T 検出対象核酸鎖
1 インターカレーター性蛍光色素
2 非インターカレーター性蛍光色素
Ex1 励起光(インターカレーター性蛍光色素)
Ex2 励起光(非インターカレーター性蛍光色素)
Em1 蛍光(インターカレーター性蛍光色素)
Em2 蛍光(非インターカレーター性蛍光色素)
Claims (2)
- 検出対象核酸鎖に対し相補的な塩基配列を有し、前記検出対象核酸鎖との二本鎖形成によって蛍光特性が変化する蛍光標識オリゴヌクレオチドを、濃度未知の前記検出対象核酸鎖を含むPCR反応液に添加し、前記蛍光標識オリゴヌクレオチドの蛍光に係る情報を検出することによって、前記蛍光標識オリゴヌクレオチドと前記検出対象核酸鎖との二本鎖形成に関わる情報を得て、前記検出対象核酸鎖の濃度を求める検出対象核酸鎖の定量方法であって、
蛍光共鳴エネルギー移動現象のドナーとなるインターカレーター性蛍光色素と、前記蛍光共鳴エネルギー移動現象のアクセプターとなる非インターカレーター性蛍光色素とが、同一末端に標識された前記蛍光標識オリゴヌクレオチドを、前記PCR反応液に添加し、
前記蛍光標識オリゴヌクレオチドを前記検出対象核酸鎖のプライマーとしてPCR反応を行い、前記PCR反応のアニーリング反応中において、励起光EX1の照射によって発生する蛍光EM2の強度I1、及び、励起光EX2の照射によって発生する蛍光EM2の強度I2を検出し、下記式(1)、(2)により、前記検出対象核酸鎖の濃度を算出する定量方法。

R=(I1−B1)/(I2−B2) …式(2)
(上記式(1)中、「Conc」は前記検出対象核酸鎖の濃度(mol/l)、「Kb」は前記蛍光標識オリゴヌクレオチドの結合定数、「Ri」は反応溶液中に前記検出対象核酸鎖がない状態のR、「Ra」は前記検出対象核酸鎖を蛍光標識オリゴヌクレオチドに対して過剰量投入したときのRを示す。上記式(2)中、「I1」は前記励起光EX1による蛍光EM2の強度、「B1」は前記励起光EX1によるバックグランド値、「I2」は励起光EX2による蛍光EM2の強度、「B2」は励起光EX2によるバックグランド値であり、「バックグランド値」とは、反応溶液中に前記蛍光標識オリゴヌクレオチドがない状態において、励起光EX1、EX2の照射により得られる蛍光EM2の強度である。) - 検量線を作成せずに、前記検出対象核酸鎖の濃度を測定する請求項1記載の定量方法。
