本発明は、バチルス(Bacillus) 属に属する新規微生物に関する。

【従来の技術】
排水浄化菌としては、各種のバチルス属微生物が知られている。しかしながら、排水浄化においては、単に浄化に止まらずに、その脱臭や、金属物質の除去も大切な要素である。

しかしながら、現在までに知られているバチルス属微生物では、これらの全ての要素を満足する微生物は発見させれおらず、そのような微生物が強く望まれていた。

【発明が解決しようとする課題】
そこで、本発明は、排水の単なる浄化とともに、その脱臭や金属除去もできる新規な微生物を提供することを目的とするものである。

【課題を解決するための手段】
本発明らは、上記のような要素を全て満足することのできる有用な新菌株を求めて鋭意探索したところ、浄化、脱臭、金属除去の全ての機能を有する一菌株を土壌から分離することに成功した。この一菌株の菌学的性質の試験および分類は、下記の文献に従い行なった。

その結果、その微生物は、耐熱性の胞子を形成するグラム陽性桿菌であることからバチルスであると同定されたが、菌体成分の分析結果が、上記の文献に記載されているいずれの菌種とも一致せず、バチルスに属する新菌であることが確認された。 当該微生物は、Bacillus sp. CCC-a(寄託番号:FERM P−16175)として工業技術院生命工学研究所微生物寄託センターに寄託されている。

以下、本発明のバチルスに属する新規微生物の菌学的性質を説明する。

本発明の新規微生物は、一般栄養培地であるならいかなる培地でも良好に成育する。

培養は、好ましくは5〜30℃程度で良好に増殖する。

本発明の新規微生物は、水や液体培地に分散させ分散液の状態で各種用途に用いることができるが、好ましくは微生物群を担体に固定し、取扱を容易にした担体固定化微生物群として用いる。

微生物群を固定する担体としては、微生物の保持容量が大きく、また微生物の活性化が容易であることがこのましい。

担体としては、岩石(例えば、真珠岩、珪藻土)またはその粉砕物、砂利、砂、プラスチックス、セラミックス(例えば、アルミナ、シリカ、天然ゼオライト、合成ゼオライト)、タルク等も好ましく、特に、多孔質セラミックス、多孔質プラスチックスのような連続通気孔を有する多孔質材料が好ましい。例えば、真珠岩の粉砕物を高温高圧で保持し熱処理した後、急激に減圧して多孔質にした微細粉(商品名:パーライト)は、菌類を着生させる性質があるため、好適である。このような多孔質材料において、通気孔の径は、2〜10μm程度であることが好ましい。また、このような担体の性状としては、塊状物、粒体、粉体、微粉末またはいた状物、針状物等いかなるものでもよいが、好ましくは平均粒径が2mm以下、特に50μm〜1mm程度の粉粒体がよい。なお、このような担体固定化微生物群は、必要に応じて、布やネット等の通水性や通気性のよい容器に収納して使用してもよい。

上記担体としては、これらの他、リボン状やシート状の織布や不織布を用いることができる。

このような担体に微生物群を担持させるには、担体と本発明の微生物群分散液とを混合した後、乾燥すればよいが、担体で直接微生物群を培養してもよい。

担体の微生物群の担持量は、担持させるときの条件によっても異なるが、好ましくは、5個〜200億個/cm3 、好ましくは10個〜100億個/cm3 である。

本発明の担体固定化微生物群は、排水の浄化に有効である。また、本発明の新規微生物は、養豚所、公衆便所等に用いて、脱臭効果が確認された。さらに、本発明の新規微生物においては、リン等の金属の回収機能が確認された。さらにまた、土壌の改質にもちいても効果が確認された。

なお、本発明の新規微生物は、マウスによる経口投与試験によりその安全性が確認された。

【実施例】
次に、本発明の実施例について説明する。
まず、次のような組成の培地を作成した。

ペプトン 5g
肉エキス 5g
NaCl 2.5g
水 500ml(pH7.0)
以上の組成の培地を、1000ml容器にいれ、これにBacillus sp. CCC-aを接種し、よく撹拌した後、静置して、30℃で3日間培養した。

得られた培養液を遠心分離機かけ、菌体を分離し、これをよく洗浄し、担体に付着させてパウダーとした。担体としては珪藻土焼いて、粉砕したものを用いた。

このパウダー状のBacillus sp. CCC-aを、計量対象へ接種してのCOD、全窒素量および全リン量の経時変化を計量した。試験方法は、CODはJIS 0102−17、全窒素量はJIS 0102−45.2、そして全リン量JIS 0102−46.2に従っておこなった。計量対象としては、海水での牛乳100倍液に、1リットル当たり0.2gの澱粉を加えたものを用いた。そのままのものを比較例とし、これにBacillus sp. CCC-aを接種したものを実施例とした。試験は、20℃に保って行なった。また、各量の測定にあたっては、計量対象液を2時間静置し、その上澄み液を試験液とした。

試験の結果を表1に示した。なお、試験は、それぞれサンプルを2つずつ作製し、その平均を測定値とした。

表1から分かるように、本発明の新規微生物であるBacillus sp. CCC-aは、水の浄化機能を有し、金属を除去する作用があることが判明した。さらに、1本のままの腐った鰯にBacillus sp. CCC-aのパウダーを振りかけ2時間経過後にその臭いをかいだところ、明らかに臭気が減少していた。また、新鮮な鰯にBacillus sp. CCC-aを振りかけ、25℃で保管したところ48時間を経過しても腐臭はしなかった。一方、Bacillus sp. CCC-aを振りかけずにそのままの状態で25℃で保管した鰯は、24時間経過でかなり臭いがきつくなった。

The present inventors have eagerly searched for a useful new strain that can satisfy all of the above-mentioned elements, and have isolated one strain having all functions of purification, deodorization, and metal removal from the soil.
