Description
複雑な外来病原体によってコードされる何千もの可能なペプチドのうち、MHCクラスI抗原に結合可能な、したがってT細胞により認識可能なペプチド型の割合はほんのわずかである。この現象は、マルチエピトープワクチンの開発に明らかに潜在的影響を及ぼすもので、免疫優性(非特許文献8)として知られる。免疫優性にはいくつかの主要な変数が寄与している。本明細書において、本発明者らは、細胞内プロセシングの結果として適切なペプチドの生成に定性的にも定量的にも影響する変数について述べる。
接合エピトープは、別の2つのエピトープが並置したために生成するエピトープと定義される。新しいエピトープは、第1のエピトープに由来するC末端セクションと第2のエピトープに由来するN末端セクションからなる。接合エピトープの生成は、以下の理由により、クラスIおよびクラスII限定エピトープ用マルチエピトープミニ遺伝子ワクチンの設計における潜在的な問題である。第1に、HLAトランスジェニック実験動物において免疫原性が一般に試験されるヒトエピトープからなるミニ遺伝子、またはそれを含むミニ遺伝子を開発するときに、ネズミエピトープの生成は、有害な免疫優性効果を生じる恐れがある。第2に、ヒトHLAクラスIまたはクラスII分子に対する新しい意図しないエピトープの生成は、ワクチンのレシピエントに、標的によって誘発されるT細胞応答であり感染細胞または腫瘍によっては発現されない新しいT細胞特異性を誘発する恐れがある。これらの応答は、明らかに不適切で効果がなく、有害な免疫優性効果を生じて逆効果でさえあり得る。
クラスI限定エピトープは、複雑なプロセスによって生成する(非特許文献8)。エンドプロテアーゼと、エキソプロテアーゼによる潜在的なトリミングを含むタンパク質限定加水分解には、抗原プロセシング関連トランスポーター(Transfers associated with antigen processing)(TAP)分子により小胞体(ER)膜を通るトランスロケーションが続く。CTL前駆体のERトリミングも示されたが(非特許文献13)、抗原性ペプチドの生成に関わる主要な細胞質ゾルプロテアーゼ複合体およびそれらの前駆体はプロテオソームである(非特許文献12)。エピトープのC末端およびN末端に隣接する残基が、エピトープの生成効率に影響を及ぼすか否かは長年議論されてきた。
本明細書で開示する発明の好ましい実施形態のいくつかを当業者が理解できるようにするために以下の定義を与える。しかし、これらの定義は例示的なものにすぎず、特許請求の範囲に定める本発明の範囲を限定するために使用するのでは決してないことを理解されたい。当業者は、以下の定義をわずかに改変し、そのように改変された定義を利用して本明細書で開示する本発明を理解し実行することができる。当業者に明白なそのような改変は、以下に定める特許請求の範囲に適用することができ、本発明の範囲内にあると考えられる。

(i)マルチエピトープ構築物に組み込むべきエピトープを分類して、形成される接合エピトープの数を最少化する順番を提供する。この分類操作を図11および12を参照して以下により詳細に考察する。エピトープを順位付けるのに2番目に検討すべき事項として、エピトープのN末端にあるCTL免疫原性を高める残基を別のCTLエピトープのC末端に並置するようにエピトープを配置することが好ましい。
(ii)免疫原性を高めるフランキング残基を、エピトープのフランキング位置(flanking position)に挿入することができる。特定の実施形態では、CTLエピトープのC+1位にフランキング残基を挿入する。
(iii)エピトープ間にスペーサー配列を挿入して接合エピトープが発生するのを防止することができる。特定の実施形態において、スペーサー配列は免疫原生を高める残基を、この残基がCTLエピトープのC末端の側面に位置するように、リンカーのN末端に含むこともできる。
マルチエピトープ構築物の設計における検討事項の1つは、エピトープを互いに隣接して配置したときに接合エピトープが意図せずに生成することである。マルチエピトープ構築物におけるこのようなエピトープの存在は、性能に有意な影響を及ぼし得る。この望ましくない効果に対する防御戦略を、マルチエピトープワクチンの開発に適用するために本明細書に開示する。接合エピトープは、エピトープを分類して接合エピトープ数が最少になる順番を特定することによってまず最少化できる。このような分類操作はコンピュータを用いて、または、必要であれば、マルチエピトープ構築物中に含まれるエピトープ数に応じて目視により実施することができる。
接合エピトープを除去する別の要素として、並置したときに接合エピトープを生成する2つのエピトープ間にスペーサー配列を挿入することができる。
マルチエピトープ構築物を設計する際に検討すべき別の因子は、CTLエピトープのC末端の側面の位置に免疫原性に有利な残基を挿入することである。
HIVマルチエピトープDNAワクチンHIV−FT(図3a)は、20個のHIV由来CTLエピトープをコードする。これら20個のエピトープのうち、8個はHLA−A*0201、9個はHLA−A*1101、3個はHLA−B*0702によって限定される。すべてのエピトープがそれらの該当する限定エレメント(restriction element)に高い親和性または中程度の親和性で結合した。HLA−A*0201限定エピトープのすべてが、精製HLA−A*0201分子に、ほぼ同じ親和性で結合し、IC50値は19〜192nMの範囲であった(図3a)。HIV−FTに含まれるように選択されたHLA−A*0201エピトープは、HIV−1感染個体に認識され、IFAと共に乳化し、HLA−A*0201/Kbトランスジェニックマウスを初回抗原刺激するのに利用したとき、CTL応答を復活させるための初回抗原刺激に極めて有効でもあった。この構築物は、複数のエピトープをそれらの間にスペーサー配列を介在させずに順次コードするように設計されており、コンセンサスIgκシグナル配列がこの構築物の5'端に融合して、コードされた抗原が小胞体中に輸送されるのを促進した(非特許文献7)。
次に、HIV−FT中のHLA−A*0201エピトープの免疫原性の差を評価した。特異なMHC結合親和性は、エピトープのすべてがHLA−A*0201に高い親和性で結合したので除外することができた(図3a)。また、IFA中で乳化し最適に前処理したペプチドでHLAトランスジェニックマウスを免疫した後、エピトープすべてが同等のCTL応答を示したことから、HLA−A*0201/KbトランスジェニックマウスにおけるTCR特異性の適切なレパートリーの欠如は除外することができた。T細胞認識に対して提示される各エピトープの相対量の変化は、エピトープの免疫原性の差に原因がある可能性がある。
本明細書に記載するように、個々のCTLエピトープの側面に位置する特定のアミノ酸は、タンパク質分解的な切断に対する抗原の感受性を変えることによって、エピトープがプロセシングを受ける効率に影響を及ぼすか、またはそれを促進する1つの因子となる。エピトープ免疫原性に対するフランキングアミノ酸の影響を調べるために、配列を中断することなく最小限のCTLエピトープをコードするいくつかの無関係な実験的マルチエピトープDNA構築物で免疫したHLA−A*0201、−A*1101および−B*0701トランスジェニックマウスから免疫原性データを得た。94個の異なるエピトープ/フランキング残基の組合せを表わすデータベースを編集して、エピトープ免疫原性に関し、すぐ側面に位置するアミノ酸の可能な影響を決定した。所与のエピトープとフランキングアミノ酸の組合せは、重複性により解析が人為的に歪められるのを防止するために1つしか含めなかった。HLAトランスジェニックにおけるエピトープ免疫原性は、測定した培養物の少なくとも30%で100SUまたは20LUを超えれば最適とみなした。CTL応答は、4つのカテゴリー、すなわち(+++)、優−200LUまたは1000SUを超える;(++)、良−20〜200LUまたは100〜1000SU;(+)、中間−2から20LUまたは10から100SU;および(+/−)、弱またはネガティブ−2LUまたは10SU未満の1つに通常採点される。最適と最適以下の各応答数は、フランキング位置のアミノ酸の化学的タイプに基づいて分類され、差の有意性はカイ二乗検定を用いて決定した。
C+1フランキング位置における好ましいタイプと有害なタイプの各アミノ酸の効果を直接評価するために、HBV.1およびHBV.2(図3b)と称する2つのマルチエピトープ構築物を評価した。HIV−FTと同様、これらのHBV構築物は、スペーサーを介在せずに順次エピトープをコードする。実際、HBV.1およびHBV.2は、すでに特性の決定された実験的マルチエピトープ構築物pMin1(非特許文献7)においてHIV−1エピトープを類似のHBV由来のエピトープで置換することによって生成された。
異なるC+1残基を伴うCore 18の免疫原性の変動が、タンパク質分解的な切断に対する特異な感受性の結果であるならば、エピトープ提示レベルの大きな差を、異なる構築物で検出できるはずである。これを試験するため、トランスジェニックマウスで発現されるのと同じHLA−A*0201/Kb遺伝子を発現するジャーカット細胞にHBV.1またはHBV.1Kのいずれかを発現するエピソームベクターをトランスフェクションした。C+1位にKがあるとき、同じ位置にFが存在するのに比べてCore 18エピトープが>105高いレベルで提示された(図7)。Pol 455の提示は10倍未満しか変動しないことから、このCore 18の提示の差を標的細胞系間の遺伝子発現の差に帰することはできそうにない。これらのデータは、マルチエピトープDNAワクチンにおいてC+1位のアミノ酸がエピトープ提示の効率に影響を及ぼすことができるという際立った効果を実証している。したがって、これらのデータは、エピトープ提示レベルに影響を及ぼすという設計検討事項を通して、DNAワクチンにおけるCTLエピトープの免疫原性を最適化できることを示している。この効果は抗原が転写され翻訳された後に発揮されるので、このタイプの最適化は、ウイルスベクター、当業者に既知の他の発現ベクターなど他の方式により送達されたエピトープ系のワクチンに適用可能である。
本発明を用いてマルチエピトープ構築物を開発するために、マルチエピトープ構築物中に含まれるエピトープを、本明細書で規定するパラメータを用いて分類し最適化する。分類および最適化はコンピュータを用いて、またはより少ない数のエピトープに対してはコンピュータを用いないで実施することができる。
マルチエピトープ構築物の開発は、MHCとのペプチド結合が種特異的なので、無類の難題である。異なる種からの異なるタイプのMHCは、異なるペプチドセットと結合する傾向がある(非特許文献65)。その結果、ヒトエピトープからなる構築物を通常の実験動物で試験することは不可能である。この制限を克服する代替方法が一般に利用可能である。それらには、1)非ヒトMHC限定エピトープを組み込んだ類似の構築物を試験すること、2)非ヒトMHC限定コントロールエピトープへの依存、3)ヒトと非ヒトMHCの交差反応性への依存、4)HLAトランスジェニック動物の使用、および5)ヒト細胞を用いたインビボの抗原性アッセイなどが含まれる。以下は、抗原性および免疫原性を解析するための技術開発の簡明な概説である。
エピトープの同定(非特許文献66〜70)およびワクチン開発(非特許文献71)のためにHLAトランスジェニックマウスを利用することが確立されている。発表された報告のほとんどは、HLA A2.1/Kbマウスの使用を検討したが、B*27およびB*3501マウスも利用可能であることに留意すべきである。さらに、HLA A*11/Kbマウス(非特許文献72)およびHLA B7/KbおよびHLA A1/Kbマウスも作製された。
いくつかの独立の系統の実験から、細胞表面のクラスI/ペプチド複合体の密度が、T細胞の初回抗原刺激レベルと相関し得ることが示されている。したがって、エピトープがAPC表面に生成し提示されるレベルを測定することで、ヒト細胞におけるマルチエピトープ核酸ワクチンのインビトロでの効力を間接的に評価する手段が提供される。HLAクラスIトランスジェニックマウスの使用を補完するものとして、この手法はヒト細胞におけるプロセシングを調べる利点を有する(非特許文献71)。
好ましい実施形態では、ワクチン構築物が最適化されてクラスII限定免疫応答を誘発する。クラスIIエピトープを含めてマルチエピトープ構築物を評価する一方法は、HLA−DRトランスジェニックマウスを使用することである。いくつかのグループがHLA−DRトランスジェニックマウスを作製しその特徴を明らかにした(非特許文献83)。
ワクチン性能を評価する重大な要素は、インビボでの免疫応答誘発能力を評価することである。免疫原ならびに一般的な復活抗原に対するCTLおよびHTL応答の解析は、一般的に使用され当分野では既知である。使用されるアッセイには、クロミウム放出、リンホカイン分泌、リンパ球増殖アッセイが含まれる。
ヒト細胞におけるエピトープのプロセシングおよび提示を評価するために抗原性アッセイを実施した。ヒト標的細胞にマルチエピトープ核酸ワクチンを効率的にトランスフェクションするエピソームベクターを用いて、このような解析を実施した。
マルチエピトープ構築物中に含まれるエピトープは、PCT出願(特許文献4または5)に記載されているように、典型的には、HLAクラスIまたはクラスII結合モチーフを有する。マルチエピトープ構築物は、たとえば、非特許文献7に記載される方法に従って調製することができる。
本発明のマルチエピトープ構築物は、マルチエピトープポリペプチドをコードする核酸を含む発現ベクターとして一般に提供される。このような発現ベクターの構築は、たとえば、特許文献9に記載されている。発現ベクターは、抗原が発現され適切なHLA分子を標的とするように、生物体の適切な細胞中で核酸をコードする転写単位を発現することが可能な少なくとも1つのプロモーターエレメントを含む。たとえば、ヒトに投与する場合、ヒト細胞中で機能するプロモーターエレメントが発現ベクター中に組み込まれている。
本発明の発現ベクターは、MHC標的配列に作動可能に連結された1つまたは複数のMHCエピトープをコードすることができ、本明細書では「標的核酸」または「標的配列」と称する。MHC標的配列を使用すると、MHC分子アセンブリー部位にペプチドエピトープを誘導し細胞表面に輸送することによって、抗原に対する免疫応答が抗原単体を送達するよりも亢進され、それによってT細胞に結合しそれを活性化するのに役立つMHC分子−ペプチドエピトープ複合体の数が増加する。
本発明は、本発明の発現ベクターを個体に投与することによって、免疫応答を刺激するための方法も提供する。本発明の発現ベクターはMHCエピトープをMHC分子に導き、したがって発現ベクターがコードする抗原によって活性化されるCTLおよびHTLの数が増加するので、本発明の発現ベクターを投与して免疫応答を刺激することは有利である。
抗原性アッセイ
DNA、ペプチド/IFAまたはリポペプチドで初回抗原刺激したトランスジェニックマウスのひ細胞から高親和性ペプチド特異的CTL系を生成させることができる。簡潔に述べると、トランスジェニックマウスから採取したひ細胞をペプチド0.1μg/mlおよびLPS芽細胞で刺激する。初回刺激から10日後およびその後毎週、ペプチド0.1μg/mlと共にLPS芽細胞を1時間間欠投与して細胞を再刺激する。再刺激から5日後に上述したin situ IFNγ ELISAでCTL系を評価する。あるいは、たとえば、標的病原体に感染した患者または標的疾患、たとえば、癌に罹った患者から誘導されるCTL系を使用することができる。トランスジェニックマウスまたは患者のいずれからも入手不可能な特異的CTL系を当技術分野の専門技術を利用して正常なドナーのPBMCから生成させる。
マウス、免疫化および細胞培養物
この試験に使用するHLA−A2.1/Kb(非特許文献60)およびA11/Kb(非特許文献72)トランスジェニックマウスの誘導についてはすでに記述されている。HLA B7 Kbトランスジェニックマウスは、社内で入手可能である(Epimmune Inc.、San Diego、CA)。HLA DR2、DR3およびDR4トランスジェニックマウスは、C.David(Mayo Clinic)から得られる。非トランスジェニックH−2bマウスをCharles River Laboratoriesまたは他の業者から購入する。免疫化を、たとえば、非特許文献7に記載のとおりに実施した。細胞すべてを、10%FBS、L−グルタミン4mM、2−ME50μM、ピルビン酸ナトリウム0.5mM、ストレプトマイシン100μg/mlおよびペニシリン100U/mlを補充したHEPES含有RPMI 1640培地(Gibco Life Technologies)からなる培地で増殖させた。
増殖アッセイ
HTLエピトープの免疫応答誘導能力を評価するために、増殖アッセイのようなアッセイを実施することが多い。たとえば、マウスCD4 Tリンパ球は、ひ臓の単細胞懸濁物からDynaBeads Mouse CD4(L3T4)(Dynal)を用いて免疫磁気により単離される。手短に述べると、2×107個のひ細胞を5.6×107個の磁気ビーズと共に4℃で40分間インキュベートし、次いで3回洗浄する。磁気ビーズをDetachaBead Mouse CD4(Dynal)を用いて引き離す。単離したCD4 Tリンパ球(2×105細胞/ウェル)を5×105個の照射された(3500rad)同系ひ細胞と共に平底96ウェルマイクロタイタープレート中で3組培養する。精製したペプチドを、最終濃度20、1、0.05および0μg/mlでウェルに添加し、細胞を合計4日間培養する。回収する約14時間前に、3H−チミジン(ICN)1μCiを各ウェルに添加する。ウェルを、Filtermate Harvester(Packard)を用いてUnifilter GF/Bプレート(Packard)上に回収する。TopCount(商標)マイクロプレートシンチレーションカウンター(Packard)を用いて液体シンチレーションカウンティングにより3H−チミジン取りこみ量を測定する。
51クロミウム放出アッセイ
CTL活性を測定するこのアッセイは当分野で周知である。このアッセイは、51Cr標識標的集団から放出される51Crの百分率を測定することによってT細胞集団の溶解活性を定量する(非特許文献121)。クロミウム放出アッセイから導出されるデータは、通常、CTL頻度/106細胞(限界希釈法、LDA(非特許文献122))として、またはこれより煩雑でないバルクCTL活性の定量評価(細胞溶解単位、LUアッセイ)によって表される。LUアッセイでは、51Cr放出アッセイで作成される標準E:T比対細胞障害性百分率のデータ曲線を、106エフェクター細胞当たりの細胞溶解単位(LU)に変換する。1LUは、標的細胞の30%を溶解するのに必要な溶解活性として定義される(非特許文献123)。LUを計算することによって、応答を定量化することが可能になり、したがって異なる実験値を容易に比較することができる。
in situ IFNγELISA
in situ IFNγELISAアッセイが開発され、新たに単離したひ細胞とペプチドで再刺激したひ細胞(非特許文献124)IFNの両方に対して最適化された。このアッセイは、ELISPOTアッセイを基にするが、可溶色素原(chromagen)を利用して、ハイスループット分析に容易に適用できるようになっている。1次および再刺激アッセイの両方で、この技術は、これら他のアッセイでは検出不可能な応答がin situ ELISAで観測されるという点で、従来の上清のELISAまたは51Cr放出アッセイのいずれよりも感度が高い。1個の細胞当たりを基準にして、in situ ELISAの感度は約1IFNγ分泌細胞/104培養細胞である。
ELISPOTアッセイ
ELISPOTアッセイでは、誘導されて特異的リンホカイン、通常はIFNγを産生し放出する個々の細胞の能力を測定することによって、所与のペプチドに特異的なT細胞の頻度を定量する。ELISPOTアッセイの感度が高くなったことで、感染ヒトまたは実験動物から新たに単離した細胞からの応答を研究者が検出できるようになった(非特許文献88、90)。IFNγELISAにおいて上述したように、ELISPOTアッセイを最終ステップまで実施する。ただし、HRP−ストレプアビジンの代わりにExtrAvidin−AP(Sigma、1:500希釈)を用いる。基質5−BCIP(BioRad)を用いて製造者の指示に従って発色させる。位相差顕微鏡を用いてスポットを数える。あるいは、Zeiss KS ELISPOTリーダーを利用してスポットを数える。この場合、BCIP/NBT基質を使用する。
四量体染色
四量体染色は、ペプチドエピトープと、クラスI抗原と、エピトープ特異的T細胞受容体との相互作用に基づいてエピトープ特異的ヒトCD8+Tリンパ球を検出するフローサイトメトリー技術である。このアッセイは、新たに単離した血液試料中のエピトープ特異的ヒトCD8+Tリンパ球を迅速に定量することができる。HIVペプチド/HLAの様々な組合せに対するMHC四量体は、たとえば、NIHリポジトリ(非特許文献125)から得られる。エピトープ特異的細胞を標識するために、体積100μl中1×106個のPBMCを、適切な四量体5μg/mlならびにヒトCD3およびCD8を認識するモノクローナル抗体(PharMingen、San Diego、CAまたはBioSource、Camarillo、CAを含めた商用供給源から様々な蛍光色素複合体が入手可能である)と共に暗所で40分間インキュベートする。細胞を洗浄し、パラホルムアルデヒドで固定した後、FACsanまたはFACSCaliburフローサイトメーター(Becton Dickinson Immunocytometry Systems、San Jose、CA)を用いて分析する。試料データをCellQuestソフトウエアを用いて解析する。
臨床試料のアッセイ
患者またはボランティアからの凍結PBMC試料中の特異的CD8+CTL活性を評価するために様々なアッセイを使用することができる。ELISPOT、クロミウム放出、in situ IFNγ放出、増殖アッセイおよび四量体アッセイすべてが、様々な実験モデル、たとえば、ネズミおよび/または霊長類起源の実験モデルからの応答を評価するのに有用である。
トランスジェニック動物
体積10〜100μl中最高100μgの用量のDNAまたはペプチドを前けい骨筋内に筋肉内投与して、または尾部つけ根に皮下投与して、トランスジェニックマウス(HLA−A2.1/Kb H2b、HLA−A11/Kb、HLA−B7/Kb)を免疫する。DNAは生理食塩水に、ペプチドはIFAに希釈して処方する。11〜21日後、動物をCO2窒息を用いて屠殺し、そのひ臓を摘出し、CTL機能をインビトロで決定するための細胞源として使用する。一般に、1実験グループ当たり3〜6匹のマウスを使用する。また、CTL培養物の再刺激のためのAPC源として非免疫マウスのひ臓を使用する。8〜12週齢のオスとメスを使用する。
複数のCTLおよびHTLエピトープに対する応答の同時誘導の実証
CTLエピトープ鎖(string)の構築および試験
この実施例では、複数のCTLおよびHTLエピトープを試験する例を提供する。たとえば、単一のプロモーターの制御下にある10〜12個の異なるCTLエピトープを含むエピトープ鎖を合成し、標準プラスミドpcDNA 3.1(Invitrogen、San Diego)に組み込む。これらの構築物は、標準シグナル配列および汎用HTLエピトープPADRE(登録商標)を含んでいる。各エピトープセットを、集団をバランス良く網羅するように選択する。試験および最適化を容易にするために、トランスジェニックマウスにおいて免疫原性であり、および/またはヒトにおいて抗原性であることが示されたエピトープのバランスの取れた集合体(representation)を含めてある。
単一のプロモーターの制御下にある3〜20個の異なるHTLエピトープを含むエピトープ鎖を合成し、標準プラスミドpcDNA 3.1(Invitrogen、San Diego)に組み込む。試験および最適化を容易にするため、IAbマウスにおいて免疫原性であることがすでに知られているエピトープのバランスの取れた提示が提供されるように、所与の構築物に対する各エピトープセットを選択する。また、接合部に対応するペプチドすべてを合成し、IAbへの結合について、さらに最も重要なことには、世界で最も一般的なDR対立遺伝子(非特許文献130)を代表する14個の異なるDR分子のパネルへの結合について試験する。したがって、対象とする抗原向けではないHTLエピトープは、これらのプラスミド中では生成しない。しかし、良好なDR結合潜在能力(したがって、インビボでの潜在的DR限定免疫原性)を有する接合領域が検出された場合には、GPGPGなどのスペーサーを導入してこれらを除去する。構築物すべてにおいて、本明細書に記載するように、クラスI接合モチーフの数も最少化される。
上述の作業により、解析可能なエピトープすべてに対する同時応答/抗原性を実証するために利用されるエピトープの小さな機能性ブロックが生じる。これらのブロックは、次の実施例に記載するさらなる最適化の対象となる。これらの同じ構築物を用いて、免疫優性を評価する。具体的には、CTLエピトープ構築物すべてを一緒に混合し、あるいはHTLエピトープ構築物すべてを一緒に混合する。次いで、構築物のプールから得られる結果を、同じ構築物を別々に注射して得られる結果と比較する。
CTLおよびHTLマルチエピトープ構築物の最適化
この実施例では、実施例10に記載したCTLおよびHTL各構築物の最適化について述べる。抗原性および免疫原性に対するフランキング残基の潜在的影響も、最適化ミニ遺伝子構築物において評価する。これらの試験には、効果的なプロセシングを促進するように設計したフランキング残基(これに対する類義語は「スペーサー」である)を含有することが含まれる。
選択したエピトープを効果的に送達可能な最も簡単なプラスミドの構成の決定
実施例11および12に記載した実験を、マルチエピトープ核酸ワクチン設計に関する変数を扱うように設計する。理想的には、ヒトに使用できるベクターをプログラム全体を通して使用するが、ワクチンエピトープの最適化試験用に1つのDNAワクチンプラスミドを使用し、次いで、ヒトの使用に適切なベクターに切り換えることができる。実際のベクター選定は、いくつかの変数に依存する。たとえば、National Gene Vector Laboratory(University of Michigan)のような信頼のおける供給源を通した、ヒトの使用に適切なベクターの利用可能性が1つの因子である。
マルチエピトープワクチンによる免疫化後の、CD8+リンパ球応答の誘導の評価およびキャラクタリゼーション
主にELISPOT法によりCD8+リンパ球応答を測定した。ELISPOTアッセイは、当分野で既知であり、本発明者らの研究室で普通に使用されている。本明細書に記載する自動化されたZeiss ELISPOTリーダーも使用する。CD8+応答を測定するのに利用するアッセイは、主に、新たに単離した細胞について、およびペプチドを用いてインビトロで再刺激した細胞についてのIFNγELISPOTアッセイである。また、特定の例では、クロミウム放出アッセイを利用する。その結果は、ELISPOTアッセイで観測された結果と相関があった。選択したペプチド/MHCの組合せに対する四量体染色も実施した。
最適化されたマルチエピトープDNAベースワクチン構築物の設計
接合エピトープ形成の最少化とC+1プロセシング効率の最適化を同時に行う上述のコンピュータ支援方法を利用して最適化構築物を設計した。以下のモチーフ、すなわち、ネズミKb(XXXX(FY)X2-3(LIMV))、Db(XXXXNX2-3LIMV))、ヒトA2(X(LM)X6-7V)、ヒトA3/A11(X(LIMV)X6-7(KRY))およびヒトB7(XPX6-7(LIMVF))を接合の最少化に利用した。C+1傾向値(propensity values)を図6に示すデータから計算し、以下のようになった。K=2.2;N=2;G=1.8;T=1.5;A、F、S=1.33;W、Q=1.2;R=1.7;M、Y=1;I=0.86;L=0.76;V、D、H、E、P=0。4個までのアミノ酸の挿入が許容された。この手順および本明細書に記載する他の手順によって設計される構築物の例を図18に示す。これらの構築物のいくつかの特性をインビトロおよびインビボでの免疫原性試験により明らかにし、以下に説明する。図19に、マルチエピトープ構築物中の特定の核酸配列によってコードされるアミノ酸エピトープ配列を列挙する。
マルチエピトープCTL構築物の免疫原性試験およびフランキングアミノ酸の影響
様々なマルチエピトープ構築物の免疫原性試験にHLAトランスジェニックマウスを使用した。1グループのマウスを、前けい骨筋の両側に10μM心臓毒50μlを注射して前処置し、4または5日後、PBSに希釈したDNA構築物100μgを同じ筋肉に投与した。別のグループでは、CFA中に乳化したペプチド(ペプチドは、DNA注射グループのマウスに投与したDNA構築物内のエピトープに対応する)を各マウスに注射した。免疫化後11から14日目に、DNAを接種した動物およびペプチドを接種した動物からひ細胞を回収し、CTL活性を、標準51Cr放出アッセイ、ペプチドエピトープ特異的再刺激を用いない精製CD8+Tリンパ球によるγ−IFN産生を測定するELISPOTアッセイおよびペプチドエピトープによるインビトロでのエピトープ特異的再刺激ステップを含むin situ ELISAを含めたいくつかのアッセイの1つで測定した。ペプチドエピトープでの刺激に関して、EP−HIV−1090構築物によって誘導されるCTL活性の例を図14Aに、ペプチドエピトープでの刺激に関して、PfCTL.1、PfCTL.2およびPFCTL.3構築物によって誘導されるCTL活性を図14Bに示す。
マルチエピトープHTL構築物の免疫原性試験およびスペーサー配列の影響
HTLエピトープを分離し、それによって接合エピトープを破壊するGPGPGからなる汎用スペーサーを開発した。このスペーサー設計の背後にある論理は、GもPも、HTLペプチドエピトープのコア領域中の1および6位の第1のアンカーとして、任意の既知のネズミまたはヒトMHCクラスIMHCクラスII分子によって使用されないことである。このスペーサーにより導入される5アミノ酸のギャップによって、隣接するエピトープが分離され、2つのエピトープのアミノ酸は、1および6位におけるアンカーとして物理的に働くことができない。1つは3個のスペーサーを有し、それ以外はスペーサーがなく、マウスIAbに結合することが知られている4個のHIV−1エピトープからなる合成ペプチドを用いて、GPGPGスペーサーの有用性を試験した。HIV75量体は3個のGPGPGスペーサーを有する構築物であり、HIV60量体は3個のスペーサーを欠く構築物であった。CFAで希釈したペプチドによるCB6F1マウスの免疫化によって、スペーサーの非存在下で4個のうち3個のエピトープに対するHTL応答が誘導されたが、スペーサーが存在するときにはエピトープすべてが免疫原性であった(図15)。この事実は、スペーサーがマルチエピトープ構築物の性能を改善できることを示している。
Claims (2)
- 2つ以上のCTLエピトープ核酸を含む最適化されたマルチエピトープ構築物をコードするポリヌクレオチドを設計するための方法であって、該方法が、
(i)ヒト白血球抗原(HLA)対立遺伝子特異的モチーフ又はスーパーモチーフを含むエピトープをコードする5つ以上の核酸配列を選択することであって、前記エピトープは、HLAクラスI細胞障害性Tリンパ球(CTL)エピトープであること、
(ii)マルチエピトープ構築物におけるエピトープの順序を決定または設計することにより、CTLエピトープ核酸を分類して接合エピトープ数を最少化すること、
(iii)前記CTLエピトープ核酸を前記マルチエピトープ構築物に組み込むことであって、K、AK、KA、KK又はAから選択される少なくとも一つのフランキング又はスペーサーアミノ酸残基が、少なくとも一つの前記CTLエピトープ核酸のC+1位に導入され、前記フランキング又はスペーサーアミノ酸残基は、CTL接合エピトープの発生を防止すること、
の各ステップを含み、
前記最適化されたマルチエピトープ構築物を含むワクチンの免疫原性は最適化されていることを特徴とする方法。 - 2つ以上のHTLエピトープ核酸を含む最適化されたマルチエピトープ構築物をコードするポリヌクレオチドを設計するための方法であって、該方法が、
(i)ヒト白血球抗原(HLA)対立遺伝子特異的モチーフ又はスーパーモチーフを含むエピトープをコードする5つ以上の核酸配列を選択することであって、前記エピトープは、HLAクラスIIヘルパーTリンパ球(HTL)エピトープであること、
(ii)マルチエピトープ構築物におけるエピトープの順序を決定または設計することにより、前記HTLエピトープ核酸を分類して接合エピトープ数を最少化すること、
(iii)前記HTLエピトープ核酸を前記マルチエピトープ構築物に組み込むことであって、GPGPGである少なくとも一つのフランキング又はスペーサーアミノ酸残基が、前記核酸エピトープの間に配置され、前記フランキング又はスペーサーアミノ酸残基は、HTL接合エピトープの発生を防止すること
の各ステップを含み、
前記最適化されたマルチエピトープ構築物を含むワクチンの免疫原性は最適化されていることを特徴とする方法。
