Description
実施例I−1:純粋シルクフィブロインの精製
本発明に使用した絹蛋白質は、家蚕(Bombyx mori)を全齢桑葉で飼育して得た蚕繭を精練して使用した。まず、セリシンを除去するために繭内に残っている蛹を除去して、水8l、炭酸ナトリウム0.45g及びマルセル石鹸0.75gを混合した溶液に蚕繭150gを入れて、40分間ずつ2回沸騰処理した。その後、適量の水でさらに20分間ずつ2回沸騰処理した後、3回水洗して乾燥すると、水溶性のセリシンが溶解されて除去される。この時の減少重量は、37g(約25%減少)であった。水洗後、自然乾燥して、純粋なフィブロイン蛋白質を得た。
前記実施例I−1の方法により得られたフィブロイン35gを基準に、塩化カルシウム(CaCl2、1級、Mw=110.99)・H2O・エタノール(1:8:2モル含量)を添加し、90℃で5時間反応して溶解した後、ガーゼ及び不織布で異物を完全に濾過した後、蒸留水で2倍希釈した。直径10cm、長さ1mのGradiFacシステム(Pharmacia Biotech, Sephadex G-25 Media, HiLoad P-50 Pump UV-1 Monitor, スウェーデン)を使用したゲル濾過装置で中性塩を除去した。その後、得られたフィブロイン溶液に対し、フレーバーザイム及びスミザイム(sumizyme;NOVA、米国)が混合(1:1)された蛋白質加水分解酵素を1%加えて、55℃で5時間加水分解を行った。その後、100℃で5〜10分間熱処理して酵素の活性をなくし、凍結乾燥により粉末を製造して、これを‘BG101’と命名した。
前記実施例I−1の方法により得られたフィブロイン113gを基準に、25%の塩酸水溶液3,400mlを添加して、110℃で12時間加水分解を行った。これを4M水酸化ナトリウム水溶液でpH5.0〜5.5に調節した。次いで、紙濾過により残留物を除去した後、活性炭で充填されたフィルターを通過させた後、電気透析装置内のサンプル槽に1,000mlを入れて、15V、20mAの電圧及び電流が自動に調節される装置を利用して、電気脱塩を行った。その後、スプレードライヤーにより粉末を製造して、これを‘BG201’と命名した。
実施例II−1:分子量分析
前記実施例Iで製造されたBG101及びBG201に対して、ゲル透過クロマトグラフィにより絶対分子量を計算した。0.2N NaNO3を緩衝溶液として、約0.5%に濃度を調節した水溶液状態の試料を屈折率(RI)、光散乱(LS)、回折圧及び紫外線(280nm)吸光度値から絶対分子量の分布が自動に計算されるGPCシステム(Viscoteck、米国)装置を使用した。標準試料としてポリエチレンオキシド(PEO、Mw=110,000)を使用して、再現性を確認した。実験結果、BG101の重量平均分子量(weight average molecular weight, Mw)は、1070であって、BG201は、850であった。
前記実施例Iで製造したBG101とBG201の溶解度を幾つかの溶媒を使用して測定した。0.1gのシルクペプチド粉末を10mlの蒸留水、エタノール、メタノール、アセトン及びジメチルホルムアミドに溶解して、溶解度(単位%)を測定して、その結果は、表1に示した。表1から確認できるように、溶解度は、BG101粉末とBG201粉末の両方とも、蒸留水では完全に溶解されたが、エタノール、メタノールなどの有機溶媒では、溶解度が著しく低下した。
前記実施例Iで製造したBG101とBG201の溶解度をpHを変化させながら測定した。0.1gのシルクペプチド粉末を10mlの蒸留水に溶解して、溶解度を測定した。蒸留水のpHは、0.1N塩酸と水酸化ナトリウムでpH3、5、7、9及び11に調節した。実験結果、表2から分かるように、溶媒のpHに関係なく、シルクペプチドはよく溶解された。
前記実施例Iで製造したBG101とBG201のアミノ酸組成分析を行った。各サンプル0.05%を取って、6N塩酸水溶液1mlに入れて窒素処理した後、110℃で18時間加水分解を行った。塩酸を完全に蒸発させた後、pH2.2のローディング緩衝液に希釈した後、全自動アミノ酸機械装置(Amersham Pharmacia Biotech、モデル:Biochrom 20 Plus、スウェーデン)を利用してアミノ酸分析を行った。得られたアミノ酸組成(単位:mol%)は、表3に示した。
前記実施例で製造されたシルクペプチドが脳神経細胞に及ぼす影響を調べるために、以下のような実験を行った。
神経細胞の一次培養は、Okudaらの方法(Okuda S. et al., Neuroscience 63(3):691-9(1994))を利用して、妊娠15日(E15)目のラット胎児の線状体(striatum)を分取して、0.25%トリプシンと0.01%DNase I酵素を処理して、個別細胞に分離した後、ピペットでさらに分離し、PEI−コーティングプレートに1×105細胞/cm2の密度で敷いて、三日後、ara−Cで神経膠細胞の培養を抑制し、純粋神経細胞を得た。また、必要に応じて、神経幹細胞株であるSK−N−SHまたはSHS−Y5Y(ATCC、米国)をPEI−コーティングプレートに80%密度で敷いて、DMEMまたはRPMIに10%FBSを補充した培養液で培養した。下記実験例のように、アミロイドβ(Aβ)、6−ヒドロキシドーパミン(6−OHDA)、セラミド、H2O2または3−ヒドロキシキヌレニン(3−HK)を処理する時は、2時間低血清培地(1%FBS含有)で前処理した後、適合した時間処置した。
BG101及びBG201が神経細胞の細胞死滅に及ぼす影響を調べるために、既存報告された方法(Shearman et al., Proc. Natl. Acad. Sci. 91(4):1470-4(1994), Shearman et al., J. Neurochem. 65(1):218-27(1995),及びKaneko et al., J. Neurochem. 65(6):2585-93(1995))を少々変形して、次のようにMTT還元実験を行った:BG101またはBG201の10pMを前記実験例で選別及び培養された神経細胞に6時間処理した後、10μMのAβ、10μMの6−OHDA、10μMのセラミド、300μMのH2O2、または250μMの3−HKを添加した。前記添加される物質は、細胞毒性により細胞死滅を誘導するようになるが、前記表示された濃度で36時間以内に50%程度の細胞死滅を誘導する。
前記実施例I−2と同様な方法によりBG101またはBG201を6時間処理して、培養された細胞に、前記実施例I−2と同一な濃度のAβ、セラミド、H2O2、6−OHDAまたは3−HKを処理した。次いで、4%パラホルムアルデヒド(in PBS, pH 7.4)で15分間固定し、PBSで洗浄した後、8μg/mlヘキスト33258溶液(Sigma,米国)で5分間それぞれの処理細胞を染色した。その後、蒸留水で2回洗浄して、グリセロール/PBS(9:1)でマウントした後、蛍光顕微鏡(Olympus Microscope、日本)で観察した(図3及び図4参照)。その結果、図3及び図4において、アポトーシス誘発物質(insult)のみを処理した細胞は、染色体の凝縮や核酸断片化(nuclear fragmentation)のような典型的なアポトーシス形状(morphology)を確認することができた。反面、BG101とBG201を前処理してインサルトを処理した場合、インサルトを単独処理した実験群に比べ、染色体凝縮や核酸断片化が効果的に阻害されていることを確認した。モック(mock)で表示されたものは、対照群であって、正常的な核の成長を示している。図2から分かるように、本発明のBG101及びBG201は、アポトーシスを効果的に抑制することが分かって、前記実験例I−2の結果の信頼性を再確認することができた。
前記実験例I−2及びI−3の結果において、BG101またはBG201がアポトーシスを抑制する効果が、カスパーゼ活性抑制によるものであるかどうかを調べるために、次のような実験を行った:まず、前記実験例I−2と同一な方法により、BG101またはBG201を6時間処理して、培養された細胞に、前記実験例I−2と同一な濃度のアミロイドβ、6−OHDA、セラミド、H2O2、FeSO4または3−HKを処理した。P100プレート当たり10×106細胞を1mlの細胞溶解緩衝液(50mM Tris-Hcl, 0.03% IGEPAL, 1mM DTT, pH 7.5)で破壊して細胞溶解液を得た。その後、細胞溶解液50μlにHEPES緩衝液(40mM HEPES, pH 7.5, 20%グリセロール, 4 mM DTT)内にあるカスパーゼの蛍光生成基質であるAc−DEVD−AMC(Enzyme Systems Products, カナダ)を500μM添加して、1時間37℃で反応させた。また、カスパーゼ活性抑制に対する陽性対照群として活用するために、広範囲(Pan)−カスパーゼ抑制剤であるzVAD−FMK(Enzyme Systems Products, ESP, カナダ)を10μM前処理した試料を用意した。
活性酸素は、老化の最も主要な原因であるばかりか、各種疾患の直・間接的な原因でもある。そこで、本発明のシルクペプチドが活性酸素に及ぼす影響を以下のように調査した。
前記実施例で製造された本発明のシルクペプチドBG101またはBG201が虚血による脳梗塞面積に及ぼす影響を調べるために、次のような実験を行った。
A.薬物処理及び局部的虚血誘導動物モデルの製作
体重200〜250gの雄性白鼠(Sprague-Dawley rat)を利用して局部的虚血誘導動物モデルを製作した。薬物は、1g/kgのBG101を、虚血誘導の1時間前に1回または虚血誘導の1時間後に1回口腔投与して(口腔投与群、n=6)、虚血対照群(n=6)は、薬物処理群と同じ容量の生理食塩水を投与した。実験動物にケタミン(ketamine)30〜40mg/kgを筋肉注射して麻酔した後、仰臥位で頚部に皮膚切開を加え、頚動脈(common carotid artery)、外頚動脈(external carotid artery)、及び内頚動脈(internal carotid artery)を探して、周囲組織と分離させた。まず、外頚動脈の分枝である上部甲状腺動脈と後頭動脈を電気焼灼して、内頚動脈の分枝である翼口蓋窩(pterygopalatine)動脈を電気焼灼して、外頚動脈を切断し、4−0ナイロン糸(ETHICON, INC, 米国)を外頚動脈を通じて内頚動脈に挿入した後、頚動脈分枝からナイロン糸の16〜18mmが入るように位置させて、中大脳動脈(Middle Cerebral Artery)基始部を閉塞した。
虚血誘導6時間後、動物を断頭して脳を摘出した。摘出された脳は、前極(anterior pole)から2mm間隔で切片して、2%トリフェニルテトラゾリウムクロライド(TTC、Sigma、米国)溶液で37℃で30分間反応した後、4%パラホルムアルデヒド(Sigma、米国)溶液に入れて固定した。染色された組織切片を撮影した後、写真(図9及び12参照)を、MCID image processing system (Imaging Research Inc., カナダ)を利用して、赤色に染色された正常部位と違って、脳梗塞が発生し白色に変わった部位の面積を測定し、全体脳切片の面積に対する脳梗塞面積の比率を求めた後、平均値を計算した(図10)。
A.薬物処理及び局部的虚血−再潅流誘導動物モデルの製作
200〜250gの雄性白鼠にケタミン30〜40mg/kgを筋肉注射して麻酔させた。 頚動脈、外頚動脈、及び内頚動脈を周囲組織と分離させた。まず、外頚動脈の分枝である上部甲状腺動脈と後頭動脈を電気焼灼して、内頚動脈の分枝である翼口蓋窩(pterygopalatine)動脈を電気焼灼して、外頚動脈を切断し、4−0ナイロン糸を外頚動脈を通じて内頚動脈に挿入した後、頚動脈分枝からナイロン糸の16〜18mmが入るようにして位置させた。皮膚切開を復元させた後、麻酔から自然回復させて、1時間後、挿入されたナイロン糸を除去し、再び血液を再潅流させた。
B−1.受動回避テスト(Passive avoidance test)
実験装置としては、自動化されたシャトルボックス(Model PACS-30, Columbus Instruments International Company)を利用した。シャトルボックスは、仕切り戸(3″L x 2.625″W)により同じ大きさ(19″L x 9″W x 10.875″H)の2つの部屋に分かれており、部屋の床は、電流を流し得るように装備されている。ヒンジプレキシガラスリッド(hinged plexiglass lid)に載せておいた20W電球により各ルームを照明できるようにした。白鼠は、仕切り戸を通って暗い部屋に入ることができる。騒音が60dB以下であり、照明を暗くしたルームで実験を行った。白鼠を、最初は明りのある部屋に入れて、仕切り戸を開けると、部屋の中をあちこち見回しながら、暗い部屋に入るようになるが、この時、仕切り戸が自動に閉まって照明が消えるようになる。このような訓練課程を、白鼠が20秒内に暗い部屋に入れるようになるまで行い続ける。訓練課程を終えた24時間後、再び白鼠を照明のある部屋に入れて、白鼠が暗い部屋に入ると、仕切り戸が閉まって、シャトルボックスの床に3秒間電流(1mA)が流れるようにした。局部的虚血−再潅流誘導7日後、再びこの白鼠をシャトルボックスの照明のある部屋に入れた後、暗い部屋に移動するまでの時間を測定した。最大に暗い部屋に入らない時間を5分として行った(図13)。
実験装置として8方放射形迷路(Etho Vision, オランダ)を使用した。床から45cm離れており、8角状の中央(半径34cm)から8個のアーム(長さ60cm及び幅12cm)が延出されており、アームと中央部位は、壁(高さ40cm)からなっている。各アームの末端は、餌コップが設けられている。補償としてヒマワリの種を使用した。迷路の周りを暗くして50Wの光を照らし、ビデオカメラでモニターした。学習訓練の1週間前から餌を80%に減少させて供給した。雄性のラットを迷路に、1日3回、30分間ずつ放置して、三日間適応させた後、一匹ずつ迷路に入れて、餌コップにあるヒマワリの種を食べきれるまでの各アームを訪問する回数と時間を測定して、2分内にエラー数が2回以内になるまで訓練をさせた。一度入ったアームに再び入る回数をエラー数とした。
実験動物において、脳虚血による記憶と学習能力の低下現象が海馬の損傷と密接な関連があるという研究結果がたくさん報告されている(Hodges et al., Neuroscience, 72(4), 959-88, 1996)。したがって、海馬の特定部位(CA1, CA2, DG)において、本発明の効能を測定した。
前記実験例II−2Bの測定を済ませた実験動物をクロラル水化物(chloral hydrate: 400 mg/ml)で麻酔した後、心臓を通じて0.1Mリン酸塩緩衝塩水(PBS, pH 7.4)200mlを潅流させて血管内の血液成分を除去し、固定液(4% paraformaldehyde/PBS)250〜300mlを潅流させた。脳を摘出して同一固定液で15〜24時間4℃で後固定(post-fixation)を行った。PBSで固定液をきれいに洗浄した後、10%、20%及び30%スクロース(sucrose)溶液を順に浸透させて、凍結時に生じる結氷顆粒(ice crystal)を防止した。脳組織を包埋液で包埋して、液体窒素で予め冷却させたイソペンタンに入れて急速冷凍させた後、、ミクロトーム(Cryostat; Reichert Frigocut model 2000)を使用して10μm厚に連続冠状切片を行って、ゼラチンでコーティングされたスライドガラスに直に付着し、1時間室温で乾燥した後、−70℃で保管した。
保管されたスライドガラスを取り出して、蒸留水で洗浄し、ヘマトキシリンで10分間反応した。反応後、スライドガラスを、1%HClを含む70%エタノールで処理し剰余のヘマトキシリンを除去して、アンモニアを処理して固定する。再び組織を、エオシンを利用して約20秒間染色した後、蒸留水で洗浄する。その後、70%、90%及び100%エタノールを利用して脱水して、キシレンで処理した後、カバーガラスとカナディアンバルサム(Canadian balsam)で封入して、光学顕微鏡で観察した。
BG101による海馬におけるH&E染色結果、虚血のみを誘発した実験対照群において、細胞死滅過程で典型的に現れる、細胞核に沈着されたエオシンが固まっている様相(eosinophilic)が海馬の全般的な部分(CA1, CA2, DG)で発現されることが確認された(図15、矢印)。反面、本発明のシルクペプチドを投与した実験群(BG101処置群)では、実験対照群に比べ、正常形態の細胞が有意に増加されていることが観察された。
パーキンソン病の誘発薬物として広く使用されている水酸化ドーパミン(6-hydroxydopamine, 6-OHDA)は、カテコールアミン性神経細胞の細胞膜を通じて吸収されて、選択低にカテコールアミン性神経細胞に毒性を示す。水酸化ドーパミンを片側の脳に注入してドーパミン神経細胞を破壊した動物モデルは、その反対側の脳を対照群として使用することができて、自発的な運動及び薬物により誘発される回転運動を比較することができ、非常に広く使用されている。
水酸化ドーパミンを白鼠の脳の片側線状体(striatum)に注入して、ドーパミン神経細胞の漸進的退行変性を起こすJooらの方法(Joo WS et al., Neuroreport, 9(18), 4123-4126, 1998)の方法を利用して、動物モデルを次のように製作した。
水酸化ドーパミンを利用して製作したパーキンソン病動物モデルにおいて、時間経過による行動学的変化を、病変製作後14日目にアポモルフィン(0.5mg/kg)を後頚に皮下注射して、60分間一側性回転反応を測定した(図16)。本実験は、ドーパミン性神経細胞が死滅され、線状体内のドーパミンの濃度が減少すると、線状体内のドーパミン受容体の過敏性が誘発されて、アポモルフィンはドーパミン受容体に活性剤として作用し、過敏性の誘発された線状体を過度に興奮させ、動物は、損傷側と反対方向に回転運動をするようになる原理を利用したものである(Ungerstedt, Brain Res. 24, 485-493, 1970)。回転運動は、Ungerstedtの前記論文に記載の自動化された回転運動測定器(rotometer)を利用して、各回転数は、[純回転数(net turns)=非病変側回転数(contralateral)−病変側回転数(ipsilateral)]で算出した。
本発明のBG101及びBG201が、パーキンソン病の重要な原因であるドーパミン神経伝達物質の濃度に及ぼす影響を調べるために、前記実験例III−2と同様にBG101またはBG201を投与して病変を製作し、2週間後、線状体でドーパミン(DA)と3,4−ジヒドロキシフェニルアセト酸(3,4-dihydroxyphenylacetic acid: DOPAC)濃度を高性能液体クロマトグラフィ(HPLC, Gilson, フランス)を利用して測定した。
白鼠を頚椎脱臼して犠牲させた後、直に脳組織を摘出して、脳切片器(brain slicer; ZIVIC MILLER、米国)を利用して視神経交叉(optic chiasma)から2mm間隔の切片を取って、組織穿孔器(tissue punch)を利用して黒質を分離した。黒質分離後、残りの前(anterior)部の脳を、冷却されたガラス板上で大脳正中裂に沿って左右大脳半球を分離した後、脳梁(corpus callosum)を境として、視床及び大脳皮質から線状体のみを分離した。これをドライアイスを利用して急速凍結させた後、−70℃で保管した。
凍結された検体に、冷却された0.1M過塩素酸(perchloric acid)と1mMEDTAを加えて、超音波破砕器で組織均等液を製造した後、12,500gで20分間遠心分離し、その上層液を取った。上層液をニトロセルロース膜濾紙(孔大きさ0.2μm)で濾過して、HPLCに注入した。分離条件は、WATERS uBondapakTM C18 3.9×300mmカラム(粒子大きさ10μm)を使用して、移動相は、0.07M リン酸ナトリウムモノベーシック、1mM オクタンスルホン酸ナトリウム、0.1mM EDTA及び8%アセトニトリル(pH4.0)溶液を流速0.7ml/分として使用した。各物質の濃度測定のために、組織均等液の製造時、一定量のジヒドロキシベンジルアミンを添加して、内部標準物質として使用して、分離された物質は、電気化学検出器で検出した。
図17から分かるように、線状体内のドーパミン比率(病変側/非病変側、%)は、シャム対照群に比べ、水酸化ドーパミンのみを投与した対照群で有意に減少した(P<0.05)。一方、本発明のシルクペプチドを投与した実験群では、水酸化ドーパミンのみを処理した対照群に比べ、ドーパミン濃度が有意に増加したことが分かり、投与量が増加するにつれて、その効果も増加することが分かる(P<0.05)。図17において、各測定値は、平均±標準偏差を示したものである。
前記実験例III−2で口腔投与した本発明のBG101またはBG201が脂質過酸化を抑制するかどうかを調べるために、前記実験例III−2と同様にBG101またはBG201を投与して病変を製作した後、2週間後に線状体内のマロンジアルデヒド(MDA)の量を測定した。
上述の方法により線状体を取って、クレブスーリンゲル緩衝溶液(Krebs-Ringer Buffer, NaCl 120mM, KCl 4.8mM, CaCl2 1.3mM, MgSO4 1.2mM, NaHCO3 25mM, Glucose 6mM, pH 7.6)を添加して、超音波破砕器(sonicator)で組織均等液を製造して実験に使用した。
脂質過酸化度合いの尺度であるTBARS(Thiobarbituric acid reactive substances)の濃度測定は、Ohkawaら(Anal Biochem. 95(2):351-8(1979))の方法に従って測定した。一定量の組織均等液に8.1% SDS, 20% acetic acid (pH 2.5), 0.8% Thiobarbituric acid (TBA)を加えて、自己酸化を抑制するために、BHT(2,6-di-t-butyl-p-cresol, 200uM)を加えた後、100℃で30分間反応した。反応後、氷水に容器を浸して、急速に温度を下げ、反応を中断した後、吸光光度計により532nmの波長で試料を測定し、TBARSの濃度を計算した。
線状体内におけるMDA生成比率((病変側/非病変側、%)は、TBARSの濃度に対するMDA−TBA complexの相対的な濃度(%)で表示して、その値は、図18に示されている。図18から分かるように、線状体内の脂質過酸化度合いは、シャム対照群に比べ、水酸化ドーパミンのみを投与した対照群で有意に増加した(P<0.05)。一方、本発明のシルクペプチドを投与した実験群では、対照群に比べ、脂質過酸化度合いが有意に減少したことが分かり、投与量が増加するにつれて、その効果も増加することが分かる(P<0.05)。図18において、各測定値は、平均±標準偏差を示したものである。
本発明のBG101またはBG201がパーキンソン病動物モデルに及ぼす影響を組織学的変化により確認するために、TH免疫組織化学法を行った。前記実験例III−2と同様にBG101またはBG201を投与して病変を製作し、2週間後、黒質切片にチロシンヒドロキシラーゼ(TH)に対する免疫組織化学染色を施して、高倍率で観察し、染色されたTH免疫陽性細胞を計数した。
前記実験例II−2と同様にBG101またはBG201を投与して病変を製作した動物をクロラル水化物(chloral hydrate, 400mg/ml)で麻酔した後、心臓を通じて0.1M PBS(pH7.4)200mlを潅流させて血管内の血液成分を除去し、固定液(4%パラホルムアルデヒド/PBS)250〜300mlを潅流させた。次いで、脳を摘出して、同一固定液で15〜24時間4℃で後固定を行った。その後、PBSで固定液をきれいに洗浄した後、10%、20%及び30%スクロース溶液を順に浸透させて、凍結時に生じる結氷顆粒(ice crystal)を防止した。その後、脳組織を包埋液で包埋して、液体窒素で予め冷却させたイソペンタンに入れて急速冷凍させた後、ミクロトーム(Cryostat; Reichert Frigocut model 2000)を使用して40μm厚に連続冠状切片を行って、30%グリセロール、30%エチレングリコール及び10%リン酸塩緩衝液(PB)が含有された保存液に保管した。
保存液に入っている組織を取り出して、PBSで10分間ずつ3回洗浄して、5%過酸化水素で10分間反応した。次いで、10%ヤギ血清(normal goat serum; NGS)、3%牛血清アルブミン(BSA, Sigma)及び3%Triton X-100で30分間反応した。第1抗体としてラットαTH(希釈倍数1:500, Boehringer Mannheim)を使用し一晩中反応して、翌日バイオチン化(biotinylation)された2次抗体(Vector laboratories、米国)で室温で1時間反応した。30分前に予め1:100で希釈混合したアビジン−バイオチン−ペロキシダーゼ複合体(ABC, Vector laboratories、米国)で室温で1時間反応した。その後、DAB(diaminobenzidine 0.05%, H2O2 0.003%)に入れて5分間発色させて、ゼラチンでコーティングされたスライドに組織を付けて乾燥させた。その後、70%、90%、及び100%エタノールを利用して脱水させて、キシレンで処理した後、カバーガラスとカナディアンバルサム(Canadian balsam)で封入して、光学顕微鏡で観察した。以上の各培養段階で、過剰の試薬はPBSで10分間ずつ3回洗浄して、対照染色のために、第1抗体または第2抗体の代わりに、血清(normal serum)を入れて、他の組織と共に処理した。
BG101またはBG201による線状体及び黒質におけるTH免疫組織化学染色結果と、黒質におけるTH免疫陽性細胞比率(病変側/非病変側、%)の変化は、それぞれ図19と図20に示されている。
アミロイドβ処理により脳損傷を被り、アセチルコリンの量が減少された実験動物のラットにおいて、本発明のBG101またはBG201がアセチルコリン量の減少を抑制することにより、認知機能向上及び退行性脳疾患抑制機能を果たすことができるのかを検査した。
実験動物は、雄性白鼠(Sprague Dawley、140-180g、大韓実験動物センター)であって、一定な環境(室内温度25±1℃、相対湿度60±10%)下で一週間、1つの飼育箱当たり4匹ずつ入れて、水と餌を制限無く摂取するようにして環境に適応させた後、実験に使用した。
アセチルコリンの濃度変化に及ぼす影響を確認するために、IslaelとLesbatsの方法(J Neurochem. 37(6):1475-83(1981))により、化学発光(chemiluminescence)を利用してアセチルコリンの濃度を測定した。前記測定法は、アセチルコリンがアセチルコリンエステラーゼによりコリンに加水分解されて、再びコリン酸化剤(oxidase)によりベスタイン(bestaine)と過酸化水素に変化される反応を利用する方法である。過酸化水素は、化学的にルミノール(5-amino-1,2,3,4-tetrahydro-1,4-phthalazinedione, luminol; Merck, Darmstadt, Germany)とペロキシダーゼ(Sigma, USA)と反応すると、光を発散するため、化学発光を利用して光の量を測定すると、間接的にアセチルコリンの濃度を測定することができる。
実権例V−1:実験動物の準備
実験動物は、雄性白鼠(Sprague Dawley、140-180g、大韓実験動物センター)であって、一定な環境(室内温度25±1℃、相対湿度60±10%)下で一週間、1つの飼育箱当たり4匹ずつ入れて、水と餌を制限無く摂取するようにして環境に適応させた後、実験に使用した。白鼠の中で、一般飼育状態で動きが少なく、発育状況が劣り、強制水泳で非正常的な常同症的行動を示すか、水泳能力が著しく劣る場合は、除外させて、本実験では総50匹を使用した。
本実験では、絶望行動検査(behavioral despair test)とも言われる、標準化された検査法の強制水泳検査(forced swimming test: FST)を利用した。強制水泳検査法は、薬物開発時の抗鬱効果を検査する基本的な実験として知られている。FSTの過程は、最初の提案者であるPorsoltら(Porsolt et al., Eur. J. Pharmacol. 51(3), 291-294, 1978)の方法に従って、次のように行った。
本発明のBG101またはBG201を100mg/mlの溶液に製造して、口腔投与した。全ての試料は、蒸留水に溶解して口腔投与して、1回投与時には、二次強制水泳の1時間前に投与して、7日間繰り返し投与する場合は、一次強制水泳の30分後に投与して、7日間繰り返し投与した。
本発明のBG101またはBG201の1回急性処置時(1g/kg)の抗鬱効果を確認した。陽性対照群としての既存抗鬱剤としては、三環系抗鬱剤の基本とされる薬物のイミプラミン((Sigma、米国)を使用して、投与量は、既存研究結果(Eur. J. Pharmacol. 138(3), 413-416, 1987; Neuropharmacology 28(3), 229-233, 1989)を参照して、1回投与時、20mg/kgとした。実験結果は、図22に示した。
本発明のBG101またはBG201の長期繰り返し処置時の抗鬱効果を確認した。BG101またはBG201を50mg/kgで一日一回、7日間口腔投与して、1回処置時と同様な実験を行った。実験結果は、図23に示されている。
臨床研究対象者は、本発明のシルクペプチドBG−101が100mg内包されたカプセル(以下、BF-7という)を服用する群の67名(BF-7 200mg/day群の33名及び400 mg/day群の34名)と、偽薬を服用する群の32名とに分けて、朝/夕の食後に一回ずつ、1日2回、3週間服用させた。
(1)繰り返し試験
単語を1秒当たり1つずつ聞かせた後、被検者に単語を言わせて、試験は、5回繰り返した。
20分の遅延時間後、被検者に聞かせた単語を言わせた。
遅延想起の実施後、被検者に紙を渡し、聞かせた単語のみに丸を付けるように指示した。
(1)描きテスト
検査の前にCFT図形と反応紙を被検者に渡し、CFT図形を描くように指示した。被検者が描き終えると、CFT図形と反応紙を被検者が見られない位置においた。
(2)即時想起テスト
描きテストの終結後、CFT図形を描くように指示した。
(3)遅延想起テスト
20分の遅延時間後、被検者にCFT図形を描くように指示した。
(4)CFTの評価
各18個の項目を形態と位置を考慮して、CFT採点基準表により、最高2点から最低0点として採点した。各テスト当たり最高点数は、36点であり、最低点数は、0点となる。
1.記憶指数(MQ):記憶力の度合いに対する最も直接的な指標となる記憶指数である。
2.記憶維持度:被検者の記憶がどれくらいよく維持されるかに係る能力を測定する。
3.想起効率性:被検者の記憶がどれくらい正確且つ効率的に活用されるかに係る能力を測定する。
4.描き/記憶一致度:被検者の認知能力の向上が記憶力の増進によるものであるかを測定する。
5.知能/記憶一致度:被検者の認知能力の向上が記憶力の増進によるものであるかを測定する。
服用前と服用後の記憶指数の差異をpaired t-testを使用して検証し、偽薬群とBF−7容量群別(BF-7 200mg及びBF-7 400mg)記憶指数の変化幅の有意な差が存在するかをANOVAを使用して検証した。
(1)BF−7の記憶指数の改善効果
記憶力の度合いに対する最も直接的な指標とされる記憶指数(MQ)の各服用群別の差異値を算出した結果、服用前と服用後の記憶指数の差が、偽薬服用群の場合は、3.1、BF−7 200mg服用群は、11.6、400mg服用群は、20.6であって、各服用群の記憶指数改善値の差異は、どれも有意な差異を示した(図24)。これは、BF−7が容量依存的に有意な記憶指数の改善効果を示すことを意味するものである。また、各服用群の服用前と服用後の記憶指数の値を比較してみると、偽薬服用群は、106から110に、BF−7 200mg服用群は、106.5から118に、BF−7 400mg服用群は、106から126に向上した(図25)。
百分率が高いほど、想起効率性が高いことを意味する。偽薬服用群の場合、服用前と服用後の想起効率性に有意な差がなかった反面、BF−7 200mg服用群の場合は、31%から58.9%に、400mg服用群の場合は、41.5%から66.5%に向上されて、より正確且つ効率的な記憶の活用がなされることを確認した(図26)。
百分率が低いほど、記憶力の低下がその原因であることを意味する。偽薬服用群の場合、服用前の38%から40%に、有意な変化を示さなかった反面(P<0.05)、200mg服用群は、36.8%から56.5%に、400mg服用群は、24.7%から65.2%に有意に増加されて、認知能力の向上が、描きの能力によるものではなく、記憶力の向上によるものであることが分かった(図27)。
百分率が高いほど、ほぼ等しいIQを有する同一年齢代より記憶力がさらに高いことを示す。図28から分かるように、偽薬服用群の場合、55.5%から63.4%に、BF−7 200mg服用群の場合は、52.9%から78.9%に、有意な増加を示した(P<0.05)。BF−7 400mg服用群の場合、52.5%から91.1%に、知能記憶一致度が非常に向上されたことが分かる(P<0.05)。
百分率が低いほど、時間が経つにつれて忘れる量が多いことを意味する。BF−7 400mg服用群の場合、53.2%から61.3%に、記憶維持度指数が増加したが、統計的に、偽薬服用群とBF−7服用群では、有意な変化を示さなかった(P>0.05)(図29)。
Claims (16)
- 有効成分として、シルク蛋白質から分解された200〜100,000の重量平均分子量を有するシルクペプチドを含み、脳疾患の予防または治療効能を奏して、前記脳疾患は、パーキンソン病、憂鬱症、または虚血性脳卒中であることを特徴とする、脳疾患の改善のための組成物。
- 有効成分として、シルク蛋白質から分解された200〜100,000の重量平均分子量を有するシルクペプチドを含み、パーキンソン病または虚血性脳卒中の脳疾患により損傷した頭脳機能または正常人の頭脳機能を改善するための組成物であって、前記頭脳機能は、記憶力及び学習能力であることを特徴とする、頭脳機能改善のための組成物。
- 前記シルク蛋白質は、シルクフィブロインであることを特徴とする、請求項1または2に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜15,000の重量平均分子量を有して、カルシウム塩溶液による加水分解、酸加水分解、蛋白質分解酵素による加水分解、または前記加水分解方法の組み合せにより製造されることを特徴とする、請求項1または2に記載の脳疾患または頭脳機能改善のための組成物。
- 前記蛋白質分解酵素は、トリプシン、ペプシン、アルカラーゼ、サーモアーゼ(thermoase)、フレーバーザイム、スミザイム(sumizyme)、プロタメックス(protamex)、プロチン(protin)、及びこれらの混合物から構成された群から選択されることを特徴とする、請求項4に記載の脳疾患または頭脳機能改善のための組成物。
- 前記蛋白質分解酵素は、サーモアーゼ(thermoase)、フレーバーザイム、スミザイム(sumizyme)、及びこれらの混合物から構成された群から選択されることを特徴とする、請求項5に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜4,000の重量平均分子量を有して、(i)カルシウム塩溶液による加水分解段階、及び(ii)サーモアーゼ(thermoase)、フレーバーザイム、スミザイム(sumizyme)、及びこれらの混合物から構成された群から選択される蛋白質分解酵素を利用した加水分解段階を順に行って製造されることを特徴とする、請求項4に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜2,000の重量平均分子量を有して、(i)カルシウム塩溶液による加水分解段階、及び(ii)フレーバーザイム及びスミザイム(sumizyme)の混合物を利用した加水分解段階を順に行って製造されることを特徴とする、請求項7に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜1,200の重量平均分子量を有することを特徴とする、請求項8に記載の脳疾患または頭脳機能改善のための組成物。
- 前記カルシウム塩は、塩化カルシウムであることを特徴とする、請求項8に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、酸加水分解により製造されて、200〜100,000の重量平均分子量を有することを特徴とする、請求項1または2に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜3,000の重量平均分子量を有することを特徴とする、請求項11に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、200〜1,500の重量平均分子量を有することを特徴とする、請求項12に記載の脳疾患または頭脳機能改善のための組成物。
- 前記シルクペプチドは、神経保護活性を有することを特徴とする、請求項1または2に記載の脳疾患または頭脳機能改善のための組成物。
- 前記神経保護活性は、神経細胞の死滅を抑えてなることを特徴とする、請求項14に記載の脳疾患または頭脳機能改善のための組成物。
- 前記組成物は、薬剤学的組成物であることを特徴とする、請求項1または2に記載の脳疾患または頭脳機能改善のための組成物。
