Description

(i)いわゆる自殺遺伝子の遺伝子産物は、前駆体を細胞障害性分子へと変換する。例は、ガンシクロビルの投薬と関連した単純ヘルペスウイルス(HSV)のチミジンキナーゼ(TK)である。特に有利な点は、毒性のあるガンシクロビル三リン酸が隣接細胞に拡散し、それによってバイスタンダー効果が起こることである。近年、この酵素の活性がさらに増加した。HSV TKは、現在、腫瘍細胞並びに移植されたベクター産生細胞を排除するための非常に効率的な可能性のあるものである。
(ii)IL−4などの免疫刺激サイトカインの発現は、腫瘍細胞に対する自然免疫を刺激することができる。
(iii)エンドスタチンなどの抗血管新生タンパク質の分泌により血管が欠失し、それ故、代謝的に非常に活発な腫瘍組織における栄養分の供給が欠乏する。腫瘍は、実質的に「飢餓」する。
(iv)最後に、これらの細胞の制御されていない増殖を阻害するために、腫瘍細胞のシグナル伝達または細胞周期に関与する一連の遺伝子が、記載された。しかしながら、臨床においてこれらの遺伝子を使用する可能性は、限定されている。なぜなら、これらの遺伝子は、遺伝子の改変された細胞自体においてしか作用せず、そして最初に記載した活性原理としてのバイスタンダー効果は有さないからである。このことは、治療効果を達成するために、実質的に全ての悪性細胞を遺伝子的に改変しなければならないことを意味し、これは理想的なベクター系を用いてさえも無理である。
(i)それらは、腫瘍特異的な指向性を有するべきであり、それによってウイルス複製および細胞溶解は、腫瘍組織に限定されて留まる。ウイルスエンベロープの改変によって、または腫瘍特異的プロモーターの使用によってこの特性を増強させることができる。神経膠腫細胞のほんの一部しか処置中に分裂していないと想定されるので、増殖中の細胞だけでなく休止細胞にも感染するウイルスが有利である。
(ii)安全性に関連する面に関しては、高い遺伝子的安定性および腫瘍組織外における低毒性が、臨床使用のために特に適している。このことは、GMP条件下における高いウイルス力価およびベクターの精製を可能とする。理想的には、OVはヒトに対して非病原性であるべきであり、そして個体群間で低い感染率を有するべきである。既存免疫は時期尚早なウイルスの中和をもたらし、これにより効率的な療法が可能とならないだろう。
それ故、本発明の主題は、リンパ球性脈絡髄膜炎ウイルス(LCMV)の糖タンパク質GPをコードする遺伝子を含み、VSVのエンベロープタンパク質Gをコードする機能的遺伝子を全く含まない、水疱性口内炎ウイルス(VSV)シュードタイプベクター(VSV−LCMV−GPシュードタイプベクター)である。LCMVのGP(LCMV−GP)は、LCMVの糖タンパク質GP−1またはGP−2であり得る。
本発明は、組換え水疱性口内炎ウイルス(VSV)およびVSVベクターに関する。VSVゲノムは、タンパク質L、M、N、PおよびGをコードし、そしてウイルスの繁殖に必須である5つの遺伝子l、m、n、pおよびgを含む。Nは、VSVゲノムRNAをパッケージングする核タンパク質であり;VSVゲノムは、RNA−タンパク質複合体としてしか複製することができない。LおよびPは一緒に、ポリメラーゼ複合体を形成し、これは、VSVゲノムを複製し、そしてVSVmRNAを転写する。Mは、脂質エンベロープとヌクレオカプシドとの間に一種のキットを形成する基質タンパク質であり、そしてこれは細胞膜における粒子の出芽に重要である。Gは、ウイルスエンベロープに取り込まれたエンベロープタンパク質であり、そしてこれはウイルスの感染性に必須である。
(i)VSVベクターは、腫瘍退縮性であり、そして他の腫瘍退縮性ウイルスベクターと比較して特に高い腫瘍退縮活性を有する:
(ii)VSVベクターは、優先的に腫瘍細胞において複製し、そして他の腫瘍退縮性ウイルスベクターと比較して特に高い複製能を有する。
(iii)VSVベクターは、活発に分裂している細胞と同様に、休止腫瘍細胞にも感染する。
(iv)VSVベクターは、強力な自然体液性および細胞性免疫応答を誘導する。
(v)VSVベクターは、純粋に細胞質で複製し、すなわち、RNAウイルスのようにそれらは宿主細胞のゲノムに組み込んだり、または複製能を有するウイルス中に組換えたりすることができない。
(vi)VSVベクターはパッケージングし易い。
(vii)VSV糖タンパク質は、外来のエンベロープタンパク質と交換可能である。VSVエンベロープに以前に取り込まれた糖タンパク質の例は、HIVgp160(OwensおよびRose 1993)、HCVE1/E2(Tani et al., 2007)、SARS S(Ge et al., 2006)、およびLassa GP(Garbutt et al., 2004)である。
(i)LCMV−GPは、細胞毒性ではない。
(ii)LCMV−GPシュードタイプベクターを、感染性を失うことなく超遠心分離によって濃縮することができる。
(iii)LCMV−GPシュードタイプベクターは、神経膠細胞に対して好ましい指向性を示し、一方ニューロンは非効率的に感染される。
実施例1:VSV−LCMV−GPシュードタイプベクターによる神経膠腫細胞の形質導入
GFPをコードするがウイルスエンベロープタンパク質はコードしない欠陥VSVベクター(VSVgfp−ncp−ΔG)を、今度はLCMVのGPを安定に発現するBHK−21およびvero細胞に形質導入した。24ウェルプレートの1つの窪みあたり1×10E5個の細胞(BHK−21、−GP、vero、−GP)を播種し、そして4時間後にMOI=5でVSVgfp−ncp−ΔGベクターを用いて形質導入した。24時化後、培養上清を回収し、そしてBHK−21における力価を、GFP発現のFACS分析によって決定した。図1は、形質導入細胞がLCMV−GP−シュードタイプ化VSVベクターを産生し、使用する細胞型に依存して、シュードタイプ力価は2〜7×10E5TU/mlの間で変動したことを示す。
本発明者らは、C. Verfaillie (Jiang et al., 2002)のプロトコールに従って単離された幹細胞が、腫瘍浸潤特性を有することをすでに実証している。その遊走能だけを調べ、その分化能は調べなかったので、これらの細胞をBM−TICと呼んだ(骨髄由来種腫瘍浸潤細胞、bone marrow derived tumor infiltrating cells)。VSVベクターについてのBM−TICのパッケージング能を試験するために、LCMV−GP−発現BM−TICを、欠陥VSV-GFPベクターを用いて形質導入した。24ウェルプレートの1つの窪みあたり1×10E5個の細胞(BM−TIC、−GP、BHK−21)を播種し、そして4時間後にMOI=5でVSVgfp−ncp−ΔGベクターを用いて形質導入した。24時間後、培養上清を回収し、そしてBHK−21細胞上で滴定した。図3は、VSV−LCMV−GPシュードタイプをまたBM−TIC中でも調製することができることを示す。しかしながら、力価は、例えばBHK−21細胞の場合よりも低い(図1参照)。
繁殖することのできるウイルスを用いて高い形質導入率を、達成することができる。神経膠腫への遺伝子導入率を高め、そして同時に高度の安全性を確実にするために、限定された程度で複製能を有するVSVシュードタイプベクターを、確立した。補完的で欠陥のあるウイルスを用いた系が、レトロウイルス(Trajecevski et al., 2005)および種々のフラビウイルス(Riepl & Mandl, 2007)のために記載されている。このような系の原理は、2つの不完全なレプリコンの間にウイルスゲノムを分配することである。1つの同じ細胞が両方のベクターで感染され、そしてこの同時感染を通してのみウイルスゲノムをパッケージングするために必要な全ての成分を含む場合にのみ、感染性ウイルスが産生される。
その力価およびその形質導入効率に関して個々のベクターを特徴付けた後、in vitroにおけるベクターの伝播を調べる。このために、BHK−21細胞を、両方のベクターで同時感染させ、そしてネイティブなBHK−21上での培養上清の連続継代による新たなベクターの複製および従って生成を調べる。これらの実験を、単層並びにスフェロイドとして培養される種々の神経膠芽腫細胞系(G44、G62、U87)を用いて繰り返す。
遊走VSV−LCMV−GP生産細胞を、crVSVベクターおよび複製能を有するVSV−LCMV−GPベクターのために確立する。
VSV Mタンパク質はVSVサイクル内に2つの必須の仕事を有する。一方で、それは、ウイルスの会合および出芽のためのビリオンの構造成分として必須である。他方で、それは、宿主のタンパク質合成に向けられるその「宿主シャットオフ」活性を通して、およびアポトーシスの誘導を通してウイルスの病態発生に著しく寄与する。これに関して、Mの重要な機能は、IFN mRNAの核−細胞質輸送の阻害である。それにより、ウイルス感染に対する細胞の最初の防御機序が、抑制される。
腫瘍細胞は主に、VSV−LCMV−GPシュードタイプの神経膠腫特異的指向性に因り形質導入される。それ故、依然として特定の細胞病原性を示すM変異体を用いて作業することも、可能である。BM−TICにおける毒性作用を低く維持するために、抗ウイルス性MxAタンパク質を、Tet誘導性プロモーターを介して細胞中で発現させるべきである。Tet誘導性プロモーターを有するレトロウイルスベクターは公知である(Loew et al., 2006)。Tet応答性エレメント(tTA)も、レトロウイルスベクターを介してBM−TICに導入する。MxAは、RNAウイルスに対して抗ウイルス活性を有するインターフェロン誘導GTPアーゼである(Pavlovic et al., 1992; StaeheliおよびPavlovic, 1991)。それは細胞質中で大きなオリゴマーの形態で存在し、毒性ではなく、そして今まで知られていない機序によってVSVの複製を阻害する。LCMV−GPを発現するBM−TICを、Tetにより調節されるMxAおよびtTAベクターを用いて形質導入し、そして続いてVSVベクターを用いて感染させる。抗生物質が培地中に存在する限り、細胞は、MxAの存在に因りVSV(LCMV−GP)を用いての増殖性感染に対して抵抗性である。in vivoにおける適用中にのみ、抗生物質が除去され、そしてVSV(LCMV−GP)のウイルス複製および産生が、次第に始まる。2日間の半減期を有するMxAは非常に安定であるので、突然変異体MxA(L612K)を代替的に使用することができる。MxA(L612A)は、野生型タンパク質と同じ有効性でVSVの感染を阻害するが、オリゴマーを形成することはできない。これによってそれは不安定化され、そして僅か2時間という短い半減期を有する(Janzen et al., 2000)。
(i)ベクター力価およびベクター産生持続時間を決定する。
(ii)ベクター力価に対する細胞の凍結および解凍の影響を分析する。
(iii)細胞が依然として腫瘍浸潤特性を有し、そしてその形質導入効率が腫瘍組織において達成されるかどうかを、最初にスフェロイドモデルにおいてそしてその後は動物モデルにおいて分析する。
(iv)産生されたベクター上清を、複製性VSVの存在について確認する。
自殺遺伝子を、本発明に従って治療遺伝子として使用することができる。このような遺伝子の例は、臨床において最も多くの経験を有する単純ヘルペスウイルスのチミジンキナーゼ(HSV−TK)である。HSV−TKの恒常的発現を通して、パッケージング細胞および形質導入細胞を療法の終了時に確実に排除することができ、これによって有糸分裂の活発なパッケージング細胞から危険が生じる可能性が全くなくなる。

組換えVSVベクターを最初にin vitroにおいて特徴付け、そしてその後、その有効性について動物モデルにおいて調査する。この目的のために、2つの特に意味のあるラットモデルを使用する。
(i)それは、個体間のばらつきの少ない再現性の良好な系を示す。
(ii)腫瘍は、ヒト神経膠腫と類似した特徴、例えば、侵襲性および活動性の増殖挙動並びに免疫抑制因子としてのTGFβ2の産生などを有する。
(iii)SCID/ヌードマウスモデルまたはラット異種移植片モデルとは対照的に、前記系は、免疫学的局面の検査または免疫治療法の試験を可能とする。
(i)腫瘍は、ヒト神経膠腫に類似した特徴、例えば侵襲性および活動性の増殖挙動などを有する。神経膠腫細胞系ではなく原発性の患者の材料を、腫瘍の確立のために使用する。
(ii)腫瘍の連続継代を通して、悪性腫瘍発達の種々の時期をマッピングすることができ、そして療法の成功を確認することができる。第1世代の腫瘍は、ゆっくりと成長し、高度に侵襲性であり、そして低度の神経血管新生を有する。強力な増殖および低下した侵襲性を有する高度に血管新生化した表現型は継代を通して発達する。
(iii)高度に侵襲性で非血管新生性の神経膠腫の細胞表現型は、腫瘍幹細胞の1つに類似している。従って、この動物モデルは、ヒトGBMの幹細胞成分に対する我々の概念の治療有効性の試験を可能とする。
(i)ゆっくりとした高度に侵襲性の増殖挙動を有する初期腫瘍。これらの腫瘍は、低度の血管新生しか有さない。
(ii)血液が十分に供給されそして急速に増殖する後期腫瘍。この表現型は、侵襲性が低い。
Claims (24)
- リンパ球性脈絡髄膜炎ウイルス(LCMV)の糖タンパク質GPシュードタイプ化水疱性口内炎(VSV)ウイルスベクター(VSV−LCMV−GPシュードタイプベクター)が、LCMVの糖タンパク質GPをコードする遺伝子を含み、そしてVSVのエンベロープタンパク質Gをコードする機能的遺伝子を欠失していることを特徴とする、前記VSV−LCMV−GPシュードタイプベクター。
- エンベロープタンパク質Gが、LCMVの糖タンパク質GPによって置換されていることを特徴とする、請求項1に記載のVSV−LCMV−GPシュードタイプベクター。
- VSVのMタンパク質が、VSVの細胞病原性を減少させる突然変異を含み、ここにおいて突然変異がM33A、M51A、V221F、S226Rおよび 37 PSAP 40 領域におけるアミノ酸交換からなる群より選択されることを特徴とする、請求項1または2に記載のVSV−LCMV−GPシュードタイプベクター。
- ベクターが、少なくとも1つの導入遺伝子を含むことを特徴とする、請求項1〜3のいずれか1項に記載のVSV−LCMV−GPシュードタイプベクター。
- 導入遺伝子が、自殺タンパク質、免疫刺激タンパク質またはマーカータンパク質をコードすることを特徴とする、請求項4に記載のVSV−LCMV−GPシュードタイプベクター。
- 自殺タンパク質が、単純ヘルペスウイルスのチミジンキナーゼ(HSV−TK)、シトシンデアミナーゼ、FKBP−FASまたはFKBP−カスパーゼ9であることを特徴とする、請求項5に記載のVSV−LCMV−GPシュードタイプベクター。
- 免疫刺激タンパク質が、IL−2、IL−4、IL−12、中和性抗TGFβまたはFlt3Lであることを特徴とする、請求項5に記載のVSV−LCMV−GPシュードタイプベクター。

- ウイルス産生細胞が、VSVのタンパク質N、L、PおよびMをコードする遺伝子n、l、pおよびmならびにLCMVの糖タンパク質GPをコードする遺伝子gpからなる群より選択される少なくとも1つの遺伝子の発現のための1つ以上の発現カセットを含むことを特徴とする、請求項12〜14のいずれか1項に記載の前記ウイルス産生細胞。
- ウイルス産生細胞が、シュードタイプ化VSVビリオン中にパッケージングするための遺伝子導入用ベクターを含み、前記遺伝子導入用ベクターは導入遺伝子を含むことを特徴とする、請求項12〜15のいずれか1項に記載のウイルス産生細胞。
- 導入遺伝子が、自殺タンパク質、免疫刺激タンパク質および/またはマーカータンパク質をコードする遺伝子であることを特徴とする、請求項16に記載のウイルス産生細胞。
- 細胞を、請求項11に記載のシュードタイプ化VSVビリオンを用いて形質導入し、前記ビリオンは導入遺伝子を含むことを特徴とする、in vitroにおいて細胞に導入遺伝子を導入するための方法。
- 細胞に、請求項16または17に記載のウイルス産生細胞を接触させることを特徴とする、in vitroにおいて細胞に導入遺伝子を導入するための方法。
- 細胞が、腫瘍細胞であることを特徴とする、請求項18または19に記載の方法。
- 固形腫瘍の療法のための薬学的組成物の調製のための、請求項1〜7のいずれか1項に記載のVSV−LCMV−GPシュードタイプベクター、請求項8〜10のいずれか1項に記載のVSV−LCMV−GPシュードタイプベクター系、請求項11に記載のシュードタイプ化VSVビリオン、または請求項12〜17のいずれか1項に記載のウイルス産生細胞の使用。
- 腫瘍が、脳腫瘍であることを特徴とする、請求項21に記載の使用。
- 腫瘍が、悪性脳腫瘍であることを特徴とする、請求項22に記載の使用。
- 薬学的組成物が、請求項1〜7のいずれか1項に記載のVSV−LCMV−GPシュードタイプベクター、請求項8〜10のいずれか1項に記載のVSV−LCMV−GPシュードタイプベクター系、請求項11に記載のシュードタイプ化VSVビリオン、または請求項12〜17のいずれか1項に記載のウイルス産生細胞を含むことを特徴とする、前記薬学的組成物。
