本発明は、黄色ブドウ球菌の除菌方法及びブラキバクテリウム(Brachybacterium)属に属する新規微生物に関する。

【従来の技術】
ブラキバクテリウム属に属する細菌としては、現在のところ下記のようなものが知られている。

Brachybacterium nesterenkovii
Brachybacterium paraconglomeratum
Brachybacterium rhamnosum
しかしながら、これらのブラキバクテリウムについては、その産業的利用性については、全く研究がなされていないのが現状である。

【発明が解決しようとする課題】
そこで、本願の発明者らは、ブラキバクテリウムについて鋭意研究を重ねたところ、ブラキバクテリウムは、黄色ブドウ球菌に対する抗菌性を持つことを知見した。
本発明の課題は、この知見に基づくものであり、上記ブラキバクテリウムを用いた黄色ブドウ球菌の除菌方法を提供することにある。
さらに、本発明者らは、ブラキバクテリウム属に属する有用な新菌株を求めて鋭意探索したところ、他のブラキバクテリウムと同様に黄色ブドウ球菌に対して抗菌機能を有する一菌株を土壌から分離することに成功した。
本発明の他の課題は、ブラキバクテリウム属に属する新規微生物を提供することにある。

【課題を解決するための手段】
上記課題は、下記構成(1)および2によって達成することができる。
(1) 寄託番号:FERM P-16951として独立行政法人産業技術総合研究所特許生物寄託センターに寄託されているものであって、黄色ブドウ球菌に対する抗菌性、脱臭機能および鮮度保存作用を備えたバクテリウム属に属する新規微生物。
(2)上記(1)に記載された ブラキバクテリウム属に属する微生物を黄色ブドウ球菌に接種して、黄色ブドウ球菌を除菌することを特徴とする黄色ブドウ球菌を除菌方法。

以下、本発明のブラキバクテリウムに属する新規微生物の菌学的性質を説明する。

本発明の新規微生物は一般栄養培地であるならいかなる培地でも良好に成育する。

培養は、好ましくは5〜30℃、最も好ましくは18〜27℃程度で良好に増殖する。

本発明の新規微生物の形態的性質は下記の通りである。
(1)細胞の形及び大きさ:
▲1▼ 肉汁寒天培地を用いて培養した場合
・30℃、6時間培養では1.2〜1.4×1.5〜1.9ミクロンの多形性の短桿菌である。
・30℃、24時間培養では1.2〜1.4×1.2〜1.5ミクロンの球菌様を示す。
▲2▼ 肉汁液体培地を用いて培養した場合
30℃、24時間培養で1.0〜1.2×1.3〜1.4ミクロンの球菌様を示す。
(2)細胞の多形性:
多形性、及び、桿状−球状 サイクルを認める(図1および2を参照)。
(3)運動性は認められない。
(4)胞子の形成は認められない。

各培地における本発明の新規微生物の培養的性質は下記の通りである。
(1)肉汁寒天平板培地を用いて培養した場合:
・集落は平滑、全縁で光沢が認められる。
・黄色系の集落色素の生成が認められる。
(2)肉汁液体培地を用いて培養した場合:
・培地全体における生育及び沈殿が認められる。
・表面皮膜の形成は認められない。
(3)肉汁ゼラチン穿刺を用いて培養した場合:
・培地の上部に生育が認められるが、液化は認められない。
(4)リトマス・ミルクを用いて培養した場合:
・培地全体に生育が認められるが、凝固及び液化は認められない。
・酸の生成がわずかに認められる。
本発明の新規微生物は、水や液体培地に分散させ分散液の状態で各種用途に用いることができるが、好ましくは微生物群を担体に固定し、取扱を容易にした担体固定化微生物群として用いる。

微生物群を固定する担体としては、微生物の保持容量が大きく、また微生物の活性化が容易であることが好ましい。

担体としては、岩石(例えば、真珠岩、珪藻土)またはその粉砕物、砂利、砂、プラスチックス、セラミックス(例えば、アルミナ、シリカ、天然ゼオライト、合成ゼオライト)、タルク等も好ましく、特に、多孔質セラミックス、多孔質プラスチックスのような連続通気孔を有する多孔質材料が好ましい。例えば、真珠岩の粉砕物を高温高圧で保持し熱処理した後、急激に減圧して多孔質にした微細粉(商品名:パーライト)は、菌類を着生させる性質があるため、好適である。このような多孔質材料において、通気孔の径は、2〜10μm程度であることが好ましい。また、このような担体の性状(形状)としては、塊状物、粒体、粉体、微粉末または板状物、針状物等いかなるものでもよいが、好ましくは平均粒径が2mm以下、特に50μm〜1mm程度の粉粒体がよい。なお、このような担体固定化微生物群は、必要に応じて、布やネット等の通水性や通気性のよい容器に収納して使用してもよい。

上記担体としては、これらの他、リボン状やシート状の織布や不織布を用いることができる。

このような担体に微生物群を担持させるには、担体と本発明の微生物群分散液とを混合した後、乾燥すればよいが、担体で直接微生物群を培養してもよい。

担体の微生物群の担持量は、担持させるときの条件によっても異なるが、好ましくは、5個〜200億個/cm、更に好ましくは10個〜100億個/cmである。

本発明の担体固定化微生物群は、黄色ブドウ球菌の除菌に有効である。また、本発明の新規微生物は、養豚所、公衆便所等に用いて、脱臭効果が確認された。

なお、本発明の新規微生物は、マウスによる経口投与試験によりその安全性が確認された。

【実施例】
次に、本発明の実施例について説明する。
まず、次のような組成の培地を作成した。

培地のpH 6.8±0.2
以上の組成の培地にブラキバクテリウム菌株AAA−aを接種し、30℃にて振とう培養を行った。対数期に達した培地を滅菌した生理食塩水にて希釈し、生菌数約1×10/mlになるように調整し、この調整液を直接ブドウ球菌に振り掛けることにより接種したところ、ハローの特性が認められた。上記の他のブラキバクテリウムについても同様の実験を行ったところ、上記新規菌株よりは機能が劣るものの、黄色ブドウ球菌に対してハローの特性が認められた。

一方、得られた調整液を遠心分離機にかけ、菌体を分離し、これをよく洗浄し、担体に付着させてパウダーとした。担体としては、珪藻土を焼いて粉砕したものを用いた。

このパウダー状のブラキバクテリウム菌株AAA−aを、1本のままの腐った鰯に振りかけ 25℃で保管した。2時間経過後にその臭いをかいだところ、明らかに臭気が減少していた。また、ブラキバクテリウム菌株AAA−aを、新鮮な鰯に振りかけ25℃で保管したところ、48時間を経過しても腐臭はしなかった。一方、ブラキバクテリウム菌株AAA−aを振りかけずにそのままの状態で25℃で保管した鰯は、24時間経過でかなり臭いがきつくなった。

このように、本発明のブラキバクテリウム菌株AAA−aは、脱臭作用および鮮度保存作用も備えている。

Furthermore, the present inventors have eagerly searched for useful new strains belonging to the genus Brachybacterium, and isolated one strain having antibacterial function against Staphylococcus aureus as well as other Brachybacterium from the soil. Succeeded in doing.

The morphological properties of the novel microorganism of the present invention are as follows.
