アスパラガス(秋田産)1kgを5mm以下にブレンダーで細断し、水2Lを導入して、オートクレーブで100℃、1時間熱処理を行った。得られた処理液は濾過助剤に珪藻土を用い、ろ紙(ADVANTEC東洋製No.5C)を用いて吸引濾過を行い、薄茶色の抽出液を得た。得られた濾液を卓上エバポレーター(EYELA社製)により10倍に減圧濃縮した後、凍結乾燥して、アスパラガス抽出物37.7gを得た。アミノ酸及びGABAの含有量は表1に示したとおりであった。
アスパラガス若茎(秋田産)1kgをミキサーで破砕し、水2Lを導入した。そこにグルタミン酸ナトリウム50gを添加してよく攪拌して溶解した。これを振とう式インキュベーターを用いて25℃、24時間振とうしGABA富化処理を行った。得られた処理液は濾過助剤に珪藻土を用い、ろ紙(ADVANTEC東洋製No.5C)を用いて吸引濾過を行い、得られた濾液を卓上エバポレーター(EYELA社製)により10倍に減圧濃縮した後、凍結乾燥して、アスパラガス抽出物74.4gを得た。GABA及びアミノ酸の含有量は表1に示したとおりであった。
実験動物:Std(クリーン):Wistar系ラット(雄)(日本エスエルシー社、約180g)、6週齢、30匹
通常試料:ラボMRストック(日本農産工業)
CCr(ml/min/kg体重)={尿中Cr(mg/dl)×尿量(ml)/血清中Cr(mg/dl)}×{1000/体重(g)}×{1/1440(min)}
尿素窒素(UN)とクレアチニン(Cr)は含窒素物質の最終代謝産物であり、主として腎臓を通じて体外に排出される。よってUNやCr濃度は、腎臓機能が低下すると、血清中の濃度が高くなる。クレアチニンクリアランス(CCr)は、単位体重、単位時間当たり排出される血中クレアチニン量を表す指標であるため、この値が低下すれば、腎機能低下と評価される。
結果:血清中のUN、Cr濃度及びCCrの結果は表3の通りである。なお、結果は平均値±標準偏差で示した。
実験動物:Dahl食塩感受性ラット(雄)(成和実験動物研究所、約180g)、6週齢、30匹
通常試料:食塩添加(8%)粉末食(CE−2)(日本クレア)
方法:ラットは、室温25℃±1℃、湿度60%±5%、12時間明期12時間暗期(明期7時〜19時)の条件下で飼育した。試料投与前に1週間高食塩食を与え、また、生理食塩水を1日1回2ml/kgで胃ゾンデで投与し、馴化を行った。その後、5群に分け、表2にあるA〜Eの試料を2ml/kgになるよう生理食塩水に溶解し、胃ゾンデで1日1回投与した。実験開始30日目に尿を採取し、直後に採血を行った。実施例1に従って、尿中および血清中の尿素窒素(UN)とクレアチニン(Cr)の濃度、クレアチニンクリアランスを算出した。
結果:血清中のUN、Cr濃度及びCCrの結果は表4の通りである。なお、結果は平均値±標準偏差で示した。
製造例2で製造したGABA富化アスパラガス抽出物を凍結乾燥した後粉砕した。得られた粉末を用いて、以下の配合割合でよく混合、造粒した後、1粒500mgで打錠し、錠剤を製造した。なお、%は、質量%を示す。
結晶セルロース 15%
還元麦芽糖 30%
ショ糖脂肪酸エステル 5%
製造例2と製造したGABA富化アスパラガス抽出物を凍結乾燥した後粉砕した。得られた粉末100gを日本粉末薬品(株)製「ウーロン茶エキスパウダー」900gと混合し、GABA含有肝機能障害予防・改善ウーロン茶粉末を製造した。このGABA含有ウーロン茶粉末5gを水500mlに溶解し、GABA含有腎機能障害予防・改善ウーロン茶を製造した。
Claims (2)
- アスパラガスから得られる組成物を有効成分とすることを特徴とする腎機能低下予防・改善剤。
- アスパラガスから得られる組成物が、アスパラガスにγ−アミノ酪酸の含有量を高めるための富化処理を施したものである請求項1記載の腎機能低下予防・改善剤。
