Description

本発明は、核移植技術を利用して作製した胚性幹細胞(nuclear transfer embryonic stem cells;ntES細胞と略記する場合がある)を提供する。成体における神経細胞は非増殖性であり、複雑な細胞構築体である脳から遺伝的に均一な細胞集団を得ることは従来の技術では極めて困難であった。本発明では、単一細胞核ゲノムを胚発生プログラムに沿った形で増幅させ、そこに由来する培養細胞株を樹立することにより、無限に増殖させることに成功し、この問題を根本的に解決したのである。すなわち、本発明は、大脳皮質由来成体神経細胞核を移植した卵母細胞に由来する胚性幹細胞を提供する。このような胚性幹細胞は、大脳皮質由来成体神経細胞核を卵母細胞に移植し、得られた核移植卵母細胞から発生する胚盤胞を使用して胚性幹細胞株を樹立することができる。
本明細書中、「胚性幹細胞(ES細胞)」とは、胚盤胞の内部細胞塊に由来する細胞で、二倍体胚盤胞にインジェクションすることにより、または桑実胚と凝集させることによって胚に導入されたときに、生殖細胞を含む全ての細胞系に発生することのできる全能細胞のことをいう。「核移植」とは、ある種の細胞から抜き取った核を除核した他の細胞に移植することをいう。"nuclear transfer"という英語表記から「NT」と略記される場合がある。「クローン」とは、細胞の場合、単一細胞に由来する細胞の集団を意味する。したがって、「クローン動物」とは、単一細胞に由来する動物個体をさす用語である。本発明において「クローン化」または「クローン化する」とは、単一細胞に由来する細胞の集団または動物個体を作出することをいう。
本発明では、核のドナー細胞として神経細胞を使用する。神経系は中枢神経系と末梢神経系に分けられる。本発明において、中枢神経系、末梢神経系いずれに由来する細胞もドナー細胞として用いることができるが、特に中枢神経系神経細胞を用いることが好適である。中枢神経系神経細胞でも成体神経細胞は最終分化型細胞であるので、特に好適である。一般に、第二減数分裂のメタフェーズで停止した卵母細胞がサイトプラストレシピエントとして使用される。DNAまたはDNAを含む核のようなドナー遺伝物質は、(1)細胞融合、(2)無傷の細胞、溶解した細胞または核の注入の方法、等によりレシピエント細胞質に導入される。遺伝物質の移植は、レシピエントがどのような状態にあっても可能である。また、再構築された胚の倍数性は、細胞周期の適した段階でのドナー遺伝物質の使用によって維持されなければならない。
本発明の胚性幹細胞を樹立する場合は、まず、大脳皮質由来成体神経細胞(ドナー細胞)の核を核移植技術によって、適当な除核卵母細胞(レシピエント細胞)に移植する。本発明で用いられる中枢神経系成体神経細胞は、好ましくは、大脳皮質由来成体神経細胞である。使用するドナー細胞とレシピエント細胞は、特に限定されないが、通常は同種の動物由来のものを用いる。本発明において使用される大脳皮質由来成体神経細胞及び卵母細胞は、特に限定されないが、好ましくは哺乳動物由来であり、より好ましくはマウス又はラット由来である。本発明で特に好ましく用いられる中枢神経系成体神経細胞及び卵母細胞は、C57BL系統とDBA/2系統とのF1ハイブリッド系統(B6C3F1)のマウス由来のものが好ましい。
本発明において用いられる核移植技術は、マイクロマニュピレータを用いるマイクロインジェクション法、in vitro 胚培養法、胚移植法などの公知の技術が用いられる。核移植およびin vitroの培養は、例えば、Wakayama, T., Perry, A.C.F., Zuccotti, M., Johnson, K.R., & Yanagimachi, R. (1998) Nature 394, 369-374に記載の方法によって実施することができる。
上記のようにして得た核移植卵母細胞から発生する胚盤胞中の内部細胞塊の細胞を培養することによって胚性幹細胞を樹立することができる。場合によっては、桑実胚の解離細胞や着床を遅らせた胚盤胞を使用することもできる。胚性幹細胞は胚盤胞から単離することができ、その後、適切な培養条件下で培養されれば永久細胞株として樹立することができる。具体的には、例えば文献(Hogan, B., Geddington, R., Constantini, F., & Lacy, E. (1994) Manipulating the mouse embryos.Eds 3. (Cold Spring Harbor Press. New York))の記載にしたがって、ES細胞の樹立を行うことができる。さらに、前述の文献(Hochedlinger, K., & Jaenisch, R. (2002) Nature 415, 1035-1038;Li, J., Ishii, T., Feinstein, P., & Mombaerts, P. (2004) Nature 428, 393-399)の記述にしたがい、四倍体胚相補発生試験によるクローンマウスの作出を行うことができる。
なお、本発明のES細胞が多分化能力を保持しているか否かは、例えば、哺乳動物(例えば、マウス)で確立されている公知の各種確認方法により、確認することができる。多分化能の確認方法としては、例えば、培養系で神経細胞に分化させ、GFAP(glial fibrillary acidic protein)、ネスチン(Nestin)抗体マーカー等で染色してその多分化能を確認する方法、あるいは、除核した体外培養由来の未受精卵にES細胞を核移植し、更に体外培養することにより胚盤胞に発達させ、これを受胚雌の子宮に移植することにより妊娠させることで確認する方法などを挙げることができる。
本発明はさらに、上記胚性幹細胞を用いて作出される形質転換体、分化細胞・組織・臓器、トランスジェニック動物などにも関する。本発明の形質転換体は、例えば、本発明のES細胞に、所望の外来遺伝子を導入することにより得られた形質転換体である。本発明のES細胞への外来遺伝子の導入方法としては、公知の遺伝子導入法、例えば、市販の遺伝子導入試薬[例えば、Effecten(キアゲン)又はFuGENE(ロシュ)]を用いる方法を挙げることができる。外来遺伝子は、ES細胞の分化能力を検証するために導入される。外来遺伝子の好適な例としては、green fluorescent protein (GFP)遺伝子、β-galactosidase (lacZ)遺伝子、DsRed遺伝子、等を挙げることができる。本発明のES細胞由来分化細胞・組織・臓器としては、例えば、神経、筋肉(例えば、骨格筋又は心筋等)、肝臓、皮膚、血球、又は血管等を挙げることができる。
なお、本発明においては、四倍体胚相補発生法によってもクローン化した個体を得ることができる。四倍体細胞は移植された後に限られた発生能しか持たないことが知られている。四倍体胚が二倍体胚と凝集するとき、四倍体細胞の分化が原始内胚葉および栄養外胚葉に強く制限され、これらはその後、胚体外組織を形成し、一方二倍体細胞は実際の胚を形成することができる。四倍体胚盤胞は、例えば、Eggan, K. et al., Proc. Natl. Acad. Sci. USA 98, 6209-6214 (2001)に記載される既知の方法により、例えば二細胞胚のエレクトロフュージョンおよびそれに続く培養により得ることができる。つまり、本発明において「四倍体胚相補発生」とは、上記のようにして得られた四倍体胚盤胞に本発明の胚性幹細胞を注入し、クローン化した個体を得ることである。
以上のようにして得られる本発明の分化細胞・組織・臓器、トランスジェニック動物などは、例えば、再生医療に関する種々の遺伝工学的実験に有効に利用することができる。
以下、実施例に基づいて、本発明をより詳細に説明するが、これらは本発明の範囲を限定するものではない。
(1)トランスジェニックマウスの作製
Cre遺伝子を有するベクタープラスミドを、aCamKIIプロモーター配列を含むpMM279の8.5kbの断片を使用して構築した(Tsien J.Z., Chen, D.F., Gerber, D., Tom, C., Mercer, E.H., Anderson, D.J., Mayford, M., Kandel, E.R., Tonegawa, S. (1996) Cell 87, 1317-1326)。トランスジェニック第一代マウスは、C57BL/6マウスの接合体へプラスミドを前核注入することにより作出した。作出した3系統のトランスジェニックマウスのうち、一つのマウスの系統(Cam-Cre)が前脳部特異的にCreタンパク質の効果的な発現を示した。内在的Nex-1遺伝子にCre遺伝子が挿入され(Nex-Cre)、GFP遺伝子がGAD67遺伝子に挿入されている(GAD67-GFP)、トランスジェニックマウスの系統が作出された。C57BL/6のバックグランドを有するCAG-CAT-EGFPマウスは、DBA/2マウスと戻し交配された。戻し交配の第2と第3の世代が、本発明において使用された。
ドナー細胞を、Cam-CreおよびCAG-CAT-EGFP対立遺伝子(Cam/CAG)、Nex-CreおよびCAG-CAT-EGFP対立遺伝子(Nex/CAG)およびGAD67-GFP対立遺伝子のそれぞれを有するCam/CAG、Nex/CAGおよびGAD67-GFPマウスの大脳皮質から分離した。トランスジェニックマウス(3-40日)の大脳皮質を、37℃でパパイン(12mg/ml、Wharrington)を含むB27(Invitrogen)を添加したNeurobasal-A培地中で20分間激しく振盪することにより分離した。数回ピペッティングした後に、細胞懸濁液をOPTI-prep細胞分離試薬(Invitrogen)に載せ、説明書に従い、20分間、1000gで遠心した。神経細胞に富んだ画分を分離し、ソート前にB27を添加したNeurobasal-A培地に再懸濁した。
再懸濁した細胞を、説明書にしたがってEPICS ALTRA(BECKMANN COULTER)によりソートした。すべての時間、試料をソートする前に、ソートの確実性をFlow-check Fluorosphere(BECKMANN COULTER)で確認した。
サイズが大きい(ほぼ8-10μm)、または特徴的な神経細胞の形態のいずれかを備えた細胞が、これらの細胞がEGFPとNeuNにそれぞれ陽性だったので、ソートされた細胞から注入された。その結果を図2(B)に示す。クローン胚についての核移植実験およびin vitroの培養を文献(Wakayama, T., Perry, A.C.F., Zuccotti, M., Johnson, K.R., & Yanagimachi, R. (1998) Nature 394, 369-374)に記載の方法に従って行った。文献(Hogan, B., Geddington, R., Constantini, F., & Lacy, E. (1994) Manipulating the mouse embryos.Eds 3. (Cold Spring Harbor Press. New York))の記載にしたがって、ES細胞の樹立を行った。前述の文献(Hochedlinger, K., & Jaenisch, R. (2002) Nature 415, 1035-1038;Li, J., Ishii, T., Feinstein, P., & Mombaerts, P. (2004) Nature 428, 393-399)の記述にしたがい、四倍体胚相補発生試験によるマウスの作出を行った。
avartinによる麻酔後、マウスを屠殺し、組織を2-6時間、灌流とインキュベーションにより、4%のPFAを含むPBSで固定化した。25-30%のショ糖で平衡化した後、組織をOTC化合物中で凍結した。5-12μmおよび50μmの厚さの切片を、三色の免疫蛍光検査法分析および共焦点顕微鏡分析のためにそれぞれ調製した。本研究に使用した抗体は以下のとおりである:ウサギ抗GFP(1:10-200, Clonetech)、マウス抗Cre(1:4000, Chemicon)、マウス抗aCamKII(クローン3E11、Yamauchi博士(愛媛大学)から贈られた)、マウス抗NeuN(免疫組織化学用1:400(Chemicon)、免疫細胞化学用1:100(CHEMICON))(Mullen, R.J., Buck, C.R., & Smith, A.M. (1992) Development 116, 201-211)、マウス抗GAD67(1:400, Chemicon)、マウス抗CNP(1:500, Chemicon)、マウス抗GFAP(1:200, Sigma)。
pEGP-N1(Clontech)のEGFP cDNAを含むEcoRI-NotI断片が、サザンブロッティング(Kawakado S., Niwa, H., Tashiro, F., Sano, S., Kondoh, G., Takeda, J., Tabayashi, K., & Miyazaki, J-I. (2000) FEBS let. 470, 263-268)のプローブとして使用された。PCR分析は説明書(Takara)にしたがって行われた。上記Kawakadoらの文献に記述されるように、CAGプロモーター、CAT遺伝子およびEGFP遺伝子の配列に特異的なプライマーセットを使用した。Dietrich, W., Katz, H., Lincoln S.E., Shin, H-S., Friedman, J., Dracopoli, N.C., & Lander, E.S. (1992) Genetics 131, 423-447に記述されたように、PCRタイピングのSSLPプライマーセットを調製した。
新皮質中の神経細胞を可視化するために、in vivoのCre/loxP部位特異的組換え系を利用した。図1にCam/CAGトランスジェニックマウスの大脳皮質中の遺伝学的マーカーを有する細胞の特性を示す。図1(A)はCam/CAGおよびNex/CAGのトランスジェニックマウスにおけるCreを用いた組換えのスキームであり、神経細胞遺伝子プロモーター(Np)により動くCreの発現により、lox-P配列に隣接するCAT/ストップカセットが切り出される。図1(B-M) は、遺伝子操作されたCam/CAGおよびNex/CAGトランスジェニックマウスの新皮質中のGFP発現細胞の共焦点顕微鏡による解析を示す(赤、NeuN免疫反応性(B、D)およびGABA免疫反応性(C、E);f-i、緑、GFP(F-I)蛍光;合体イメージ(J-M);バーは10μmを表わす(J-M))。
αカルシウム/カルモジュリ依存性キナーゼII(aCamKII)遺伝子のプロモーター配列(Cam-Cre)の支配下でCreリコンビナーゼ遺伝子を発現する、トランスジェニックマウスを最初に作出した。Cam-Creマウスを、リポーターマウス系統(CAG-CAT-EGFP)と交配すると、EGFP遺伝子は、NeuN陽性の成体神経細胞の中で優性に発現し、得られたマウス中のGABA陽性のGABAergicな神経細胞では発現されない(図1(B-M))。
細胞数から、EGFP+細胞がNeuN免疫反応性に優性的に陽性であることが示された(535個のNeuN+EGFP+/552個のEGFP+(96.9%))。比較的大きな、EGFP陽性の生細胞をセルソーターで濃縮した画分では、大多数のこれらの細胞が神経細胞の起源であることが明らかとなった(1562個のNeuN+細胞/1575個のソートされた細胞(99.2%))。実験方法の確実性は、Nex-1遺伝子の内在性プロモーターにより動く神経細胞のEGFP発現をする別のトランスジェニックマウスの系統によって評価された。組織切片およびセルソートの細胞数から、Nex/CAGマウス由来のEGFP発現細胞は選択的にピラミッド形の神経細胞であることが明らかとなった(組織切片中では648個のNeuN+細胞/650個のEGFP+細胞(99.7%);セルソート後では309個のNeuN+細胞/310個のEGFP+細胞(99.7%))。したがって、選別過程により、CAM/CAGおよびNex/CAGマウス由来の核ドナーとして使用されるEGFP発現細胞はすべてピラミッド形の成体神経細胞であると保証された。
GFPを発現するGABAergicな神経細胞は単独で分離された(1052個のNeuN+細胞/1057個のEGFP+細胞(99.5%);112個のGAD67+細胞/112個のEGFP+細胞(100%))。選択データは、GAD67ノックインマウス由来のドナー細胞として使用されるEGFP+細胞がGABAergicな神経細胞であることを強く示唆している。
まず、非遺伝学的マーカーを有する脳細胞(出生後18-40日(P))を用いて、再構築された卵母細胞から発生した胚盤胞由来のES細胞系の生成を試験した。結果を表1に示す。
図4は、成体神経細胞の核を用いたクローンマウスの形態学的解析の結果を表す。図4(A)は、 正常な妊娠能を有する6週齢のCAM-2 ES細胞核に由来する神経細胞のマウスクローン「Cerebro」(挿入部分は尾部の情報を表す)を示す。明視野(上)および蛍光(下)により、イメージを写真撮影した。挿入部のバーは1mmを表す。図4(B)は、 Cam/CAGマウスの新皮質DNA(レーン1)および肝臓DNA(レーン2)、核移植により産生されたCam-2(レーン3)およびNex-1(レーン4)に由来する神経細胞クローンの尾部DNA、および四倍体胚相補発生試験によって産生されたGad67-2 ES細胞由来の2クローンの尾部DNA(レーン5、6)のサザンブロッティングの結果を示す。図4(C-H)は、 Cam-2 ES細胞(C-E)およびGad67-2 ES細胞(F-H)由来の神経細胞マウスクローンの大脳皮質切片を示す(Nissl染色(C、F)および、抗NeuN(D、G)および抗GAD67(E、H)抗体を使用する間接免疫蛍光法を用いた切片)。明白な細胞質構造の変化は検出されなかった。バーは1mm(AとBの挿入部)および50μm(C-H)をそれぞれ表す。図4に示されるように、サザンブロット分析から、Creを用いたDNA組換えが生じたドナー神経細胞に由来する4系統のES細胞系すべてにおいて、組換えを起こした対立遺伝子の存在が確認された(図4(H))。
EGFP遺伝子が挿入されるGAD67遺伝子の対立遺伝子は、ドナーのGAD67ノックインマウスおよびドナー核に由来するES細胞のゲノムDNAをEcoRIで消化することにより検出された(図2(H))。CAG-CAT-EGFPマウスがDBA/2と戻し交配されたので、C57BL/6およびDBA/2の近交系の染色体セグメントで差異分配がPCRタイピングにより示された。これらのデータから、単一の神経細胞核に由来する遺伝学的に独立したES細胞系の樹立が実証される。
神経細胞由来のES細胞から神経細胞起源のクローンを生成するために、まず核移植ES細胞(ntES細胞)を四倍体胚盤胞に注入した(四倍体胚相補発生試験)(図3(A))。ここで、図3は、ES細胞中間体を用いたマウスクローニングを示す図である。図3(A)は、神経細胞核に由来するES胎児およびESクローンを産出するためのスキームを示す。図3(B、C)は、四倍体胚相補発生試験によるCam-1 ES細胞に由来する子を示す(GFP発現は胎盤ではなく、胚組織だけに制限されることに注意(矢印))。図3(D、E)は、核移植によるCam-2 ES細胞に由来する子を示す。四倍体胚相補発生試験による子とは対照的に、胚外組織(胎盤)はGFP蛍光(矢印)を発現する。イメージは、明視野(B、D)および蛍光(C、E)により写真撮影された。棒は1cm(B-E)を表す。
3系統のntES細胞系(Cam-1、Cam-2およびGAD67-2)から、誕生時に19匹の子クローンが得られた。結果を表2に示す。
四倍体胚相補発生試験(Cam-1 ES細胞系からの1匹、Cam-2からの2匹およびGAD67-2からの3匹)および核移植(Cam-2 ES細胞系からの1匹)の両方から、開いた眼瞼をした合計7匹の子を得た(図3(D、E))。7匹のうち、CAM-2およびGad67-2由来のそれぞれ1匹が、四倍体胚相補発生試験の結果として成体となった。しかし、表現型の異常は遺伝学的に伝わらなかった。核移植の間に、CAM-2およびGad67-2の細胞核に由来するマウスクローンは、思春期に入った(図4(A)のCAM-2 ES細胞由来の「Cerebro」、Gad67-2 ES細胞由来の「Cerebra」)。サザンブロット分析から、四倍体胚相補発生試験および核移植により産出されたクローンマウスがドナーES細胞の遺伝子型と同一の組換え対立遺伝子を有することが確認された(図4(B))。組織学的試験から、四倍体胚相補発生試験および核移植により得られたP0時の子からの大脳皮質の総体的形態および細胞構造の明白な欠損は明らかにされなかった(図4(C-H))。これらの結果は、核移植により、皮質の新生を含む十分な発生をサポートするために、新皮質の成体神経細胞がリプログラミングされることを示している。
Claims (3)
- 大脳皮質神経細胞に特異的なプロモーターであるaCamKII、Nex-1またはGAD67が作動して大脳皮質由来神経細胞でのみ特異的にDNA組換を行って、green fluorescent protein(GFP)遺伝子、β-galactosidase(lacZ)遺伝子またはDsRed遺伝子である外来遺伝子のマーカーを発現するよう組み換えられた非ヒトトランスジェニック動物より、該マーカーに基づき大脳皮質成体神経細胞を単離し、
単離された細胞の核を非ヒト卵母細胞に移植し、
得られた核移植卵母細胞から発生する胚盤胞を使用して、全能性の胚性幹細胞株を樹立すること
を特徴とする胚性幹細胞の樹立方法。 - 前記DNA組換が前記プロモーターの支配下で発現するCreリコンビナーゼによって行われる、請求項1に記載の胚性幹細胞の樹立方法。
- 前記大脳皮質由来成体神経細胞及び前記卵母細胞が、マウス又はラット由来である、請求項1または2に記載の胚性幹細胞の樹立方法。
