接種時に添加する。200 rpmで操作される回転振とう機で、フラスコを30℃、64時間培養する。
上の手順は、この技術で周知の手順により、大発酵装置まで容易に規模拡大できる。
上の発酵で得られるB.t.胞子及び結晶は、この技術で周知の手順により単離できる。しばしば用いられる手順は、取り入れた発酵液を分離手順、例えば遠心分離にかけることである。
本発明の毒素タンパクの給源として使用できるバシラス・スリンギエンシス(B.t.)分離体は、例えばB.t.PS17、B.t.PS52A1、B.t.PS33F2、B.t.PS63B、B.t.PS69D1、B.t.PS80JJ1、B.t.PS158D5、B.t.PS167P、B.t.PS169E、B.t.PS177F1、B.t.PS177G、B.t.PS204G4、及びB.t.PS204G6でありうる。分離体を実施例1に記述されたとおりに、又はこの技術で知られたその他の標準的な培地及び発酵手法を用いて培養できる。毒素タンパク封入体は標準的な沈降遠心分離によって取り入れられる。回収されたタンパク封入体は、臭化ナトリウム(28-38%)密度勾配平衡遠心によって部分的に精製できる。[ファネンスチール・エム・エイ(Pfannenstiel, M.A.)、イー・ジェイ・ロス(E.J. これらの配列データからオリゴヌクレオチドプローブが、他のB.t.毒素遺伝子の入手可能な配列から組立てられたコドン頻度表を用いて設計された。プローブは、アプライド・バイオシステムズ社のDNA合成機で合成された。

B.t.PS17細胞を低光学密度(OD600=1.0)に生育させ、細胞を遠心分離によって回収して、全細胞DNAを調製した。20%庶糖と50 mg/mlリゾチームを含有するTES緩衝液(30 mMトリス-Cl、10 mM エチレンジアミン四酢酸[EDTA]、50 mM NaCl、pH=8.0)中で細胞をプロトプラスト化させた。プロトプラストを4%の最終濃度までドデシル硫酸ナトリウム(SDS)添加によって溶菌化した。細胞材料を最終濃度100 mMの中性塩化カリウム中、4℃で一夜沈殿させた。上澄み液をフェノール/クロロホルム(1:1)で2回抽出した。DNAをエタノール中で沈殿させ、塩化セシウム/臭化エチジウム勾配上の密度平衡バンディングによって精製した。
600 nmで1.0の光学密度まで生育させたバシラス・スリンギエンシスPS52A1細胞から全細胞DNAを調製した。細胞を遠心分離によってペレット化し、プロトプラスト緩衝液(0.3M庶糖、25 mM トリス-Cl[pH8.0]、25 mM EDTA中リゾチーム20 mg/ml)中に再懸濁させた。37℃で1時間培養後、2回の冷凍/解凍サイクルでプロトプラストを溶菌させた。溶菌を終了させるため、0.1M NaCl、0.1%SDS、0.1Mトリス-Clの溶液9倍量を加えた。透明になった溶菌液をフェノール:クロロホルム(1:1)で2回抽出した。核酸を2倍量のエタノールで沈殿させ、遠心分離によってペレット化した。ペレットをTE中で再懸濁させ、RNアーゼを50μg/mlの最終濃度に添加した。37℃で1時間培養後、フェノール:クロロホルム(1:1)とTE-飽和クロロホルムでそれぞれ1回ずつ抽出した。1/10量の3M NaOAcと2倍量のエタノールの添加によって、DNAを水相から沈殿させた。DNAを遠心分離によってペレット化し、70%エタノールで洗い、乾燥し、TE中で再懸濁した。

実施例2は、標準エドマンタンパク配列決定法によって決定されるとおりに、PS63B毒素タンパクのアミノ末端及び内部ポリペプチド配列を示す。これらの配列から、δ-エンドトキシンをコード化したB.t.遺伝子から組立てられたコドン頻度表を用いて、二つのオリゴヌクレオチドプライマーが設計された。順プライマー(63B-A)の配列は、遺伝子の5’末端で推定されるDNA配列と相補的であった。

これらのプライマーは、DNAクローニングプローブとして使用される63B毒素遺伝子の約460 bp断片を増幅するために、標準的なポリメラーゼ連鎖反応(シータス・コーポレーション)で使用された。PS63Bからの全細胞DNAのサザンブロットを放射性標識つきPCRプローブとハイブリッド形成させた。ハイブリッド形成帯は約4.4 kbpXbaI断片、約2.0 kbpHindIII断片、及び約6.4 kbpSpeI断片を含んでいた。
S-基本培地1 ml、アンピシリン0.5 mg、及びコレステロール0.01 mgを含有するコーニング(コーニング・ガラス・ワークス、ニューヨーク州コーニング)の24穴組織培養基プレート中で、シンプキン及びコールズ(J. Chem. Tech. Biotechnol. 31巻66-69頁、1981年)に記述のとおりにカエノルハブジチス・エレガンス(CE)を培養した。各穴は、大腸菌菌株OP-50(ウラシル栄養要求株)の細胞約108個も含有した。各穴当たり100-200個のCEを穴に接種し、20℃で培養した。タンパク試料(野性型B.t.又は組替えB.t.から得られたもの)を連続希釈によって穴に添加した。水は、対照として、またタンパクを穴に導入するビヒクルとしての役目を果たした。

B.t.タンパクを発現する野性型B.t.分離体及び組替え微生物が生産する毒素を、その殺吸虫活性について試験した。
生体外培養基(37℃、5%CO2)は、イバラ(Ibarra)及びジェンキンズ(Jenkins)[Z. Parasitenkd. 70巻655-661頁、1984年]の変法によった。培養基培地は、50%v/vうさぎ血清及び2%v/vうさぎRBCを加えたRPMI(pH 7.5)からなっていた。
〔試験化合物〕 下記の実験で、肝臓吸虫への種々の物質の影響を試験し、比較した。本明細書で使用される化合物PS17は、上記のように、B.t.PS17の培養基から回収精製される毒素のことである。化合物PS17aは、B.t.PS17aと指定されるB.t.遺伝子で形質転換された組替え微生物の培養基から回収精製された毒素のことである。化合物Cは、陰性対照として使用された処方剤ブランクである。ABZは、スミスクリン・ビーチャム社(ネブラスカ州リンカーン)から得られる薬剤アルベンダゾールのことである。化合物PS17、PS17a、及びCは100 ppmで培地に直接添加され、ABZは培地添加に先立って、無水エタノール100μlに溶解された。
〔効力の基準〕 倒立顕微鏡を40Xで使用し、吸虫を未処理対照吸虫と比較して、死亡、致死率の乱れ、又は形態的変化によって証拠づけられる薬剤処理の効果について、3-8時間に毎時間、次いで1日1回又は2回の検査を行なった。
実験的に感染させたうさぎの肝臓から取り出した生後3週間のファスキオラ・ヘパチカを、1.6 cmのリンボ培養基プレート穴(培地2 ml、各穴に4-6匹)中で5日間、生体外で培養した。5日間の培養後、化合物PS17a(100 ppm、5匹)、化合物PS17(100 ppm、6匹)を含有する培地と未処理対照培地(4匹)に吸虫を移した。化合物PS17a中では、18時間までに全吸虫が死亡した。化合物PS17中では、培地対照中の生存吸虫3匹に比べ、24時間に1匹のみ生存した(動きはにぶい)。形態的検査のため全吸虫を固定し、染色した。死亡吸虫の分析に起因するものを除いて、明白な形態的変化は観察されなかった。5日の培養前期間中、吸虫は活発な状態にあり、明らかに活発な消化機能をもって培地からRBCを摂取するのが見られた。

廃棄処分の子牛肝臓の胆管から吸虫を回収し、無菌のRPMI中で10-20匹ずつの群で2-3時間洗った。6穴リンブロ組織培養基プレート(サイズ10 ml)の、処理又は未処理培地4 mlを含有する各穴で、1匹ずつの吸虫を培養した。知られた殺吸虫活性をもたない処方ブランク(化合物C)も試験した。潜在的汚染源を確認するために、2%RBCを加えた培地対照と2%RBCなしの培地対照を包含する追加の対照を加えた。ABZ濃度を25 ppmに高めた。汚染源の制御のため、薬剤又は培地のみを含有する対照穴も含まれた。
12レプリカのうち8匹、及び対応培地対照の6匹のうち2匹については、暴露後24時間で、吸虫を入れた穴と吸虫を入れない穴とも、培地を選択的に変更した。この余分の取扱い手順で、培地の混濁と黄変から証拠づけられるように、明らかに汚染のため、8レプリカのうち6匹が58時間までに死亡する結果となった。24時間で培地を変えた2レプリカは、汚染されなかった。6培地対照(吸虫なし)レプリカは、115時間の培養中、汚染されなかった。

本明細書で明らかにされている新規な殺虫剤毒素をコードした新規な遺伝子は、アグロバクター・ツメファシエンス(Agrobacter tumefaciens)からのTiプラスミドを使用して、植物細胞へ挿入できる。次に植物細胞を植物へ再生させる[ザンブリスキ・ピー(Zambryski, P.)、ジョース・エッチ(Joos, H.)、ジェンテロ・シー(Gentello, C.)、リーマンス、ジェイ(Leemans, J.)、バン・モンタギュー・エム(Van Montague, M.)、及びシェル・ジェイ(Schell, J.)(1983年)Cell 32巻1033-1043頁]。この点で、特に有用なベクターはpEND4Kである[クリー・エッチ・ジェイ(Klee, H.J.)、ヤノフスキー・エム・エフ(Yanofsky, M.F.)及びネスター・イー・ダブリュー(Nester, E.W.)(1985年)Bio/Technology 3巻637-642頁]。このプラスミドは、植物細胞と細菌中で複製でき、パッセンジャー遺伝子に対して複数のクローニング位置をもっている。例えば毒素遺伝子はpEND4KのBamHI位置へ挿入され、大腸菌中で増殖し、適当な植物細胞へ形質転換される。
本発明の新規な遺伝子を、オートグラファ・カリフォルニカ(Autographa californica)核ポリヘドロシスウイルス(AcNPV)のようなバクロウイルスへクローン化できる。pUC8のような市販のクローニングベクターにクローン化されたAcNPVゲノムを含有するプラスミドを構築できる。AcNPVゲノムを変更し、ポリヘドリン遺伝子のコード領域が除かれて、パッセンジャー遺伝子の特異なクローニング位置がポリヘドリンプロモータの真後ろに置かれるようにする。このようなベクターの例はペノックら[ペノック・ジー・ディー(Pennock,G.D.)、シューメーカー・シー(Shoemaker, C.)及びミラー・エル・ケイ(Miller,L.K.)(1984年)Mol.Cell Biol.4巻399-406頁]に記述されたpGP-B6874、及びスミスら[スミス・ジー・イー(Smith,G.E.)、サマーズ・エム・ディー(Summers, M.D.)及びフレーザー・エム・ジェイ(1983年)Mol. Cell Biol.3巻2156-2165頁]に記述されたpAC380である。本発明の新規なタンパク毒素をコードした遺伝子は、コード領域から上流及び下流の適当な領域で、BamHIリンカーによって変更でき、AcNPVベクター類の一つのもののパッセンジャー位置に挿入できる。
Claims (2)
- 以下の群、すなわち
(a) 配列番号8又は配列番号8に示す配列の殺線虫活性を保持している上記配列の任意の部分に示すアミノ酸配列からなる殺線虫毒素をコードしたヌクレオチド;
(b) 殺線虫毒素をコードした、配列番号7に示す配列又はその断片、からなるヌクレオチド;
(c) 上記(b)に記載のヌクレオチド又はその相補鎖とストリンジェントな条件下でハイブリダイズし、かつ、殺線虫毒素をコードする、ヌクレオチド;
からなる群から選ばれるDNA。 - 請求項1に記載のDNAによりコードされるアミノ酸配列からなる、殺線虫毒素。
