Description
ファフィア・ロドジーマATCC96594を、YPD培地(DIFCO、Detroit、U.S.A.、試験管中10 mL)に接種し、20℃において2日間、振とう培養した。0.5mLの培養物を、30g/Lのグルコース、ならびに、各々0μg/mL、0.2μg/mL、0.5μg/mL、1.0μg/mL、2.0μg/mL、5.0μg/mL、10.0μg/mLおよび20.0μg/mLの[3-(3-アリル-ビフェニル-4-イルオキシ)-プロピル]-イソプロピル-アミンを含む新鮮なYPD培地(試験管中10mL)に接種し、20℃において5日間、振とう培養した。
ファフィア・ロドジーマATCC96594を実施例1のようにYPD培地に接種し、20℃において2日間、振とう培養した。2.5mLの培養物を、22g/Lのグルコース、ならびに、各々0μg/mL、0.5μg/mL、1.0μg/mL、2.0μg/mLおよび5.0μg/mLの[3-(3-アリル-ビフェニル-4-イルオキシ)-プロピル]-イソプロピル-アミンを含む新鮮なYPD培地(フラスコ中50mL)に接種し、20℃において7日間、振とう培養した。培養2日目に、50g/Lのグルコースを培養物に添加した。培養の途中における阻害剤の添加も同様に試験された。培養物のアリコートを、培養の4日目および7日目に採取し、660nmにおける吸光度(実施例1に記載されたのと同じ方法を用いることにより)、および培養物中のアスタキサンチン含量を測定した。

Claims (5)
- 有酸素条件下の水性栄養培地において、ファルネシルピロリン酸由来のステロール生合成阻害剤の存在下で、カロテノイドを産生できる微生物を培養する段階を含み、ファルネシルピロリン酸由来のステロール生合成阻害剤が、アンモニウムイオンに基づくスクアレン合成酵素阻害剤からなる群より選択されるカロテノイドを作製するための生物学的方法であって、
培養する段階が、スクアレン合成酵素阻害剤の非存在下で培養を開始し、その後、スクアレン合成酵素阻害剤を添加するものであり、
微生物が、キサントフィロマイセス・デンドロロス(Xanthophyllomyces dendrorhous)(ファフィア・ロドジーマ(Phaffia rhodozyma))ATCC96594であり、
スクアレン合成酵素阻害剤が[3-(3-アリル-ビフェニル-4-イルオキシ)-プロピル]-イソプロピル-アミンであり、
スクアレン合成酵素阻害剤が5.0μg/mLの濃度を有し、
カロテノイドがアスタキサンチンであり、
培養が、pH4〜8の範囲で、15℃〜26℃の温度範囲で24時間〜500時間実施される、方法。 - 有酸素条件下の水性栄養培地において、カロテノイドを産生するための基質であるファルネシルピロリン酸由来のステロール生合成阻害剤の存在下で、キサントフィロマイセス(Xanthophyllomyces)(ファフィア(Phaffia))属に属し、カロテノイドを産生できる微生物を培養する段階、および微生物の細胞からまたは培養されたブロスから得られたカロテノイドを単離する段階を含み、ファルネシルピロリン酸由来のステロール生合成阻害剤が、アンモニウムイオンに基づくスクアレン合成酵素阻害剤からなる群より選択されるカロテノイドを作製するための生物学的方法であって、
培養する段階が、スクアレン合成酵素阻害剤の非存在下で培養を開始し、その後、スクアレン合成酵素阻害剤を添加するものであり、
微生物が、キサントフィロマイセス・デンドロロス(Xanthophyllomyces dendrorhous)(ファフィア・ロドジーマ(Phaffia rhodozyma))ATCC96594であり、

培養が、pH4〜8の範囲で、15℃〜26℃の温度範囲で24時間〜500時間実施される、方法。 - 阻害剤の濃度が、カロテノイド産生条件下で、細胞成長を50%未満低下させる濃度の範囲内である、請求項1または2記載の方法。
- 阻害剤の濃度が、カロテノイド産生条件下で、細胞成長の30%未満低下させる濃度の範囲内である、請求項3記載の方法。
- 培養が、pH5〜7の範囲で、18℃〜22℃の温度範囲で48時間〜350時間実施される、請求項1記載の方法。
