Description
本発明は、マトリクス支援レーザー脱離イオン化(MALDI)質量分析法の分析において有用なプレート、ならびにこのプレートを作製および使用するためのプロセスに関する。より詳細には、本発明は、疎水性表面を有するMALDIプレート、ならびにこのプレートを作製および使用するためのプロセスに関する。
本発明の1つの局面では、物質(例えば、合成ろう(例えば、パラフィンろう)、自然のろう(例えば、蜜蝋)、脂質、エステル、有機酸、ケイ素オイル、またはシリカポリマー)の一体型の、容易に除去可能な疎水性コーティングを有する、MALDIサンプルプレートが提供される。この前記の物質は、純粋な化合物として、または互いに混合されてもしくは市販の化学的組成物(例えば、金属艶出しペーストまたは植物油)の一部としてのいずれかで、サンプルプレートに適用される。1つの実施形態では、金属艶出し剤の適用は、サンプルプレートの表面疎水性を作製および回復するために有効である。
本発明の1つの実施形態によれば、取り外し可能なミクロン以下の厚さのパラフィン層で完全にコーティングされた導電性基板を有する、MALDI MSプロセス用のサンプルプレートが提供される。「完全にコーティングされた」(または「完全に結合した」または「一体型の」)により、本発明者らは、種々の力の相互作用(例えば、疎水性相互作用、イオン相互作用、ファンデルワールス力など)によって作製された、基板上の薄い(1ミクロン以下の厚さ)の物理的コーティングであって、基板からインタクトに分離も引き剥がしもできず、むしろ、このコーティングは、化学的処理(例えば、溶媒の使用による)または機械的(剥離)処理によって取り外し可能である、物理的コーティングを意味する。パラフィン溶液は、基板(例えば、ステンレス鋼製サンプルプレート)の表面に、噴霧、浸漬などにより適用される。次いで、この溶液の溶媒は、適切な条件下で蒸発されて、この表面に完全に結合した、パラフィンの薄い均質なコーティングが残される。
鏡面仕上げを有するMALDI用のステンレス鋼製の2.25”×2.25”の矩形プレートに、50マイクロリットルのヘキサン/ヘプタン(50:50v/v)中の60マイクログラムのパラフィンを噴霧した。得られた表面は、20nmの厚さのパラフィンで均質にコーティングされていた。
鏡面仕上げを有するMALDI用のステンレス鋼製の2.25”×2.25”の矩形プレートに、50マイクロリットルのヘキサン/ヘプタン(50:50v/v)中の60マイクログラムのパラフィンを数分で噴霧した。得られた表面は、10nmの厚さのパラフィンで均質にコーティングされていた。
鏡面仕上げを有するMALDI用のステンレス鋼製の2.25”×2.25”の矩形プレートを、水中のRBS−35の10%溶液を用いたこのプレートのスクラビング、水でのリンスおよびくずのないティッシュを用いた乾燥によって準備した。このプレートを、最少量(ピンヘッドのサイズのビーズ)の金属艶出し剤を用いて艶出しした。この艶出し剤は、石油エーテル、灯油(石油)、ココナッツ脂肪酸ジエタノールアミン、酸化アルミニウム、アンモニア溶液および水から構成されていた。曇りが完全に除去され、新しいくずのないティッシュ上に黒い残存物が検出されなくなったら、このプレートを、イソプロパノールで洗浄し、そしてこのプレート全体に空気を吹き付けて乾燥した。

Claims (6)
- MALDIサンプルプレートであって、該MALDIサンプルプレートは、導電性基板を備え、該基板は、疎水性コーティングの薄いフィルムで完全にコーティングされており、該疎水性コーティングは、合成ろう、天然のろう、脂質、疎水性有機酸、疎水性エステル、ケイ素オイル、もしくはシリカポリマー、または前記の物質の互いの混合物のうちの少なくとも1つを含むか、あるいは化学的組成物の成分として前記の物質の混合物を含み;
該薄いフィルムは、疎水性相互作用、イオン相互作用、またはファンデルワールス力のうちの一つ以上を含む力により該表面に完全に結合した疎水性コーティングの単層を含み;そして
該薄いフィルムは、化学的処理または剥離処理によってのみ該物質から取り外し可能である、
サンプルプレート。 - 前記合成ろう疎水性コーティングが、パラフィンを含む、請求項1に記載のMALDIサンプルプレート。
- 前記脂質疎水性コーティングが、トリパルミチンを含む、請求項1に記載のMALDIサンプルプレート。
- 前記疎水性コーティングが、金属表面をクリーニングおよび保護するように設計された組成物の混合物を含む艶出し剤を含む、請求項1に記載のMALDIサンプルプレート。
- 前記疎水性コーティングが、約5nmと約50nmとの間の厚さを有する、請求項2、3または4に記載のMALDIサンプルプレート。
- 前記疎水性コーティングが、約5nmと20nmとの間の厚さを有する、請求項2、3または4に記載のMALDIサンプルプレート。
