Th(b0)、198Th(LbS)、および201Th(HbS)で、質量調節基がオクチルの場合にはそれぞれ286Th(b0)、212Th(LbS)、および215Th(HbS)で観測された。前記結果より、本発明で製造されたエチル(C2)、プロピル(C3)、ブチル(C4)、ペンチル(C5)、ヘキシル(C6)、ヘプチル(C7)またはオクチル(C8)を質量調節基として用いたMBIT試薬が正常的に合成されたことが分かる。
製造されたMBIT試薬とペプチドとの反応性を確認するために、それぞれのMBIT試薬をアンジオテンシンII(1046.5Th)で反応させて質量分析した。図17は互いに異なる質量調節基RTを有する7種のMBIT試薬対それぞれをモデルペプチドとしてのアンジオテンシンIIと反応させてMALDI−TOF質量分析した結果である。MBIT試薬において、質量調節基が(a)エチル、(b)プロピル、(c)ブチル、(d)ペンチル、(e)ヘキシル、(f)ヘプチル、および(g)オクチルの場合をそれぞれ示した。図17から分かるように、質量調節基がエチルの場合には1247.7Th、質量調節基がプロピルの場合には1261.7Th、質量調節基がブチルの場合には1275.7Th、質量調節基がペンチルの場合には1289.7Th、質量調節基がヘキシルの場合には1303.7Th、質量調節基がヘプチルの場合には1317.7Th、質量調節基がオクチルの場合には1331.8Thでそれぞれ分析体が観測された。また、各分析体に対してタンデム質量分析によって標識シグニチャーおよび定量信号の質量値を確認した。その結果より、反応の行われていないペプチドまたはMBITが2つ以上付いているペプチドなしで、アンジオテンシンIIに一つのMBITが付いている反応結果物のみが存在するので、反応がよく行われたことが分かる。
Claims (20)
- 前記R T は直鎖または分枝鎖のエチル、プロピル、ブチル、ペンチル、ヘキシル、ヘプチルまたはオクチル(ただし、イソプロピルを除く。)であることを特徴とする、請求項1に記載の可変質量ラベリング剤。
- 前記RSおよびRBはそれぞれメチルであり、RSおよびRBの少なくとも一つは一つ以上の重水素を含むことを特徴とする、請求項1または2に記載の可変質量ラベリング剤。
- 前記RSおよびRBはそれぞれCH3およびCD3であり、或いはそれぞれCD3およびCH3であることを特徴とする、請求項1〜3のいずれか1項に記載の可変質量ラベリング剤。
- 前記リンカーは、N−ヒドロキシスクシンイミジル基、N−ヒドロキシスルホスクシンイミジル基、ベンゾトリアゾール−1−イルオキシル基、ペンタハロベンジル基、および4−ニトロフェニル基よりなる群から選ばれることを特徴とする、請求項1〜4のいずれか1項に記載の可変質量ラベリング剤。
- 請求項1の化学式1で表される可変質量ラベリング剤を2種以上含む、可変質量ラベリング剤セット。
- 前記2種以上の可変質量ラベリング剤はそれぞれのRSとRBに含まれた重水素数が相異し、
前記2種以上の可変質量ラベリング剤は互いに重水素の数が同一であることを特徴とする、請求項6に記載の可変質量ラベリング剤セット。 - 請求項6または7に記載の可変質量ラベリング剤セットを2種以上含む、多重可変質量ラベリング剤セット。
- 請求項1〜8のいずれか一項に記載の可変質量ラベリング剤で標識された分析体を含む混合物、その塩またはその水和物。
- 前記分析体はタンパク質、炭水化物または脂質であることを特徴とする、請求項9に記載の混合物、その塩またはその水和物。
- 前記分析体はペプチドであることを特徴とする、請求項9に記載の混合物、その塩またはその水和物。
- 前記分析体は核酸または核酸誘導体であることを特徴とする、請求項9に記載の混合物、その塩またはその水和物。
- 前記分析体はステロイドであることを特徴とする、請求項9に記載の混合物、その塩またはその水和物。
- 請求項6〜8のいずれか1項に記載の可変質量ラベリング剤セットを分析体に結合させる段階と、
前記可変質量ラベリング剤セットが結合している分析体を分解して前記分析体を定量する段階とを含むことを特徴とする、ペプチド配列およびタンパク質定量同時分析方法。 - 前記定量のための分解法はタンデム質量分析法であることを特徴とする、請求項14に記載のペプチド配列およびタンパク質定量同時分析方法。
- 前記タンデム質量分析法は、ラベリング剤のRTに応じて、定量信号質量を与える定量信号の位置が変わることを特徴とする、請求項15に記載のペプチド配列およびタンパク質定量同時分析方法。
- 前記定量信号質量を与える定量信号はbSイオン、aSイオン、およびRBイオンよりなる群から選択される1種以上の内部断片イオンによることを特徴とする、請求項16に記載のペプチド配列およびタンパク質定量同時分析方法。
- 1)前記RTがエチル基の場合、定量信号質量(bS)が128と131Thで現れ、別の定量信号質量(aS)が100と103Thで現れ、標識シグニチャーが202Thで現れ、
2)前記RTが直鎖または分枝鎖プロピル基の場合、定量信号質量(bS)が142と145Thで現れ、別の定量信号質量(aS)が114と117Thで現れ、標識シグニチャー(b0)が216Thで現れ、
3)前記RTが直鎖または分枝鎖ブチル基の場合、定量信号質量(bS)が156と159Thで現れ、別の定量信号質量(aS)が128と131Thで現れ、標識シグニチャー(b0)が230Thで現れ、
4)前記RTが直鎖または分枝鎖ペンチル基の場合、定量信号質量(bS)が170と173Thで現れ、別の定量信号質量(aS)が142と145Thで現れ、標識シグニチャー(b0)が244Thで現れ、

7)前記R T が直鎖または分枝鎖オクチル基の場合、定量信号質量(bS)が212と215Thで現れ、別の定量信号質量(aS)が184と187Thで現れ、標識シグニチャー(b0)が286Thで現れることを特徴とする、請求項14〜17のいずれか1項に記載のペプチド配列およびタンパク質定量同時分析方法。 - 請求項6〜8のいずれか1項に記載の多重可変質量ラベリング剤セットを分析体に結合させ、分解して該当分析体を定量する、ペプチド配列およびタンパク質定量同時分析方法。
- 請求項6〜8のいずれか1項に記載の多重可変質量ラベリング剤セットを分析体に結合させて定量する過程で、1種の試料と残りのそれぞれ異なる試料との比率を請求項18に該当する方法でそれぞれ別々に定量する、多重定量分析方法。
