Description
本発明は、無機材料と結合することができる有機材料に関し、特に、半導体材料に結合し秩序構造を形成することができるバクテリオファージに関する。
本願において実施された研究は、米国科学財団(National Science Foundation)からのグラントにより一部支援されており、政府は一定の権利を所有し得る。
本発明者らは、構築物を設計し、調節された精巧な構造への無機材料の集合を指図し調節する生物学的材料を作製した。興味深い電気的又は光学的な特性を有する材料を作出し設計するための生物学的材料の使用は、フィーチャーのサイズを減少させ、例えば材料の光電気特性の調節を改良するために使用され得る。半導体材料は、典型的には、硫化亜鉛、砒化ガリウム、リン酸インジウム、硫化カドミウム、砒化アルミニウム、アルミニウム・アンチモン(alminum stibinide)、及びシリコンから作製される。これらの半導体材料は、しばしば、II-V族半導体材料及びII-VI族半導体材料へと分類される。
本発明の様々な態様の作製及び使用が、以下に詳細に記述されるが、本発明は極めて多様な特定の情況で具体化され得る多くの適用可能な発明概念を提供することを認識されたい。本明細書に記述された特定の態様は、本発明を作製し使用するための特定の様式を例示するものに過ぎず、本発明の範囲を画定するものではない。
ペプチドの作出、単離、選択、及び特徴決定
ペプチド選択
表面上のファージ-ファージ相互作用を低下させるため、0.1%TWEEN-20を含有しているトリス緩衝生理食塩水(TBS)の中で、ファージ・ディスプレイ又はペプチド・ライブラリーを半導体又は他の結晶と接触させた。室温で1時間の振とうの後、トリス緩衝生理食塩水(pH 7.5)及び0.1%から0.5%(v/v)へと増加するTWEEN-20濃度に10回曝すことにより、表面を洗浄した。グリシン-HCl(pH 2.2)の添加により10分間、表面からファージを溶出させ、新しいチューブへ移し、次いでトリス-HCl(pH9.1)で中和した。溶出したファージを力価測定し、結合効率を比較した。
基板の配向をX線回折により確認し、適切な化学的特異的エッチングにより自然酸化膜を除去した。エッチング時間1分間及び10分間で、以下のエッチング剤を、GaAs及びInPの表面上で試験した:NH40H:H20 1:10、HCl:H20 1:10、H3PO4:H202:H20 3:1:50。GaAs及びInPのエッチングされた表面のための最良の元素比及び最小の酸化膜形成(XPSを使用)は、HCl:H2Oを1分間使用した後、脱イオン水で1分間濯ぐことにより達成された。しかしながら、ライブラリーの一次スクリーニングにおいてGaAsのために水酸化アンモニウム・エッチング剤を使用したため、その他の全てのGaAs基板例にこのエッチング剤を使用した。Si(100)ウェハーは、HF:H20 1:40の溶液中で1分間エッチングし、その後、脱イオン水で濯いだ。全ての表面を濯ぎ溶液から直接採取し、直ちにファージ・ライブラリーへ導入した。AFM及びXPSにより、ファージに曝されていない対照基板の表面を、エッチング過程の有効性及び表面の形態学に関して特徴決定し、マッピングした。
XPS、SEM、及びAFMの実施例に関しては、基板を、トリス緩衝生理食塩水中で1時間ファージに曝し、次いでfdファージのpIIIタンパク質に対する抗体である抗fdバクテリオファージ-ビオチン・コンジュゲート(リン酸緩衝液中1:500、Sigma)へ30分間導入し、次いでリン酸緩衝液で濯いだ。ストレプトアビジン/20nmコロイド金標識(リン酸緩衝生理食塩水(PBS)中1:200、Sigma)を、ビオチン-ストレプトアビジン相互作用を介してビオチン結合ファージに接着させ;表面を30分間標識に曝し、次いでPBSで数回濯いだ。
XPSにおいて見られた金シグナルが、ファージに結合した金によるものであり、非特異的な抗体のGaAs表面との相互作用によるものではないことを保証するため、以下の対照をXPS実施例のために行った。調製された(100)GaAs表面を、(1)抗体及びストレプトアビジン-金標識に曝し、ファージには曝さなかった、(2)G1-3ファージ及びストレプトアビジン-金標識に曝し、抗体には曝さなかった、(3)ストレプトアビジン-金標識に曝し、G1-3ファージにも抗体にも曝さなかった。
使用したAFMは、Gスキャナーによりチップ・スキャニング・モード(tip scanning mode)で作動するゼイス・アキシオバート(Zeiss Axiovert)100s-2tvに設置されたデジタル・インストルメンツ・バイオスコープ(Digital Instruments Bioscope)であった。画像はタッピング・モードを使用して空気中で得られた。AFM探針は、200±400kHzのそれらの共振周波数の近くで駆動された、125mmのカンチレバー及び20±100Nm-1のバネ定数を有するエッチングされたシリコンであった。走査速度は約1±5mms-1であった。試料の傾斜を除去するため、一次平面を使用して画像を水平にした。
TEM画像は、60kVでフィリップス(Philips)EM208を使用して得られた。G1-3ファージ(TBSで1:100希釈)を、GaAs片(500mm)と共に30分間インキュベートし、未結合のファージから粒子を分離するために遠心分離し、TBSで濯ぎ、TBSに再懸濁させた。2%酢酸ウラニルで試料を染色した。
G12-3ファージ(TBSで1:100希釈)を、新規に切断されたヘテロ構造表面と共に30分間インキュベートし、TBSで濯いだ。G12-3ファージを、20nmコロイド金でタグ化した。SEM画像及び元素マッピング画像は、5kVで、ヒタチ(Hitachi)4700電界放射型走査型電子顕微鏡に設置されたノリアン(Norian)検出システムを使用して収集した。
本発明者らは、有機-無機ハイブリッド材料が、新規の材料及びデバイスのための新経路を提供することを認識した。サイズ調節されたナノ構造は、光学的かつ電気的に調整可能な半導体材料の特性を与え、有機添加剤は、無機形態学、相、及び核生成方向を修飾する。生物学的材料の単分散性のため、そのシステムは、高秩序スメクチック秩序化構造との適合性を有する。本発明の方法を使用して、特異的半導体表面の認識モエティを有する遺伝学的に操作された自己集合性生物学的分子、例えば、M13バクテリオファージを使用した、II-VI族半導体材料のナノメートル・スケール及びマルチレングス(multilength)スケールの高秩序アラインメントが作出された。

M13ファージ懸濁物を、偏光顕微鏡により特徴決定した。各懸濁物を、直径0.7mmのガラス毛管へ充填した。高度に濃縮された懸濁物(127mg/ml)は、図10(a)のように、平行偏光の下では虹色[5]を示し、交差偏光の下ではスメクチック・テクスチャーを示した。コレステリック・ピッチ、図10(b)は、表1のように、懸濁物の濃度を変動させることにより調節され得る。試料の調製物から24時間後に、ピッチ長が測定され、顕微鏡写真が撮影された。
AFM観察のため、M13バクテリオファージ懸濁物のM13懸濁物(濃度:30mg/ml)5ulを、デシケーターで4時間3-アミノプロピルトリエチルシランによりシレート化(silated)された8mm×8mm雲母基板上で24時間乾燥させた。画像は、タッピング・モードを使用して空気中で得られた。自己集合した秩序化構造が、長さ880nm及び幅6.6nmのM13バクテリオファージの異方形状により観察された。図10(c)において、M13ファージは、写真の平面内にあり、スメクチックアラインメントを形成している。
SEM観察のため、バクテリオファージの臨界点乾燥試料及びZnSナノ粒子スメクチック懸濁物(バクテリオファージ懸濁物の濃度127mg/ml)を調製した。図10(d)において、ナノ粒子リッチな区域及びバクテリオファージ・リッチな区域が観察された。ナノ粒子とバクテリオファージとの間隔は、バクテリオファージの長さに相当する。TEMにより、スメクチック懸濁物の希釈試料を使用して、電子回折パターンにより、ZnSウルツ鉱型結晶構造が確認された。
バクテリオファージ・ペレットを、トリス緩衝生理食塩水(TBS、pH7.5)400ul、及び、1mM Na2Sが添加された1mM ZnCl2 200ulに懸濁させた。室温で24時間の振とうの後、1mlエッペンドルフ・チューブに含有されていた懸濁物を、1週間、デシケーターで徐々に乾燥させた。厚さ約15umの半透明フィルムが、チューブの内側に形成された。図11(a)のこのフィルムを、ピンセットを使用して注意深く採取した。
A7-ZnSフィルムのナノスケール・バクテリオファージアラインメントを、SEMを使用して観察した。SEM分析を実施するため、フィルムを切り取り、次いで真空蒸着によりアルゴン雰囲気中でクロミウム2nmをコーティングした。高密構造、図11(d)が、試料全体に観察された。個々のファージの平均長895nmは、ファージの長さ880nmと合理的に類似している。フィルムは、ナノ粒子層とバクテリオファージ層との周期性を示すスメクチック様のA又はC様の層状形態学を示した。周期性の長さは、バクテリオファージの長さに相当した。ナノ粒子の平均サイズは、個々の粒子のTEM観察と類似した約20nmである。
ZnSナノ粒子アラインメントを、TEMを使用して調査した。フィルムを1日間、エポキシ樹脂(LRホワイト)で包埋し、促進剤10ulを添加することにより重合させた。加硫後、樹脂を、ライカ・ウルトラミクロトーム(Leica Ultramicrotome)を使用して薄片化した。これらの約50nmの切片を蒸留水に浮遊させ、ブランクの金グリッド上に選出した。概略図中のx-z平面に相当する一列に平行に整列したナノ粒子が観察された(図11(e))。各バクテリオファージはA7モエティを5コピー有するため、各A7は1個のナノ粒子(2〜3nmのサイズ)を認識し、約20nmの幅で整列し、2マイクロメートル超の長さにまで及んでいた。20nmのバンドによる2マイクロメートルが、平行に約700nm隔離された各バンドを形成していた。TEMにおける観察に関するこの不一致は、Marvinのグループにより報告されている、ファージ層の傾斜したスメクチックアラインメントから来るのかもしれない。y-z軸様のナノ粒子層平面も、図3(f)のように観察された。整列した粒子のSAEDパターンは、ZnS粒子がウルツ鉱型六方構造を有することを示した。
ウイルス・フィルムの表面配向を、AFMを使用して調査した。図11(c)において、ファージは、スメクチックOと命名される、表面の大部分における隣接ダイレクター標準(バクテリオファージ軸)間のほぼ直角の角度を有する平行アラインメントヘリングボーン・パターンを形成していることが示された。フィルムは、数十マイクロメートルに持続性の標準ダイレクターの広域の秩序化を示した。2個のドメイン層が相互に出会う区域のうちのいくつかにおいては、2個又は3個のマルチレングス・スケールのバクテリオファージが、平行に持続性にスメクチックC秩序化構造へと整列した。
Claims (7)
- (i)一つまたは複数のバクテリオファージ、および(ii) バクテリオファージのそれぞれに結合している一つまたは複数のペプチドを含む、組成物であって、ペプチドが異なる組成または面を有する標的結晶表面構造に選択的であり、ペプチドが、ナノ結晶をさらに核生成して結合させることができ、かつペプチドがナノ結晶に結合しており、組成物が巨視的フィルム形態である、組成物。
- 標的結晶が単結晶である、請求項1記載の組成物。
- 標的結晶が無機結晶である、請求項1記載の組成物。
- 標的結晶が半導体結晶である、請求項1記載の組成物。
- 標的組成物を含む無機相をさらに含む、請求項1記載の組成物。
