Description

図1は、本発明者が、突然変異体の対によって機能の再構成を起こすことができ、その結果機能的オリゴマーを形成するということをどのように想定したかの説明を示す。図に示すように、機能的オリゴマーは、天然GPCR及び突然変異Gタンパク質を含む第一融合タンパク質(a)、突然変異GPCRと天然Gタンパク質を含む第二融合タンパク質(b)から構成される。これらの2つの融合タンパク質が組合わさるときに、GPCRシグナル伝達カスケードの機能的活性化が、第一融合タンパク質の天然GPCRに結合するアゴニストリガンドと第二融合タンパク質のGタンパク質を介する機能的な結合とシグナル伝達によって起こる。

また、図3は、例えば突然変異するのに適切な、及びGPCRを実質的に非機能的にするのに適切な、GPCRの第二細胞内ループの高保存残基を示す。これらの残基の突然変異は、GPCRを不活性にするがいまだリガンドと結合できるようにするのに特に有効であることが本発明者によって示された。更に、疎水性残基は、高く保存され、すべてのクラスAのGPCRを、この方法で突然変異して不活性にすることができると考えられる。典型的には、残基は疎水性残基であり、例えばこの分野で公知の部位特異的突然変異誘発技術によって酸性残基に変異してもよい。しかしながら、GPCRを機能的に不活性又は実質的に機能的に不活性にする別の突然変異を行ってもよく、それらの活性/活性の欠失を、ヘテロ二量体活性のアッセイに関する後述のアッセイを使用してテストしてもよい。つまり、突然変異誘発後に活性の程度がどのくらい残るかを確認するために、天然のGPCRに対する機能的アッセイを突然変異誘発されたGPCRで行ってもよい。突然変異誘発の第一ラウンドがGPCRを不活化するのに十分でなければ、突然変異誘発の更なるラウンドを行い、その後活性をテストしてもよい。
おそらく、今日の薬剤開発における非常にやりがいのある工程は、標的の検証に関連している。大きな公的な及び私的な、ここ数年で達成されたヒトゲノム配列決定の試みは、医薬品開発の前例のない数の遺伝子ターゲットを提供した。機能性の解析及び、最も重要なことに、遺伝子ターゲットの潜在的な治療の関連性は、次の世代の治療の開発の基本として高い優先度を有する。この課題は、多数の新規遺伝子が同定されているGPCR遺伝子ファミリーよりも重要性は大きくない。目的とする組織内で高く又は選択的な発現を示すGPCRを同定することにより、細胞レベルの分析を更に洗練することが可能である。これには、RNAプローブ及び抗体の両方を使用して、目的とするGPCRを発現する組織内の細胞集団をマッピングすることを含めることができる。
様々な細胞を、本発明の融合タンパク質をコードする核酸構築物を発現するのに使用することができる。改変されたGPCRを発現するために形質移入され得るすべての細胞は、本発明の範囲内に入ると考えられる。このような細胞の例として、新生児の細胞、内分泌細胞、腫瘍細胞、腺房細胞、島細胞、免疫細胞、神経内分泌細胞、ニューロン細胞及び下垂体細胞が挙げられる。また、本発明に従って改変されたGPCRを発現することができる他の細胞系統、例えばCHO(チャイニーズハムスター卵巣)細胞(CHO-K1)及びHEK(ヒト胎児由来腎臓)細胞を決定することは、当業者の能力の十分に範囲内である。
改変されたGPCRをコードする核酸構築物で細胞を同時形質移入した場合、少なくとも機能的(第二の)GPCRに特異的な、少なくとも1つのリガンドの結合を評価することができる。本発明によるGPCRオリゴマーの機能的活性は、たくさんの標準物質によって測定することができ、周知の技術である。GPCRは、アデニリルシクラーゼ活性に影響を与え、電位によるゲート開閉型カルシウムチャンネルの伝導性を調節し、カリウムチャンネルを調節し、MAPキナーゼ経路を活性化し、ホスホリパーゼC(PLC)-IP3経路を活性化し、ホスホチロシンホスファターゼを活性化し、有糸分裂誘発を刺激し、開口分泌を刺激し、細胞分裂停止を刺激し、走化作用を刺激し、アポトーシスを誘発することが知られている。これらの活性を、日常的な標準的方法を使用して当業者は測定することができる。また機能的活性を、GPCRのリガンドを決定するためのアッセイにおいてこれらの技術を使用して測定することができる(以下を参照のこと)。
GPCRオリゴマーからのシグナル伝達は、様々なレポーター系を使用して評価することもできる。細胞内cAMPレベルの変化を測定するためには、環状AMP応答エレメント(CRE)を含むプロモーターによって駆動されるレポーター構築物が好ましいであろう。プロテインキナーゼCの活性化に導くGq経路から放出された応答を測定するためには、PKC感受性レポーター系が好ましく、PKCプロモーター配列にあるAP1部位を含む。MAPキナーゼ活性化に感受性の他のレポーター及び血清応答エレメント(SRE)を含むレポーターもまたGPCRオリゴマーからの応答を測定するのに使用できる。レポーター分子自体は、動くことができ、これらの例には、ルシフェラーゼ、β-ガラクトシダーゼ、β-ラクタマーゼ、緑色蛍光タンパク質、黄色蛍光タンパク質及びその他が含まれる。
本発明は、オリゴマー形成の結果としてGPCRに結合する新しいリガンドを決定するのに特に適切である。当業者に周知の天然のリガンド及び合成リガンドをテストすることができる。適切な試験リガンドは、コンビナトリアルライブラリー、ペプチド及びペプチド模倣物、確定した化学物質、オリゴヌクレオチド及び活性についてスクリーニングできる天然物ライブラリーからのものである。1つの可能なアプローチにおいて、候補物質は、バッチで、例えば1反応につき10個の物質で、また個別に試験された効果を示すこれら物質のバッチで、最初のスクリーニングを行うのに使用することができる。
組合わせGαq/Gα11ダブルノックアウトマウス(Offermansら、1998)由来の線維芽細胞細胞系統(EF88)(Gohlaら、1999)が、Dr. M.I. Simon、 California Institute of Technology, Pasadena CAから恵与された。組織培養のためのすべての材料は、Life Technologies Inc. (Paisley, Strathclyde, UK)により供給された。[3H]プラゾシン(80Ci/mmol),[3H]メピラミン(30Ci/mmol)及び[35S]GTPγS(1250Ci/mmol)はNEN/Perkin Elmerから得た。オリゴヌクレオチドは、Cruachem (Glasgow, Strathclyde, UK)から購入した。時間分解蛍光共鳴エネルギー移動用の試薬はWallacから得た。受容体リガンドは、RBI(Gillingham, Kent,英国)から購入した。抗Gq/G11抗血清CQの製造と特徴づけは、Mitchellら, 1991; Mitchellら, 1993によって説明されたGq/G11の広範囲に及ぶ分布は、予想されたC末端デカペプチドに対する抗ペプチド抗血清によって免疫学的に検出された(FEBS Lett. 287, 171-174)。
野生型及び突然変異型α1b-アドレナリン作用性受容体-Gα11融合タンパク質の製造及びサブクローニングは、Carrilloら、2002に説明されているように行った。ヒトヒスタミンH1受容体-G11α融合タンパク質の製造及びサブクローニングを2つの分かれたステージで行った。第一の工程では、アミノ末端プライマー5'-GATACTGGGCTATCCAAGCTTATGAGCCTCCCCAATTCCTC-3'を使用し、HindIII制限酵素認識部位(下線)をヒトヒスタミンH1受容体のコード配列の上流にPCRで導入した。カルボキシル末端プライマー5'-AAGGAAAAAAGCGGCCGCTGGAGCGAATATGCAGAATTCTCT -3'を使用して、3つのアミノ酸スペーサー(Ser-Gly-Arg)とNotI制限酵素認識部位を停止コドンのすぐ上流に導入した。同様に、マウスG11α配列を、アミノ末端プライマー5'-AAGGAAAAAAGCGGCCGCATGACTCTGGAGTCCATGATGGC-3'及びカルボキシル末端プライマー5'-ATGAAACCGCTCGAGTCACACCAGGTTGTACTCCTTCAG-3'を使用してPCRによって増幅した。これにより、G11αコード配列にそれぞれ隣接したNotI及びXhoI制限酵素認識部位を導入した。第二の工程では、増幅された受容体フラグメントをHindIII/NotIで消化し、G11αフラグメントをNotI/XhoIで消化した。これらのフラグメントを精製して、フラグメントをあらかじめHindIII/XhoIで消化したpcDNA3ベクター(Invitrogen)に連結した。細胞内ループ2LeuからAspへの突然変異体への選択は、Greasleyら、2001の研究に基づいた。また、クラスAのGPCRの大部分において、同等の位置に疎水性のアミノ酸がある(図2を参照のこと)。このような突然変異を、部位特異的突然変異誘発の標準的な方法によってPCR突然変異誘発を用いて融合タンパク質構築物の中に導入した(Sambrookら及びCarrilloら, Manufacturers Kit等)。
HEK293細胞を、37℃、5%CO2の加湿した大気中で、0.292g/lのL-グルタミン及び10%(v/v)の仔ウシ血清を補ったDMEM中に維持した。細胞を一時的な形質移入の前に60mm皿に60-80%の集密度(confluency)に増殖した。形質移入を、製造者の指示に従ってリポフェクタミン(LipofectAMINE)試薬(Life Technologies, Inc.)を用いて行った。
[35S]GTPγS結合の実験を、融合構築物の規定された量を含む膜(詳細は結果を参照のこと)を指示された濃度の受容体リガンドを含むアッセイバッファー(20mM HEPES (pH 7.4), 3mM MgCl2, 100mM NaCl, 1 μMグアノシン5'-二リン酸, 0.2mM アスコルビン酸, 50nCi [35S]GTPγS)に添加することにより開始した。非特異的結合を、100μM GTPγSの存在下以外は同じ条件で測定した。反応物を15分30℃でインキュベートし、20 mM HEPES (pH7.4), 3mM MgCl2及び100mM NaClを含む0.5mlの氷冷バッファーを添加して停止した。サンプルを16,000gで15分、4℃で遠心分離し、得られたペレットを可溶化バッファー(100mM Tris, 200mM NaCl, 1mM EDTA, 1.25%Nonidet P-40)に0.2%のドデシル硫酸ナトリウムを加えたものに再懸濁した。サンプルをパンソルビン(Calbiochem)で澄明にし、続いてCQ抗血清で免疫沈降を行った(Mitchellら, 1993)。
構築物を含むα1b-アドレナリン作用性受容体の発現をモニターするため、[3H]プラゾシン結合の研究をCarilloら、2002に説明されるように行った。構築物を含むヒスタミンH1受容体の発現をモニターするため、[3H]メピラミン結合アッセイを、[3H]メピラミン(0.1-10nM)を含むアッセイバッファー(50mM Tris-HCl, 100mM NaCl, 3mM MgCl2, pH7.4)に3μgの細胞膜を添加して開始した。非特異的結合を100μMメピラミンの存在下で測定した。反応物を30分25℃でインキュベートし、GF/Bフィルターを通して真空ろ過により結合したリガンドを遊離のものから分離した。フィルターを2回アッセイバッファーで洗浄し、結合したリガンドを液体シンチレーション分光分析により評価した。
EF88細胞を、95%空気及び5%CO2の雰囲気中で37℃で、10%(v/v)の熱不活化ウシ胎児血清とL-グルタミン(1mM)を補ったDMEMで増殖した。トリプシン処理中に回収した細胞の一部をカバーガラスに載せて、24時間の増殖期間後、細胞をリポフェクタミンを使って(Life Technologies Inc.)製造業者の指示に従って形質移入した。3時間後、細胞を2回OPTIMEM1で洗浄して、更に24時間DMEM増殖培地で培養した。関連のcDNA種を含む合計3μgのpcDNA3を各カバーガラスで形質移入するのに使用した。色素の膜浸透性アセトキシメチルエステル(1.5μM)を含むDMEM増殖培地で、減少した光の下でインキュベーション(15-20分、37℃)することにより、形質移入されたEF88細胞をCa2+感受性色素Fura-2でロードした。画像化研究の詳細及びその分析については、Liuら、2002に説明されている。
α1b-アドレナリン作用性受容体及びヒスタミンH1受容体構築物のFLAG及びc-myc標識形態を使用した同時免疫沈降の研究を、McVeyら、2001で述べられているように行った。ヒスタミンH1受容体での研究において、30U/mlのエンドグリコシダーゼFを添加した。
時間分解(time resolved)蛍光共鳴エネルギー移動を、McVeyら、2001で説明されているように、Eu3+-標識抗c-myc抗体とアロフィコシアニン標識抗FLAG抗体を使用して、無傷のHEK293細胞で行った。
本発明者は、α1b-アドレナリン作用性受容体と、[3H]プラゾシンを含むアゴニスト及びアンタゴニストリガンドの両方に結合するG11のαサブユニットとの間に融合タンパク質をあらかじめ生成した。この構築物発現するように形質移入したHEK293細胞の膜にアゴニストフェニレフリンを添加することにより、G11αのC末端デカペプチドに対する抗血清を使用した免疫沈降のアッセイの終了につづいてモニターされた[35S]GTPγSの結合の大きな刺激が生じた(図4Aを参照のこと)。
それぞれのクローン系統由来の標的受容体(実験用)及びpCMV(陰性コントロール)で安定に形質移入した細胞を、次の日のアッセイのために、ポリ-D-リジン前処理96ウエルプレート(Becton-Dickinson,#356640)に、完全培地(10%FBS、2mM L-グルタミン、1mMピルビン酸ナトリウムの入ったDMEM)に5.5×104細胞/ウエルで蒔く。Fluo4-AM(Molecular Prove、#F14202)インキュベーションバッファーストックを調製するために、1mgのFluo4-AMをDMSO467μlとプルロン酸(Pluoronic acid)467μl(Molecular Probe, #P3000)に溶解し、1ヶ月間-20℃で保存できるように1mMストック溶液を得る。Fluo4-AMは、蛍光カルシウム指示色素である。
メラニン保有細胞は下等の脊椎動物に見られる皮膚細胞である。該細胞は、メラノソームと呼ばれる色素性細胞小器官を含む。メラニン保有細胞は、Gタンパク質結合受容体(GPCR)活性化で微小管網状構造に沿ってこれらのメラノソームを再分配することができる。この色素移動の結果、細胞を見かけ上明るくしたり、暗くしたりする。メラニン保有細胞では、Gi結合受容体の活性化の結果、細胞内cAMPのレベルが減少することにより、メラノソームが細胞の中心に移動し、劇的に色が明るくなる。次に、cAMPレベルが上がると、Gs結合受容体の活性化に続いて、メラノソームは再分散されて、細胞が再び暗くなったように見える。Gq結合受容体の活性化の結果、レベルが上昇したジアシルグリセロールは、この再分散を誘発することもできる。更に、この技術はまた、特定の受容体チロシンキナーゼの研究に適切である。メラニン保有細胞の反応は、数分内で受容体活性化を起こし、単純な、しっかりとした色変化を起こす。この反応は、従来の吸光度マイクロプレートリーダー又は中程度のビデオ画像システムを用いて簡単に検出できる。他の皮膚細胞とは違って、メラニン保有細胞は神経堤に由来し、シグナル伝達タンパク質の全部を発現すると思われる。特に細胞は非常に広い範囲のGタンパク質を発現し、ほとんどすべてのGPCRを機能的に発現することができる。
α1b-アドレナリン作用性受容体及びヒスタミンH1受容体の両方とGタンパク質G11αとの間の融合タンパク質を使用することによって、本発明らは、いまや、これらのGPCRが二量体化すること、及びこれはGPCRのC末端尾部にGタンパク質を付加することによって危うくならないことを示す。同等に、無傷の細胞でGPCR二量体/オリゴマーを検出するtrFRETを使用することにより、無傷の細胞は、細胞表面にGPCR二量体/オリゴマーの複合体の存在を示すことができた。これはまた、GPCRにGタンパク質配列を付加することによって危うくならなかった。更に、GPCR又はGタンパク質のどちらかにGタンパク質のアゴニスト活性化を防ぐ突然変異を導入することによって、本発明者らは、同時発現した場合にアゴニスト介在機能を回復させることができる非機能的融合タンパク質の対を製造した。機能的再構成を2つの方法でモニターした。第一は、アゴニストは、単離状態でそれぞれ非機能的であった2つの突然変異体の同時発現後にのみ、EF88細胞で細胞内[Ca2+]の上昇を起こすことができた。EF88細胞は、ホスホリパーゼC結合Gタンパク質の発現を欠き、従って、Ga2+シグナルを発生させるにはEF88細胞に適切なGPCRとGタンパク質の両方を導入する必要がある。このアッセイは、本発明者にCa2+画像化が単一細胞での機能的二量体化をモニターすることを可能にしたという明らかな利益を有する。シグナル伝達カスケードで測定することができる最初の工程のひとつは、アゴニストで誘発されたGタンパク質でのグアニンヌクレオチド交換である。これは、[35S]GTPγSの結合によって便利にモニターできる。[35S]GTPγS結合アッセイのすべてにおいて、本発明者らは、最初に、飽和[3H]アンタゴニスト結合の研究によってGPCR Gタンパク質融合体のそれぞれの発現レベルを測定した。これにより、構築物の決められた量を含む膜量をアッセイに添加できるようになった。本発明者らは、増加した量のGPCR-G11α融合タンパク質の添加で、アゴニストで刺激された[35S]GTPγS結合の直線的増加があることを先に示した(Stevensら、2001)。ヒスタミンH1受容体-Gly208AlaG11α及びLeu133AspヒスタミンH1受容体-G11α融合タンパク質を同時発現したとき、野生型ヒスタミンH1受容体-G11α融合タンパク質のみを発現したときとほぼ同量の、アゴニストで刺激された[35S]GTPγS結合を生成するためには、[3H]アンタゴニスト結合部位の数の2倍の存在が必要であった。これは、機能的要素が二量体又はより高次のオリゴマーであるというよい証拠を提供する。機能的ヒスタミンH1受容体が二量体である場合、確率論的には、2つの非機能的突然変異融合体を同時発現するとき、生成された二量体の半分は非機能的であるだろう。なぜならば、この二量体は、ヒスタミンH1受容体-Gly208Ala G11α又はLeu133AspヒスタミンH1受容体-G11αのどちらかのホモ二量体であろうからである。二量体の50%だけが、1つのヒスタミンH1受容体-Gly208Ala G11αの複製物、及び1つのLeu133AspヒスタミンH1受容体-G11αを含む機能的なヘテロ二量体であると予想される(図10)。またこれらの研究は、クラスCのGPCRに関して、トランス活性化メカニズムを介したアミン作用性のクラスAのGPCRの機能を有するという考えを支持する。GPCRの機能的な形態は非機能的Gタンパク質に結合しているのに対して、活性化され得る二量体のGタンパク質の複製物は、GPCRの非機能的な形態に結合する。
ているため、非特異的「傍観者(bystander)」(Mercierら, 2002 J. Biol. Chem 277, 44925-44931)作用が報告された範囲内の非常に高いレベルで発現した。EF88細胞の使用により、内生的Gタンパク質との相互作用の可能性が排除され、EF88細胞がGqα又はG11αを発現しないと、効果は融合されたGタンパク質の活性化を反映せざるを得ない。更に、Gly208Ala突然変異を融合体のGタンパク質要素に導入した後で、形質移入されたHEK293の膜の[35S]GTPγS結合のアゴニスト刺激はほとんど廃止された。これは、達成された発現のレベルにおいて、HEK293細胞の内生的Gqα又はG11αの活性化が、両方が発現されたにもかかわらず、ほとんどなかったことを示す。HEK293細胞のヘテロ二量体化実験において、過剰なGタンパク質は、融合体により規定された1:1の化学量論のGPCR及びGタンパク質のために、第二GPCRと1:1モル比で導入された。この結果は、単に余分なGタンパク質の存在に起因しうるかどうかを評価するために、本発明者は代わりのストラテジーを通して余分なGタンパク質を提供した。そのようにすることによって、本発明者は、α1b-アドレナリン作用性受容体のN末端及び第一膜貫通領域に結合したG11αの形成物を作製した。他のGタンパク質に関する同等の構築物は、既に使用されており(Leeら, 1999 Biochemistry 38, 13801-13809, Guzziら, 2001 Biochem J. 355, 323-331, Molinariら, 2003 J. Biol. Chem 278, 15778-15788)、膜貫通αへリックスへの連結により、活性化に対して特に効果的な方向性でGタンパク質が提供されることを示唆していた(Molinariら, 2003 前述)。この構築物は、α1b-アドレナリン作用性受容体-G11α融合体よりも著しく高いレベルに発現できるにもかかわらず、単独で発現しても、また、単離されたα1b-アドレナリン作用性受容体又はα1b-アドレナリン作用性受容体-G11α融合体のどちらかとの組合わせて発現しても、Gタンパク質はフェニレフリンによって活性化されなかった。これらの研究により、GPCR-Gタンパク質融合体の非機能的な対の同時発現によりシグナルを再構成することは、再構成された二量体内での内部トランス活性化(internal transactivation)を反映するはずであることが確認された。

を有するヘテロ二量体からなる複合体を含む生物学的試薬であって、

(a)第一GPCR及び第一Gタンパク質の融合タンパク質をコードする第一核酸構築物を製造する工程であって、該第一GPCRは野生型と比較して突然変異されており、それにより該第一GPCRはリガンド結合後に該融合Gタンパク質を活性化することができない、上記工程、
(b)第二GPCR及び第二Gタンパク質の融合タンパク質をコードする第二核酸構築物を製造する工程であって、該第二Gタンパク質は野生型と比較して突然変異されており、それにより該第二Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記工程、及び
(c)細胞内で第一及び第二核酸構築物を同時発現して第一及び第二GPCRを含むヘテロ二量体からなる複合体を生成する工程、
を含み、第一GPCRの野生型が第二GPCRと異なり、該ヘテロ二量体が、それと結合するリガンドに反応してシグナル伝達カスケードを開始することができる、請求項1〜12のいずれか1項に記載の生物学的試薬の製造方法。 - (d)前記複合体を含む細胞膜の一部を単離する工程、
を更に含む、請求項13又は14に記載の方法。 - 第一及び第二GPCRがGPCRオリゴマーを形成するように互いに親和性を有する第一及び第二GPCRを同定する方法であって、以下の工程、
(a)第一GPCR及び融合タンパク質として第一GPCRに結合したGタンパク質をコードする第一核酸構築物を製造する工程であって、該第一GPCRは野生型GPCRと比較して突然変異されており、それにより該第一GPCRはリガンド結合後に該結合したGタンパク質を活性化することができない、上記工程、
(b)第二GPCR及び融合タンパク質として第二GPCRに結合したGタンパク質をコードする第二核酸構築物を製造する工程であって、該Gタンパク質は野生型Gタンパク質と比較して突然変異されており、それにより該Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記工程、
(c)細胞内で第一及び第二核酸構築物を同時発現させる工程、及び
(d)第一及び第二GPCRを含むヘテロ二量体からなる複合体の存在を同定する工程
を含み、第一GPCRの野生型が第二GPCRと異なり、該ヘテロ二量体が、それと結合するリガンドに反応してシグナル伝達カスケードを開始することができる、上記方法。 - 前記GPCRオリゴマーが、請求項1〜12のいずれか1項に記載の生物学的試薬である、請求項16に記載の方法。
- 前記細胞を第二GPCRに対するリガンドと接触させること及び前記第一Gタンパク質が活性化されているかどうかを測定することにより、複合体の存在を決定する、請求項16または17に記載の方法。
- 前記野生型第一及び第二GPCRを、少なくとも1つの細胞型により内生的に同時発現する、請求項16〜18のいずれか1項に記載の方法。
- GPCRオリゴマー形成により生じた新しい又は変化したリガンド結合部位の存在を測定する方法であって、以下の工程、
a)化合物を、GPCRオリゴマーを発現する第一細胞に接触させる工程であって、ここでGPCRオリゴマーは、(i)Gタンパク質に結合した第一GPCRであって、該第一GPCRは改変されており、それによって該第一GPCRはリガンド結合後に該Gタンパク質を活性化することができない、上記第一GPCR、及び(ii)Gタンパク質に結合した第二GPCRであって、Gタンパク質は改変されており、それによって該Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを有する、上記工程、
b)該化合物を未改変の第一GPCRを発現する第二細胞に接触させる工程及び/又は該化合物を未改変の第二GPCRを発現する第二細胞に接触させる工程、及び
c)第一細胞及び第二細胞に対する該化合物の作用を比較して、GPCRオリゴマーにより作られた新しい又は改変されたリガンド結合部位の存在を測定する工程を含み、該第一GPCRの野生型が第二GPCRと異なる、上記方法。 - 前記GPCRオリゴマーが、請求項1〜12のいずれか1項に記載の生物学的試薬である、請求項20に記載の方法。
- GPCRオリゴマーの形成の結果としての、リガンド結合後にGタンパク質を活性化するGPCRの機能の変化を測定する方法であって、以下の工程、
a)化合物を、GPCRオリゴマーを発現する第一細胞に接触させる工程であって、ここでGPCRオリゴマーは、(i)Gタンパク質に結合した第一GPCRであって、該第一GPCRは改変されており、それによって該第一GPCRはリガンド結合後に該Gタンパク質を活性化することができない、上記第一GPCR及び(ii)Gタンパク質に結合した第二GPCRであって、該Gタンパク質は改変されており、それによって該Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを有する、上記工程、
b)該化合物を、未改変の第一GPCRを発現する第二細胞及び/又は未改変の第二GPCRを発現する第二細胞に接触させる工程、及び
c)GPCRオリゴマーの機能を、未改変の第一GPCRの機能及び/又は第二GPCRの機能と比較して、オリゴマー形成により生じた受容体の機能の変化を測定する工程、
を含み、該第一GPCRの野生型が第二GPCRと異なる、上記方法。 - 前記GPCRオリゴマーが、請求項1〜12のいずれか1項に記載の生物学的試薬である、請求項22に記載の方法。
- リガンド結合後にGタンパク質を活性化する前記受容体の機能の変化が化合物の効力の変化である、請求項22または23に記載の方法。
- 前記受容体の機能の変化が、前記化合物による受容体の活性化により生じた細胞シグナル伝達経路の変化である、請求項24に記載の方法。
- 化合物がGPCRオリゴマーに対して有する、細胞シグナル伝達カスケードを開始する作用を測定する方法であって、以下の工程、
a)該化合物を、GPCRオリゴマーを発現する第一細胞に接触させる工程であって、GPCRオリゴマーが、(i)Gタンパク質と結合した第一GPCRであって、該第一GPCRは野生型GPCR配列と比較して改変したアミノ酸配列を有しており、それによって該第一GPCRはリガンド結合後に該Gタンパク質を活性化することができない、上記第一GPCR、及び(ii)Gタンパク質と結合した第二GPCRであって、該Gタンパク質は改変されており、それによって該Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを有する、上記工程、
b)該化合物と該GPCRオリゴマー間の接触により生じた細胞シグナルの存在を検出する工程、及び
c)化合物がGPCRオリゴマーに対して有する作用を測定する工程、
を含み、第一GPCRの野生型が第二GPCRと異なる、上記方法。 - 前記GPCRオリゴマーが、請求項1〜12のいずれか1項に記載の生物学的試薬である、請求項23に記載の方法。
- 前記作用を、化合物と未改変の第一GPCR間の接触により生じる作用及び/又は化合物と未改変の第二GPCR間の接触により生じる作用と比較する工程を更に含む、請求項26または27に記載の方法。
- GPCRオリゴマーと結合して、細胞シグナル伝達カスケードを開始することができる化合物を同定する方法であって、以下の工程、
a)請求項1〜12のいずれか1項に記載の生物学的試薬を発現する細胞を提供する工程、
b)生物学的試薬を該化合物に接触させる工程、及び
c)該化合物がGPCRオリゴマーと結合して、細胞シグナル伝達カスケードを開始するかどうかを測定する工程、
を含む、上記方法。 - 前記化合物と前記GPCRオリゴマー間の相互作用を、相互作用により生じる細胞シグナルの存在を同定することにより測定する、請求項29に記載の方法。
- 前記細胞シグナルを、Ca2+、cAMP、イノシトール1,4,5-三リン酸レベル、プロテインキナーゼもしくはプロテインC活性、またはMAPキナーゼ活性の細胞内レベルにより測定する、請求項30に記載の方法。
- 前記細胞シグナルを、レポーターアッセイを使用して測定する、請求項30に記載の方法。
- 前記細胞が色素細胞であり、前記細胞シグナルを細胞中の色素の凝集状態の変化により測定する、請求項30に記載の方法。
- 前記色素細胞がメラニン形成細胞である、請求項33に記載の方法。
- 前記化合物が、アゴニスト、アンタゴニスト又はインバースアゴニストとして前記GPCRオリゴマーと相互作用する、請求項29〜34のいずれか1項に記載の方法。
- GPCRオリゴマーとそのリガンド間の結合を調節する能力を有する化合物を同定する方法であって、以下の工程、
a)GPCRオリゴマーを発現する細胞を提供する工程であって、該GPCRオリゴマーが、(i)第一Gタンパク質と結合した第一GPCRであって、該第一GPCRはリガンド結合後にGタンパク質を活性化することができない、上記第一GPCR、及び(ii)第二Gタンパク質に結合した第二GPCRであって、該第二Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを含む、上記工程、
b)該細胞をリガンドの存在下で試験化合物と接触させる工程、及び
c)GPCRオリゴマーと結合する該リガンドの能力を、同等の条件下であるが該化合物の非存在下でGPCRオリゴマーと結合する該リガンドの能力と比較する工程、
を含み、該第一GPCRの野生型が第二GPCRと異なる、上記方法。 - 前記化合物がタンパク質である、請求項36に記載の方法。
- 前記タンパク質が第三GPCRである、請求項37に記載の方法。
- 前記第一、第二及び第三GPCRを少なくとも1つの細胞型により内生的に同時発現する、請求項38に記載の方法。
- 前記リガンドが、請求項20に記載の方法によりオリゴマー上に存在すると決定された新しい又は変化したリガンド結合部位に結合する、請求項36〜39のいずれか1項に記載の方法。
- 前記GPCRオリゴマーが、請求項1〜12のいずれか1項に記載の生物学的試薬である、請求項36〜40のいずれか1項に記載の方法。
- 示差的Gタンパク質共役を評価する方法であって、以下の工程、
(a)GPCRオリゴマーを発現する第一細胞を準備する工程であって、GPCRオリゴマーが、(i)第一Gタンパク質と結合した第一GPCRであって、ここで該第一GPCRは野生型GPCR配列と比較して改変されたアミノ酸配列を有しており、それによってリガンド結合後に該第一Gタンパク質を活性化することができない、上記第一GPCR、及び(ii)第二Gタンパク質と結合した第二GPCRであって、該第二Gタンパク質は、該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを含む、上記工程、
(b)GPCRオリゴマーを発現する第二細胞を準備する工程であって、GPCRオリゴマーが、(i)第一Gタンパク質と結合した第一GPCRであって、ここで第一GPCRは機能的であり、かつ該Gタンパク質は該第一GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第一GPCR、及び(ii)第二Gタンパク質と結合した第二GPCRであって、第二GPCRはリガンド結合後にGタンパク質を活性化することができない、上記第二GPCRを含む、上記工程、
(c)第一Gタンパク質と結合した第一GPCRの単量体を発現する対照細胞を準備する工程であって、GPCR及びGタンパク質の両方が機能的である、工程、
(d)第一、第二及び対照細胞を、第一及び/又は第二GPCRに存在するリガンド結合部位に結合できる化合物と接触させる工程、
(e)異なる第一及び/又は第二Gタンパク質を用いて、工程(a)〜(d)を繰り返す工程、及び
(f)示差的Gタンパク質共役を評価する工程、
を含み、該第一GPCRの野生型が第二GPCRと異なる、上記方法。 - GPCRに対する示差的Gタンパク質共役を評価する方法であって、以下の工程、
(a)GPCRオリゴマーを発現する第一細胞を準備する工程であって、ここでGPCRオリゴマーが、(i)第一Gタンパク質と結合した第一GPCRであって、該第一GPCRは野生型GPCR配列と比較して改変されたアミノ酸配列を有しており、それによってリガンド結合後に該第一Gタンパク質を活性化することができない、上記第一GPCR、及び(ii)第二Gタンパク質と結合した第二GPCRであって、該第二Gタンパク質は該第二GPCRからの刺激後に細胞シグナル伝達カスケードを開始することができない、上記第二GPCRを含む、上記工程
(b)第一Gタンパク質と結合した第二GPCRを発現する第二細胞を準備する工程であって、第二GPCRと第一Gタンパク質の両方が機能的である、工程、
(c)第一及び第二細胞を、第二GPCRに存在するリガンド結合部位に結合できる化合物と接触させる工程、
(d)異なる第一Gタンパク質を用いて、1回以上、工程(a)〜(c)を繰り返す工程、
(e)場合により、異なる第一GPCRを用いて、1回以上、工程(a)〜(d)を繰り返す工程、及び
(f)第二GPCRにより、示差異的Gタンパク質共役を評価する工程、
を含み、該第一GPCRの野生型が第二GPCRと異なる、上記方法。 - 前記第一及び第二Gタンパク質を、Gs、Gi、Gq、G11、G12、G13、G15、G16、Go又はGzから選択する、請求項42または43に記載の方法。
- 前記第一及び第二Gタンパク質を、Ca2+、cAMP、cGMP、イノシトール1,4,5-三リン酸、ジアシルグリセロール、プロテインキナーゼC活性又はMAPキナーゼ活性から選択した細胞内レベルを調節するGタンパク質から選択する、請求項42または43に記載の方法。
- 前記第一GPCRが、野生型GPCR配列と比較して改変したアミノ酸配列を有しており、それによって該第一GPCRはリガンド結合後に第一Gタンパク質を活性化することができず、かつ第一GPCRの野生型が第二GPCRと異なる、請求項42〜45のいずれか1項に記載の方法。
- 前記野生型第一及び第二GPCRを、少なくとも1つの細胞型で内生的に同時発現する、請求項42〜46のいずれか1項に記載の方法。
- 前記第一GPCRを、少なくとも1つのアミノ酸残基を置換することにより、野生型GPCRと比較して突然変異させる、請求項42〜47のいずれか1項に記載の方法。
- 前記少なくとも1つのアミノ酸がGPCRの第二細胞内ループに存在する、請求項48に記載の方法。
- 前記少なくとも1つのアミノ酸が、疎水性アミノ酸残基であって、かつ酸性アミノ酸残基に置換される、請求項49に記載の方法。
- 前記第二Gタンパク質を、少なくとも1つのアミノ酸残基を置換することにより、野生型Gタンパク質と比較して突然変異させる、請求項42〜51のいずれか1項に記載
の方法。 - 前記置換されるアミノ酸残基がグリシンである、請求項52に記載の方法。
- 前記第一及び第二GPCRがクラスAのGPCRである、請求項42〜53のいずれか1項に記載の方法。
- 前記クラスAのGPCRを明細書記載の図3に提供されたグループから選択する、請求項54に記載の方法。
