Description
本発明は、癌(例えば、前立腺癌)の診断に関する。特に、本発明は、前立腺腫瘍においてアップレギュレートされた発現を示すヒト内因性レトロウイルス(HERV)の亜群、およびこれらのウイルスによって発現されるスプライシングされたmRNAによってコードされるポリペプチドに関する。
参考文献1および2は、HERV−KファミリーのHML−2亜群のヒト内因性レトロウイルス(HERV)が、前立腺腫瘍においてアップレギュレートされた発現を示すことを開示している。参考文献1および2の内容は、本明細書中で参考として援用される。
HERVは、長年にわたって知られており、そしてHERV−Kファミリーゲノム配列は、1986年以降公知である{参考文献187}。通常のgag、prt、polおよびenvのレトロウイルスタンパク質が、cORFまたはRecとして公知のHIV RevまたはHTLV Rexのアナログを有するとして、HERV−Kについて同定されているが{3}、他の調節タンパク質(例えば、HIV Tatタンパク質またはHTLV Taxタンパク質)のアナログは同定されていない。

N1およびN4は存在するが、N2、N3、N5およびポリAは、必要に応じてである。
診断がmRNA検出に基づく場合、本発明の方法は、好ましくは、以下の初期工程を包含する:RNA(例えば、mRNA)を患者サンプルから抽出する工程;(b)mRNAを除去することなく、患者サンプルからDNAを除去する工程;および/または(c)配列番号4を含むRNAではなく、配列番号4を含むDNAを、患者サンプルから除去もしくは破壊する工程。これは必要である。なぜなら、正常な細胞および癌細胞の両方のゲノムは、複数のPCAV DNAテンプレートを含み、一方、PCA−mRNAレベルの増加は、癌細胞においてのみで見出されるからである。代替として、相同なDNAの存在により影響を受けないRNA特異的アッセイが使用され得る。
サンプル中の特定のRNA配列の存在または非存在を検出するための種々の技術が利用可能である(例えば、参考文献8および17)。サンプルが、ゲノムPCAV DNAを含む場合、検出技術は、一般に、RNA特異的であり;サンプルが、PCAV DNAを含まない場合、検出技術は、RNA特異的であってもなくてもよい。
RNAを直接検出するよりむしろ、RNAに由来する分子を検出すること(すなわち、RNAの間接検出)が好ましくあり得る。mRNAを検出する代表的な間接方法は、逆転写によってcDNAを調製することであり、次いでcDNAを直接検出することである。cDNAの直接検出は、一般的にRNAを直接するための、上述された技術と同じ技術を使用する(が、RT−PCRのような方法は、DNA検出については適切ではないこと、およびcDNAが二本鎖であり、そのため検出技術が、配列、その相補体、または二本鎖分子に基づき得ることが理解される)。
本発明は、例えば、PCAV核酸の検出において使用するためのポリヌクレオチド物質を提供する。
ハイブリダイゼーション反応は、異なる「ストリンジェンシー」の条件下で実行され得る。これは、当該分野において広く公知であり、かつ公開されているハイブリダイゼーション反応のストリンジェンシーを増大する条件である(例えば、参考文献17の7.52頁を参照のこと)。関連する条件の例としては、(ストリンジェンシーを増大する順で):25°C、37°C、50°C、55°Cおよび68°Cのインキュベーション温度;10×SSC、6×SSC、1×SSC、0.1×SSCの緩衝液濃縮物(ここで、SSCは、0.15M NaClおよび15mMクエン酸緩衝液である)および他の緩衝系を使用する等価なもの;0%、25%、50%、および75%のホルムアミド濃縮物;5分間〜24時間のインキュベーション時間;1回、2回、またはそれ以上の洗浄工程;1分間、2分間、または15分間の洗浄インキュベーション時間;ならびに6×SSC、1×SSC、0.1×SSC、または脱イオン水の洗浄溶液が挙げられる。ハイブリダイゼーション技術は、当該分野において周知である(例えば、参考文献8、17、18、19、20などを参照のこと)。当業者によって理解されるように、特定のポリヌクレオチド配列およびこのポリヌクレオチド配列によってコードされる特定のドメインに依存して、本発明のポリヌクレオチドと既知のポリヌクレオチドとを比較する際のハイブリダイゼーション条件が異なり得る。
本発明は、PCAV核酸内に含まれるテンプレート配列を増幅するためのプライマー(例えば、PCRプライマー)を含むキット、第1プライマーおよび第2プライマーを含むキットを提供し、ここで第1プライマーは、該テンプレート配列に対して実質的に相補的であり、そして第2プライマーは、該テンプレート配列の相補体に対して実質的に相補的であり、ここで、実質的に相補性を有する該プライマーの部分が、増幅されるべきテンプレート配列の末端を規定する。第1プライマーおよび/または第2プライマーは、検出可能な標識を含み得る。
本方法がポリペプチドの検出に基づく場合、本方法は、スプライシング事象(envコード領域の5’領域および開始コドンが、ゲノムにおけるenvのコード領域に対して、リーディングフレーム+2において下流コード領域に結合される)によって産生される転写物によってコードされるHML−2ポリペプチドの発現を検出する工程を包含する。このポリペプチドは、ウイルスの生活環において機能的であっても、機能的でなくでもよい。
サンプル中の特定のポリペプチドの存在または非存在を検出するために、種々の技術が利用される。これらは、一般的に、抗体とポリペプチドにおける抗原性アミノ酸配列との間の特定の相互作用に基づく免疫アッセイ技術である。適切な技術としては、標準的な免疫組織学的方法、免疫沈降、ELISA、RIA、FIA、免疫蛍光検査法などが挙げられる。
ポリペプチドを直接的に検出するより、ポリペプチドに応答して生体によって産生される分子を検出することが好ましくはあり得る(すなわち、ポリペプチドの間接検出)。これは、代表的には抗体の検出に関し、そのため患者サンプルは概して血液サンプルである。抗体は、従来の免疫アッセイ技術によって(例えば、本発明のPCAVポリペプチドを使用して)検出され得、これは、代表的には固定される。

HML−2転写物は、腫瘍においてアップレギュレートされる。このようなアップレギュレーションを検出するために、基準点(すなわち、コントロール)が必要とされる。コントロールサンプルの分析は、患者のサンプルが比較され得る標準レベルのRNAおよび/またはタンパク質発現を与える。

前立腺癌細胞株のノーザンブロットは、それらがいくつかのサイズ(全長ウイルスゲノム配列およびサブゲノムのスプライシングされた転写物の両方に対応する)のPCAV転写物の発現することを示す(図14)。そのような転写物の発現はまた、図14のレーン1において示されるように、奇形癌細胞株においても観察される(4)。さらに、PCAVのスプライシングパターンを特徴付けるために、取り込まれたプロウイルス配列の隣接するLTR間の任意のスプライス事象を検出し得るRT−PCRストラテジーを使用した。統合PCAVゲノムの一般的な特徴に対して、このアプローチにおいて使用された順方向プライマー(配列番号15)および逆方向プライマー(配列番号16および17)のおよその位置は、図15中の矢印によって示される。
レンチウイルスの明確な特徴は、それらがウイルスLTRプロモーターからの転写を活性化し得るポリペプチドをコードすることである。HIVのtatポリペプチドは、これらのアクチベーターの最も理解される例である。tat遺伝子は、HIVにおけるrev遺伝子およびenv遺伝子と物理的に重複し、そしてenv領域にわたるHIVのmRNAの変更スプライシングを介して作られる。Tatポリペプチドは、TARと呼ばれる特定部位でHIVのmRNAの5’末端に結合し、そしてHIVに特定の活性化を提供する。
PCAVのenv領域内の最終エキソン内において、リーディングフレーム1およびリーディングフレーム2は、それぞれenvおよびcORFをコードする(図4および5)。配列番号11、配列番号28、配列番号29および配列番号31は、PCAP2であり、それは、同一の5’領域および開始コドンをenvとして共有するが、1型ウイルスで発見された欠失は、下流3’スプライス部位に連結する5’スプライス部位を誘導する(図4)。
PCAP2の細胞成分の局在を試験するために(その役割をより理解するために)、C末端V5タグ(配列番号60)を用いてPCAP2を発現するアデノウイルスを、始原前立腺上皮細胞に感染に使用した。このタンパク質は、高度に発現しなかったが、抗V5を用いて、より広汎的に、全ての細胞にわたる核小体内を明らかにした(図30)。この細胞内に位置する小タンパク質の濃度は、核内にあるものと特に相互作用することを示す。
PWPE1細胞を、ヒトパピローマウイルス18(203)を用いて正常前立腺内皮細胞を不朽化することによって、作製した。この細胞は,ヌードマウスにおいては非腫瘍形成性であり、そして、正常前立腺内皮細胞のマーカーおよび増殖特性を有する。
PCAP2発現は、正常非形質転換細胞には関連していないが、各種の腫瘍組織および形質転換細胞株に関連することが発見されている(204)。特に、発現は、乳癌細胞株および乳癌患者組織において認められている。

Claims (15)
- 前立腺腫瘍細胞を検出する方法であって、該方法は、患者からのサンプルにおけるHML−2発現産物の存在または非存在を検出する工程を包含し、ここで、該発現産物は、スプライシング事象によって生成され、ここで、HML−2 envコード領域の5’領域の一部およびenvコード領域の開始コドンが、envのリーディングフレームに対して+2のリーディングフレームにおける下流コード領域に連結され、ここで、非腫瘍細胞に比較してより高いレベルの該HML−2発現産物は、前立腺腫瘍細胞の指標である、方法。
- 請求項1に記載の方法であって、前記サンプルが、前立腺組織サンプル、胸部組織サンプル、または血液サンプルである、方法。
- 請求項1または2に記載の方法であって、前記発現産物が、mRNA転写物またはポリペプチドである、方法。
- 請求項3に記載の方法であって、前記mRNA転写物が、配列番号19、20、21、24、25、26、38、40および/または42のうちの1つ以上に対して少なくとも90%の配列同一性を有する配列を含む、方法。
- 請求項3に記載の方法であって、前記mRNA転写物が、配列番号19、20、21、24、25、26、38、40および/または42のうちの1つ以上に対して少なくとも95%の配列同一性を有する配列を含む、方法。
- 請求項3〜5のいずれか1項に記載の方法であって、前記mRNA転写物が、配列番号7、8、9、10、11、12、28、29、30、31、34、35、36、39、41、43および69のうちの1つ以上に対して少なくとも90%の配列同一性を有するポリペプチドをコードする、方法。
- 請求項3〜5のいずれか1項に記載の方法であって、前記mRNA転写物が、配列番号7、8、9、10、11、12、28、29、30、31、34、35、36、39、41、43および69のうちの1つ以上に対して少なくとも95%の配列同一性を有するポリペプチドをコードする、方法。
- 請求項1〜7のいずれか1項に記載の方法であって、該方法が、前記サンプルからRNAを抽出する工程を包含する、方法。
- 請求項1〜8のいずれか1項に記載の方法であって、前記発現産物が、ハイブリダイゼーションによって検出される、方法。
- 単離されたポリヌクレオチドであって、以下:(a)配列番号19、20、21、24、25、26、38、40、51、52および/または42のヌクレオチド配列;(b)(a)に対して少なくとも95%の同一性を有するヌクレオチド配列;または(c)(a)、もしくは(b)の相補体、を含み、ただし、該ポリヌクレオチドは、HREV−Kの核酸配列からなるものではない、単離されたポリヌクレオチド。
- スプライシング事象により生成される転写物によりコードされる単離されたポリペプチドであって、ここで、HML−2 envコード領域の5’領域の一部およびHERV−K envコード領域の開始コドンが、HERV−Kゲノムにおけるenvのリーディングフレームに対して+2のリーディングフレームにおける下流コード領域に連結される、ポリペプチドであって、以下:配列番号7、8、9、10、11、12、28、29、30、31、34、35、36、39、41、43、66、67および68からなる群から選択されるアミノ酸配列;または(b)(a)に対して少なくとも90%の同一性を有するポリペプチド配列、を含み、ただし、該ポリペプチドは、cORFのアミノ酸配列からなるものではない、ポリペプチド。
