Description
Claims (18)
- 検体中における酵素又は酵素阻害剤の存在又は量を検出するための側方流動分析装置であって、
前記検体が塗布される塗布領域、前記塗布領域の下流に位置する共役領域、及び前記共役領域の下流に位置する検出領域を画定するクロマトグラフ媒体と、
前記塗布領域に拡散可能に固定され、前記酵素又は前記酵素阻害剤と接触することによって触媒反応を行って生成物を作成可能な分子基質と、
前記共役領域に拡散可能に固定され、前記生成物又は前記分子基質のいずれか一方と接触することによって検出信号を生成可能な検出可能物質とを含み、
前記検出領域は、前記生成物又は前記分子基質のいずれか一方と結合した検出可能物質を捕捉可能であり、
酵素又は酵素阻害剤の存在又は量が、前記検出領域における前記検出信号から測定可能であることを特徴とする装置。 - 請求項1に記載の側方流動分析装置であって、
前記分子基質が第1の特異的結合部分を有し、
前記生成物が第2の特異的結合部分を有することを特徴とする装置。 - 請求項2に記載の側方流動分析装置であって、
前記検出可能物質が、第3の特異的結合部分と直接的又は間接的に結合された状態で前記共役可能領域に拡散可能に固定されており、前記第3の特異的結合部分は、前記第1の特異的結合部分又は前記第2の特異的結合部分のいずれか一方と結合可能であることを特徴とする装置。 - 請求項3に記載の側方流動分析装置であって、
前記第3の特異的結合部分が、前記第1若しくは第2の特異的結合部分のいずれか一方に対して親和性を有する、又は、前記第1若しくは第2の特異的結合部分のいずれか一方と同一である若しくはその類似体であることを特徴とする装置。 - 請求項1乃至4のいずれかに記載の側方流動分析装置であって、
前記検出領域内に、前記分子基質又は前記生成物のいずれか一方と結合する第1の受容物質が固定されることを特徴とする装置。 - 請求項1乃至5のいずれかに記載の側方流動分析装置であって、
前記クロマトグラフ媒体が、第2の検出信号を生成可能な第2の検出領域をさらに画定することを特徴とする装置。 - 請求項6に記載の側方流動分析装置であって、
前記第2の検出領域内に、前記第2の検出信号を生成可能な検出可能物質と直接的又は間接的に結合可能な第2の受容物質が固定されることを特徴とする装置。 - 請求項7に記載の側方流動分析装置であって、
前記検体中の酵素の量が、前記第2の検出信号の強度と直接的に比例することを特徴とする装置。 - 請求項1乃至8のいずれかに記載の側方流動分析装置であって、
前記酵素が加水分解酵素であり、前記加水分解酵素がプロテアーゼ又はペプチダーゼであることを特徴とする装置。 - 請求項1乃至9のいずれかに記載の側方流動分析装置であって、
前記分子基質が、カゼイン、アルブミン、ヘモグロビン、ミオグロビン、ケラチン、ゼラチン、インスリン、プロテオグリカン、フィブロネクチン、ラミニン、コラーゲン、エラスチン又はそれらの誘導体であることを特徴とする装置。 - 請求項1乃至10のいずれかに記載の側方流動分析装置であって、
前記塗布領域をその下流側の隣接する領域から一時的に分離させる、取り除き可能なバリアを更に含むことを特徴とする装置。 - 検体中における酵素又は酵素阻害剤の存在又は量を検出するための方法であって、
塗布領域、前記塗布領域の下流に位置する共役領域、及び前記共役領域の下流に位置する検出領域を画定するクロマトグラフ媒体、前記塗布領域に拡散可能に固定され、かつ前記酵素又は前記酵素阻害剤と接触することによって触媒反応を行って生成物を生成可能な分子基質、並びに前記共役領域に拡散可能に固定され、かつ前記生成物又は前記分子基質のいずれか一方と接触することによって検出信号を生成可能な検出可能物質を含む側方流動分析装置を提供するステップと、
前記塗布領域に前記検体を塗布させるステップと、
前記分子基質に前記酵素又は酵素阻害剤を接触させることによって触媒反応を行わせ、前記生成物を生成させるステップと、
前記検出可能物質に前記生成物又は前記分子基質を接触させることによって前記検出信号を生成させるステップと、
前記検出領域で、前記前記生成物又は前記分子基質のいずれか一方と結合した検出可能物質を捕捉するステップと、
前記検出領域内における前記検出信号の存在又は強度を測定するステップとを含むことを特徴とする方法。 - 請求項12に記載の方法であって、
前記分子基質が第1の特異的結合部分を有し、
前記生成物が第2の特異的結合部分を有することを特徴とする方法。 - 請求項13に記載の方法であって、
前記検出可能物質が、第3の特異的結合部分と直接的又は間接的に結合しており、
前記検出信号を生成させるステップが、前記第1又は第2の特異的結合部分のいずれか一方を前記第3の特異的結合部分に結合させるステップを含むことを特徴とする方法。 - 請求項12乃至14のいずれかに記載の方法であって、
前記検体中の酵素の量が、前記検出信号の強度と直接的又は間接的に比例することを特徴とする方法。 - 請求項12乃至15のいずれかに記載の方法であって、
前記酵素が加水分解酵素であり、前記加水分解酵素がプロテアーゼ又はペプチダーゼであることを特徴とする方法。 - 請求項12乃至16のいずれかに記載の方法であって、
前記分子基質が、カゼイン、アルブミン、ヘモグロビン、ミオグロビン、ケラチン、ゼラチン、インスリン、プロテオグリカン、フィブロネクチン、ラミニン、コラーゲン、エラスチン又はそれらの誘導体であることを特徴とする方法。 - 請求項12乃至17のいずれかに記載の方法であって、
前記側方流動分析装置が、前記塗布領域をその下流側の隣接する領域から一時的に分離させる取り除き可能なバリアを更に含み、
前記生成物を生成させるステップに続いて行われる、前記バリアを取り除くステップを更に含むことを特徴とする方法。
