Description
格子に基づくセンサーは、100nm未満の精度で被処理物の正確な沈着と腐食を可能にする半導体製造装置の最近の発達によって開発された新規な光学的装置群である。

米国特許第6707561号明細書には、当該分野においてはしばしば「エバネッセント共鳴(ER;evanescent resonance)」と呼ばれている格子に基づくバイオセンサー技術が記載されている。この技術においては、サブミクロンスケール(sub-micron scale)の格子構造体を使用して蛍光信号を増幅させた後、格子表面上に結合した分子であって蛍光性標識を保有する1つの分子へ結合させる。ER技術は、非増幅アッセイに比べて著しく低い濃度の被検体の結合検出を可能にする蛍光体に基づくアッセイの感度を増大させる。
下記の実施態様とこれらの特徴は、範囲を制限しない例示的なシステム、装置及び方法によって例証的に説明するものである。種々の実施態様においては、前述の1又は複数の問題点を軽減又は解消し、また、その他の実施態様は別の改良に向けられたものである。
i)バイオセンサーからER検出データを得るために適合した第1光源、
ii)標識不使用検出データを得るために適合した第2光源、
iii)第1光源からの光と第2光源からの光を合成することによって、バイオセンサーを照射するための照射ビームを発生させる光学系、
iv)バイオセンサーからの反射光を検出するための少なくとも1つの検出器、及び
v)少なくとも1つの検出器からのデータを使用することによって試料からERデータと標識不使用データを得るための分析モジュール。
1)2つの技術の結合により、混合試料の母集団(population)中に存在する蛍光体で標識化された物質の百分率が識別される。標識の存在しないシグナルは、センサーに結合したマス全体の定量的な尺度を提供する。一方、ERシグナルは、標識の存在を定量化する。
2)これらの技術の結合は、異種源からの2つの結合シグナルを提供することによって、細胞、タンパク質及び小さな分子間における相互作用の測定における統計的厳密性も高める。
4)2つの技術の結合により、付加的な情報を提供することができる。標識のないシグナルは細胞の付着を定量化することができ、発光シグナルは、細胞に結合した蛍光体標識化リガンドの量を定量化することができる。もちろん、上述のような標識のない生物学的物質及び標識化した生物学的物質が本発明によるバイオセンサーにおいて検出される場合には、その他の方法も可能である。
5)2つの技術の結合により、結合した全マスと分子数を識別することによって分子量の測定をおこなってもよい。
6)さらに、結合されたバイオセンサーは、抑制結合の研究のような他の方法を実施するための相互に独立した2種の定量試験を可能とする。さらに、抑制剤リガンドの結合性の直接的定量化能を含むタンパク質と支持体との間の抑制結合相互作用に関するより完全な理解と特徴付けが可能となる。付加的な例としては、バイオセンサーが非常に強い結合性相互作用に関する研究であって、弱い結合親和性を有する既知の競合性抑制剤を使用することによって、より強い結合性物質の摂動と観察をおこなう該研究を促進することが挙げられる。
図2は、従来技術によるバイオセンサー/検出システムであって、該バイオセンサーを照明すると共に、該バイオセンサーからの反射光のピーク波長のシフトを測定するための該システムの模式図である。
図3は、底のないマイクロタイタープレートの基底部へ固定された図1に示す構造を有するバイオセンサー装置のウェルの全列を読み取る8個の照明ヘッドを具備する装置の模式的構成図である。

図4は、格子に基づくセンサーのER検出と標識不使用検出に対する商業的な要求に適合することが期待される格子構造100を有する一次元的センサーの第1の実施態様を示す模式的断面図である。図4は、一次元又は一方向に延びる格子構造100の1つの周期を示す。図4においては、寸法の尺度は一定ではない。図4に示す格子構造100は積層状態で透明な材質、例えば、ポリエチレンテレフタレート(PET)若しくはその他のプラスチック、ガラス又はその他の材質から成る基材シート(図示せず)へ接合される。
図5は第2の実施態様の模式的断面図である。この図においては、一次元の格子構造の1つの周期並びにUV硬化層102、高屈折率層104及び試料媒体106の構造を示す。この図には寸法と遷移点も示すが、寸法の目盛りは示さない。図5に示すデザインは、図4に示すデザインに比べて、いくつかの点で相違する。
a)格子の周期がより短い。
b)格子の溝又は窪みがより狭い。図5における装荷率(duty cycle)(単位セル中の上部面における格子の百分率)は88%である(0〜0.85及び0.97〜1.0)。装荷率が70%〜95%の狭い溝は、狭い溝の実施態様の例示である。一般的には、狭い溝は良好な標識不存在下での検出結果をもたらす。狭い溝の特徴は、TE共鳴ピークの幅を狭めることであり、このことは場の強度を高めることを示す。ER効果の実用的な用途に対しては十分に広い共鳴が必要であるが、過度に広い共鳴は、有用な蛍光信号の増幅をもたらすには不十分な場の強度を示す。
c)半周期に対する格子深さの比は1:1である。
ER方式と標識不使用方式の両方に対して適した二次元的(2−D)格子構造の可能性も考慮することができ、このような構造は好ましいものである。二次元的格子は、二次元において高い領域と低い領域が交互に配列されたチェス盤、ワッフル(ホール)又はワッフル焼き型(ポスト)のような形態を有する。二次元的格子は、X方向とY方向において異なる周期を有する。このような特徴を有する格子はZ方向において種々の側面(例えば、段状側面又は湾曲状側面等)を有する。ワッフル状パターンの場合、窪み又はウェルは正方形の形態よりも長方形の形態を有していてもよい。これによって付与される可撓性に基づいて、標識不使用検出方式及びER検出方式に対する共鳴位置が異なる波長において発生するように調整することができる。また、この可撓性は、ER共鳴を異なる励起波長に対して発生するように調整することができると共に、現存する標識不使用検出の計測系との適合性を維持することができるという重要な利点をもたらす。例えば、X方向の周期性は、CY3蛍光体(緑色光)又はCY5蛍光体(赤色光)を励起するように調整される波長を有する垂直入射角又はこれに近い入射角における共鳴をもたらし、Y方向の周期性は、820nm〜850nmの範囲(近赤外領域)に固定される共鳴をもたらす。
複合バイオセンサーの2−D「ホール(hole)」の実施態様の特定の実施例を図15〜図19に基づいて説明する。このセンサーは、単一の装置を用いるER方式と標識不使用方式(BIND)の両方による検出に対して最適化されるように二次元的に設計されたものである。
ポスト(post)によって特徴付けられる反復単位セルを用いる二次元的格子構造について、図20〜図24に基づいて説明する。図20A及び図20Bは、センサーの表面上に形成された周期的なポスト220によって特徴付けられる二次元的格子デザインを有する単位セルの斜視図及び断面図をそれぞれ示す。各々の単位セルは1つのポスト220を有する。ポスト220は、基材シート(図示せず)上に塗布された支持体材料103(例えば、UV硬化ポリマー)内に隆起した突起である。これらの図に示すように、高い屈折率を有する被覆層(例えば、TiO2)が該突起と支持体上に形成される。図示される構造体は、X方向に偏光された光を用いる水環境下でのBIND(標識不使用)検出方式、及びY方向に偏光された光を用いる空気環境下でのER検出方式に対して最適化されている。
図28A〜図28Cは、ERと標識不使用(BIND)の組合せ検出用に設計されて製造されたバイオセンサー用の単位セル500の別の実施態様を示す3つの斜視図である。この構造のいくつかの特徴を理解するためには、エバネッセント共鳴(ER)センサーと標識不使用(BIND)センサーに適合したデザインの観点について概括することが有用である。この種のセンサーは3つの基本的なデザインの観点、即ち、共鳴波長、共鳴幅及び格子の深さの観点から相違する。
ERセンサーは、励起波長の数(約+/−2)nm内で共鳴を発生することが好ましい。励起光が一般的にレーザーから発生されてその帯域が非常に狭い場合には、この要件は、ER共鳴の波長位置に関して高い特異性となる。操作のBIND方式にはこの制限がないので、別の波長(例えば、外部の周辺光の波長の範囲)で共鳴するという利点があり、また、BIND信号をER励起源からスペクトル的に分離させることによって潜在的な検出の重なりによる矛盾を回避することができる。
ERセンサーは、可変因子(例えば、生物学的被覆物の厚さ及び照射の開口数)の存在下で励起波長に重なるのに十分な共鳴幅を有していなければならない。実際上は、ER共鳴は、約5nm未満、より好ましくは10〜15nm未満の半値全幅(FWHM)を有しているべきではない。一方、ピークの幅が狭くなるとピーク位置の不確定性が低減するので、BINDの感度は約1/sqrt(FWHM)増大する。
BINDセンサーは、格子上により多くの生物学的物質が付着すると、より大きな共鳴波長シフトを示す。より深い格子は、生物学的物質の結合に対して、より大きな表面積を提供する。ERの格子深さが深くなると、ER効果は必ずしも改善されず、低減するかもしれない。
以下に説明する抑制剤結合シナリオについて検討した。次式に示すように、このシナリオにおいては、被検出物(例えば、タンパク質)の一部を格子の支持体(標識不存在)上へ直接結合させ、タンパク質の別の一部を、蛍光性標識を有する抑制剤へ結合させる。式中、Ks及びKiは支持体及び抑制剤に対する平衡結合定数をそれぞれ示す。

別の実施態様として、ERと標識不使用方式を組み合わせたバイオセンサーは、生物学的分子の有する天然の蛍光を利用するアッセイ(即ち、結合された蛍光性標識の使用を必要としないアッセイ)であって、他の生物学的分子や低分子量の被験分子との相互作用に際してのこれらの折り畳み、積み重ね及び変化と変化速度に関する生物物理学的特性決定測定をおこなうアッセイに対して特に有用である。このような特性決定測定は結合した蛍光性標識を用いておこなうことはできるが、この種の結合標識は必ずしも必要ではなく、特に固有の蛍光特性を有する生物学的分子に対しては不要である。
本明細書に開示されるERとBINDを組み合わせたセンサーを結合蛍光体からの蛍光又は天然蛍光を検出するために使用する場合、バイオセンサーの構成物質の内部から放出される背景蛍光を低減させることは有用であり、これによって、信号対雑音比のより高い条件下での蛍光測定が可能となる。これを達成するための1つの方法は、付加的な低蛍光性SiO2層をUV−硬化ポリマー層上へ沈着させた後、SiO2層上にさらに屈折率の高い物質(例えば、TiO2)から成る最上層を沈着させる。この種のバイオセンサーの断面図を図30に示す。このバイオセンサーは、UV−硬化ポリマー層524(該層は図示されていない支持体シート上に結合されている)、中間のSiO2層700及び上部のTiO2層522を具備する。空気又は水に基づく媒体中の試料はTiO2層上に存在させる。付加的なSiO2層の厚さは、格子の装荷率や要求される背景蛍光レベル等の要因によって左右されるが、一般的には、500〜5000Åである。
本発明によるER/BINDセンサーの新規な用途の1つの別の実施例は、時間分解蛍光(TRF)と蛍光偏光(FP)を測定するための該センサーの使用である。TRFとFPは、本明細書に開示された標識不使用装置とERを組み合わせた装置から誘導だれる増強信号から大きな利点をもたらす2つの方法である。これらの2つの方法は、背景信号及び結合事象に関与しない分子からの結合事象に関する特定のER信号の分離に対して有用である。蛍光体は、センサーの波長増強能に適合させる必要がある。
本発明によるバイオセンサーの1つの可能な用途においては、ERとBINDセンサーを使用することによって、多数の位置における試料の分析がおこなわれる。この場合、各々の位置は、直径が約10〜500ミクロンのスポット(spot)のマイクロアレイ(microarray)を規定する。
ER法と標識不使用法を組み合わせたバイオセンサーに関する上記の説明を参酌しながら次の実施態様について説明する。即ち、センサーに問い合わせをした後で、検出器上の単一の結合サイトから標識不使用データとERデータの両方を獲得するために有用な読取り/検出システムのいくつかの実施態様について説明する。
図25に示す実施態様と図26に示す実施態様においては次の点に留意すべきである。即ち、前者においては、BIND測定用光源350とER測定用光源340が別々に配設されており、後者においては、BIND測定用光源402とER測定用光源406が別々に配設されている。1つの可能な変形態様においては、BIND測定とER測定の両方に対して単一光源を使用してもよい。このような光源としてはいくつかの光源が使用可能であり、例えば、波長可変レーザー、又は広帯域スペクトルを示す高強度閃光電灯等が挙げられる。光源からの出力は所望により平行化し、ビーム拡大器を用いて拡大し(光源として波長可変レーザーを使用する場合)、モノクロメーター又はフィルターを通過させ(閃光電灯を使用する場合)、次いでバイオセンサーの表面上へ誘導される。ER信号とBIND信号を検出するために使用される光学装置(optics)は図25〜図27に示す態様及び前述の態様の装置であってもよい。

Claims (10)
- センサー上に載置される試料を検査するためのセンサーであって、下記の光照射1)及び2)の両方に対して対応できるように、周期と深さを含む格子パラメーターを有する構造に設計された周期的な表面格子構造を有する支持体を具備し、
(i)該格子構造が二次元的な周期的格子構造を含み、該周期的格子構造が第1次元と第2次元において周期的であり、これらの次元が相互に直交し、
(ii)該周期的格子構造の第1次元が標識不使用検出用に設計される格子構造を含み、該周期的格子構造の第2次元がER検出用に設計される格子構造を含み、第1次元における格子構造の深さが、第2次元における格子構造の深さよりも実質上大きい深さである該センサー:
1)エバネッセント共鳴(ER)検出方式において、該センサーから応答を得るための該センサーへの光照射、及び
2)標識不使用検出方式において、該センサーから応答を得るための該センサーへの光照射。 - 格子構造が積層体をさらに具備し、該積層体が支持体、該支持体に結合される格子構造を有する層、格子構造を有する該層上に沈着されるSiO2層、及び該SiO2層上に沈着される比較的高い屈折率を有する屈折物質の層を具有する請求項1記載のセンサー。
- 第1次元における格子構造が、260nm〜1500nmの周期及び100nm〜3000nmの格子深さを有し、第2次元における格子構造が、200nm〜1000nmの周期及び10nm〜300nmの格子深さを有する請求項1記載のセンサー。
- SiO2層が、500Å〜5000Åの厚さを有する層を含む請求項2記載のセンサー。
- 高屈折率を有する屈折物質層に結合される発光性物質をさらに含む請求項2記載のセンサー。
- 格子構造が、2段状の2次元的形態を有する単位セルのアレイを含む請求項1記載のセンンサー。
- 格子構造が単位セルのアレイを含み、各々の単位セルが、標識不使用検出方式用に設計される構造で第1次元方向へ延びる交互の高い領域と低い領域を有する第1の周期的格子構造、及びER検出方式用に設計される構造で該第1次元方向に対して直交する第2次元方向へ延びて第1の周期的格子構造上に重ね合わされる交互の高い領域と低い領域を有する第2の周期的格子構造を含む2段状の2次元形態を有する請求項1記載のセンサー。
- 直交する第1次元と第2次元内の格子構造が第1次元と第2次元において異なる周期性を有し、第1次元における周期性が、試料と結合する蛍光体を励起させるように同調される波長を有して第1光源から直角又はほぼ直角に入射する入射光に対して広域共鳴をもたらすように調整され、第2次元における周期性が、スペクトルの近赤外領域の第2光源からの光照射に対して鋭い共鳴をもたらすように調整される請求項1記載のセンサー。
- 下記の過程(i)〜(iii)を含む少なくとも1種の試料の分析方法:
(i)1)エバネッセント共鳴(ER)検出方式において、バイオセンサーから応答を得るための該バイオセンサーへの光照射及び2)標識不使用検出方式において、バイオセンサーから応答を得るための該バイオセンサーへの光照射の両方に対して対応できるように、周期と深さを含む格子パラメーターを有する構造に設計された周期的な表面格子構造を有する支持体を具備するセンサーであって、該格子構造が2次元的格子構造を含み、周期的な格子構造の第1次元が標識不使用検出用に設計される格子構造を含み、周期的格子構造の第2次元がER検出用に設計される格子構造を含む該センサー上に少なくとも1種の試料を載置させ、
(ii)ER検出方式用に設計される少なくとも1つの光源からの光を用いて読取検出器内の試料を照射すると共に、標識不使用検出方式用に設計される少なくとも1つの光源を用いてセンサーを照射し、次いで
(iii)センサーからの反射光を分析する。 - 第1次元における格子構造が、260nm〜1500nmの周期及び100nm〜3000nmの格子深さを有し、第2次元における格子構造が、200nm〜1000nmの周期及び10nm〜300nmの格子深さを有する請求項9記載の方法。
